The aim of this study was to determine the first outbreaks of nontoxic Gonyaulax polygramma Stein in Yeosu waters in place of harmful Cochlodinium polykrikoides Margalef, which has occurred annually in the same coastal region since 1995. The observation of cellular arrangement and structure by electron microscopy showed that G. polygramma isolated from Yeosu waters had a few spines connecting with mem-branes and prominent longitudinal ridges on the cell surface, with a cingular dis-placement 1.5 times their cell width. Furthermore, the location of the nucleus wasposterior of large oval formation according to electron microscopy. On 6 August, 2004,the first bloom of G. pozygramma occurred, the date of own its disappearance was with a maximum cell density of 8,000 cells ml$^{-1}$ on 21 August, 2004. During the period of this study, the horizontal distribution of sea water temperature and salinity showed a strong coastal front, whereas the front of DIN (Dissolved Inoganic Nitrogen) was significantly different between the occurrence and disappearance of G. polygramma blooms. These results suggested that the process of the breakdown of stratification by wind and a low level of inorganic nitrogen play important roles in the rapid growth of G. polygrmma, which is associated with a greater robustness in growth against DIN than that of C.polykrikoides in nature.
Streptococcus pneumoniae, pneumococcus, is the most common cause of community-acquired pneumonia (CAP). CAP is an important infectious disease with high morbidity and mortality, and it is still one of the leading causes of death worldwide. Many genetic factors of the host and various environmental factors surrounding it have been studied as important determinants of the pathophysiology and outcomes of pneumococcal infections. Various cytokines, including transforming growth factor $(TGF)-{\beta}1$, are involved in different stages of the progression of pneumococcal infection. $TGF-{\beta}1$ is a cytokine that regulates a wide range of cellular and physiological functions, including immune and inflammatory responses. This cytokine has long been known as an anti-inflammatory cytokine that is critical to preventing the progression of an acute infection to a chronic condition. On the other hand, recent studies have unveiled the diverse roles of $TGF-{\beta}1$ on different stages of pneumococcal infections other than mitigating inflammation. This review summarizes the recent findings of the role of $TGF-{\beta}1$ on the pathophysiology of pneumococcal infections, which is fundamental to developing novel therapeutic strategies for such infections in immune-compromised patients.

In this study, 6-10 clinically healthy farmed abalone, Haliotis discus hannai were obtained from Jeollanam-Do Wando Soan-myeon, Bogil-myeon, and Nohwa-eup monthly for 1 year 7 months and were examined histopathologically. As a result, the adductor muscle had severe cellular swelling and myonecrosis. These lesions were stained with hematoxylin and eosin and Giemsa. The lesions were not caused by infection of bacteria or parasites. We investigated the relationship between lesion frequency and water temperature and discovered that increased water temperature was associated with increasing lesion frequency. As water temperature is related to growth rate, increased growth rate was closely related to increased lesion frequency. We considered that these lesions could be a useful factor in measuring shellfish health.

The Ubiquitin proteasome system (UPS) plays roles in protein degradation, cell cycle control, and growth and inflammatory cell signaling. Dysfunction of UPS in cardiac diseases has been seen in many studies. Cholesterol acts as an inducer of cardiac hypertrophy. In this study, the effect of proteasome inhibitors on the cholesterol-induced hypertrophic growth in H9c2 cells is examined in order to observe whether UPS is involved in cardiac hypertrophy. The treatment of proteasome inhibitors MG132 and Bortezomib markedly reduced cellular surface area and mRNA expression of β-MHC in cholesterol-induced cardiac hypertrophy. In addition, activated AKT and ERK were significantly attenuated by MG132 and Bortezomib in cholesterol-induced cardiac hypertrophy. We demonstrated that cholesterol-induced cardiac hypertrophy was suppressed by proteasome inhibitors. Thus, regulatory mechanism of cholesterol-induced cardiac hypertrophy by proteasome inhibitors may provide a new therapeutic strategy to prevent the progression of heart failure.

