Observations of Infection Structures after Inoculation with Colletotrichum orbiculare on the Leaves of Cucumber Plants Pre-inoculated with Two Bacterial Strains Pseudomonas putida or Micrococcus luteus

  • Infection structures were observed at the penetration sites on the leaves of cucumber plants inoculated with Colletotrichum orbiculare using a fluorescence microscope. The cucumber plants were previously drenched with suspension of bacterial strains Pseudomonas putida or Micrococcus luteus. The plants pre-inoculated with both bacterial strains were resistant against anthracnose after inoculation with C. orbiculare. To investigate the resistance mechanism by both bacterial strains, the surface of infected leaves was observed at the different time after challenge inoculation. At 3 days after inoculation there were no differences in the germination and appressorium formation of conidia of C. orbiculare as well as in the callose formation of the plants between both bacteria pre-inoculated and non-treated. At 5 days, the germination and appressorium formation of the fungal conidia were, however, significantly decreased on the leaves of plants pre-inoculated with M. luteus at the concentration with $1.0{\times}10^7\;cfu/ml$. Furthermore, callose formation of plants cells at the penetration sites was apparently increased. In contrast, there were no defense reactions of the plants at the concentration with $1.0{\times}10^6\;cfu/ml$ of M. luteus. Similarly, inoculation P. putida caused no plant resistance at the low concentration, whereas increase of callose formation was observed at the higher concentration. The results of this study suggest that the resistant mechanisms might be differently expressed by the concentration of pre-treatment with bacterial suspension.

Glyceraldehyde-3-Phosphate Dehydrogenase, an Immunogenic Streptococcus equi ssp. zooepidemicus Adhesion Protein and Protective Antigen

  • Streptococcus equi ssp. zooepidemicus (Streptococcus zooepidemicus, SEZ) is an important pathogen associated with opportunistic infections of a wide range of species, including pigs and humans. The absence of a suitable vaccine makes it difficult to control SEZ infection. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) has been previously identified as an immunogenic protein using immunoproteomic techniques. In the present study, we confirmed that the sequence of GAPDH was highly conserved with other Streptococcus spp. The purified recombinant GAPDH could elicit a significant humoral antibody response in mice and confer significant protection against challenge with a lethal dose of SEZ. GAPDH could adhere to the Hep-2 cells, confirmed by flow cytometry, and inhibit adherence of SEZ to Hep-2 cells in an adherence inhibition assay. In addition, real-time PCR demonstrated that GAPDH was induced in vivo following infection of mice with SEZ. These suggest that GAPDH could play an important role in the pathogenesis of SEZ infection and could be a target for vaccination against SEZ.

Intake of Korean Red Ginseng Extract and Saponin Enhances the Protection Conferred by Vaccination with Inactivated Influenza A Virus

  • Vaccination is the main strategy for preventing influenza infection. However, vaccine efficacy is influenced by several factors, including age and health status. The efficacy of the influenza vaccine is much lower (17% to 53%) in individuals over 65 yr of age compared with young adults (70% to 90%). Therefore, increasing vaccine efficacy remains a challenge for the influenza vaccine field. In this study, we investigated the impact of supplementing vaccination with the dietary intake of Korean red ginseng (RG) extract and RG saponin. Mice were immunized two times intranasally with inactivated influenza A (H1N1) virus. Mice received RG extract or RG saponin orally for 14 d prior to the primary immunization. After the primary immunization, mice continued to receive RG extract or RG saponin until the secondary immunization. Mice vaccinated in combination with dietary intake of RG extract and RG saponin showed elevated serum anti-influenza A virus IgG titers and improved survival rates in lethal influenza A virus infection: 56% and 63% of mice receiving RG extract or RG saponin survived, respectively, while 38% of mice that only received the vaccine survived. Moreover, mice receiving RG extract supplementation recovered their body weight more quickly than those not receiving RG extract supplementation. We propose that the dietary intake of RG extract and RG saponin enhances the vaccine-induced immune response and aids in providing protection against influenza virus infection.

Toxoplasma gondii Induces Apoptosis via Endoplasmic Reticulum Stress-Derived Mitochondrial Pathway in Human Small Intestinal Epithelial Cell-Line

  • Toxoplasma gondii, an intracellular protozoan parasite that infects one-third of the world's population, has been reported to hijack host cell apoptotic machinery and promote either an anti- or proapoptotic program depending on the parasite virulence and load and the host cell type. However, little is known about the regulation of human FHs 74 small intestinal epithelial cell viability in response to T. gondii infection. Here we show that T. gondii RH strain tachyzoite infection or ESP treatment of FHs 74 Int cells induced apoptosis, mitochondrial dysfunction and ER stress in host cells. Pretreatment with 4-PBA inhibited the expression or activation of key molecules involved in ER stress. In addition, both T. gondii and ESP challenge-induced mitochondrial dysfunction and cell death were dramatically suppressed in 4-PBA pretreated cells. Our study indicates that T. gondii infection induced ER stress in FHs 74 Int cells, which induced mitochondrial dysfunction followed by apoptosis. This may constitute a potential molecular mechanism responsible for the foodborne parasitic disease caused by T. gondii.

