An organic solvent stable lipase from solvent-tolerant Pseudomonas sp. BCNU 171 had an optimal pH of 8 and an optimal temperature of 37℃. This crude extracellular lipase from BCNU 171 exhibited increased stability in the presence of various types of solvents at high concentrations (25%, v/v). The lipase stability was found to be highest in the presence of xylene (137%), followed by toluene (131%), octane (130%), and butanol (104%). Overall, BCNU 171 lipase tended to be more stable than immobilized commercial lipase (Novozyme435) in the presence of organic solvents. Furthermore, BCNU 171 lipase maintained about 90% of its enzyme original activity in the presence of NH4+, Na+, Ba2+, Hg2+, Ni2+, Cu2+, and Ca2+ion and significantly increased its enzyme activity in the presence of various emulsifying agents. Thus, the organic solvent stable lipase from Pseudomonas sp. BCNU 171 could be usable as a potential whole cell biocatalyst and for synthetic applications of enzymes for industrial chemical processes in organic solvents without using immobilization.
A liquefied seasoning material was manufactured by using the enzymatic hydrolysis for the benefit of highly effective utilization of cockle shell by-product, and their quality was investigated. The weight ratio of by-product to whole cockle shell was 32.7%, and the contents of moisture and crude protein in the raw cockle shell by-product were 83.1% and 10.7%, respectively. The optimal concentrations of protease such as Protease N. P.(Pacific Chemical Co.) and Alcalase(Noo co), used in order to reduced the hydrolysis period, were effective at 4%(w/w), and optimal hydrolyzing time was 8 hours and after 8 hours were little changed. To improve flavor of the liquefied seasoning material, by Maillard reaction used thermal treatment, addition of glucose was very effective. And addition in hydrolysate with 10% glucose, 9% table salt, 2% starch and 0.5% caramel were suitable for promotion of taste. Total nitrogen and amino type nitrogen in the product were 1,607mg% and 1,264mg%, respectively. And the ratio of amino type nitrogen to the total nitrogen was 78.6%. The major free amino acid were glutamic acid, lysine, leucine, valine and aspartic acid, and content of glutamic acid was 1,027.5mg%.
A ditch is a facility for managing washed-off runoff from parking lot area. Washed-off runoff inflows into ditches where it is retained for a short period of time. At this point, it is assumed that a ditch is a preliminary unit for runoff treatment. This research carries out the distribution of particle size and chemical compound for sediment in parking lot ditch. This work is important to understand the amount of generated sediment from this area to be able to determine different particle size ranges for treatment. Metal concentrations for sediment according to particle size are analyzed. From the distribution of particle size, the weight ratio with the range of $425-850{\mu}m$ is the highest. Considering its weight ratio, the metal concentration of coarser particles is high, otherwise metal concentration increases as particle size decreases. Metal load of the range is higher and the ratio of total metal load in the case of Cu, Pb, Zn is nearly 30%. Moreover metal concentration associated with particle size depends on particle ratio. To manage non-point source pollution for parking lot area, these results can be used with this ditch unit.

In this study, the effect of physico-chemical variables on sludge conditioning was determined to enhance dewaterability of effluent produced from the thermophilic anaerobic digestion of food waste. The gas production rate and methane content during the anaerobic digestion of food waste were $1.1m^3/kg$ VS and 63%, respectively, and the biodegradability of volatile solids was 87.5%. The concentrations of CODcr, TKN and TP of effluent from digestor were 18,500mg/L, 2,800mg/L, and 582mg/L, respectively. At the jar test to screen the flocculant for the dewatering of effluent from digestor, $FeCl_3$ and strong cationic polymer were effective on making flocs in the effluent. The condition of flocculation of effluent were 500mg/L of $FeCl_3$ and 50-100 mg/L of strong cationic polymer, respectively. As the result of measuring of dewaterability potential of effluent to determine the mixing ratio between $FeCl_3$ and polymer by capillary suction time(SCT), optimum condition was 500mg/L of $FeCl_3$ and 80mg/L of strong cationic polymer.
