오미자를 첨가한 스펀지케이크의 품질특성 (Quality Characteristics of Sponge Cake added with Schizandra chinensis)

  • 이 연구 목적은 기능성이 있는 오미자의 이용확대를 위한 오미자 분말 5~20%를 첨가한 스펀지케이크를 제조하여 일반성분, 오미자첨가 스펀지케이크의 반죽의 비중, pH, 부피, 무게, 색도, texture, 관능검사를 실시하였다. 대조군의 비중과 무게는 각각 0.50, 144.3 g으로 가장 낮게 나타났으며, 오미자 분말 첨가량이 증가할수록 유의적으로 증가하였다. 굽기 손실률과 높이, 부피, 비용적은 대조군이 각각 10.0(%), 5.10 cm, 935, 24, $6.47cm^3/g$으로 가장 높게 나타났으며, 오미자분말 첨가량이 증가할수록 유의적으로 감소하게 나타냈다. 색도 측정은 L(명도)와 b(황색도) 오미자 분말첨가량이 증가할수록 감소하였다. 조직감의 경도(hardness), 탄력성(springiness), 응집성(cohesiveness)은 오미자 분말의 첨가량이 증가할수록 감소하는 경향을 보였다. 관능검사는 향(flavor), 종합적인 기호도(overall acceptability)는 오미자 분말을 5% 첨가하는 것이 우수하게 나타났다. 이러한 결과를 토대로 오미자 분말을 5% 첨가하면 맛과 향이 가미된 기능성 스펀지케이크를 상품을 개발할 수 있다고 판단되었다.

감송향(甘松香) 물추출물이 U937 백혈병 세포의 분화유도에 미치는 영향 (Effect of Nardostachys chinensis on Induction of Differentiation in U937 Monomyelocytic Cells)

  • Nardostachyts chinensis (N. chinensis) belonging to the family Valerianaceae has been used to elicit stomachic and sedative effects. The MAPKs are serine/threonine kinases involved in the regulation of various cellular responses, such as cell proliferation, differentiation and apoptosis. The PKC also plays a key role in regulating the response of hematopoietic cells to both physiological and pathological inducers of proliferation and differentiation. This study investigated the signaling pathways on the U937 cell differentiation induced by N. chinensis. N. chinensis induced the differentiation of U937 cells, as shown by increased of differentiation surface antigen CD11b. Activation of ERK increased time-dependently in differentiation of U937 cells induced by N. chinensis, but activations of JNK and p38 were unaffected. Inhibitor of ERK (PD98059) significantly reduced CD11b expression induced by N. chinensis in U937 cells. In addition, N. chinensis increased protein level of PKC ${\beta}$I and PKC ${\beta}$II isoforms, but the protein level of PKC ${\alpha}$ and PKC ${\gamma}$was constant. PKC inhibitors (GF 109203X and H-7) inhibited U937 cell differentiation and the ERK activation induced by N. chinensis. These results indicated that PKC and ERK may be involved in U937 cell differentiation induced by N. chinensis.

오미자약주 제조 및 품질특성 (Brewing and Quality Characteristics of Schisandra chinensis Yakju)

  • 약주의 소비증대와 품질증진을 위해 오미자(Schisandra chinensis) 과실을 0-20% 농도 별로 약주에 첨가하여 발효를 통한 품질증진에 미치는 효과를 연구하였다. 당농도와 알코올 생성량은 오미자 첨가구와 무첨가구에서 유사한 경향을 나타났다. pH는 무첨가구가 4.17로 가장 높았고, 오미자 첨가비율이 증가함에 따라 최종 3.57-3.90로 낮아지는 경향을 보였다. 유기산은 citric acid가 무첨가구에서 9.22 mg/100 mL, 오미자 첨가구에서 161.38-634.96 mg/100 mL 검출되었다. 무첨가구에 비하여 오미자 첨가구에서 약 17.50-68.87배로 높은 비율로 함유되어 있으며, citric acid는 오미자 첨가구의 pH에 농도 의존적으로 영향을 주기 때문에 농도가 높은 오미자 첨가구의 pH가 가장 낮은 값을 가진 것과 연관이 있는 것을 확인하였다. 무첨가구의 색도는 명도 18.31, 적색도 -0.58 및 황색도 -0.38을 나타내었으며, 오미자 첨가구는 명도 13.55-15.14, 적색도 0.12-5.16 및 황색도 -0.78-0.02로 오미자 첨가로 인해 어두운 색조로 바뀌었으나 적색도가 증가하여 관능적인 요인을 향상시켰다.