Studies were made to remove the toxic phytohormone, 2,4-D, in suspension cultures of Panax vietnamensis. Cells grown in normal MS medium with 2,4-D were inoculated and grown in the MS medium without hormone. Not a big difference was observed in growth characteristics between media with and without 2,4-D. The 2,4-D in the culture, however, was completely removed. During the culture, the residual 2,4-D was consumed rapidly at the early growth stage. The intra-cellular 2,4-D was consumed first and the 2,4-D in the medium was used afterward.
At the fifth day after right lung pneumonectomy in New-Zealand white rabbits $(0.8{\sim}1.1\;kg\;B.W.)$, phospholipid and protein concentration in the left lung lavage fluid were measured for clarification of the effect of unilateral pneumonectomy on the secretory function of the type II pneumocytes in growing rabbits. In an attempt to evaluate the effect of unilateral pneumonectomy on the compensatory growth of the residual lung, left lung weight and left lung weight-body weight ratio and DNA concentration, RNA/DNA and total DNA content in the left lung tissue were measured in pneumonectomized and in sham operated control rabbits. The lung weight of pneumonectomized rabbit was approximately two times heavier than that of the control rabbits. DNA concentration and RNA/DNA of the lung tissue were not changed but total DNA content was increased significantly. Phospholipid concentration in the lung lavage fluid of the pneumonectomized rabbits was over two times higher than that of control rabbits. from these experimental results, It is concluded that unilateral pneumonectomy in growing rabbits might cause to increase the secretion of pulmonary surfactant from type II pneumocyte of the residual lung. The cellular hyperplasia seems to be the primary response of the compensatory growing lung in unilateral pneumonectomized growing rabbits.
Labyrinthulids are unicellular heterotrophic marine microalgae. Two labyrinthulid strains, L4 and L75, which produce generous amounts of polyunsaturated fatty acids(PUFAs), were isolated from leaves floating in the coastal areas of Quang Ninh province, Vietnam, and Fukuoka Prefecture, Japan, respectively. Both strains had spindle-shaped cells surrounded by ectoplasmic networks. Numerous oil bodies were observed in each cell, mostly in the vicinity of cell membranes. When soybean oil or soybean lecithin was used as a carbon source with peptone as a nitrogen source, the proportion of PUFAs reached 25-30% and 50-56% for L4 and L75, respectively. After 14 days of growth at $25^{\circ}C$, L4 produced 0.3 mg PUFAs/g-agar in PYA-SBO medium and 0.6 mg PUFAs/g-agar in PYA-SBL medium. In comparison, L75 produced 0.2 mg PUFAs/g-agar in both types of media. The differences between the two strains included changes in cellular morphology and the capacity for attaching tightly to fibers when cultured in liquid PYA medium containing 2% SBL. In addition, when the strains were grown under the same conditions, L4 had a higher growth rate and produced more PUFAs than L75.

Abalone immortal cell lines can be used to study the physiological properties and disease mechanisms of abalone at the cellular and molecular level. As a first step for the final goal to establish abalone immortal cell lines, we examined various initial culture conditions for primary cell populations derived from Haliotis discus hannai radula tissue. The survival rate after cell isolation procedures using the enzymatic method was as low as $9.95{\pm}2.37%$. Based on three different experimental conditions for H. discus hannai radula-derived cell culture, we found that the salinity of the media and the presence of growth-promoting factors were important to support radula-derived primary cell populations during the initial culture. The growth factor-containing media adjusted to 35 psu salinity could induce 100% (8 out of 8 trials) initial cell attachment, and the rate of cell attachment reached 50-70%. The data obtained from this study will provide useful information for developing immortal cell lines from abalone species.
Fibroblast growth factor-2 (FGF-2) is known to modulate numerous cellular functions in various cell types, including cell proliferation, differentiation, survival, adhesion, migration, and motility, and also in processes such as wound healing, angiogenesis, and vasculogenesis. FGF-2 regulates the expression of several molecules thought to mediate critical steps during angiogenesis. This study examines the mechanisms underlying FGF-2-induced cell migration, using human umbilical vein endothelial cells (HUVECs). FGF-2 induced the nondirectional and directional migration of endothelial cells, which are inhibited by MMPs and plasmin inhibitors, and induced the secretion of matrix metalloproteinase-3 (MMP3) and MMP-9, but not MMP-l and MMP-2. FGF-2 also induced the secretion of the tissue inhibitor of metalloproteinase-l (TIMP-I), but not of TIMP- 2. Also, the pan-PKC inhibitor inhibited FGF-2-induced MMP-9 secretion. It is, therefore, suggested that FGF-2 induces the migration of cultured endothelial cells by means of increased MMPs and plasmin secretion. Furthermore, FGF-2 may increase MMP-9 secretion by activating the PKC pathway.
AL072 is a potent anti-Legionella antibiotic produced by Streptomyces strain AL91. The minimum inhibitory concentration (MIC) of AL072 against Legionella pneumophila was 0.2$\mu$g/ml. Bacterial growth was rapidly inhibited at the dose range between the MIC and 20 times of the MIC when the antibiotic was added at the mid-exponential phase. Ultrastructural changes in L. pneumophila were observed upon treatment with AL072. Under electron microscopical observation, the organisms treated with AL072 exhibited characteristic morphological changes in the cellular outer coat. Also irregular morphological changes, such as the formation of filamentous materials in the cytoplasm, an increase in the size and number of cytoplasmic vacuoles, the extruding of cytoplasmic contents, the formation of spheroplast and ghost cells, and blebbings in the cell wall were observed. Furthermore, immunoelectron microscopical observation of the group treated with the MIC showed that the immune complex attached mainly to the cell wall. The results of these experiments indicate that AL072, like the inhibitors of cell wall synthesis, act selectively on the cell wall of L. pneumophila.