Oil pollution was world-wide prevalent treat to the environment, and the physic-chemical remediation technology of the TPH (total petroleum hydrocarbon) contaminated soil had the weakness that its rate was very slow and not economical. Bioremediation of the contaminated soil is a useful method if the concentrations are moderate and non-biological techniques are not economical. The aim of this research is to investigate the influence of additives on TPH degradation in a diesel contaminated soil environment. Six experimental conditions were conduced; (i) diesel contaminated soil, (ii) diesel contaminated soil treated with microbial additives, (iii) diesel contaminated soil treated with microbial additives and the mixture was titrated to the end point of pH 7 with NaOH, (iv) diesel contaminated soil treated with microbial additives and accelerating agents and (v) diesel contaminated soil treated with microbial additives and accelerating agents, and the mixture was titrated to the end point of pH 7 with NaOH. After 10 days, significant TPH degradation (67%) was observed in the DSP-1 soil sample. The removal of TPH in the soil sample where microbial additives were supplemented was 38% higher than the control soil sample during the first ten days. The microbial additives were effective in both the initial removal rate and relative removal efficiency of TPH compared with the control group. However, various environmental factors, such as pH and temperature, also affected the activities of microbes lived in the additives, so the pH calibration of the oil-contaminated soil would help the initial reduction efficiency in the early periods.
In this study, the rhizoremediation of petroleum and heavy metal-contaminated soil was characterized employing Zea mays and two plant-growth promoting rhizobacteria, Gordonia sp. S2RP-17 and Serratia sp. SY5 which have petroleum-degrading activity and heavy metal-resistance, respectively. After 51 days, the average dry weights of Zea mays' root without and with the inoculation of rhizobacteria were $1.9{\pm}0.2$ and $5.6{\pm}0.7\;g$, respectively. Compared with initial TPH concentration in soil ($21,576{\pm}3,426\;mg-TPH{\cdot}kg-dry\;soil^{-1}$), the residual TPH concentrations were $220{\pm}98\;mg-TPH{\cdot}kg-dry\;soil^{-1}$ in soil planted with Zea mays, and $20{\pm}41\;mg-TPH{\cdot}kg-dry\;soil^{-1}$ in soil planted with Zea mays and inoculated with rhizobacteria. These results indicated that the inoculation of S2RP-17 and SY5 could promote TPH removability in soil as well as the growth of Zea mays' root. There was little positive effect of the rhizobacteria inoculation on the removability of heavy metal such as Cu, Cd and Pb in soil planted with Zea mays.

Genus Nitrospira and Nitrobacter species are the key nitrite-oxidizing bacteria(NOB) in nitrifying wastewater treatment plants. It has been hypothesized that genus Nitrospira are K-strategists(low $K_6$ value) that can exploit low amounts of nitrite more efficiently than Nitrobacter. In contrast, Nitrobacter species are r-strategists(high $V_{max}$) that can grow faster than Nitrospira. It has also been known that the availability of organic compounds and dissolved oxygen as well as nitrite affects the distribution of NOB. In this study, we determined the distribution and competition of NOB in wastewater nitrification systems where nitrite, organic compounds, and dissolved oxygen concentrations were compositively varied. For the purpose, several compounds of the laboratory-scale nitrificaiion bioreactor and full-scale $A_2O$ wastewater treatment plant and their distribution of NOB were analyzed and compared. The analysis showed that Nitrobacter was the dominant NOB in nitrification bioreactor where average nitrite was maintained at 5 mg-N/L with very low organic concentration in aerobic condition, whereas Nitrospira was the dominant NOB in full-scale $A_2O$ plant where nitrite was maintained very low and organic compounds were maintained relatively high in alternating aerobic-anoxic condition. The result indicates that nitrite concentration is more critical factor than organics and dissolved oxygen which determines the dominant NOB in nitrification system and it is confirmed that Nitrospira and Nitrobacter showed the characteristics of r-strategist and K-strategist, respectively.

