Clinical Experience of Basosquamous Cell Carcinoma (기저편평세포암의 치험례)

  • Purpose: Basosquamous carcinoma is a rare malignancy, with features of both basal cell carcinoma and squamous cell carcinoma. It is considered as aggressive tumor with a high risk of recurrence and metastases. Authors report a case of basosquamous cell carcinoma. Methods: A 72 year-old man, who had an erythematous ulcer on his left auricle, described a slow growing lesion, starting at the posterior surface of the superior helix with a steady increase in size during the past 10 years. At operation, auricular cartilage was grossly invaded by the tumor and was, therefore, amputated with tumor-free margins. Results: Histopathologic examination was revealed a basosquamous cell carcinoma. On positron emission tomography/computed tomography (PET/CT) and neck CT were negative for signs of further nodal involvement or metastases to other organs. At follow-up 6 months later, his wounds were noted to be well healed, with no evidence of local recurrence or identifiable metastases. Conclusion: Because basosquamous cell carcinoma has a significant potential to recur and metastasize, surgical excision for this type of carcinoma should be more extensive than that performed for conventional basal cell carcinoma or squamous cell carcinoma. In addition, regional lymph nodes should be monitored and close follow-up should be carried out.

Oligosaccharide-Linked Acyl Carrier Protein, a Novel Transmethylase Inhibitor, from Porcine Liver Inhibits Cell Growth

  • We have previously reported on the identification of the endogenous transmethylation inhibitor oligosaccharide-linked acyl carrier protein (O-ACP), In this study, the role of the transmethylation reaction on cell cycle progression was evaluated using various transmethylase inhibitors, including O-ACP. O-ACP significantly inhibited the growth of various cancer cell lines, including NIH3T3, ras-transformed NIH3T3, MDA-MB-231, HT-1376, and AGS. In addition, exposure of ras-transformed NIH3T3 to O-ACP caused cell cycle arrest at the $G_0/G_1$ phase, which led to a decrease in cells at the S phase, as determined by flow cytometry. In contrast, transmethylase inhibitors did not affect the expression of $p21^{WAF1/Cip1}$, a well known inhibitor of cyclin dependent kinase, indicating that the cell cycle arrest by transmethylase inhibitors might be mediated by a $p21^{WAF1/Cip1}$-independent mechanism. Therefore, O-ACP, a novel transmethylase inhibitor, could be a useful tool for elucidating the novel role of methylation in cell proliferation and cell cycle progression.

Microscopic Image-based Cancer Cell Viability-related Phenotype Extraction (현미경 영상 기반 암세포 생존력 관련 표현형 추출)

  • During cancer treatment, the patient's response to drugs appears differently at the cellular level. In this paper, an image-based cell phenotypic feature quantification and key feature selection method are presented to predict the response of patient-derived cancer cells to a specific drug. In order to analyze the viability characteristics of cancer cells, high-definition microscope images in which cell nuclei are fluorescently stained are used, and individual-level cell analysis is performed. To this end, first, image stitching is performed for analysis of the same environment in units of the well plates, and uneven brightness due to the effects of illumination is adjusted based on the histogram. In order to automatically segment only the cell nucleus region, which is the region of interest, from the improved image, a superpixel-based segmentation technique is applied using the fluorescence expression level and morphological information. After extracting 242 types of features from the image through the segmented cell region information, only the features related to cell viability are selected through the ReliefF algorithm. The proposed method can be applied to cell image-based phenotypic screening to determine a patient's response to a drug.

  • Stem cells are the foundational cells for every organ and tissue in our body. Cell-based therapeutics using stem cells in regenerative medicine have received attracting attention as a possible treatment for various diseases caused by congenital defects. Stem cells such as induced pluripotent stem cells (iPSCs) as well as embryonic stem cells (ESCs), mesenchymal stem cells (MSCs), and neuroprogenitors stem cells (NSCs) have recently been studied in various ways as a cell-based therapeutic agent. When various stem cells are transplanted into a living body, they can differentiate and perform complex functions. For stem cell transplantation, it is essential to determine the suitability of the stem cell-based treatment by evaluating the origin of stem, the route of administration, in vivo bio-distribution, transplanted cell survival, function, and mobility. Currently, these various stem cells are being imaged in vivo through various molecular imaging methods. Various imaging modalities such as optical imaging, magnetic resonance imaging (MRI), ultrasound (US), positron emission tomography (PET), and single-photon emission computed tomography (SPECT) have been introduced for the application of various stem cell imaging. In this review, we discuss the principles and recent advances of in vivo molecular imaging for application of stem cell research.

