Electrochemical Behaviors of Pt-Ru Catalysts on the Surface Treated Mesoporous Carbon Supports for Direct Methanol Fuel Cells (직접메탄올 연료전지용 표면처리된 중형기공 탄소지지체에 담지된 백금-루테늄 촉매의 전기화학적 거동)

  • In this work, the effect of surface treatment on mesoporous carbons (MCs) supports was investigated by analyzing surface functional groups. MCs were prepared by a conventional templating method using mesoporous silica (SBA-15) for using catalyst supports in direct methanol fuel cells (DMFCs). The MCs were treated with different phosphoric acid ($H_3PO_4$) concentrations i.e., 0, 1, 3, 4, and 5 M at 343 K for 6 h. And then Pt-Ru was deposited onto surface treated MCs (H-MCs) by chemical reduction method. The characteristics of Pt-Ru catalysts deposited onto H-MCs were determined by specific surface area and pore size analyzer, X-ray diffraction, X-ray photoelectron, transmission electron microscopy, and inductive coupled plasma-mass spectrometer. The electrochemical properties of Pt-Ru/H-MCs catalysts were also analyzed by cyclic voltammetry experiments. From the results of surface analysis, an oxygen functional group was introduced to the surface of carbon supports. From the results, the H4M-MCs carbon supports surface treated with 4 M $H_3PO_4$ led to uniform dispersion of Pt-Ru onto H4M-MCs, resulting in enhancing the electro-catalytic activity of Pt-Ru catalysts.

Groundwater Quality Characteristics of Pollution Concerned Area in Gyeongnam Using Groundwater Quality Monitoring Data (지하수수질측정망 자료를 활용한 경남 오염우려지역의 지하수 수질 특성)

  • This study analyzed the groundwater quality characteristics according to the main source of pollution and quarter (season) by using data from the pollution exclusive monitoring network in the Gyeongsangnam-do area for five years (2013-2017). The main source of pollution was the industrial complex areas, waste mines, and sewage treatment facilities. The analysis items were field measurement items (water temperature, pH, electrical conductivity, dissolved oxygen, oxide reduction potential), positive ions, and negative ions. Water temperature and pH did not vary significantly according to the main source of pollution. In industrial complex areas, the value of electrical conductivity was the highest, and dissolved oxygen value was the lowest. The mean concentration of positive and negative ions was the largest in industrial complex areas, followed by sewage treatment facilities and waste mines. It was shown that the concentration of sodium ion was the highest in industrial complex areas and calcium ion in waste mines and sewage treatment facilities. The concentration of bicarbonate ion was the highest in all main sources of pollution. Water temperature, pH, and concentrations of cations and anions did not vary significantly from quarter to quarter. Of the water quality types, the Na-HCO3 type accounted for the highest proportion, but the Na-Cl type, which has a high possibility of external contamination, accounted for about 20% of the total data in the pollution exclusive monitoring network.

Transfer of Arsenic from Paddy Soils to Rice Plant under Different Cover Soil Thickness in Soil Amendments in Abandoned Coal Mine (폐탄광지역 비소오염 농경지(논) 개량 시 복토두께에 따른 비소의 벼 전이 및 토양용액 특성)

  • This study was carried out to investigate the feasibility of reducing clean cover soil using a flooded column test in arsenic-contaminated farmland reclamation of abandoned coal mine area that shows generally low or about worrisome level (25 mg/kg) of Korea soil environment conservation act unlike abandoned metal mine. During the monitoring period of soil solution for 4 months, chemical properties (pH, EC, ORP, Fe, Mn, Ca, and As) in each layer (clean soil cover and contaminated/stabilized soil) showed different variation. This result revealed that soil solution in stabilized or contaminated soil rarely affected that in cover soil. Whether stabilized or not, arsenic concentrations in the rice roots grown in the soil covers with the thickness of 40 cm decreased by 98% in compared with the that grown in the control soil. In case of the soil covers with 20 cm thickness on stabilized soil, it decreased by 80% and this was 22 percentage point higher than when the soil of lower layer was not stabilized. Thus, reducing clean cover soil could be possible in contaminated farmland soil reclamation if appropriate stabilization of contaminated soil is carried.

Rare Earth Elements of Atmospheric Particulates (PM2.5) in Northeast Asia: Beijing and Gwangju (동북아시아 초미세먼지의 희토류 원소 특성 비교: 북경과 광주)

  • Rare earth elements (REEs) have been used as one of power tracers for understanding geological and environmental changes due to their similar physico-chemical properties. In this study, we investigated the characteristics of rare earth elements in atmospheric particulates(PM2.5) collected in Beijing and Gwangju during January 2018. The total concentrations of REEs in the Beijing samples were about 16X higher than those in Gwangju samples, and both samples are enriched in light REE than heavy REE, up to 8-10 times. The Post Archean Australian Shale (PAAS)-normalized pattern showed that both samples are enriched in Eu, Tb, and Er, and displayed positive Eu but negative Ce anomalies. The elemental correlations indicate that both samples originated from China desert and Loess plateau as well as cities surrounding Beijing. This study suggests that REEs in PM2.5 can be used as a powerful proxy of revealing the difference between China and Korea, and provide basic information on the source and transport of PM2.5.

