To evaluate the effects of lactic acid bacteria (LAB) on Peyer's patch cells, mice were treated with a high dose of kanamycin to disturb the gut microbial environment. The overarching goal was to explore the potential of LAB for use as a dietary probiotic that buffers the negative consequences of antibiotic treatment. In vitro, LAB stimulated the production of immunoglobulin A (IgA) from isolated Peyer's patch cells. Inflammation-related genes (TNF-α, IL-1β, and IL-8) were up-regulated in Caco-2 cells stimulated with lipopolysaccharide (LPS), while tight-junction-related genes (ZO-1 and occludin) were down-regulated; the effects of LPS on inflammatory gene and tight-junction gene expression were reversed by treatment with LAB. Mice treated with a high dose of kanamycin showed increased serum IgE levels and decreases in serum IgA and fecal IgA levels; the number of Peyer's patch cells decreased with kanamycin treatment. However, subsequent LAB treatment was effective in reducing the serum IgE level and recovering the serum IgA and fecal IgA levels, as well as the number of Peyer's patch cells. In addition, ZO-1 and occludin mRNA levels were up-regulated in the ileum tissues of mice receiving LAB treatment. Lactic acid bacteria can enhance the intestinal immune system by improving the integrity of the intestinal barrier and increasing the production of IgA in Peyer's patches. Lactic acid bacteria should be considered a potential probiotic candidate for improving intestinal immunity, particularly in mitigating the negative consequences of antibiotic use.
In an attempt to improve the productivity of ${\beta}-galactosidase$ from Bacillus sp. A1, which was isolated from soil and has remarkably higher transgalactosylation activity than lactose hydrolysis activity, a chemical mutation procedure using N-methyl-N'-nitro-N-nitrosoguanidine followed by selection was conducted. The final selection, designated as Bacillus sp. A4442, turned out to show a substantially increased enzyme productivity. Catabolite repression by glucose and lactose requirement as an inducer for the enzyme biosynthesis, which were shown in the parent strain, was markedly diminished; instead it was found out that galactose acts as another inducer. Because pH of medium, one of the most important factors for cell growth as well as enzyme production, is closely related with the sugar concentration during culture, it was kept in the optimum range of $6.5{\sim}7.5$; for this the initial glucose concentration was adjusted to be 0.5% which was thereafter maintained by the controlled pumping-in of lactose using the pH-stat technique. By doing so, we were able to increase the productivity of ${\beta}-galactosidase$ with high transgalactosylation activity up to $44\;unit/m{\ell}-broth$.

An accurate prediction of budburst in grapevines is indispensable for vineyard frost warning system operations in spring because cold tolerance depends heavily on phonology. However, existing frost warning systems utilize only daily minimum temperature forecasts since there is no way to estimate the site-specific phonology of grapevines. A budburst estimation model based on thermal time was used to project budburst dates of two grapevine cultivars (Kyoho and Campbell Early), and advisories were issued depending on phonology as well as temperature. A 'warning' is issued if two conditions are met: the forecasted daily minimum temperature falls below $-1.5^{\circ}C$ and the estimated phonology is within the budburst period. A 'watch' is issued for a temperature range of -1.5 to $+1.5^{\circ}C$ with the same phonology condition. Validation experiments were done at 8 vineyards in Anseong in spring 2005, and the results showed a good agreement with the observations. This method was applied to the climatological normal year (1971-2000) to determine sites with high frost risk at a 30 m grid cell resolution. Among 608,585 grid cells constituting Anseong, 1,059 cells were identified as high risk for growing Kyoho and 2,788 cells for Campbell Early.

