Antitumor profiles and cardiac electrophysiological effects of aurora kinase inhibitor ZM447439

  • Aurora kinases inhibitors, including ZM447439 (ZM), which suppress cell division, have attracted a great deal of attention as potential novel anti-cancer drugs. Several recent studies have confirmed the anti-cancer effects of ZM in various cancer cell lines. However, there have been no studies regarding the cardiac safety of this agent. We performed several cytotoxicity, invasion and migration assays to examine the anti-cancer effects of ZM. To evaluate the potential effects of ZM on cardiac repolarisation, whole-cell patch-clamp experiments were performed with human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) and cells with heterogeneous cardiac ion channel expression. We also conducted a contractility assay with rat ventricular myocytes to determine the effects of ZM on myocardial contraction and/or relaxation. In tests to determine in vitro efficacy, ZM inhibited the proliferation of A549, H1299 (lung cancer), MCF-7 (breast cancer) and HepG2 (hepatoma) cell lines with $IC_{50}$ in the submicromolar range, and attenuated the invasive and metastatic capacity of A549 cells. In cardiac toxicity testing, ZM did not significantly affect $I_{Na}$, $I_{Ks}$ or $I_{K1}$, but decreased $I_{hERG}$ in a dose-dependent manner ($IC_{50}$: $6.53{\mu}M$). In action potential (AP) assay using hiPSC-CMs, ZM did not induce any changes in AP parameters up to $3{\mu}M$, but it at $10{\mu}M$ induced prolongation of AP duration. In summary, ZM showed potent broad-spectrum anti-tumor activity, but relatively low levels of cardiac side effects compared to the effective doses to tumor. Therefore, ZM has a potential to be a candidate as an anti-cancer with low cardiac toxicity.

  • Hepatotoxicity has many facets. Those to be discussed in this review include the mechanism for the hepatotoxic effects, nature of the injury, and animal models of hepatotoxicity suitable for the detection of chemical injury. Some therapeutic drugs used for treatment of hepatitis are also presented. In addition, as an important and serious problem in future, alternative toxicity testing is discussed.

  • A peroxopolyoxotungsten-based ionic hybrid was synthesized by anion-change of peroxopolyoxometalate (POM) $PW_4O{_{24}}^{3-}$ with dicationic long-chain alkyl imidazolium ionic liquids. The characterization was conducted by FT-IR, TGA, $^1H$-NMR and CHN Elemental analyses. Its catalytic performance was evaluated by the epoxidation of soybean oil with $H_2O_2$ under solvent-free condition, including testing of organic cations influence, catalytic reusability and reaction conditions. The catalyst was proved to be a highly efficient recyclable catalyst for epoxidation of various vegetable oils with $H_2O_2$, showing high $H_2O_2$ utilization efficiency, high catalytic activity, convenient recovery and good reuse ability.

Four-week Repeated Dose Toxicity Test for Myelophil in SD Rats

  • Aim : To evaluate the pharmaceutical safety of the herbal formula Myelophil, composed of Astragali Radix and Salviae Radix, via systemic subacute toxicological study using SD rats. Methods : Forty male and 40 female SD rats were fed with Myelophil (5000, 2500 or 1250 mg/10 mL/kg) or distilled water for four weeks. Adverse effects were examined intensively by comparing the differences between normal and drug-administered groups using clinical signs, necropsies, histopathologic findings, hematology, urinalysis, and blood biochemical analysis. Results : No altered clinical symptoms including body weight, diarrhea, anorexia, death, and abnormal necropsy of major organs were observed in male or female rats. No drug-induced abnormalities in histopathological finding, hematological values, urinalysis, and blood biochemical values were found at any doses of Myelophil. Conclusion : Myelophil should be very safe when used in a clinical application with a wide therapeutic index.

Mineral Contents of Edible Seaweeds Collected from Gijang and Wando in Korea (기장산과 완도산 식용해조류 중의 미네랄 함량)

  • Mineral and heavy metal contents were determined in three brown edible seaweeds (sea mustard, Undaria pinnatifida; seaweed fusiforme, Hizikia fusiforme; sea tangle, Laminaria japonica) collected from Gijang and Wando in Korea, and two green seaweeds (sea lettuce, Enteromorpha prolifera; green laver, Monostroma nitidum) collected from Gijang. Seaweeds contained high proportions of ash (13.8-24.8%) and sulfate (1.08-3.25%). In brown seaweeds, ash contents (24.5-24.8%) were higher than in green seaweeds (13.8-15.2%). The differences of mineral contents were remarkable in both Gijang and Wando and in 5 various kinds of seaweeds. Brown seaweeds contained higher amounts of both macro-minerals (657-13,947 mg/100 g; Ca, Mg, Na, K) and trace elements (0.27-12.98 mg/100 g; Fe, Al, Zn, Mn, Cr, Cu, Ni), and green seaweeds also had relatively higher amounts of both macro-minerals (233-5,279 mg/100 g) and trace elements (0.26-10.61 mg/100 g).

  • The photovoltaic modules installed in the actual field are affected by various external environments and the electrical performance output value is generally lowered compared to initial output value. The most of photovoltaic modules consists of low iron glass, encapsulant (EVA), back sheet, frame and junction box assembly based on the solar cells. In this paper, the characteristics of encapsulant which is an important constituent material of photovoltaic module were verified by maximum power determination, electro luminescence images, yellowness index measurement, and gel content measurement after ultraviolet (UV) irradiation exposure. The most commonly installed 72 cells crystalline photovoltaic modules were tested after various UV exposure of 0, 15, 30, and $60kWh/m^2$ and compared with the reference module. After UV exposure of $15kWh/m^2$, which is the current international test condition, a small amount of change was observed in yellowness index and electroluminescence, while a gell content rapidly increased. At a cumulative dose of $60kWh/m^2$, which will be a new international test condition in the near future, however, the yellowness index increased sharply and showed the greatest output power drop.