Effect of serotonin on the cell viability of the bovine mammary alveolar cell-T (MAC-T) cell line

  • 5-Hydroxytryptamine (5-HT), a monoamine, as a local regulator in the mammary gland is a chemical signal produced by the mammary epithelium cell. In cows, studies have shown that 5-HT is associated with epithelial cell apoptosis during the degenerative phase of the mammary gland. However, studies in other tissues have shown that 5-HT can effectively promote cell viability. Whether 5-HT could have an effect on mammary cell viability in dairy cows is still unknown. The purpose of this study was to determine: (1) effect of 5-HT on the viability of bovine mammary epithelial cells and its related signaling pathways, (2) interaction between prolactin (PRL) and 5-HT on the cell viability. The bovine mammary alveolar cell-T (MAC-T) were cultured with different concentrations of 5-HT for 12, 24, 48 or 72 hours, and then were assayed using cell counting kit-8, polymerase chain reaction (PCR) and immunobloting. The results suggested that 20 μM 5-HT treatment for 12 or 24 h promote cell viability, which was mainly induced by the activation of 5-HT receptor (5-HTR) 1B and 4, because the increase caused by 5-HT vanished when 5-HTR 1B and 4 was blocked by SB224289 and SB204070. And protein expression of mammalian target of rapamycin (mTOR), eukaryotic translation elongation factor 2 (eEF2), janus kinase 2 (JAK2) and signal transducer and activator of transcription 5 (STAT5) were decreased after blocking 5-HT 1B and 4 receptors. When MAC-T cells were treated with 5-HT and PRL simultaneously for 24 h, both the cell viability and the level of mTOR protein were significantly higher than that cultured with 5-HT or PRL alone. In conclusion, our study suggested that 5-HT promotes the viability of MAC-T cells by 5-HTR 1B and/or 4. Furthermore, there is a reciprocal relationship between PRL and 5-HT.

Desalinization Effect of Pennisetum Alopecuroides and Characteristics of Leachate Depending on Calcium Chloride (CaCl2) Concentration

  • Background and objective: Calcium chloride (CaCl2) and sodium chloride (NaCl) are commonly used as a deicing agent in South Korea and penetrate the soil on the roadside, causing damage to plants. This study was conducted to investigate the salinity reduction effect of Pennisetum alopecuroides and the chemical characteristics of soil leachate. Methods: The plants were treated with five different concentrations of CaCl2 (0, 1, 2, 5, and 10g·L-1) and were grouped into the Cont., C1, C2, C5, and C10 groups. CaCl2 of 200 m·L-1 was sprayed to each plant once every two weeks. The growth of P. alopecuroides (plant height, leaf length, leaf width and the number of leaves) was measured. The level of EC and pH, and exchangeable cations (K+, Ca2+, Na+, and Mg2+) in the leachate of soil was monitored. Results: The pH of soil leachate decreased as the CaCl2 concentration increased, and the EC increased significantly. The content of K+ did not change significantly until the concentration of CaCl2 reached 5 g·L-1, but the content of Ca2+, Na+, and Mg2+ significantly increased. The plant height, leaf length, and leaf width of P. alopecuroides showed the highest value in CaCl2 1 g·L-1 followed by CaCl2 2 g·L-1 and the control group. Root fresh weight was the highest in CaCl2 2 g·L-1. On the other hand, there was no change in the shoot fresh weight, dry weight and root dry weight, and P. alopecuroides growth inhibition at the concentration of 5 g·L-1 or higher in the plant height and leaf length. Conclusion: P. alopecuroides is relatively highly salt-tolerant and can improve the salt damaged soil by lowering the content of the salt-based exchangeable K+ ions.

Mulberry Low-Fat Ice Cream Supplemented with Synbiotic: Formulation, Phytochemical Composition, Nutritional Characteristics, and Sensory Properties

  • For this study, we designed and produced mulberry low-fat ice cream supplemented with synbiotics (MLF-ISS). The sensory characteristics and physical, chemical, and microbiological qualities of MLF-ISS were then determined. Mulberry juice inoculated with or without probiotic (Lactobacillus plantarum TISTR 926 and Saccharomyces boulardii CNCM I-745) was also tested at 37℃ for 24 h to determine probiotic growth rate, pH, total anthocyanin content (TAC), total phenolic content (TPC), and antioxidant activity (AA). Only the TAC of mulberry juice inoculated with S. boulardii CNCM I-745 increased considerably (p < 0.05) among these parameters. MLF-ISS was produced with varied mulberry fruit concentrations (0, 10, 20, 30, or 40%) (w/w). The MLF-ISS prepared with 30% mulberry fruit (w/w) (30-MLF-ISS) had a higher score in appearance, color, and sweetness (p < 0.05) when sensory qualities were measured using the 9-point hedonic scale method. In the CIE lab system (L*, a*, b*), the color values of 30-MLF-ISS were 27.80 ± 0.26, 12.99 ± 0.59, and 1.43 ± 0.05, respectively. The 30-MLF-ISS was also subjected to a proximate analysis. The melting rate of 30-MLF-ISS was 0.29 ± 0.03 g/min and the time it took for the first drop to fall was 37.00 ± 7.00 min. TAC, TPC, and AA of 30-MLF-ISS were observed to alter significantly (p < 0.05) during varied intervals of storage at - 18℃ (0, 30, and 60 days). The viability of probiotics in 30-MLF-ISS slightly decreased after storage at -18℃ for 8 weeks, but remained about 6 log CFU/g. During storage at -18℃ for 0 and 120 days, no pathogenic bacteria were detected in 30-MLF-ISS. These findings show that 30-MLF-ISS has nutritional and functional value, is free of foodborne pathogenic bacteria, is safe for consumers' health, and is suitable for application in the ice cream and related food industries.

