Involvement of Cytochrome P450 in (-)-(4R)-Isopiperitenone Oxidation by Cell Suspension Cultures of Mentha piperita

  • Biotransformation of exogenous (-)-(4R)-isopiperitenone in cell suspension cultures of Mentha piperita resulted in oxidized products, with (-)-7-hydroxyisopiperitenone being the major compound. The mass of products obtained $unde^{18}O_2$, atmosphere was two units higher than that under normal atmosphere. The biotransformation was inhibited by several cytochrome P450-specific inhibitors as well as by carbon monoxide. Carbon monooxide inhibition was substantially overcome by irradiation of cells with blue light including light at 450nm wavelength. These results suggested that a cytochrome P450-type monooxygenase was involved in the biotransformation.

Charged Cluster Model as a New Paradigm of Crystal Growth

  • A new paradigm of crystal growth was suggested in a charged cluster model, where charged clusters of nanometer size are suspended in the gas phase in most thin film processes and are a major flux for thin film growth. The existence of these hypothetical clusters was experimentally confirmed in the diamond and silicon CVD processes as well as in gold and tungsten evaporation. These results imply new insights as to the low pressure diamond synthesis without hydrogen, epitaxial growth, selective deposition and fabrication of quantum dots, nanometer-sized powders and nanowires or nanotubes. Based on this concept, we produced such quantum dot structures of carbon, silicon, gold and tungsten. Charged clusters land preferably on conducting substrates over on insulating substrates, resulting in selective deposition. if the behavior of selective deposition is properly controlled, charged clusters can make highly anisotropic growth, leading to nanowires or nanotubes.

Crystal Structure of LysB4, an Endolysin from Bacillus cereus-Targeting Bacteriophage B4

  • Endolysins are bacteriophage-derived enzymes that hydrolyze the peptidoglycan of host bacteria. Endolysins are considered to be promising tools for the control of pathogenic bacteria. LysB4 is an endolysin produced by Bacillus cereus-infecting bacteriophage B4, and consists of an N-terminal enzymatic active domain (EAD) and a C-terminal cell wall binding domain (CBD). LysB4 was discovered for the first time as an L-alanoyl-D-glutamate endopeptidase with the ability to breakdown the peptidoglycan among B. cereus-infecting phages. To understand the activity of LysB4 at the molecular level, this study determined the X-ray crystal structure of the LysB4 EAD, using the full-length LysB4 endolysin. The LysB4 EAD has an active site that is typical of LAS-type enzymes, where $Zn^{2+}$ is tetrahedrally coordinated by three amino acid residues and one water molecule. Mutational studies identified essential residues that are involved in lytic activity. Based on the structural and biochemical information about LysB4, we suggest a ligand-docking model and a putative endopeptidase mechanism for the LysB4 EAD. These suggestions add insight into the molecular mechanism of the endolysin LysB4 in B. cereus-infecting phages.

  • Recently the Elderly with dementia is increasing in Korea with rapid aging of the population. In addition to increasing a number of nuclear family and women's advancement in society, people are less capable of supporting family with dementia. In response, the Seoul city increased welfare facilities for the elderly with dementia about two times for one year. I think that the quantitative portion of facility get to meet the requirements but it is lack of qualitative one in fact. Therefore, the space for the elderly can keep up with the falloff of mental and physical functions, dementia and geriatric diseases according to aging and careful consideration about the space considering stability and convenience is required. At this point, this study has the purpose to plan a daycare center which reflects the characteristics of the elderly to protect them within the community safely and comfortably. As for the research method, case study was conducted to understand the overall situation on the space plan of the daycare center for 3 daycare centers located in Seoul and found out problems. From the generalization of the above study, a space plan for a daycare center considering the characteristics of the elderly was provided. The composition of Dementia welfare facility should be carefully planned to fit characteristics of the patient's behaviors.

Role of 'Spot Sign' on CT Angiography to Predict Hematoma Expansion in Spontaneous Intracerebral Hemorrhage

  • Objective : Patients with spontaneous intracerebral hemorrhage (ICH) presenting within 24 hours of symptom onset are known to be increased risk of hematoma expansion which is closely correlated with morbidity and mortality. We investigated whether tiny enhancing foci ('Spot sign') on axial view of 3-dimensional computed tomography angiography (3D-CTA) source images can predict subsequent hematoma expansion in spontaneous ICH. Methods : During a 2-year period (March 2007-March 2009), we prospectively evaluated 3D-CTA of 110 patients with spontaneous ICH. Based on source images of 3D-CTA, patients were classified according to presence or absence of 'Spot sign'; 'Spot sign' (+) group, 'Spot sign' (-) group. Radiological factors and clinical outcomes were compared between two groups. Results : Hematoma expansion occurred in 16 patients (15%). Mean Glasgow Coma Scale (GCS) score of patients with hematoma expansion was significantly different compared to score of patients without hematoma expansion (5 vs. 9, P < 0.001). Nineteen patients (16%) of 110 ICH patients demonstrated 'spot sign' on 3D-CTA. Among the 'spot sign' (+) group, 53% of patients developed hematoma expansion. Conversely 7% of patients without 'spot sign' demonstrated the hematoma expansion (p < 0.001). Initial volume and location of hematoma were significantly not associated with hematoma expansion except shape of hematoma. Conclusion : Our study showed that patients with hematoma expansion of spontaneous ICH had significant clinical deterioration. And the fact that 'spot sign' (+) group have higher risk of hematoma expansion suggests the presence of 'spot sign' on source images of 3D-CTA can give a clue to predict hematoma expansion in spontaneous ICH.
