It has been well documented from animal and human studies that conjugated linoleic acid (CLA) has numerous beneficial effects on health. In chickens, CLA exerts many effects on performance ranging from egg quality and yolk lipids to meat quality. Although there are several CLA isomers available, not all CLA isomers have the same incorporation rates into egg yolk: cis-9,trans-11 and trans-10,cis-12 CLA isomers are more favorably deposited into egg yolk than other isomers investigated, but of the two isomers, the former has a higher incorporation rate than the latter. CLA alters the amounts and profiles of lipids in plasma, muscles and liver. Furthermore, increased liver weight was reported in chickens fed dietary CLA. As observed in egg yolk, marked reduction in intramuscular lipids as well as increased protein content was observed in different studies, leading to elevation in protein-to-fat ratio. Inconsistency exists for parameters such as body weight gain, feed intake, feed conversion ratio, egg production rate and mortality, depending upon experimental conditions. One setback is that hard-cooked yolks from CLA-consuming hens have higher firmness as refrigeration time and CLA are increased, perhaps owing to alterations in physico-chemistry of yolk. Another is that CLA can be detrimental to hatchability when provided to breeders: eggs from these breeders have impaired development in embryonic and neonatal stages, and have increased and decreased amounts of saturated fatty acids and monounsaturated fatty acids (MUFAs), respectively. Thus, both problems can be fully resolved if dietary sources rich in MUFAs are provided together with CLA. Emerging evidence suggests that CLA exerts a critical impact on stress and immune functions as it can completely nullify some of the adverse effects produced by immune challenges and reduce mortality in a dose-dependent manner. Finally, CLA is a key regulator of genes that may be responsible for lipid metabolism in chickens. CLA down-regulates both expression of the gene encoding stearoyl-CoA desaturase-1 and its protein activity in the chicken liver while up-regulating mRNA of sterol regulatory element-binding protein-l.
An immunogenic, high molecular weight lipopolysaccharide (LPS)-protein complex isolated from a potassium thioncyanate extract of a Pasteurella multocida (P multocida ; strain P-2383, capsular type A and somatic type 3) was characterized. Chemical analysis of the complex by gas chromatography on a capillary column demonstrated that this complex contained most of the chemical constituents characteristic of LPS extracted by the phenol-water methed from the whole bacterium. However, there was proportionately more carbohydrate than fatty acid in the complex in contrast to LPS in which fatty acid seemed to be in excess. When toxicity of the complex was evaluated in 10-day-old chicken embryos, the complex was less toxic ($LD_{50}=12.72{\mu}g$) than the purified LPS ($LD_{50}=0.44{\mu}g$). The $LD_{50}$, of the LPS moiety extracted from the complex was $5.24{\mu}g$. Composition of the complex was analyzed by SDS-PAGE with silver staining and Western immunoblotting. The complex did not migrate through the polyacrylamide gel unless dissociated with SDS. The complex dissociated with SDS contained at least 32 different protein and polysaccharide components: 18 components reacted with an antiserum against the complex. There was no significant compositional variation between the complexes from different strains, but quantitative differences in individual components were noted. When cross-protectivity of the complex was evaluated in mice, this complex provided substantial protection not only against the homologous bacteriun but also against different P multocida strains of the same serotype. LPS-protein complexes isolated by the same method from other strains also induced protection against an challenge with P-2383.
IBDV는 양계업이 전업화된 모든지역에서 발생하고 있다. 이 질병의 임상증상은 매우 특징적이기도 하지만 IBDV 감염시의 닭의 연령 또는 바이러스의 병원성에 따라 임상증상 업이 내과 하기도 한다. IBDV에 의한 BF의 종장이나 출혈성 병변은 감염 후 짧은 기간에만 관할되며 질병이 경과함에 따라 BF는 위축된다. IBDV는 감염력이 극히 높으며, 오염된 계분, 사료, 물 등에 의하여 전파된다. 이 바이러스는 외계 환경에 대하여 극히 높은 저항성을 갖고 있으며 Ribovirus군의 Diplorna 바이러스로서 크기는 55-60nm이다. IBDV는 계태아나 계태아 조직 또는 감수성이 있는 닭의 BF에서 용이하게 증식시킬 수 있다. agar gel을 사용한 면역 확산법은 IBDV 감염을 진단할 수 있는 가장 간편한 방법이다. IBDV에 대한 항체는 감염 후 1주일 후부터 증명되며 장기간 지속된다. 감수성 계군일 경우 높은 역가의 항체형성이나 지속은 계군의 연령에 비례한다. IBDV에 대한 모체이행 항체는 critical age때의 병아리를 야외 감염으로부터 보호하여 준다. IBDV 단독 감염에 의한 IBD 피해는 경미한 것이지만 면역부전증에 의하여 유발되는 복합성 질병인 Inclusion body hepatitis, 전염성 빈혈, 괴저성 피부염 등은 닭의 생산성을 크게 저하시킨다.
