The Effect of the Anthracnose Resistant Chili Pepper (PepEST gene) on the Non-target Insects, Green Peach Aphids (Myzus persicae Sulzer, Homoptera) (유전자 변형 탄저병 저항성 고추(PepEST gene)가 비표적 곤충인 복숭아혹진딧물(Myzus persicae Sulzer)에 미치는 영향)

  • To assess the environmental risks of transgenic chili pepper with PepEST gene on non-target organisms before it exposes to the agro-ecosystem environments, we conducted the three sets of green peach aphids (Myzus persicas S.) life table experiment under laboratory conditions (Temp. $25^{\circ}C$, R.H. 50-70%, Photoperiod L16 : D8) in series during 2005-2006. We measured the net reproductive rate ($R_0)^*$, the intrinsic rate of increase ($r_m$), the mean generation time ($T_c$), $fecundity^*$, life span, and reproduction period between non-transgenic chili peppers and transgenic chili peppers, respectively. The life span of green peach aphids from three sets was 31, 27, 25 days, and the period of life span was similar to the general average length of green peach aphids, 25-29 days. Although the first reproduction of transgenic pepper was similar to the non transgenic pepper (P>0.05), the fecundity and the net reproductive rate ($R_o$) by using Jackknife method of transgenic pepper were lower than those of non transgenic pepper (P<0.05). Conclusively, we observed the adverse effect from our results but we should execute further experiments to confirm the results at the fields with the similar way.

  • A species of Heterobasidion was encountered during a diversity study of endophytic fungi from healthy root tissues of chili pepper (Capsicum annuum L.) in Korea. The fungal species (CNU081069) was identified as Heterobasidion araucariae based on phylogenetic analyses of the internal transcribed spacer and translation elongation factor gene sequences. Morphological descriptions of the endophytic isolate matched well with the previous references and supported the molecular identification. The fungus Heterobasidion araucariae CNU081069 is new to Korea.

  • Evolutionary studies conducted on NAC (NAM, ATAF1&2, and CUC2) genes for all major groups of land plants, indicate the presence of the NAC subfamilies, even in the early land plants. The varied roles played by NAC proteins in plant growth and development range from the formation of shoot apical meristem, floral organ development, reproduction, lateral shoot development, and defense responses to biotic and abiotic stresses. Considering the value and importance of solanaceous crops, the study of NAC proteins in these plants needs to be intensified. This will help to identify and functionally characterize their promoters, which will subsequently aid in engineering plants with improved performance under stressful conditions. In this review, the functionally characterized NAC transcription factors specific to tomato, potato, tobacco, chili pepper and eggplant (aubergine) are summarized, clearly indicating their biological functions in the defense mechanism of the plants, against biotic and abiotic stresses.

A Comparison Study on Isoprene Emission Rates from Oak Trees in Summer (여름철 참나무속의 이소프렌 배출속도 비교에 관한 연구)

  • In order to compare the NVOC (Natural Volatile Organic Compound) emission rates from oak trees, the emission rates of isoprene were quantitatively measured in situ based on tree species and region. As a result, the emission rates from Quercus serrata T. were found to be 1000 times greate. than those from Quercus acutissima C. However, the emission rates at the Chili and Gumsung mountain sites did not show any significant differences in summer. Cuvette tests using four different oak species also supported that there was a tremendous difference in emission rates between Quercus serrata T. and Quercus acutissima C. It was found that the emission rates from the trees were highest on the order of Quercus serrata T., Quercus aliena B., Quercus acutissima C. and Quercus variabilis B.

  • Capsaicin (8-methyl-N-vanillyl-6-nonenamide), a pungent ingredient of hot chili peppers, has been reported to possess substantial anticarcinogenic and antimutagenic activities. In the present study. we investigated the effects of capsaicin on pulmonary metastasis of B 16-F10 murine melanoma cells. as well as its mechanism of action.   • Capsaicin, a pungent ingredient in chili pepper, is known to excite sensory neurons that mediate pain sensation. This effect of capsaicin is determined by unique receptors and the capsaicin receptor (transient receptor potential subfamily V, member 1 (TRPV1)) was cloned recently. TRPV1 contains six transmembrane domains and three ankyrin repeats at N-terminal. This characteristic architecture is common in other ion channel in TRPV families. (omitted)

  • The clastogenic activity of capsaicin, a major pungent and irritating constituent of hot chili pepper, was evaluated in cultured human lymphocytes. Capsaicin (125, 250, and 500 $\mu$M) caused cytogenetic damage as determined by increased frequency of chromosome/chromatid aberrations compared to the solvent control. The mitotic indices were also decreased in a concentration-related manner in capsaicin-treated cells. Moreover, capsaicin suppressed [$^3$]thymidine incorporation into lymphocytes. The clastogenicity and cytotoxicity of capsaicin towards human lymphocytes were evident without an external metabolic activation system. Taken together, these findings suggest that capsaicin is a genotoxic agent and may thus represent a potential health hazard in humans.

Linkage Map Construction and Molecular Genetic Approach in Capsicum spp.

  • We have constructed a molecular linkage map of chili pepper (Capsicum spp.) in an interspecific (C. annuum cv. TF68 x C. chinense cv. Habanero) F$_2$ population of 107 plants with 150 RFLP and 430 AFLP markers. The resulting linkage map consists of 11 large (206-60.3 cM) and 5 small (32.6- 10.3 cM) linkage groups cover-ing 1,320 cM with an average map distance between framework markers of 7.5 cM. Most (80%) of the RFLP markers were pepper-derived clones and these markers were evenly distributed across the genome. By using 30 primer combinations, 444 AFLP markers were generated in the F$_2$population. The majority of the AFLP markers clustered in each linkage group, although PstI/MseI markers were more evenly distributed than Eco RI/MseI markers within the linkage groups. Genes for biosynthesis of carotenoids and capsaicinoids were mapped on our linkage map. This map will provide the basis of studying secondary metabolites in pepper.