Potential harmful effects of viral hemorrhagic septicemia virus in mammals

  • Most of the emerging diseases that threaten humans are caused by RNA viruses which are extremely mutable during evolution. The fish RNA virus, viral hemorrhagic septicemia virus (VHSV) can infect a broad range of aquatic animal hosts, but the transmissibility of VHSV to mammals has not been thoroughly investigated. Therefore, our study aimed to investigate the potential adverse effects of VHSV in mammals. Briefly, the survival of VHSV was determined using only minimum essential media (MEM-2) and mammalian SNU-1411 and hepa-1c1c7s cells at 15℃ and 37℃. Mice (Mus musculus, 27.3 ± 1.9 g) were intravenously injected with VHSV (2.37E+05 TCID50·mice-1) in triplicate. Clinical signs and survival rates were examined at 14 days post-challenge, and infection was confirmed in the surviving mice. The 50% tissue culture infective dose (TCID50) and polymerase chain reaction analysis were used to determine viral titers and the infection rate, respectively. The titer of VHSV suspended in MEM-2 at 15℃ was reduced by only one log after 8 days, whereas the virus maintained at 37℃ was inactivated 8 days post-inoculation (dpi). There were no recognizable cytopathic effects in either SNU-1411 or hepa-1c1c7s cells inoculated with VHSV at 15℃ and 37℃. VHSV in those cell lines at 37℃ was rapidly decreased and eventually inactivated at 12 dpi, whereas virus at 15℃ remained at low concentrations without replication. In vivo experiment showed that there were no signs of disease, mortality, or infection in VHSV-infected mice. The results of this study indicate that it is highly unlikely that VHSV can infect mammals including humans.

우기종저(牛氣腫疽)에 대한 예방약(豫防藥)과 항혈청(抗血淸)의 검정(檢定)을 위한 연구(硏究) (Studies on the Biological Assay of Black leg Vaccine and Antiserum)

  • Throughout the studies the following experimental results were summarized. 1. It was impossible to infect and kill the mice, weighing 10 to 12 gm, by inoculating 0.2ml of virulent Cl. chauvoei, diluted 1 to 10 with physiological saline, via subcutaneous, intramuscular, intraperitoneal or intraveonus, route. 2. The mice which were inoculated in brain with 0.03ml of Cl. chauvoei diluted 1 : 5120 with physiological saline were resulted in all death after infection, but not in case of attenuated strain even in dilution of one to five. 3. Virulent Cl. chauvoei were diluted with each of those of whole blood, erythrocytes and serum of horse, calf, swine, sheep, rabbit, guinea pig, chicken and duck, human plasma and 2% CaCl solution, and inoculated subcutaneously 0.25 to 0.5ml in mice, weighing 12 to 15gm. It was resulted in significant increase in virulence as comparing with the case of physiological saline solution except when horse and pig sera were used. Such a phenomena were not seen in attenuated strain. 4. Virulence of virulent Cl. Chauvoei could be increased significantly in rat, as the procedures used in mice, by suspending in whole blood, erythrocytes, serum, or plasma of various animals, or 2% $CaCl_2$ solution and by inoculating subcutaneously 0.5 to 10ml in rat, weighing 30 to 60 gm, as compared with those of control group which used physiological saline solutionos diluent. 5. Mice resisted 100 and 80 percent against challenge of $10^3$ and $10^4$ M.L.D.. respectively, 24 hours after inoculation of 0.5ml black leg antiserum. 6. Immune response to the black leg living vaccine in mice could be obtained more favorably in the group of respected vaccination rather than those of single inoculation and the most profitable inoculm size of the vacine was 0.5 to 1.0ml. 7. Challenge for the immunized mice could be carried out effectively 3 weeks after first vaccination. 8. Satisfactory results could be obtained by inoculating subcutaneously for the immunization and intracerebrally or subcutaneously for the challenge. 9. Mice which were inoculated with 0.5ml of black leg living vaccine via subtaneucously two times at seven days interval and 21 days after first inoculation and challenged with 5 and 10 M.L.D. of virulent strain, resited 100 and 70 to 80 percent respectively. Same results were obtainable in black leg killed vaccine as the procedures used in living vaccine. 10. There were significantly different resistances against the definite challenge does between the mice groups which were immnuized with the living vaccine diluted five or 10 times and the undiluted. 11. For the biological assay of black leg living vaccine and antiserum, satisfactory results could be obtained using mice.