Regional assessment of crop productivity using a gridded simulation approach could aid policy making and crop management. Still, little effort has been made to develop the systems that allows gridded simulations of crop growth using ORYZA 2000 model, which has been used for predicting rice yield in Korea. The objectives of this study were to develop a series of data processing modules for creating input data files, running the crop model, and aggregating output files in a region of interest using gridded data files. These modules were implemented using C++ and R to make the best use of the features provided by these programming languages. In a case study, 13000 input files in a plain text format were prepared using daily gridded weather data that had spatial resolution of 1km and 12.5 km for the period of 2001-2010. Using the text files as inputs to ORYZA2000 model, crop yield simulations were performed for each grid cell using a scenario of crop management practices. After output files were created for grid cells that represent a paddy rice field in South Korea, each output file was aggregated into an output file in the netCDF format. It was found that the spatial pattern of crop yield was relatively similar to actual distribution of yields in Korea, although there were biases of crop yield depending on regions. It seemed that those differences resulted from uncertainties incurred in input data, e.g., transplanting date, cultivar in an area, as well as weather data. Our results indicated that a set of tools developed in this study would be useful for gridded simulation of different crop models. In the further study, it would be worthwhile to take into account compatibility to a modeling interface library for integrated simulation of an agricultural ecosystem.
Among finite filed arithmetic operations, division/inverse is known as a basic operation for public-key cryptosystems over $GF(2^{m})$ and it is computed by performing the repetitive $AB^{2}$ multiplication. This paper presents a digit-serial-in-serial-out systolic architecture for performing the $AB^2$ operation in GF$(2^{m})$. To obtain L×L digit-serial-in-serial-out architecture, new $AB^{2}$ algorithm is proposed and partitioning, index transformation and merging the cell of the architecture, which is derived from the algorithm, are proposed. Based on the area-time product, when the digit-size of digit-serial architecture, L, is selected to be less than about m, the proposed digit-serial architecture is efficient than bit-parallel architecture, and L is selected to be less than about $(1/5)log_{2}(m+1)$, the proposed is efficient than bit-serial. In addition, the area-time product complexity of pipelined digit-serial $AB^{2}$ systolic architecture is approximately $10.9\%$ lower than that of nonpipelined one, when it is assumed that m=160 and L=8. Additionally, since the proposed architecture can be utilized for the basic architecture of crypto-processor and it is well suited to VLSI implementation because of its simplicity, regularity and pipelinability.

Purpose : There are various oriental medicine therapy of treating Pelvic Inflammatory Disease(PID) in clinics. We made the PID mice model by injection Lipopolysaccharide (LPS) in vagina, and investigate anti-inflammatory effects of Hongdeung-Tang among oral medication, retention enema therapy and herbal-acupuncture treatment. Method : The ICR(20-30g) mice(♀) were used. To examine the occurrence of inflammation, LPS in different concentration was injected into the vagina of the mice, and White Blood Cell(WBC) in blood was counted. To examine anti-inflammatory effects, 6 mice were assigned to each of the normal group, the control group and the sample group. Hongdeung-Tang was medicated in oral and rectal with 1.0g/kg low dose and 3.0g/kg high dose and by herbal acupucture for 5 days, 2 days before LPS injection. After 3days from LPS injection, blood was collected from retro-orbital plexus, and WBC, Interleukin-6(IL-6), Tumor Necrosis Factor-${\alpha}$(TNF-${\alpha}$) in the blood was counted. Result : After LPS injection with each dose, WBC count showed significant increase depending on LPS concentration from $100{\mu}g/kg$, and it was maximized at 3 or 4 days after LPS injection. the Hongdeung-Tang treatment groups, The number of WBC was decreased significantly only in low dose and high dose oral medication, and IL-6 concentration showed significant decrease in oral and rectal medication as well as in herbal acupuncture treatment. TNF-${\alpha}$ concentration was decreased significantly in oral and rectal medication of low dose and high dose. herbal-acupuncture treatment group datas showed reductive tendency. Conclusion : Based on above results, we made the PID mice model by injection LPS in vagina, and demonstrated anti-inflammatory effect of Hongdeung-Tang of oral medication, retention enema therapy and herbal-acupuncture treatment.

The Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and probably the most common plant among the various herbal folk remedies being used in the treatment of abdominal pain hepatitis chronic liver disease, jaundice and coughing in Korea. Recently the biological and pharmacological actions of herb have been studied well such as antibacterial, antidiabetic and antitumor activities. This experiment was conducted to investigate antitumor and immunomodulatory effects of Artemisia capillarix extracts against Hepa-1c1c7 and Sarcoma 180 cancer cells on in vivo experimental tests. On in vivo experimental tests using 280 ICR mice the gain of body weight in the control-group mice bearing Sarcoma 180 ascites tumor was 1.5 times more than that of the normal-group mice after 33 days. However, the gain of body weight in all experimental groups administered with Artemisia capillaris extracts was significantly lower than that of the control-group mice (P<0.05). The mean survival times of mice administered with Artemisia capillaris extracts of 25 and 100 mg/kg for 28 days were shown to be 25.39% and 15.39% longer than that of the control-group mice injected with saline (P<0.05). Artemisia capillaris extracts showed the highest tumor inhibition effects, which were 42.4% and 27.2% when intraperitoneally injected with doses of 25 and 100 mg/kg once a day for 28 days in inoculated ICR mice with Sarcoma 180 solid tumor cells (P<0.05). The results suggest that Artemisia capillaris methanol extracts have prominent antitumor effects on the cancer cell lines Hepa-1c1c7 and Sarcoma 180.