PC12 Cell에 대한 토사자(?絲子)의 안정성 및 항산화작용에 대한 연구 (Safety and Antioxydative effects of Cuscuta chinensis Lam. in PC12 Cell)

  • Purpose : Cuscuta chinensis Lam. is utilized extensively as important medicines for threatened abortion habitual abortion. However, objective data related to an embryo is not existed until now. Therefore, this study is focused to find out stability of Cuscuta chinensis Lam. about an embryo during pregnancy based on data related to stability of nerve cell and antioxydative effect by using neural cell line, PC 12 cell. Methods : Experimentation concerns cytotoxic effects and antioxydative effect through methods such as MTT ssay, western botting after abstracting an undiluted solution from domestic Cuscuta chinensis Lam. Results : 1. As a result of experimentation on MTT assay according to each magnification from Cuscuta chinensis Lam. extraction solution with different abstraction methods, cytotoxic effect is not observed to all extract except an undiluted solution which is abstracted from MeOH stiring. Also, an undiluted solution in stiring with MeOH could not confirm whether come from Cuscuta chinensis Lam. or not. 2. As a result of revelation of Bax and GSK-3${\beta}$ which is responsive to the first stage from general stress in order to observe antioxydative effects of Cuscuta chinensis Lam. revelation of Bax by Cuscuta chinensis Lam. appeared to decrease. Conclusion : Cytotoxic effects with Cuscuta chinensis Lam. about PC12 cell is not discovered and it assume that it would be anti apoptotic effect by ROS as Bax and GSK-3${\beta}$ inviable effect. In the future, this study could be used as basic data for additional research on Cuscuta chinensis Lam. and effect and stability of complicated prescriptions for keeping pregnancy.

Inhibitory effects of Coptis chinensis extract on the growth and biofilm formation of Streptococcus mutans and Streptococcus sobrinus

  • Streptococcus mutans and Streptococcus sobrinus play important roles in dental caries. Coptis chinensis is a natural product with antimicrobial activity against enterobacteria; however, its effects on oral streptococci are still unknown. Therefore, the effects of C. chinensis on the growth and biofilm formation of the representative cariogenic bacteria S. mutans and S. sobrinus were investigated for the possible use of C. chinensis as an anticaries agent. The C. chinensis extract was diluted with sterile distilled water, and 0.1-2.5% of the extract was used in the experiment. The effects of the C. chinensis extract on the growth and glucan formation of S. mutans and S. sobrinus were measured by viable cell counting and spectrophotometry at 650 nm absorbance, respectively. Crystal violet staining was also carried out to confirm the C. chinensis extract's inhibitory effect on biofilm formation. The C. chinensis extract significantly inhibited the growth of S. mutans and S. sobrinus at concentrations of ≥ 0.3% as compared with the control group. The viable cell count of colonies decreased by 1.7-fold and 1.2-fold at 2.5% and 1.25%, respectively, compared with the control group. The biofilm formation of S. mutans and S. sobrinus was inhibited by > 20-fold at C. chinensis extract concentrations of ≥ 1.25% as compared with the control group. In summary, the C. chinensis extract inhibited the growth and biofilm and glucan formation of S. mutans and S. sobrinus. Therefore, C. chinensis might be a potential candidate for controlling dental caries.

오배자의 항균활성 및 세포독성에 미치는 영향 (Antimicrobial Effect and Cytotoxicity of Chinensis Galla)

  • Chinensis galla has been used as an multi-functional herb, such as anti-inflammatory, anti-virus, and antitumor agent. This study was performed to antimicrobial and cytotoxicity effect in vitro. The results were summarized as follows : Chinensis galla was antimicrobial effect on Staphylococcus aureus subsp. aureus, Bacillus cereus, Bacillus subtilis, Staphylococcus epidermidis. Chinensis galla was antimicrobial effect on Kocuria rhizophila, Corynebacterium ammoniagenes. The extracts of Chinensis galla exhibited cytotoxicity on human dermal fibroblast at $10\;{\mu}{\ell}$ but not at $5\;{\mu}{\ell}$, and the same results was known under a microscope. Accordingly the results show Chinensis galla could antimicrobial effect but exhibited cytotoxicity on human dermal fibroblast at high concentration and it needs more research.

오미자(Schizandra chinensis)의 병원성 미생물에 대한 항균효과 (Antimicrobial Effects of Schizandra chinensis Extract on Pathogenic Microorganism)

  • The growth of pathogens such as Staphylococcus aureus ATCC 29737, Bacillus subtilis KCTC 1021 and Escherichia coli ATCC 11775 was inhibited as much as 1 to 4 log cycles in tryptic soy broth containing 1% of Schizandra chinensis extract but Salmonella tuphimrium ATCC 14028 and Vibrio parahaemolyticus ATCC 17802 were not inhibited. The essential oil of Schizandra chinensis showed stronger inhibitory activity than ethanol extract against above menioned pathogens. The growth of Listeria monocytogenes was not changed in minced pork containing 1% of Schizandra chinensis extract for 12 days at 4$^{\circ}C$. The sensory quality of hamburger patty such as taste, flavor and overall accepability were not decreased by the addition of 1% Schizandra chinensis extract(p<0.05).

