In this study were evaluated the whitening and anti-oxidative activities from the extracts of Sorbus alnifolia branches, and identified the chemical structures of the active ingredients. In the whitening tests using α-MSH stimulated B16F10 melanoma cells, the 70% ethanol extract and n-butanol (n-BuOH) fractions concentration-dependently inhibited cellular melanogenesis and intracellular tyrosinase activities without causing cell toxicity. The total polyphenol content of n-BuOH and ethyl acetate (EtOAc) fractions were measured to be respectively 241.1 ± 1.1 and 222.9 ± 2.4 (mg/g GAE), and the total flavonoid content of EtOAc fraction was 75.3 ± 2.0 (mg/g QE). Upon anti-oxidant studies with DPPH and ABTS+ radicals, potent radical scavenging activities were observed in the EtOAc and n-BuOH fractions. Moreover, in the study of cell protection efficacy using HaCaT keratinocytes damaged by H2O2, the EtOAc and n-BuOH fractions showed a very positive results on prevention of oxidative stress. Phytochemical studies for this extract resulted in the isolation of four compounds; 2-oxopomolic acid (1), euscaphic acid (2), epi-catechin (3), prunasin (4). These results suggested that the extract of S. alnifolia branches containing compounds 1-4 as natural ingredients could be used as whitening and anti-oxidant ingredients in cosmetic formulations.

The pathogen Gallibacterium anatis has caused heavy economic losses for commercial poultry farms around the world. However, despite its importance, the functions of its hypothetical proteins (HPs) have been poorly characterized. The present study analyzed the functions and structures of HPs obtained from Gallibacterium anatis (NCTC11413) using various bioinformatics tools. Initially, all the functions of HPs were predicted using the VICMpred tool, and the physicochemical properties of the identified virulence proteins were then analyzed using Expasy's ProtParam server. A virulence protein (WP_013745346.1) that can act as a potential drug target was further analyzed for its secondary structure, followed by homology modeling and three-dimensional (3D) structure determination using the Swiss-Model and Phyre2 servers. The quality assessment and validation of the 3D model were conducted using ERRAT, Verify3D, and PROCHECK programs. The functional and phylogenetic analysis was conducted using ProFunc, STRING, KEGG servers, and MEGA software. The bioinformatics analysis revealed 201 HPs related to cellular processes (n = 119), metabolism (n = 61), virulence (n = 11), and information/storage molecules (n = 10). Among the virulence proteins, three were detected as drug targets and six as vaccine targets. The characterized virulence protein WP_013745346.1 is proven to be stable, a drug target, and an enzyme related to the citrate cycle in the present pathogen. This enzyme was also found to facilitate other metabolic pathways, the biosynthesis of secondary metabolites, and the biosynthesis of amino acids.
Pulsed field ablation (PFA) was recently rediscovered as an emerging treatment modality for the ablation of cardiac arrhythmias. Ultra-short high voltage pulses are leading to irreversible electroporation of cardiac cells subsequently resulting in cell death. Current literature of PFA for pulmonary vein isolation (PVI) consistently reported excellent acute and long-term efficacy along with a very low adverse event rate. The undeniable benefit of the novel ablation technique is that cardiac cells are more susceptible to electrical fields whereas surrounding structures such as the pulmonary veins, the phrenic nerve or the esophagus are not, or if at all, minimally affected, which results in a favorable safety profile that is expected to be superior to the current standard of care without compromising efficacy. Nevertheless, the exact mechanisms of electroporation are not yet entirely understood on a cellular basis and pulsed electrical field protocols of different manufactures are not comparable among one another and require their own validation for each indication. Importantly, randomized controlled trials and comparative data to current standard of care modalities, such as radiofrequency- or cryoballoon ablation, are still missing. This review focuses on the "pearls" and "pitfalls" of PFA, a technology that has the potential to become the future leading energy source for PVI and beyond.
The p97 adenosine triphosphatase is a key player in protein homeostasis, responsible for unfolding ubiquitylated substrates. It engages with various adaptor proteins through its N-terminal domain, with the p97-p47 complex attracting particular attention for its involvement in membrane remodeling. Although the structures of p97 in complex with the Ubiquitin regulatory X (UBX) domain from various adaptors have been reported, the stoichiometry is conflicting. Here, we report the crystal structure of the p97 N-D1 hexamer in complex with the p47 UBX domain at a resolution of 2.7 Å. The structure reveals a stoichiometry of 6:6 between the p97 N-D1 and the p47 UBX domain. These findings provide valuable insights into the binding stoichiometry of p97 N-D1 and p47 UBX domain, which are crucial for understanding the role of p97 and adaptor proteins in cellular processes such as the ubiquitin-proteasome pathway, membrane fusion, and cell cycle regulation.