Unfortunately, there is no technique which is stable and repetitive to produce transgenic chicken, although various ways of gene transfer including PGC-and embryonic cell-mediated gene transfer, DNA microinjection, virus inoculation and sperm cells have been employed. The aims of this study were 세 develop and establish such a stable, repetitive and efficient way of gene transfer giving a faithful gene expression during development after the reconstruction of embryo in an UV-irradiated egg. A dual reporter plasmid (pJJ9), a fusion gene containing lacZ and GFP driven by a CMV promoter was used to exploit either merits of both reporting markers. lacZ with strong signal or GFP with vital marking. Electroporated embryonic blastodermal cells (EBCs) in the presence of the pJJ9 DNA faithfully showed 377 bp PCR product and lacZ or GFP expressions in the identical cells in vitro of in vivo. Furthermore, analyses of expression pattern of the foreign DNA demonstrated that microinjected EBCs cells into the UV-irradiated recipient egg should participate in normal developmental process, for example, proliferation and differentiation into various tissues. Thirty percentages of the manipulated eggs showed lacZ expression in their tissues. These results together with the specific procedures used in this study should facilitate avian transgenesis.
본 연구에서는 4급 암모늄에 속하는 quaternary ammonium chloride와 구연산의 합제인 Citra-$Kill^{(R)}$의 조류인플루엔자에 대한 살바이러스 효과를 확인하기 위해 국립수의과학검역원의 소독제 효력시험 중 바이러스 소독제 효력시험에 따라 수행하였다. 소독제와 조류인플루엔자바이러스를 증류수, 경수, 그리고 유기물 조건에서 반응시킨 후, 중화액을 이용하여 중화시킨 다음, 중화된 용액 0.2 ml를 10일령의 계태아 요막에 주입하여 5일 동안 배양시킨 다음, 요막액을 채취하여 바이러스의 생존여부를 혈구응집반응을 통해 확인하여 소독제의 효력배수를 결정하였다. 본 연구의 결과, Citra-$Kill^{(R)}$은 증류수, 경수, 그리고 유기물 조건에서 각각 2,000, 1,500, 그리고 500배에서 조류인플루엔자바이러스를 불활성화시켜, 조류인플루엔자에 대해 뛰어난 소독효과를 갖고 있는 것이 확인되었다. 따라서 향후, 야외적용시험을 통해 실제적용에 따른 효과를 확인할 필요가 있을 것으로 사료된다.