The cultivated pearls collected for the study were pretreated through the removal of contaminants and the surface bleaching for easy dyeing. Coloring of pearls are necessary after selecting dyes adsorbable to the Conchiolin layer, a kind of hard protein formed in the seawater, covering the surface of the pretreated pearls. Dyes adsorbable to the Conchiolin layers are mostly basic dyes such as Rhodamine 6G(R6G), Rhodamine B(RB), Methylene Blue(MB) etc. and the binary and ternary competitive adsorption were performed by mixing two or three dyes together. The multi-dye adsorption data were compared with the predictions from the ideal adsorbed solution theory(IASI) combined with the single-dye adsorption model, the Langmuir or the Redlich-Peterson(RP) model. The quality of prediction was compared by using determination coefficient($R^2$) and standard deviation(SSE) values. Predictions from the IAST were found to be in good agreement with the data for the R6G/RB binary adsorption to the pearl layers not fractionated with their size, except for the adsorption data for RB at high concentrations. Among the three binary adsorption systems, R6G/RB, R6G/MB, and MB/RB, only the RB sorption data in the R6G/RB binary system was in poor agreement with the IAST prediction. Competitive adsorption data in ternay systems were in good agreement with the predictions from the IAST except for the RB data.
Corynebacterium glutamicum is one of the well-studied industrial strain that is used for the production of nucleotides and amino acids. Recently, it has also been studied as a possible producer of organic acids such as succinic acid, based on its ability to produce organic acids under an oxygen deprivation condition. In this study, we conducted the optimization of medium components for improved succinate production from C. glutamicum under an oxygen deprivation condition by Plackett-Burman design and applied a response surface methodology. A Plackett-Burman design for ten factors such as glucose, ammonium sulfate, magnesium sulfate, potassium phosphate ($K_2HPO_4$ and $KH_2PO_4$), iron sulfate, manganese sulfate, biotin, thiamine, and sodium bicarbonate was applied to evaluate the effects on succinate production. Glucose, ammonium sulfate, magnesium sulfate, and dipotassium phosphate were found to have significant influence on succinate production, and the optimal concentrations of these four factors were sequentially investigated by the response surface methodology using a Box-Behnken design. The optimal medium components obtained for achieving maximum concentration of succinic acid were as follows: glucose 10 g/l, magnesium sulfate 0.5 g/l, dipotassium phosphate ($K_2HPO_4$) 0.75 g/l, potassium dihydrogen phosphate ($KH_2PO_4$) 0.5 g/l, iron sulfate 6 mg/l, manganese sulfate 4.2 mg/l, biotin 0.2 mg/l, thiamine 0.2 mg/l, and sodium bicarbonate 100 mM. The parameters that differed from a normal BT medium were glucose changed from 40 g/l to 10 g/l, dipotassium phosphate ($K_2HPO_4$) 0.5 g/l changed to 0.75 g/l, and ammonium sulfate ($(NH_4)_2SO_4$) 7 g/l changed to 0 g/l. Under these conditions, the final succinic acid concentration was 16.3 mM, which is about 1.46 fold higher than the original medium (11.1 mM) at 24 h. This work showed the improvement of succinate production by a simple change of media components deduced from sequential optimization.

The field surveys and the seawater analyses were conducted over the southern waters of Cheju island every month from July 1997 to June 1988. The distributions of dissolved inorganic nitrogen(DIN), dissolved inorganic phosphorus(DIP), chemical oxygen demand(COD) and dissolved oxygen(DO) in the southern water area of Cheju island were simulated and reproduced by an ecosystem model. In order to estimate the environmental capacity of the southern coastal area of Cheju island, the simulations for predicting the situation of unfavorable environment in which marine water quality might not be satisfied with Korean standards were performed with an ecosystem model by controlling quantitatively the pollution loads of discharge sources including streams flowing into the sea. The more the loads of 4 major pollution sources flowing into model region increase, the more evident appear the increases of COD, DIN and DIP concentrations around the water areas adjacent to pollution sources. In case the pollution loads of all 4 discharge sources including streams become 3 times as high as the present loads, the concentration of DIN at near-shore waters appears to be increased to about 14.5mmol/m³(=0.20mg/ℓ) which is the third class criterion of Korean standards for marine water quality. In case of 10 times as high as the present loads, COD at near-shore waters appears to be increased to about 1.0mg/ℓ which is the first class criterion of Korean standards. In case of 20 times as high as the present loads, DIP at near-shore waters appears to be increased to about 0.50mmol/m³(=0.015mg/ℓ) which is the second class criterion of Korean standards.
