Preparation and Characterization of Microorganism Fermentation Celluose as Hydrogel by Radiation Crosslinking (방사선 가교에 의한 미생물 발효 셀룰로오스 하이드로겔의 제조 및 특성)

  • Hydrogels from a mixture of poly(N-vinylpyrrolidone) (PVP), ${\kappa}$-carrageenan and microorganism fermentation celluose were prepared by $^{60}Co$ gamma-ray irradiation. PVP and ${\kappa}$-carrageenan were mixed with the different ratios. Microorganism fermentation celluose were added to the mixture of PVP and ${\kappa}$-carrageenan to evaluate the effect of microorganism fermentation celluose on the gel strength. The gel strength of the hydrogel was evaluated for application of a wound dressing. The results showed that gelation and gel strength were increased with increasing the content of the microorganism fermentation celluose.

Development of Antimicrobial and Deodorizing Cellulose Fiber (항균방취 셀룰로오스섬유 제조에 관한 연구)

  • Both cellulose and chitin together were dissolved in DMAc/LiCl and these solutions were extruded into coagulant of $DMAc/H_2O$. Fibers thus obtained were treated in NaOH aqueous solution. Results showed that the fiber surface contains celluose and chitosan. This means that these fibers treated are composed of three components, ie, cellulose, chitin, and chitosan. These fiber showed secure antibacterial and mechanical properties.

Effect of Mixing Pattern of Different Types of Bioreactor on Enzymatic Hydrolysis of Cellulose (각종 섬유질 효소당화 반응조내의 현탁액의 혼합교반양상이 효소당화에 미치는 영향)

  • Celluose is an insoluble substrate, therefore, a proper mixing of the cellulose suspension is essential for an effective enzymatic hydrolysis. To study the effect of mixing motion of various enzyme reactors on enzymatic hydrolysis of cellulose, three distinct types of biroreator: vertical impeller type bioreator(VITB), horizontal paddle type bioreactor(HPTB), and tumbling drum type bioreactor(TDTB), were assembled and their performance was compared. The optimal agitation speed was 100rpm for VITB and HPTB, 200rpm for TDTB. The saccharification efficiency of each reactor was compared under the optimal agitation intensity. The highest degree of saccharification was achieved in the case of VITB, especially, at high cellulose concentration. The VITB seems to be the most suitable type of bioreactor that can maintain proper mixing pattern for effective enzyme reaction. In the view of energy consumption, the TDTB showed the lowest value: however, the energy consumption was rapidly increased at high concentration of celluose. To dertermine the most suitable type of bioreactor, the entire process, including substrate cost, substrate concentration, and feasibility of scale-up, needs to be evaluated.

  • Monodispersed alumina-zirconia fine powders were prepared by controlled hydrolysis of alkoxides. These powders and the sintered bodies were characterized. Aluminium alkoxide and zirconium alkoxide were dissolved into complex solvent with butanol and n-propanol, and by acetonitrile added hydrolytic solution, hydrolysis rate was controlled. The oil, as a dispersant, was added in hydrolytic solution, and then prepared powders were nano-sized and well-monodispersed. In the case of hydroxypropyl celluose (HPC) as a dispersant, it was added in complex solution with butanol and iso-propanol, sub-micrometer sized and well-monodispersed powders could be prepared. The value of relative density (R.D.) and tetragonal phase fraction of zirconia in the sintered body made by nano-meter sized powders were respectively higher than those in the case of sub-micrometer sized one.

  • Lactate and malate dehydrogenase isozymes in crude extracts of various tissues from 28 avian species were investigated by means of celluose acetate electrophoresis. In a given species the presence of two types of the malate dehydrogenase isozymes could be revealed in somatic tissues; one was characterized by fast electrophoretic mobility, and the other by slow mobility. Although the bird lactate dehydrogenase isozymes existed in a variety of species-specific molecular forms, they seemed to show the same pattern within the same genus and family.

  • This study was carried out to investigate changes in the cell wall components of peach during maturation and storage. The hardness of peach was decreased from $24.1\;\pm\;2.2N\;to\;14.6\;\pm\;1.3N$ during maturation and storage. Contents of alcohol-insoluble substance, water-soluble material and cell wall were decreased. Total and insoluble pectic substances were decreased while water-soluble pectin was increased during maturation and storage. Contents of cellulose were increased. Galactose and arabinose contents were decreased during maturation and storage.

  • In the present study adding hydroxypropyl celluose as dispersant to the ethanol solution of Zr-butoxide prior to starting the hydrolysis reaction, spherical and submicrometer sized hydrated zirconia powders were synthesized successfully. But syntesized hydrated zirconia powders were soluble considerably in ethanol used as washing medium. Washing once the powder with plenty amount of water after washing thrice it with acetone, reaction mother solution remained between the particles was effectively removed and the particles were converted to insoluble state to ethanol at the same time. As a result of such washing processes it was able to prevent the formation of polydispersed, agglomerated and multiplet particles almost always even when such concentrated solutions of Zr-n-butoxide as 0.5 M were hydrolyzed.

  • A novel type of sulfotransferase, arylsulfate sulfotransferase (EC 2.8.2.22) purified from Haemophilus K-12, an intestinal bacterium of a mouse, was immobilized onto AH-S epharose 4B, CH-Sepharose 4B and DEAE-celluose. The enzyme was stabilized for storage more markedly by covalent immobilization onto AH-Spharose 4B or CH-Sepharose 4B and by adsorptive immobilization onto DEAE-cellulose than the free enzyme. The optimal pH and acceptor substrate specificity of immobuized enzyme were similar to those of the free enzyme.

  • The cationic isoperoxidases were isolated from oat root tips which had been grown in treatment with $1{\times}10^{-4}$ M alachlor and purified about 30-fold by treatment with ethylalchol and ion exchange chromatography on DEAF-celluose and CM-sephadex medium. The oat root was found to contain three isoperoxidase. The major activity peak (B) represented 65% of the total isoperoxidase activity. After purification, the major peak of isoperoxidase was purified about 37-fold from the oat root. Analysis of the major peroxidase peak(column fraction 58-78) by SDS revealed a single band which corresponed to a molecular mass of 42.5 kD. In vitro, isoperoxidase activies were inhibited by IAA. Isoperoxidase(50 unit) significantly inhibited 70.2% of cell division in oat root and 54.2% of cell elongation in oat coleoptile as compared with control.
