Heterogeneous Sequences of Brain Cytoplasmic 200 RNA Formed by Multiple Adenine Nucleotide Insertions

  • Brain cytoplasmic 200 RNA (BC200 RNA), originally identified as a neuron-specific non-coding RNA, is also observed in various cancer cells that originate from non-neural cells. Studies have revealed diverse functions of BC200 RNA in cancer cells. Accordingly, we hypothesized that BC200 RNA might be modified in cancer cells to generate cancerous BC200 RNA responsible for its cancer-specific functions. Here, we report that BC200 RNA sequences are highly heterogeneous in cancer cells by virtue of multiple adenine nucleotide insertions in the internal A-rich region. The insertion of adenine nucleotides enhances BC200 RNA-mediated translation inhibition, possibly by increasing the binding affinity of BC200 RNA for eIF4A (eukaryotic translation initiation factor 4A).

  • Amphetamine-type stimulants (ATS) are a group of ${\beta}$-phenethylamine derivatives that produce central nervous system stimulants effects. The representative ATS are methamphetamine and 3, 4-methylenedioxymethamphetamine (MDMA), and abuse of ATS has become a global problem. Methamphetamine is abused in North America and Asia, while amphetamine and 3, 4-methyle nedioxym ethamphetamine (Ecstasy) are abused in Europe and Australia. Methamphetamine is also the most abused drug in Korea. In addition to the conventional ATS, new psychoactive substances (NPS) including phenethylamines and synthetic cathinones, which have similar effects and chemical structure to ATS, continue to spread to the global market since 2009, and more than 739 NPS have been identified. For the analysis of ATS, two tests that have different theoretical principles have to be conducted, and screening tests by immunoassay and confirmatory tests using GC/MS or LC/MS are the global standard methods. As most ATS have a chiral center, enantiomer separation is an important point in forensic analysis, and it can be conducted using chiral derivatization reagents or chiral columns. In order to respond to the growing drug crime, it is necessary to develop a fast and efficient analytical method.

Bioprospecting of Novel and Bioactive Metabolites from Endophytic Fungi Isolated from Rubber Tree Ficus elastica Leaves

  • Endophytic fungi are an important component of plant microbiota, and have the excellent capacity for producing a broad variety of bioactive metabolites. These bioactive metabolites not only affect the survival of the host plant, but also provide valuable lead compounds for novel drug discovery. In this study, forty-two endophytic filamentous fungi were isolated from Ficus elastica leaves, and further identified as seven individual taxa by ITS-rDNA sequencing. The antimicrobial activity of these endophytic fungi was evaluated against five pathogenic microorganisms. Two strains, Fes1711 (Penicillium funiculosum) and Fes1712 (Trichoderma harzianum), displayed broad-spectrum bioactivities. Our following study emphasizes the isolation, identification and bioactivity testing of chemical metabolites produced by T. harzianum Fes1712. Two new isocoumarin derivatives (1 and 2), together with three known compounds (3-5) were isolated, and their structures were elucidated using NMR and MS. Compounds 1 and 2 exhibited inhibitory activity against Escherichia coli. Our findings reveal that endophytic fungi from the rubber tree F. elastica leaves exhibit unique characteristics and are potential producers of novel natural bioactive products.

Biological Control Activities of Plant Growth Promoting Rhizobacteria from Organic and Nonorganic Rice Fields against Rice Sheath Blight Pathogen (Rhizoctonia solani Kühn)