This experiment was conducted on the fowl pox embryo vaccine for the production immunity, and stability, using an attenuated fowl pox virus (Nakano strin). Burnet's window method was applied, that is, 0.1 ml. of seed virus was inoculated on the chorioallantoic membrane of 12-day old chicken embryos, and incubated for 5 to 6 day, and then the result were read. Four kinds of suspensions of different embyo tissue were prepared and tested for the infectivity in chickens. Finally the suspension of chorioallantoic membrane was used as the vaccine throughout the experiment. Results obtained in this experiment are summarized as follows: (1) Of embryo tissue infected with the vaccine virus, chorioallantoic membrane had the highest virus titer of $10^{-5.4}$$EID_{50}$, and albumen the lowest titer of $10^{-0.7}$$EID_{50}$. (2) Suspensions of infected whole embryo with or without saline, and de-embryonated whole egg had about the same virus titer of $10^{-4.4}$$EID_{50}$, whereas the chorioallantoic membrane had $10^{-5.7}$ EID 50 or higher. The virus titer droped one log from $EID_{50}$ when inoculated into chickens. Takes were observed 35.6% of 500 chickens by stick method and 89% of 500 chickens by brush method. (3) The chorioallantoic membrane conferred almost perfect immunity for chickens by 10 days after vaccination. (4) Satisfactory immunity was observed in the chickens when eruption in a single follicle. (5) Eight of 10 vaccinated chickens revealed durable immunity for 307 days following vaccination. (6) The vacuum-dried vaccine maintained its infectiviy for 899 days at $5^{\circ}C$ or below and maintained the vius titer of $10^{-3.6}$$EID_{50}$. On the other hand, non-desiccated wet vaccine maintained the titer of $10^{-3.0}$$EID_{50}$ for 50 days of preservation period at $5^{\circ}$. However, in 50% glycerin-saline the infectivtiy of the same wet vaccine dropped to $10^{-1.5}$$EID_{50}$ (7) The vartation of virus titer of the vaccine before and after desiccation was $10^{-0.5}$$EID_{50}$ on the average. (8) As suspending media, 0.85 per cent saline and distilled water showed nearly the same effect on the infectivity of the vaccine by retaining the titer $10^{-3.0}$$EID_{50}$ after 50 days of preservation both at $5^{\circ}C$ and $20^{\circ}C$, while 50 percent cent glycerine-saline dropped the titer to $10^{-2.5}$ EID and $10^{-1.5}$$EID_{50}$ respectively at $5^{\circ}C$ and $2^{\circ}C$ after the same period.
이 실험은 약재(Cnidium officinale Makino와 Tabanus bovinus)를 이용한 장요막과 흰쥐 각막에서의 항신생혈관 효과를 규명하기 위한 것이다. 먼저, 배 발생과정 중 자연스러운 신생혈관이 생성되는 장요막에서의 신생혈관 억제를 위한 실험에서는 유정란을 $37^{\circ}C$, 포화습도상태인 incubator에 넣어 4일 경과 후 주사바늘을 계란의 흠집에 찔러 넣어 알부민 3ml를 추출하고 계란의 둥근 부분에 지름 2cm의 창을 내고 투명테이프로 막아 incubation시킨다. 이때, 장요막의 직경이 3~5 mm인 embryo를 신생혈관 억제 실험에 사용하였으며, retinoic acid는 양성 대조군으로 이용되었다. 혈관이 뻗어 나오고 있는 장요막에 retinoic acid($10{\mu}g/egg$)를 처리한 후 48시간이 경과되었을 때 현저하게 혈관의 분포가 감소되었음을 알 수 있다. 이와 유사한 신생혈관 억제 효과가 약재(Cnidium officinale Makino와 Tabanus bovinus) ($50{\mu}g/egg$)를 시약으로 처리했을 경우에도 나타났다. 다음은 각막을 봉합하여 투명한 각막에 인위적으로 신생혈관을 유도하는 실험으로 흰쥐를 복강 마취시킨 후 10-0nylon용 사용하여 각막의 중앙부를 전층으로 한번 봉합한 후 각막의 염증을 막고자 terramycin을 하루에 한번씩 일주일간 점안하였다. 약재를 처리하지 않은 각막에서는 봉합 후 2~3일째 많은 혈관들이 뻗어 나오는 것을 관찰할 수 있었으며, 5~6일 이내에 각막기능 손상부위까지 혈관이 뻗어 나가는 것을 볼 수 있었다. 또한 봉합 후 3주일째의 신생혈관 분포가 최고치에 달했다. 이에 비해 약재(Cnidium officinale Makino와 Tabanus bovinus) ($20{\mu}g/kg$)를 4주간 구강투여한 흰쥐의 각막에서는 신생혈관 억제에 주목할 만한 효과를 보였다. 대조군 흰쥐와 비교해 보았을 때에도 약재를 구강 투여한 흰쥐의 각막에서 신생혈관이 상당히 줄어든 것을 볼 수 있었으며, 신생혈관 억제제로 효능을 인정받은 thalidomide와 비교해 보았을 때에도 현저한 차이를 나타내지 않았다. 이러한 결과로 볼 때, 이 약재(Cnidium officinale Makine와 Tabanus bovinus) 는 항신생혈관의 특성을 가지고 있으며, 이 특성은 각막에서 뿐 아니라 다른 부위의 종양을 억제하고 그 전이를 막는 약리학적인 측면에도 이용될 수 있을 것으로 기대된다.