Activated Carbon-Embedded Reduced Graphene Oxide Electrodes for Capacitive Desalination

  • Capacitive deionization of saline water is one of the most promising water purification technologies due to its high energy efficiency and cost-effectiveness. This study synthesizes porous carbon composites composed of reduced graphene oxide (rGO) and activated carbon (AC) with various rGO/AC ratios using a facile chemical method. Surface characterization of the rGO/AC composites shows a successful chemical reduction of GO to rGO and incorporation of AC into rGO. The optimized rGO/AC composite electrode exhibits a specific capacitance of ~243 F g-1 in a 1 M NaCl solution. The galvanostatic charging-discharging test shows excellent reversible cycles, with a slight shortening in the cycle time from the ~260th to the 530th cycle. Various monovalent sodium salts (NaF, NaCl, NaBr, and NaI) and chloride salts (LiCl, NaCl, KCl, and CsCl) are deionized with the rGO/AC electrode pairs at a cell voltage of 1.3 V. Among them, NaI shows the highest specific adsorption capacity of ~22.2 mg g-1. Detailed surface characterization and electrochemical analyses are conducted.

Impact of Compost Application on Improvement of Rice Productivity and Quality in Reclaimed Soil (간척지 토양에서 퇴비처리가 벼의 생산 및 품질개선에 미치는 영향)

  • This study was conducted to reduce the dependability of farmers on chemical fertilizers for rice cultivation. Soil chemical and biological properties were monitored before experiment and at the time of harvesting. The results showed that EC, available $SiO_2$, and exchangeable $Ca^{2+}$ were decreased at the time of harvesting while pH, OM, and exchangeable $K^+$ and $Mg^{2+}$ were remain unchanged, compared with soil before experiment. Population of aerobic bacteria, Bacillus sp., and fungi were also increased at the time of harvesting in the paddy field, compared with before fertilization, in the treatment of 50% soil-testing fertilizer+ 50% compost. Concentrations of N, P, and K in rice leaves increased with the fertilizers application, maximum increase was recorded in 50% soil-testing fertilizer+ 50% compost. Non-significant difference was observed in the morphological parameters of rice among the treatments. The chlorophyll contents of rice leaf increased in a similar fashion up to 60 days, thereafter, sharp decrease was observed in all the treatments. Maximum yield (per 10a) was recorded in the field treated with 50% soil-testing fertilizer+ 50% compost followed by standard applied fertilizer, 70% soil-testing fertilizer+ 30% compost, soil-testing fertilizer and unfertilized plot. Amylose content showed non-significant difference within the treatments. Protein content increased with the use of fertilizers and best protein content was recorded in the treatment of 50% soil-testing fertilizer+ 50% compost. It was concluded that the amount of the chemical fertilizer used was directly proportional to the protein content of rice grain. However, the palatability of rice grown in unfertilized field was better than the treatments but minimum yield was obtained. Hence, the treatment of 50% soil-testing fertilizer+ 50% compost, was the best among the fertilizer combinations for rice cultivation as supported by the yield, protein and palatability index.

An Index Structure for Substructure Searching In Chemical Databases (화학 데이타베이스에서 부분구조 검색을 위한 인덱스 구조)

  • The relationship between chemical structures and biological activities is researched briskly in the area of 'Medicinal Chemistry' At the base of these structure-based drug design tries, medicinal chemists search the existing drugs of similar chemical structure to target drug for the development of a new drug. Therefore, it is such necessary that an automatic system selects drug files that have a set of chemical moieties matching a user-defined query moiety. Substructure searching is the process of identifying a set of chemical moieties that match a specific query moiety. Testing for substructure searching was developed in the late 1950s. In graph theoretical terms, this problem corresponds to determining which graphs in a set are subgraph isomorphic to a specified query moiety. Testing for subgraph isomorphism has been proved, in the general case, to be an NP- complete problem. For the purpose of overcoming this difficulty, there were computational approaches. On the 1990s, a US patent has been granted on an atom-centered indexing scheme, used by the RS3 system; this has the virtue that the indexes generated can be searched by direct text comparison. This system is commercially used(http://www.acelrys.com/rs3). We define the RS3 system's drawback and present a new indexing scheme. The RS3 system treats substructure searching with substring matching by means of expressing chemical structure aspredefined strings. However, it has insufficient 'rerall' and 'precision‘ because it is impossible to index structures uniquely for same atom and same bond. To resolve this problem, we make the minimum-cost- spanning tree for one centered atom and describe a structure with paths per levels. Expressing 2D chemical structure into 1D a string has limit. Therefore, we break 2D chemical structure into 1D structure fragments. We present in this paper a new index technique to improve recall and precision surprisingly.

A New Test Method to Evaluate Potential White Pitch Deposit - Influence of pH and calcium hardness - (백색 점착성 이물질을 측정하기 위한 새로운 시험법 - pH와 칼슘경도의 영향 -)

  • A new testing method to evaluate the deposition potential of white pitch was developed. The new method involves depositing the potential white pitch particles on the air bubble covered plastic film in the pitch deposit tester (PDT) developed by KRICT and analysing the deposited area of white pitch using an image analyzer. In addition, the effect of two important factors (pH and calcium hardness) on white pitch deposition potential was elucidated. When pH of the coated broke stock was increased from neutral to alkali or the calcium hardness of the stock was decreased, the pitch deposit area was decreased, implying that these two factors have to be controlled during the evaluation of pitch deposition potential. It was found that hydrophobicity of the surface of latex binding films repulped is a key factor influencing white pitch deposition.