Edwardsiella tarda 인위감염에 의한 넙치, Paralichthys olivaceus의 가계별 내성변이 (Family variation in survival of olive flounder Paralichthys olivaceus after Edwardsiella tarda infection in challenge tests)

  • 본 연구는 에드워드 내성 정도를 기준으로 선발된 넙치 친어를 이용하여 $4{\times}4$ 펙토리얼 교배를 통해 얻어진 16가계 넙치의 에드워드 인위 감염 결과를 이용하여 교배 그룹, 교배 암컷, 교배 수컷의 효과를 분석하였다. 내성 친어간의 교배에 의해 생산된 교배구(RR)의 내성 지수, 생존율, 생존 일수는 각각 17.98, 13.77%, 18.36일로 나타났으나, 민감 친어간의 교배에 의해 생산된 교배구(SS)는 각각 12.46, 2.71%, 12.40일로 나타났다. 이상의 결과에서, 에드워드 질병내성 형질에 대한 가계간, 개체간 내성 정도의차이가 존재하는 것을 확인하였고, 인위 감염 결과를 이용하여 질병 저항성에 대한 선발이 가능할 것으로 판단된다.

Aeromonas hydrophila 인위감염이 붕어, Carassius carassius의 생존율, 혈액학적 성상 및 혈장 성분에 미치는 영향 (Effects of Artificial Infection with Aeromonas hydrophila on Survival Rate, Hematological Parameters and Plasma Components of Crucian Carp, Carassius carassius)

  • 붕어(무게 28.1±3.7 g, 길이 10.0±1.0 cm)에 0, 2.0×104, 2.0×105, 2.0×106, 2.0×107 CFU/ml의 Aeromonas hydrophila를 주사 후 2주간 실험을 실시하였다. A. hydrophila로 인위감염한 Carassius carassius의 2주 뒤 반수치사농도는 19.776×105 CFU/ml이었다. 혈액학적 지표에서는 A. hydrophila에 의해 혈색소와 적혈구 수가 유의하게 감소한 반면, 적혈구 용적은 유의적 변화가 없었다. 마그네슘, 칼슘과 같은 혈장 무기 성분은 유의적으로 감소하였다. 유기 혈장 성분에서 A. hydrophila 인위감염에 의해 혈장 포도당과 콜레스테롤이 유의하게 증가한 반면, 총 단백질은 유의하게 감소하였다. 혈장 효소 성분인 ALP는 A. hydrophila 인위감염에 의해 유의하게 증가하였다. 본 연구의 결과는 C. carassius에 대한 A. hydrophila의 인위감염이 치명적인 병원성 세균으로 혈액학적 성상 및 혈장 성분에 유의한 생리학적 변화를 유도하였음을 의미한다.

비피도박테리아의 생존성 증진을 위한 캡슐화 기술 (Microencapsulation Technology for Enhancement of Bifidobacterium spp. Viability: A Review)

  • The intestinal microbiota has been shown to have a vital role in various aspects of human health, and accumulating evidence has shown the beneficial effects of supplementation with bifidobacteria for the improvement of human health, ranging from protection against infection to various positive effects. However, maintaining bacterial cell viability during storage and gastrointestinal transit remains a challenge. Microencapsulation of probiotic bacterial cells provides protection against adverse conditions during processing, storage, and gastrointestinal passage. In this paper, we review the current knowledge, future prospects, and challenges of microencapsulation of probiotic Bifidobacterium spp.

Dravya-A Putative Organic Treatment Against Alternaria padwickii Infection in Paddy

  • Innate defence mechanism in plants can be triggered and enhanced by certain agents, referred as inducers against broad range of pathogens. In the present study, Dravya (a sea weed extract) was highly compatible with commonly available synthetic fungicides, Bavistin and Dithane M-45. Incidence of Alternaria padwickii and Bipolaris oryzae was also reduced to a greater extent in the paddy seed samples in Dravya treatment. Dravya also enhanced the seed germination and seedling vigour. Seedlings of treated samples also showed enhanced activity of peroxidase upon challenge inoculation with Alternaria padwickii. The enzyme activity was two fold high after the inoculation of pathogen. The suppression in disease incidence in growing plants indicated the promising effect of Dravya and Dithane M-45 under green-house condition.