  • Rhizoctonia solani is one of the major pathogens that cause sheath blight disease in rice. Sheath blight is one of the most difficult diseases to control. Biological control (with the use of rhizobacteria) is one of the ways to control this disease. Plant Growth Promoting Rhizobacteria (PGPR) is a rhizosphere bacterium that can be used to enhance plant growth. The composition of the rhizobacteria in organic and nonorganic soil is affected by the chemical characteristics of the soil - which influences plant physiology and root exudation patterns. This study aimed to obtain a species of rhizobacteria which shows PGPR activity, from organic and nonorganic rice fields and test their capability to suppress R. solani growth. Out of 23 isolates screened for PGPR activity, the following isolates showed high PGPR activity and were selected for in vitro antagonistic activity testing against R. solani: ISO6, ISO11, ISO15, ISN2, ISN3, and ISN7, The six isolates produced 43,42-75,23 ppm of IAA, possessed phosphorus solubilization capability, and chitinase-producing activity. ISO6 (54.88%) and ISN7 (83.33%) displayed high inhibition capacities against R. solani, in vitro. ISO6 and ISN7 inhibited the growth of R. solani lesions on rice leaves by 89% and 100% (without lesion), respectively, after 7 days of incubation. Analysis of their 16S rRNA sequences revealed that the ISO6 isolate was Citrobacter freundii and ISN7 isolate was Pseudomonas aeruginosa.

Effect of Surface Treatment on Bioactivity of Ti-Ni Shape Memory Alloys (Ti-Ni형상기억합금의 생체활성에 미치는 표면처리의 영향)

  • Research into the replacement of injured systems and tissue in the human body is advancing rapidly. Recently, Ti-Ni shape memory alloys have shown excellent biofunctionality related to their shape memory effect and superelasticity. In this study, the effect of an acid or an alkali treatment on the bioactivity in 49Ti-Ni and 51.5Ti-48.5Ni alloys is investigated in an effort to utilize Ti-Ni alloy as a biomaterial. In addition, the biocompatibility in a SBF solution is assessed through in vitro testing. A porous surface was formed on the surface of both alloys after a chemical treatment. According to the in vitro test, apatite formed on the surfaces of both alloys. The forming rate of apatite in the Ti-rich alloy was faster that in the Ni-rich alloy. The formation of apatite provided proof of the bioactivity of the Ti-Ni alloy. A small quantity of Ni was eluted at the initial stage, whereas Ni was not found for 12 days in the Ti-rich alloy and for 8 days in the Ni-rich alloy. In the case of the treated 51.5Ti-Ni alloy, the shape memory property was worsened but the biocompatibility was improved.

  • This study conducted the measurements of air flow rate for blower systems with experiment and numerical. A new airflow rate test method is suggested, with which it is possible to accurate measurements and calculate the air flow rate for blower systems. The blower(axial fan) is an industrial fluid machine device that supplies a large amount of air by driving an impeller with an electric motor, and it is widely used throughout the industry such as steel, power plant, chemical, semiconductor, LC D, food, and cement. The airflow from the blower is for exchanging the heat in the cooling unit or heat exchanger. The temperature of coolants and hydraulic oil primarily depends on the amount of airflow rate through the cooling package so its accurate estimation is very important. Moreover, it required a larger investment in time and cost since it could not be executed until the system is actually made. Therefore, this research is intended to examine the phenomenon of air flow pattern when testing air flow rate, suggested new test method, and show the result of the validation test.

  • Nuclear power plant operations for electricity generation, rare-earth mining, nuclear medical research, and nuclear weapons reprocessing considerably increase radioactive waste, necessitating massive efforts to eradicate radioactive waste from aquatic environments. Cobalt (58Co) and strontium (90Sr) radioactive elements have been extensively employed in energy generation, nuclear weapon testing, and the manufacture of healthcare products. The erroneous discharge of these elements as pollutants into the aquatic system, radiation emissions, and long-term disposal is extremely detrimental to humans and aquatic biota. Numerous methods for treating radioactive waste-contaminated water have emerged, among which the adsorption process has been promoted for its efficacy in eliminating radioactive waste from aquatic habitats. The current review discusses the adsorptive removal of radioactive waste from aqueous solutions using low-cost adsorbents, such as graphene oxide, metal-organic frameworks, and inorganic metal oxides, as well as their composites. The chemical modification of adsorbents to increase their removal efficiency is also discussed. Finally, the current state of 58Co and 90Sr removal performances is summarized and the efficiencies of various adsorbents are compared.