Insomuch as it is important to manage water quality, from the perspective of water management, it is essential to understand the effect of the weirs on water quality and phytoplankton dynamics in various regions. The purpose of this study is to investigate the characteristics of nitro-nutrients, as well as occurrences and succession patterns of phytoplankton, in the river sections of the two weirs in the Yeongsan River for the five years (from 2012 to 2016) after the weir construction. In respect to this data, the average water temperature measured at the representative point in the section of the Seungchon Weir ($17.1^{\circ}C$) was higher than that of the Juksan Weir ($16.6^{\circ}C$) by comparison. By way of an analysis of this data, it was found that the water quality variables such as, organic matter, nitrogen nutrients and phosphorus nutrients were improved gradually during the period, but the degree of the improvement differs as noted and measured between the weirs. Under the circumstances, it is especially noted that the $NH_3-N$ concentration was higher for the point of the Seungchon Weir (2.204 mg/L) than that of the Juksan Weir (1.157 mg/L). This indicates that effluent as seen from sewage treatment plants and hydrological feature near the densely population area, could be the main cause for the incidence of water pollution in the upstream section of the Seungchon Weir. Additionally, the phytoplankton analysis showed that a relative abundance of diatoms and green algae were 56.9 % and 25.8 % respectively. However, it is noted that the cyanobacteria was measured lower as 10.7 %. Also, in the study sites cell density and occurrence frequency of cyanobacteria were relatively lower than compared to the same measurements noted in other rivers.
A gene for cellulolytic xylanase of Bacillus circulnns ATCC21365 was cloned on pUC 19 in Eschwichia coli. The recombinant plasniid pXLI80 contained an 1.8 id, inselt composed of0.5 kb and 1.3 kb PslI fragments derived from B, circulans. The 0.5 kh fragment in the upstream region of 1.3 kb one was confirmed lo be indispensable for not only expression but also hyperexpression of the cloned gene. The transformant overproduced the xylanase 135 times greater than that produced by the orlginal B circulnns. The optimum pH and temperature of the cloned enzyme we]-e pH 5.2 and $60^{\circ}C$, respectively. Heal pretl-eatment at TEX>$55^{\circ}C$C for 1 Indid not cause inhibition of the activity of this enzyme. The elm.ynie could hydl-olyre CMC and lichenan as well as xylan to produce xylose(or GI), xylohiose(or G2) and xylolnose(or G3) as inah products. Hence We defined the cloned enzyme as a cellulolytic xylanase. The SDS-PAG electrophoretic mobility and zyiiogram of this enzyme derived from whole cell extracts or c~~lture supematants or E. coli(pXL180) indicated a molecular weight of 45,000 and nonprocessing of the enzyme in the peilplasln of E. coli.

Objectives: The objective of this study was to examine the effect of non-thermal dielectric barrier discharge(DBD) plasma on decontamination of Staphylococcus aureus(S. aureus) and Escherichia coli(E. coli) as common pathogens. Methods: This experiment was carried out in a chamber($0.64m^3$)designed by the authors. The plasma was continuously generated by a non-thermal DBD plasma generator(Model TB-300, Shinyoung Air tech, Korea). Suspensions of S. aureus and E. coli of 0.5 McFarland standard($1.5{\times}10^8CFU/mL$) were prepared using a Densi-Check photometer(bio $M{\acute{e}}rieux$, France). The suspensions were diluted1:1000 in sterile PBS solutions(approximately$10^{4-5}CFU/mL$) and inoculated on tryptic soy agar(TSA) in Petri dishes. The Petri dishes(80mm internal diameter)were exposed to the non -thermal DBD plasma in the chamber. Results: The results showed that 95% of S. aureus colonies were killed after a six-hour exposure to the DBD plasma. In the case of E. coli, it took two hours to kill 100% of the colonies. The gram-negative E. coli had a greater reduction than the gram-positive S. aureus. This difference may be due to the structure of their cell membranes. The thickness of gram-positive bacteria is greater than that of gram-negative bacteria. The S. aureus is more resistant to DBD plasma exposures than is E. coli. It should be noted that average concentrations of ozone, a byproduct of the DBD plasma generator, were monitored throughout the experiment and the results were well below the criteria, 50 ppb, recommended by the Korean Ministry of the Environment. Thus, non-thermal DBD plasma is deemed safe for use in hospital and public facilities. Conclusions: There was evidence that non-thermal DBD plasma can effectively kill S. aureus and E. coli. The results indicate that DBD plasma technology can greatly contribute to the control of infections in hospitals and other public and private facilities.