Development of Regression Models for Estimation of Unmeasured Dissolved Organic Carbon Concentrations in Mixed Land-use Watersheds (복합토지이용 유역의 수질 관리를 위한 미측정 용존유기탄소 농도 추정)

  • In order to prevent water pollution caused by organic matter, Total Organic Carbon(TOC) has been adopted indicator and monitored. TOC can be divided into Dissolved Organic Carbon(DOC) and Particulate Organic Carbon(POC). POC is largely precipitated and removed during stream flow, which making DOC environmentally significant. However, there are lack of studies to define spatio-temporal distributions of DOC in stream affected by various land use. Therefore, it is necessary to estimate the past DOC concentration using other water quality indicators to evaluate status of watershed management. In this study, DOC was estimated by correlation and regression analysis using three different organic matter indicators monitored in mixed land-use watersheds. The results of correlation analysis showed that DOC has the highest correlation with TOC. Based on the results of the correlation analysis, the single- and multiple-regression models were developed using Biochemical Oxygen Demand(BOD), Chemical Oxygen Demand(COD), and TOC. The results of the prediction accuracy for three different regression models showed that the single-regression model with TOC was better than those of the other multiple-regression models. The trend analysis using extended average concentration DOC data shows that DOC tends to decrease reflecting watershed management. This study could contribute to assessment and management of organic water pollution in mixed land-use watershed by suggesting methods for assessment of unmeasured DOC concentration.

A Genetically Encoded Biosensor for the Detection of Levulinic Acid

  • Levulinic acid (LA) is a valuable chemical used in fuel additives, fragrances, and polymers. In this study, we proposed possible biosynthetic pathways for LA production from lignin and poly(ethylene terephthalate). We also created a genetically encoded biosensor responsive to LA, which can be used for screening and evolving the LA biosynthesis pathway genes, by employing an LvaR transcriptional regulator of Pseudomonas putida KT2440 to express a fluorescent reporter gene. The LvaR regulator senses LA as a cognate ligand. The LA biosensor was first examined in an Escherichia coli strain and was found to be non-functional. When the host of the LA biosensor was switched from E. coli to P. putida KT2440, the LA biosensor showed a linear correlation between fluorescence intensity and LA concentration in the range of 0.156-10 mM LA. In addition, we determined that 0.156 mM LA was the limit of LA detection in P. putida KT2440 harboring an LA-responsive biosensor. The maximal fluorescence increase was 12.3-fold in the presence of 10 mM LA compared to that in the absence of LA. The individual cell responses to LA concentrations reflected the population-averaged responses, which enabled high-throughput screening of enzymes and metabolic pathways involved in LA biosynthesis and sustainable production of LA in engineered microbes.

Optimization of Soil Contamination Distribution Prediction Error using Geostatistical Technique and Interpretation of Contributory Factor Based on Machine Learning Algorithm (지구통계 기법을 이용한 토양오염 분포 예측 오차 최적화 및 머신러닝 알고리즘 기반의 영향인자 해석)

  • When creating a soil contamination map using geostatistical techniques, there are various sources that can affect prediction errors. In this study, a grid-based soil contamination map was created from the sampling data of heavy metal concentrations in soil in abandoned mine areas using Ordinary Kriging. Five factors that were judged to affect the prediction error of the soil contamination map were selected, and the variation of the root mean squared error (RMSE) between the predicted value and the actual value was analyzed based on the Leave-one-out technique. Then, using a machine learning algorithm, derived the top three factors affecting the RMSE. As a result, it was analyzed that Variogram Model, Minimum Neighbors, and Anisotropy factors have the largest impact on RMSE in the Standard interpolation. For the variogram models, the Spherical model showed the lowest RMSE, while the Minimum Neighbors had the lowest value at 3 and then increased as the value increased. In the case of Anisotropy, it was found to be more appropriate not to consider anisotropy. In this study, through the combined use of geostatistics and machine learning, it was possible to create a highly reliable soil contamination map at the local scale, and to identify which factors have a significant impact when interpolating a small amount of soil heavy metal data.