CELL MORPHOLOGY CHANGE BY THE ULTRAVIOLET RAY IRRADIATION

  • The effect of low doses of ultraviolet (UV) irradiation on morphology changes of cell has been studied based on the observation of the cell length. It was shown that UV-irradiated cell has different behavior in comparison with non-irradiated cell. From the histogram of cell-length distribution, it was confirmed that cell cycle of non irradiated cell was 28 hours, and that cell cycle of irradiated cell with dose of $20\;Jm^{-2}$ was delayed (39 hours), while irradiated cell with $40\;Jm^{-2}$ and $60\;Jm^{-2}$ did not divide and kept growing continuously. It was supposed that in case of $20\;Jm^{-2}$ of irradiation dose, the cell cycle was delayed because the checkpoint worked in order to repair DNA damage induced by generation of pyrimidine dimer, reactive oxygen species and so on. It was also supposed that in case of $40\;Jm^{-2}$ and $60\;Jm^{-2}$ of irradiation dose, overgrowth was induced because the checkpoint was not worked well. The morphology of overgrown cell was similar to that of normally senescent cell. Therefore, it was considered that cell senescence was accelerated by UV irradiation with irradiation doses of $40\;Jm^{-2}$ and $60\;Jm^{-2}$.

Effect of n-3 Fatty Acids on Estrogen Dependency and Protein Kinase C Activity of Human Breast Cancer Cell

  • To investigate the effect of long chain n-3 polyunsaturated fatty acids on breast cancer cell growth, estrogen-dependent MCF-7 human breast cancer cells were cultured serum-free DMEM media containing 0.5$\mu\textrm{g}$/ml of differnet kinds of fatty acids; linoleic acid(LA), arachidonic acid(AA), eicosapentaenoic acid(EPA) and docosahexaenoic acid acid(DHA) and 1, 0.1, 0.2, 0.5and 1.0ng/ml 17$\beta$-estradiol as well as 10$\mu\textrm{g}$/mi insulin and 1.25 mg/ml delipidized bovine serum albumin for 3 days. Cell growth monitored by MTT assay was lower in DHA and EPA treatments as compared to LA treatment, but not with AA treatment. Estrogen concentrations at which cell growth was initially stimulated were 0.1ng/ml for LA and DHA treatments and 0.2ng/ml for EPA and AA treatments, but the degree of stimulation was 25~30% lower in DHA and EPA treatments than in LA treatment. Fatty acid analysis showed that each fatty acid in culture medium was well incoporated into celluar lipid. Protein kinase C activity of cells was most elevated in LA treatment from 2 to 8 hours of culture followed by DHA, EPA, and AA treatments. It is concluded that inhibitions of n-3 DHA and EPA on breast cancer cell growth as compard with n-6 LA is mediated via changes in membrane fatty acid composition reducing estrogen sensitivity and increasing protein kinase C activity.

  • For stationary 1kW-class fuel cell systems to be used widely, it is essential to achieve dramatic improvements in system durability as well as cost reduction. In order to address this engineering challenge, it is important to develop innovative technologies associated with BOP components. According to this background, in 2009, the Korean Government and "Korea Institute of Energy Technology Evaluation and Planning(KETEP)" launched into the strategic development project of BOP technology for practical applications and commercializations of stationary fuel cell systems, named "Technology Development on Cost Reduction of BOP Components for 1kW Stationary Fuel Cell Systems to Promote Green-Home Dissemination Project". The objectives of this project are to develop fundamental technologies to meet these requirements, and to improve the performance and functionality of BOP components with reasonable price. The project consortium consists of Korea's leading fuel cell system manufacturers, BOP component manufacturers which technologically specialized, and several research institutions. This paper is to provide a summary of the project, as well as the achievements made through the 1st period of the project(2009~2010). Several prototypes of BOPs - Cathode air blowers, burner air blowers, preferential oxidation air blowers, fuel blowers, cooling water pumps, reformer water pumps, heat recovery pumps, mass flow meters, valves and power conditioning systems - had been developed through this project in 2010. As results of this project, it is expected that a technological breakthrough of these BOP components will result in a substantial system cost reduction.

