Antibacterial Activity of Bioconverted Linoleic Acid Produced by Pseudomonas aeruginosa PR3

  • Crude extract of bioconverted linoleic acid using Pseudomonas aeruginosa PR3 was evaluated for its antibacterial activity against food-borne pathogenic bacteria. Crude extract showed antibacterial activity against four Gram-positive bacteria, Staphylococcus aureus (ATCC 6538), S. aureus (KCTC 1916), Listeria monocytogenes (ATCC 19166), and Bacillus subtilis (ATCC 6633), and one Gramnegative bacterium, Pseudomonas aeruginosa (KCTC 2004), with minimum inhibitory concentrations ranging from 750 to $1,500\;{\mu}g{\cdot}ml^{-1}$. S. aureus and B. subtilis were selected for growth inhibition assays with bioconverted linoleic acid. Major antibacterial effects occurred at lag phase.

Effect of pH on the Formation of Lysosome-Alginate Beads for Antimicrobial Activity

  • In this study, we developed lysosome-alginate beads for application as an oral drug delivery system (ODDS). The beads harboring lysosomes, which have antimicrobial activity, and various concentrations of alginate were characterized and optimized. For application as an ODDS, pH-dependent lysosome-alginate beads were generated, and the level of lysosome release was investigated by using antimicrobial tests. At low pH, lysosomes were not released from the lysosome-alginate beads; however, at neutral pH, similar to the pH in the intestine, lysosome release was confirmed, as determined by a high antimicrobial activity. This study shows the potential of such an ODDS for the in vivo treatment of infection with pathogens.

  • $Tb^{3+}-activated$ green-emitting $(Y,Gd)Ga_3(BO_3)_4$ phosphor has been investigated. The main absorption was in the $120{\sim}238$ nm and exhibited a green emission with the 545 nm and several peaks due to inner shell transition of $Tb^{3+}$ ion. With the optimized $Tb^{3+}$ concentrations, the maximum emission brightness was 90% of the $Zn_2SiO_4$:Mn phosphor.

  • The substituted pyridinium salts of oxochloromolybdates(V) have been prepared characterized by visible and infrared spectra, and magnetic moments. The electrical conductance of aqueous solutions of substituted pyridinium salts have been measured over a range of concentrations. The results indicate that the complex anions are monomeric in the solid. The magnetic moments of the complexes are similar to the spin-only values. Substituted pyridines are present as pyridinium cation in solid state.

  • We prepared gold nanoparticles (Au NPs) by reduction-oxidation reaction between $HAuCl_4$ and trisodium citrate and measured the size and morphology of Au NPs by TEM for various molar ratios of $HAuCl_4$ to citrate and for various concentrations of $HAuCl_4$. UV-vis spectroscopy was used to characterize the optical properties of Au NPs. Au NPs in the size range from 14.3 nm to 20.3 nm were prepared with monodisperse distribution.

  • Hydroxlamine effect on the determination of dissolved oxygen by the azide-modified Winkder method and polarography has been studied. It was found that hydroxylamine interference on the dissolved oxygen by the azide-modified Winklet method can be eliminated completely by using permanganate. An inexpensive and convenient polarograph device was constructed. Dissolved oxygen in an air-saturated 0.1 F KCl solution undergoes, independent of hydroxylamine concentrations a two-step irreversible reduction at the dropping electrode the $H_2O_2$ produced in the first step is reduced to $H_2$O in the second. Two waves of equal size result, the first with a half-wave potential (E1/2) at about -0.13 V and the second at about -0.91 V (vs. SCE).

  • In this study, we have attempted to determine the optimum concentration of inducers responsible for efficient laccase production by the white-rot fungus, Trametes sp. Variations in laccase activity were investigated with changing concentrations of 2,5-xylidine, syringaldazine, ABTS, and guaiacol. Enhancement of peak laccase activity was achieved via the combination of 2,5-xylidine with ABTS, syringaldazine, or guaiacol, resulting in increases of up to 359, 313, and 340%, respectively, as compared to control values. Among the tested inducers, the addition of 0.1mM of ABTS coupled with 1.0mM of 2,5-xylidine in the medium after 24 h of cultivation proved optimal with regard to laccase enzyme production.

Corrosion and Adhesion of Electrophoretic Paint on AZ31 Magnesium Alloy Pretreated in Cerium Chemical Conversion Coating Solution

  • In this study, the corrosion resistance and adhesion of electrophoretic paint (E-paint) were studied on AZ31 magnesium alloy pretreated in cerium chemical conversion coating solutions with the addition of various ethanol concentrations. It was found that with increasing ethanol concentration from 0 to 90 percent can decrease the formation of $Mg(OH)_2/MgO$ and increase the formation of nano-crystalline cerium oxides on the coating. Both corrosion resistance and adhesion of E-painted AZ31 increased with increasing ethanol concentration. The best E-paint sample was observed on the sample pretreatment in cerium chemical conversion coating solution with the addition of 80 percent of ethanol. This sample showed an excellent adhesion without paint detached after water immersion test for 500 h at $40^{\circ}C$, and only a few blisters observed at the near scratched sites after 1000 h salt-spray test.

  • The toxicity of 54 selected process effluents of 16 industry classifications in Korea was evaluated by bioassay test using fish Oryzias latipes, invertebrate Daphnia magna, algae Selenastrum capricornutum and bacteria Vibrio fischeri. Our results demonstrate that the toxicity assessment, such as bioassay test, is effective and of practical use for industrial wastewater management in 54 selected effluents with the limitation of the physico-chemical permit. Among 54 effluents, 4 sample failed physico-chemical permit limitation and 50 passed it. The physcio-chemical measurement items and permit concentrations on the present Water Quality Conservation Act are low related a ecotoxicity.

  • To easily separate chitosanoligosaccharides by size exclusion, an coencapsulating technology of substrate and enzyme was developed. The membrane was composed of alginate and a divalent cation such as calcium. Chitosan and chitosanase were enveloped in this membrane and the product released to medium by size exclusion. The capsule was stabilized in a 2% acetic acid solution (pH 5.0) containing 0.145 M CaCO$_3$. The leakage of substrate caused by the agitation speed was controlled by increasing alginate and CaCO$_3$concentrations. The lower limit of the alginate concentration and the agitation speed were 0.5% and 49rpm, respectively. Membrane thickness and capsule diameter were 10$\mu\textrm{m}$ and 2.5mm, respectively. By TLC analysis, the composition of chitosanoligosaccharides were mainly 3-6 mers. The molecular weight distribution of the released oligosaccharides ranged from 262 to 3624 Da by GPC.