오미자 추출물이 고지혈증 흰쥐에 미치는 영향 (Effect of Schizandra chinensis Extract in Hyperlipidemic Rats)

  • The purpose of this study was to investigate the effect of Schizandra chinensis extract on the serum and hepatic lipids inhyperlipidemic rats fed with high-fat diet. The Schizandra chinensis extract decreased significantly in total serum cholesterol, serum triacylglycerol and heptic triacylglycerol but not in the total hepatic cholesterol. The Schizandra chinensis extract also decreased in very low density lipoprotein increased by the highfat diet without affecting high density lipoprotein cholesterol. These results suggest that Schizandra chinensis extract may be benificial for the regulation of hyperlipidemia.

세포막손상 유발로 인한 황련의 캔디다 바이오필름 형성 억제 (Inhibition of Candida albicans Biofilm Formation by Coptidis chinensis through Damaging the Integrity of Cell Membrane)

  • Candida 바이오필름은 숙주조직이나 인체에 삽입된 의료기구의 표면에 자라는 진균의 군락으로 전통적인 항진균제에 대한 내성을 유발한다. 황련(Coptidis chinensis)의 뿌리는 극동지방에서 의료용 목적으로 널리 사용되어 왔다. 본 연구의 목적은 임상에서 분리한 C. albicans 바이오필름 형성 균주가 형성한 바이오필름에 대한 C. chinensis 수용성 추출물의 효과와 C. albicans 바이오필름 형성을 저해하는 데 기여하는 항진균활성을 평가하는 데 있다. 바이오필름에 대한 효과는 XTT [2,3-Bis-(2-Methoxy-4-Nitro-5-Sulfophenyl)-2H-Tetrazolium-5-Carboxanilide)] 환원분석법을 사용하였으며, 조사된 모든 균주에 대한 대사활성은 $98{\mu}g/ml$의 C. chinensis 수용성 추출물에 의해 현저하게 감소($57.3{\pm}14.7%$)되어 유의성 있는 항바이오필름 활성을 나타내었다. Fluorescein diacetate와 propidium iodide로 이중 염색한 결과 C. chinensis 추출물은 C. albicans의 세포막을 손상시켰다. C. chinensis 수용성 추출물은 살진균 활성을 나타냈고, C. albicans 바이오필름의 폴리스티렌 표면으로의 부착을 억제하였으며 C. albicans를 $G_o/G_1$기에 머무르게 하여 바이오필름이나 출아법에 의한 증식을 억제시켰다. 본 연구의 결과는 C. chinensis 추출물이 목표가 되는 C. albicans에 복합적으로 유해한 효과를 내어 궁극적으로는 C. albicans 바이오필름 형성을 억제함을 나타낸다. 따라서 C. chinensis 추출물은 바이오필름과 관련된 캔디다의 감염을 치료하고 제거하기 위한 천연물 기반항진균제 개발에 대한 높은 가능성을 가진다.

오미자 물 추출물이 파골세포 분화에 미치는 영향 (Effect of Water Extract of Schisandra Chinensis on Osteoclast Differentiation)

  • Bone maintains its homeostasis through balance between bone resorbing osteoclasts and bone forming osteoblasts. Thus, unusual balance between osteoclasts and osteoblasts leads to pathological bone diseases, such as osteoporosis, rheumatoid arthritis, autoimmune arthritis, periodontitis. Schisandra chinensis well known traditional herbal has been used for treatment of diseases in China, Korea, Japan, andothers. Recently, research studies have demonstrated that the lignans found in Schisandra chinensis stimulate osteoblasts and suggest that it may be helpful against osteoporosis. However, the inhibitory effect of water extract of Schisandra chinensis on osteoclast differentiation remains largely unknown. In this study, Water extract of Schisandra chinensis markedly suppressed RANKL-induced osteoclast differentiation in cultures of BMMs without cytotoxicity. The mRNA expression of c-Fos, NFATc1, and TRAP induced by RANKL was inhibited by water extract of Schisandra chinensis. It also suppressed c-Fos and NFATc1 protein expression. Taken together, these results suggest that water extract of Schisandra chinensis has the potential to serve as a treatment of bone disease such as osteoporosis.
