Somatic cell nuclear transfer is an efficient technique for the multiplication of elite livestock, engineering of transgenic animals, cell therapy and xenotransplantation, and analyzing the interactions between nucleus and cytoplasm, for various agricultural, biomedical and research purposes. Since the first somatic cell clone lamb was born, tremendous progress has been made toward developing technology for animal cloning. Viable farm animals and mice have now been produced by nuclear transfer using various fetal and adult somatic cells as nuclei donors. Transgenic clones were also produced from nuclear transfer of transfected somatic cells. In the future, somatic cell nuclear transfer will provide more numerous opportunities, both in basic and appled research as well as immediate uses in the generations of superior clone and transgenic animals. However, further technology refinement and improved understanding of the process are essential for commercial and basic research applications.
PD98059 is the specific inhibitor of extracellular signaling-regulated kinase (ERK) kinase (MEK). ERK is involved in a mitogen-activated protein kinase (MAPK) cascade controlling cell growth and differentiation. Although the inhibition of ERK is known to induce cell death in various cell lines, this effect is still controversial and the role of PD98059 on the death of HeLa $S_3$ cells, a subclone of the cervical cancer cell line, is not well understood. The apoptosis of HeLa $S_3$ cells increased after the treatment of 50 ${\mu}M$ PD98059. The induction of apoptosis by PD98059 was occurred in a time- and a dose-dependent manners. The expression of Bcl-2 was reduced in accordance with decrease of ERK2 expression. Taken together, these results indicate that PD98059 has a cytotoxicity in HeLa $S_3$ cells and it may be used as a potential target for the treatment of cervical cancer.
In this study, the cytotoxicity of commonly used local anesthetics was evaluated on odontoblasts which are essential for pulpal homeostasis in vitro. Local anesthetics, such as articaine, bupivacaine, levobupivacaine, lidocaine, mepivacaine, prilocaine, and procaine, were tested on the odontoblast cell line, MDPC-23. The concentration-and time-dependent cytotoxic effects of local anesthetics on odontoblasts were measured by MTT assay. Among local anesthetics treated for 18 h, only bupivacaine significantly showed cell death in a concentration-($LC_{50}=1.2mM$) and time-dependent manner. To confirm cell death induced by bupivacaine, the observation of cell morphology and FACS using Annexin V and propidium iodide (PI) staining were performed. As a result of Annexin V and PI staining, as well as the morphological change, only bupivacaine induced apoptotic cell death on odontoblasts when compared with levobupivacaine and lidocaine. These results suggest that bupivacaine might affect normal pulpal integrity even after uneventful local anesthesia.
Direct Methanol Fuel Cell(DMFC) using Pt-Ru electrocatlayst and Nafion menbrane can provide high performance if operating conditions are well designed. In this study, operating temperature, pressure, and fuel flow rate were changed to obtain optimum operating conditions of DHFC single cell. Performance of DMFC were increased by the increase of operating temperature. The concentration of fuel methanol was 2.0M $CH_{3}OH$ and pressure difference of cathode and anode was 2 atm were showed maximum performance of DMFC single cell with showing the current density of 160 $mA/cm^2$ at 0.2V cell voltage.
Coordinated multi-point transmission (CoMP) is a candidate technique for next generation cellular communications systems. One of the primary elements discussed in LTE-Advanced technology is CoMP, which can improve cell edge user data rate as well as spectral efficiency due to multiple input multiple output - orthogonal frequency division multiplex (MIMO-OFDM). We consider a system with multiple cells in which base stations coordinate with each other by sharing user channel state information (CSI), which mitigates inter cell interference (ICI), especially for users located at the cell edge. We introduce a new user scheduling method of ICI cancellation and the loss reduction of effective channel gain during the beamforming process, the proposed method improves the system sum rate, when compared to the conventional method by an average of 0.55bps/Hz in different number of total users per cell. It also outperforms the conventional method by approximately 0.38bps/Hz using different SNRs.