Periodontal ligament stem cells (PDLSCs) are multipotent stem cells derived from periodontium and have mesenchymal stem cell (MSC)-like characteristics. Recently, the perivascular region was recognized as the developmental origin of MSCs, which suggests the in vivo angiogenic potential of PDLSCs. In this study, we investigated whether PDLSCs could be a potential source of perivascular cells, which could contribute to in vivo angiogenesis. PDLSCs exhibited typical MSC-like characteristics such as the expression pattern of surface markers (CD29, CD44, CD73, and CD105) and differentiation potentials (osteogenic and adipogenic differentiation). Moreover, PDLSCs expressed perivascular cell markers such as NG2, ${\alpha}-smooth$ muscle actin, platelet-derived growth factor receptor ${\beta}$, and CD146. We conducted an in vivo Matrigel plug assay to confirm the in vivo angiogenic potential of PDLSCs. We could not observe significant vessel-like structures with PDLSCs alone or human umbilical vein endothelial cells (HUVECs) alone at day 7 after injection. However, when PDLSCs and HUVECs were co-injected, there were vessel-like structures containing red blood cells in the lumens, which suggested that anastomosis occurred between newly formed vessels and host circulatory system. To block the $SDF-1{\alpha}$ and CXCR4 axis between PDLSCs and HUVECs, AMD3100, a CXCR4 antagonist, was added into the Matrigel plug. After day 3 and day 7 after injection, there were no significant vessel-like structures. In conclusion, we demonstrated the perivascular characteristics of PDLSCs and their contribution to in vivo angiogenesis, which might imply potential application of PDLSCs into the neovascularization of tissue engineering and vascular diseases.
최근 이동단말기의 급속한 보급과 발전에 의하여, 통상의 시가지에서의 전자파전파의 특성을 해명해야할 뿐만 아니라, 시가지, 산림, 계곡 등의 외부 공간과 실내 공간 등의 종래에는 전파불감지대로 불려오던 공간에 있어서도 안정적으로 통신을 이용할 수 있는 무선통신환경 개선의 중요성이 높게 요구되고 있다. 더욱이, 이동통신의 디지털화와 더불어 건물 등에 의한 반사에 의해서 생기는 멀티패스에 기인하는 수신펄스의 시간지연이 고속디지털통신의 장해가 되고 있다. 이러한 도심 시가지에 대한 전자파전파의 특성을 보다 정도 높게 추정하기 위해서는 건물 벽면의 구조 및 간판 등의 영향도 고려 대상으로 포함시킬 필요성이 있다. 본 연구에서는, 먼저 무선통신환경에서의 시가지, 산림, 해면, 빌딩 벽면 등의 문제를 규칙적 및 불규칙조면(random rough surface)에 의한 산란문제로 취급하여 그 특성을 상세히 파악하고자 한다. 실제 문제에 있어서도 간판과 같은 빌딩의 부착물, 창, 테라스 등과 같은 규칙적 벽면 구조 또는 불규칙적 벽면 구조를 가지고 있는 경우가 일반화되어 있는 실정이다. 따라서 본 논문에서는 FVTD(finite volume time domain)법을 적용하여 기초적인 자료 보완을 위하여 3분류에 의한, 즉 주기적 구조에 의한 산란 특성, 불규칙적 구조에 의한 산란 특성, 주기적 및 불규칙의 복합형구조에 의한 2차원 전파환경 모델들에 대한 산란 특성을 조사하였다. 특히 주기적 벽면구조에 대한 Bragg reflection의 영향을 조사하였다.
Purpose: The periosteum is a well-known source of osteogenic precursor cells for tissue-engineered bone formation. However, cultured endothelial or endothelial-like cells derived from periosteum have not yet been investigated. This study focused on endothelial-like cell culture from the periosteum. Methods: Periosteal tissues were harvested from the mandible during surgical extraction of lower impacted third molars. The tissues were treated with 0.075% type I collagenase in phosphate-buffered saline (PBS) for 1 hr at $37^{\circ}C$ to release cellular fractions. The collagenase was inactivated with an equal volume of DMEM/10% fetal bovine serum (FBS) and the infranatant was centrifuged for 10 min at 2,400 rpm. The cellular pellet was filtered through a $100{\mu}m$ nylon cell strainer, and the filtered cells were centrifuged for 10 min at 2,400 rpm. The resuspended cells were plated into T25 flasks and cultured in endothelial cell basal medium (EBM)-2. Results: Among the hematopoietic markers, CD146 was more highly expressed than CD31 and CD34. The periosteal-derived cells also expressed CD90 and CD166, mesenchymal stem cell markers. Considering that the expression of CD146 was constant and that the expression of CD90 was lower at passage 5, respectively, the CD146 positive cells in passage 5 were isolated using the magnetic cell sorting (MACS) system. These CD146 sorted, periosteal-derived cells formed tube-like structures on Matrigel. The uptake of acetylated, low-density lipoprotein, labeled with 1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI-Ac-LDL) was also examined in these cells. Conclusion: These results suggest that the CD146-sorted positive cells can be referred to as periosteal-derived CD146 positive endothelial-like cells. In particular, when a co-culture system with endothelial and osteoblastic cells in a three-dimensional scaffold is used, the use of periosteum as a single cell source would be strongly beneficial for bone tissue engineering.