Development of Dye Natural Batik Based on Fiber Coconut Waste and Leaf Avocado through Extraction Method in Supporting Green Business

  • Purpose: The development of natural batik dyes based on a combination of coconut fiber waste and avocado leaves using the extraction method is important to support the green economy and reduce chemical waste in Indonesia. Research design, data and methodology: The research explores the use of coconut fiber and avocado leaf waste extraction as a natural batik dye and conducts market testing to assess consumer satisfaction. Results: Indonesian batik exports are growing, but synthetic dye practices are causing a decline in demand. To address this, natural dyes are being explored, including coconut fiber waste and avocado leaf waste. Conclusion: Test results from washing at 40 degrees Celsius in terms of color changes and color staining, from sweat in terms of changes in acid color and changes in base color, to sunlight in terms of color fastness value, to heat to iron in terms of color change and color staining shows a value of 3-4 (quite good) and 4-5 (good), meaning that coconut fiber and avocado leaves waste can be used as natural batik dye.

Bioactivity of Cajuput Seedling n-Hexane Extract as an Attractant for Subterranean Termite Coptotermes curvignathus Holmgren (Isoptera: Rhinotermitidae)

  • Subterranean termite attacks on cajuput (Melaleuca cajuputi) seedling roots were widespread in several of Java's Perum Perhutani Forest Management Units. This attack was suspected to be related to the chemical components of the cajunput seedling roots. This study was conducted to determine the bioactivity of cajuput seedling root extract as an attractant for the subterranean termite Coptotermes curvignathus Holmgren (Isoptera: Rhinotermitidae). The extraction process was performed according to ASTM D1108-96, and the extract was characterized using gas chromatography-mass spectrometry (GC-MS). Bioactivity testing of the extracts was carried out using attractiveness and no-choice feeding bioassays. The results showed that the average root extraction yield from cajunput seedlings was 4.94%. The attractiveness of the extract solutions at concentrations of 0.50%, 0.75%, and 1.00% were 45.33%, 62.00%, and 74.67%, respectively. The mortality rate of C. curvignathus termites ranged from 9.63% to 24.44%. Cajuput seedling root extract's lethal concentration 50 was 2.45% (non-toxic). GC-MS analysis showed that the extract contained linoleic acid, which has the potential to attract insects.

Reinforcement of mechanical properties in unsaturated polyester resin with nanosheet

  • In the oil and gas industry, composite materials should exhibit high flexibility and strength for offshore structures. Therefore, weak points in the composites should be improved, such as brittleness, moisture penetration, and diffusion of detrimental ions into nanometric pores. This study aimed to increase the strength, flexibility, and plugging of nanopores using single-layer graphene oxide (SGO) nanosheets. Therefore, SGO is added to unsaturated polyester resin at concentrations of 0.015 and 0.15 % with Normal Methyl Pyrrolidone (NMP) as a solvent for the formation of Nanographene Oxide Reinforced Polymer (NGORP). The mechanical properties of the prepared samples were tested using tensile testing (ASTM-D 638). It has been shown that incorporating SGO, approximately 0.015%, into the base resin resulted in enhanced properties such as rupture resistance forces increased by 745.61 N, applied stress tolerances increased by 4.1 MPa, longitude increased to 1.58 mm, elongation increased by about 2.38%, and rupture energy increased by about 204.51 J. Despite the decrease in tensile force strength properties in the manufactured nanocomposite with 0.15% SGO, it has exclusive flexibility properties such as a high required energy level for rupture of 5,576 times and a formability of 40% more than the base sample. It would be best to use NGORP manufactured from 0.015% nanosheets with exclusive properties rather than base samples for constructing parts and equipment, such as rebars, composite sheets, and transmission pipes, on offshore platforms.