(목적) 본 연구에서 사용된 바이러스는 HIV-1 nef유전자가 일부 삭제되고 대신 Chloramphenicol acetyltransferase(CAT)가 pSVCAT recombinant 바이러스다. 이러한 recombinant 바이러스를 사용하는 이유는 첫째, CAT activity가 매우 민감하므로 바이러스의 복제억제 정도를 정확하게 측정 할 수 있고 둘째, simian immunodeficiency virus(SIV)의 경우 nef 유전가 in vivo에서는 바이러스의 복제에 필수적이므로 HIV가 SIV와 유사한 것으로 미루어 본 연구에서 사용되는 recombinant SVCAT 바이러스가 안전한 것으로 고려되기 때문이다. (방법) 특히 화합물이 HIV-1의 복제에 얼마나 영향이 있는가는 1) 어느정 도의 virus inoculm을 넣었는지 2) 사용하는 cell line 3) 사용한 cell line의 infection kinetics 4) 실험의 지속기간 5) 테스트하는 assay의 sensitivity에 의존한다. 따라서 $10^{5}$ cell의 H9과 sup T1을 24 well plate에 넣고 sup T1 cell line의 경우 3일 후 항 화합물에 의한 syncytia 형성 및 CAT activity의 억제정도를 현재 AIDS drug으로 쓰이고 있는 Zidovudine을 control로 비교 관찰하였다. H9 cell line의 경우 3일 간격으로 media의 3/4을 fresh media로 바꾸어 주고 9일 후 CAT assay를 하였다. 이러한 assay에서 activity를 보이는 화합물을 reverse transcriptase와 P24 ELISA assay를 재확인하였다.다.
This paper proposes a multiuse detection system in a multi-cell M-ary Frequency Shift Keying(MFSK)/frequency hopping(FH)-Code Division Multiple Access(CDMA) environment, in which the channel model is an OR-channel and in the reverse link. We have proposed a multiuse detection system in a single cell. However, this sitye is not adequate to detect multiuser in a multi-cell environment. Therefore, we propose a multiuser detection system based on 3 level OR decision with two threholds. The proposed detection system can delete interference as well as intra-cell interference, receive the weakened desired signal and reject the false alarm. computer simulation shows the performance improvement.
We have developed a novel vertical-alignment(VA) -$\pi$ cell mode that provides a wide viewing angle and fast response times for nematic liquid crystal(NLC) with negative dielectric anisotropy on a homeotropic polymide(PI) surfaces. Good voltage-transmittance curves and low driving voltages were achieved with the new VA-$\pi$ cell mode without a negative compensation film. Iso-viewing angle characteristics using the new VA-$\pi$ cell mode without a negative compensation film was also successfully observed. As well a fast response time of 31.7 ms for a new VA-$\pi$ mode was measured. Consequently the iso-viewing angel fast response time and low driving voltage characteristics using a VA-$\pi$ cell can be achieved.
In general, cell transmission delay is more sensitive for real time video service rather than cell losses in a wireless ATM network. Existing handoff schemes, which are emphasizing the prevention of cell losses, have limitations in cell transmission delay to satisfy QoS. In this paper, we propose a scheme to minimize transmission delay generated during the handoff and to maintain the sequence of cells. Transmission delay can be reduced by transmitting ATM cells with low CLP(i.e., CLP=0) prior to others and by discarding cells with high CLP(i.e., CLP= 1). The simulation results show that the proposed scheme is suitable for delay sensitive real time VBR service as well as fast handoff by giving high CLP to less meaningful MPEG frames.
Fuel-cell powered Railway Vehicle Propulsion Systems (RVPSs) are highly desirable due to environment friendly characteristics, and high efficiency of fuel cell (FC). Among various types, the faster start-up and optimality to frequent starts and stops of Polymer electrolyte membrane fuel cell (PEMFC) makes it well suited for propulsion systems. A comprehensive model of PEMFC with reflection of multi-physics behavior required to identify and validate its performance in real RVPSs. Thus this paper will model and simulate the PEMFC unit cell model: a detailed reflection of governing laws and account of dynamic conditions.