둥근잎꿩의비름(Sedum rotundifolium L.) 엽육조직에 대한 초박절편 및 연속 후박절편의 시료를 제작하여 TEM 및 HVEM 고압전자현미경으로 연구하였고, 이로부터 수합된 색소체 결정체 구조의 tilting 및 연속절편 결과에 image processing을 실시하여 세포수준에서의 초미세구조 정보를 추출 3-D 입체구조로 재구현하였다. ${\pm}60^{\circ}$에서의 tilting과 $0.125{\sim}1{\mu}m$에 이르는 연속절편에서 결정체를 구성하는 미세한 관상요소(tubular elements)의 구조적 특성을 조사한 결과, 결정체는 일시적으로 분화 초기단계에서 형성되어 $4{\sim}5{\mu}m$에 이르기까지 크게 여러 형태로 발달하나, 엽육조직이 성숙하면 이들 구조는 색소체에서 완전히 사라지는 특성을 보였다. 결정체를 구성하는 관상의 요소는 절단각도에 따라 격자구조 또는 평행구조를 이루었으며, 이들 구조 내에 형성되어 있는 정교한 구조적 pattern은 회절분석에 의해 확인되었다. 결정체 내에는 규칙적으로 약 20nm의 격자간격으로 이루어진 초미세관상의 요소들이 수백-수천 개 무리지어 발달하였다. 색소체 내에는 이러한 결정체가 하나 이상 형성되기도 하며, 일부 결정체의 경우 결정구조의 말단부위가 국소적으로 융합 또는 분지되기도 하였다. 결정구조는 막으로 둘러싸이지는 않으나, 대부분 틸라코이드 막성계와 밀착하여 발달하였다. 일차적으로 수집된 HVEM 상의 2-D 결과는 디지털화 과정을 거친후 Imod와 3-D Max를 이용하여 3-D 입체구조로 재구현되었다.
본 연구는 반도체 산업에서 반도체소자로서 주목받고 있는 GaAs, InP및 InAs의 세포독성을 평가하기 위해 햄스터의 폐포 대식세포를 사용하여 in vitro 자계 측정, LDH 활성치측정 및 세포의 형태학적 관찰 등을 검토하였다. 세포자계측정 결과 GaAs, InP 및 InAs첨가군 모두 대조군(PBS첨가군)에 비해 완화곡선이 유의하게 지연되었으며, 특히 GaAs 첨가군은 농도증가에 따라 용량의존적으로 완화곡선이 지연되는 경향이었다. 자화 후 2분간의 완화계수는 대조군에 비해 GaAs 첨가군은 농도증가에 따라 유의하게 낮아지는 용량의존성이 높은 경향이었으나, InP 및 InAs 첨가군에서는 모두 유의성이 인정되지 않았다. LDH활성치는 GaAs, InP 및 InAs첨가군 모두 용량 의존적으로 점차 높아지는 경향이었다. 세포의 형태학적 관찰소견은 GaAs첨가군에서는 용량의존적으로 세포막의 현저한 파괴, 핵의 형태적 변화 등 심한 세포장해가 유발된 반면, InP첨가군과 InAs첨가군에서는 세포내의 구조는 유지되었으나 세포질의 변성이 관찰되었다. 결과적으로 GaAs는 InP나 InAs보다 폐포 대식세포의 세포독성이 강한 것으로 보인다.
일반적인 세포 배양 기술은 평평한 표면에 세포 부착을 위한 화학적, 생화학적 표면처리를 하는 것이 기본이지만, 요즘 들어 마이크로나 나노 크기의 구조체를 형성하여 세포 부착을 하는 연구들이 많이 진행되고 있다. 본 연구에서는 전도성 고분자인 피롤과 나노임프린트 기술을 이용하여 300 nm 선 패턴과 150 nm 원기둥 패턴의 나노구조체 제작 후 대표적인 암세포인 HeLa 세포를 배양하여, 주사전자현미경과 공초점 현미경을 이용하여 세포의 부착 특성을 연구하였다. 상용 페트리 접시와 평면 피롤에서는 세포들이 부정형의 형태로 부착 및 배양되었지만, 선폭 300 nm 선패턴 상에서는 길이 방향으로 세포가 부착되고 세포 내의 핵과 액틴 역시 배열되어 있고, 지름 150 nm 원기둥 패턴 상에서는 단일 세포로 고정되고 세포 내 액틴은 방사상으로 나노구조체에 고정되어 있는 것을 확인할 수 있었다.