  • Ultrastructures on the integumentary epidermis of the marbled sole, Limanda yokahamae, were examined by means of the light and transmission electron microscope. Epidermal layer consists of supporting cells, unicellular glands and accessory cells. The supporting cells were classified into superficial cell, intermediated cell and basal cell. The cytoplasm of supporting cells is divided into cortex and medullar part. In the cortex and medullar part, microfilaments and cell organelles are well developed, respectively. Gland cells are present in the superficial and middle epidermis. The cytoplasm of mucous cell reacted to blue in AB-PAS (pH 2.5). Club cell has a roundish central vacuole and well-developed microfilaments in the cytoplasm. Granular cells are occurs in the middle and basal epidermis , and the cytoplasm is occupied with membrane-bounded granules of electron dense. Chloride cells are present in the superficial epidermis , and the cytoplasm is occupied with tubular mitochondria. Three types of pigment cells can be distinguished by electron density of cytoplasmic inclusions.

Establishment of a Stable Cell Line Expressing Green Fluorescence Protein-fused Hypoxia Inducible Factor-1α for Assessment of Carcinogenicity of Chemical Toxicants

  • Hypoxia inducible factor $1\alpha$ (HIF-$1\alpha$) is a potential marker of carcicnogenesis since it is overexpresssed in many human cancers such as brain, breast, and uterus, and its role has implicated in tumor cell growth and metastasis. In this study, we established a stable cell line that express green fluorescence protein (GFP)-fused hypoxia inducible factor $1\alpha$ (HIF-$1\alpha$) and evaluated the potential use of this cell line for assessment of carcinogenicity of chemical toxicants. Western blot analysis as well as fluorescence measurements showed that protein-level of GFP-HIF-$1\alpha$ was significantly enhanced in a dose-dependent manner upon treatment of hypoxia mimicking agents such as dexferrioxamine and $CoCl_2$. Well-Known tumor promoters such as mitomycin and methyl methanesulfonate. significantly induced the fluorescence intensity of GFP-HIF-$1\alpha$, whereas the known negative controls such as o-anthranilic acid and benzethonium chloride, did not. These results indicate that HIF-$1\alpha$ could be a biological parameter for detection of tumor initiators/promoters and suggest that the GFP-HIF-$1\alpha$ cell line is a useful system for screening of carcinogenic toxicants.

The effect of Rhemanniae radix extract on allergic contact dermatitis on Mice induced by DNCB (생지황(生地黃) 추출물(抽出物)이 DNCB로 유발(誘發)된 생쥐의 Allergy성(性) 접촉피부염(接觸皮膚炎)에 미치는 영향(影響))

  • Allergic contact dermatitis is a common skin disease resulting from specific immunologic to topically applied various allergen. After Dinitrochlorobenzene (DNCB) secondary sensitization, the ICR mice administered Rhemanniae radix extract (RRE) was observed to ascerstain the effect of RRE on allergic contact dermatitis. Purpose of this study was to investigate contact hypersensitivity assay, abdominal skin morphologic changes. Including mast cells and cell-surface glycoconjugates. The change of interleukin 2 (IL-2) receptor (CD25R). ICAM in abdominal skin, lymph node of inguinal region, and electro microscope-morphologic changes of abdominal skin. The results of this study were as follows: 1. The contact hypersensitivity assay, the ear swelling in the RRE had lesser probability than in the ACD Group. 2. In the general morphologic change of skin, hyperplasia of keratinocytes, distribution of vasculogenesis and epidermal lymphocytes infiltration were decreased in the RRE group compared with the ACD group. In epidermal basal layer and prickle layer, cell damage was decreased in the RRE group compared with the group painted with ACD. 3. MasT-cell in dermis was decreased in the RRE group compared with the group painted with DNCB. 4. Distribution of interlukin-2 Receptior positive cell and ICAM positive cell in dermis was decreased in the RRE Group compared with the ACD group. 5. Distribution of helper T-lymphocyte and cytotoxic T-lymphocite in inguinal nodes was decreased in the RRE group, and was observed well in paracortical area and cortical cord. 6. Distribution of apoptotic cell was appeared in the RRE group compared with the ACD group, in skin, dermis. in inguinal nodes, paracortical area observed well. With above results, the restarint of immunosuppression occuring in Allergic contact dermatitis is resulted in the slow progress the effect of Allergic contact dermatitis, and it is thought that ithis fact has a series of relation with apoptosis.
