Hepatitis B virus X protein enhances NFκB activity through cooperating with VBP1

  • Hepatitis B virus X protein (HBx) is essential for hepatitis B virus infection and exerts a pleiotropic effect on various cellular machineries. HBx has been also demonstrated as an indirect transcriptional transactivator of various different viral and cellular promoters. In addition, HBx is involved in the development of various liver diseases including hepatocellular carcinoma. However the mechanism of HBx in hepatocellular carcinogenesis remains largely unknown. In this study, to identify possible new cellular proteins interacting with HBx, we carried out yeast two-hybrid assay. We obtained several possible cellular partners including VBP1, a binding factor for VHL tumor suppressor protein. The direct physical interaction between HBx and VBP1 in vitro and in vivo was confirmed by immunoprecipitation assay. In addition, we found that VBP1 facilitates HBx-induced $NF{\kappa}B$ activation and cell proliferation. These results implicate the important role of HBx in the development of hepatocellular carcinoma through its interaction with VBP1.

Preventive Effects of a Major Component of Green Tea, Epigallocathechin-3-Gallate, on Hepatitis-B Virus DNA Replication

  • Background: Hepatitis B virus infection is one of the major world health problems. Epigallocatechin-3 gallate is the major component of the polyphenolic fraction of green tea and it has an anti-viral, anti-mutagenic, anti-tumorigenic, anti-angiogenic, anti-proliferative, and/or pro-apoptotic effects on mammalian cells. In this study, our aim was to investigate the inhibition of HBV replication by epigallocatechin-3 gallate in the Hep3B2.1-7 hepatocellular carcinoma cell line. Materials and Methods: HBV-replicating Hep3B2.1-7 cells were used to investigate the preventive effects of epigallocatechin-3 gallate on HBV DNA replication. The expression levels of HBsAg and HBeAg were determined using ELISA. Quantitative real-time-PCR was applied for the determination of the expression level of HBV DNA. Results: Cytotoxicity of epigallocathechin-3-gallate was not observed in the hepatic carcinoma cell line when the dose was lower than $100{\mu}M$. The ELISA method demonstrated that epigallocatechin-3 gallate have strong effects on HBsAg and HBeAg levels. Also it was detected by real-time PCR that epigallocatechin-3 gallate could prevent HBV DNA replication. Conclusions: The obtained data pointed out that although the exact mechanism of HBV DNA replication and related diseases remains unclear, epigallocatechin-3 gallate has a potential as an effective anti-HBV agent with low toxicity.

세균액 및 세균단백질 추출물이 배양 세포에 미치는 영향 (EFFECTS OF HEAT-KILLED AND SONIC EXTRACTS OF MICROORGANISM ON CULTURED CELLS)

  • Dental pulp infection is most commonly caused by extensive dental caries, and some bacterial species invade root canals; bacterial components and products are thought to be associated with the pathogenesis of periapical periodontitis. A principle driving force behind pulpal disease response appears to lie in the host immune system's to bacteria and their products. We examined the production of interleukin $1{\beta}$ (IL-$1{\beta}$) and tumor necrosis factor ${\alpha}$(TNF-${\alpha}$) from human peripheral mononuclear cells, lymphocytes and monocytes stimulated by heat-killed Acitnobacillus actinomycetemcomitans (ATCC 29523), Porphyromonas gingivalis (ATCC 33277) and Prevotella intermedia (ATCC 25611), and also by their sonicated bacterial extracts (SBE), respectively. The effects of three strains of heat-killed bacteria and their SBEs on the morphology of cultured blood cell lines HL-60 (KCLB 10240) and J774A.1 (KCLB 40067) were observed under the inverted microscope. Ultrastructural changes of J774A.1 exposed to heat-killed P. intermedia and its SBE were investigated using transmission electron microscopy. Production of IL-$1{\beta}$ was reduced in human peripheral mononuclear cells after stimulation by sonic bacterial extracts of A. actinomycetemcomitans, P. gingivalis, and P. intermedia. Heat-killed and sonic extract of P. gingivalis inhibited the production of TNF-${\alpha}$ in peripheral mononuclear cells. Production of TNF-${\alpha}$ was inhibited in peripheral monocytes after stimulation by sonic extracts of A. actinomycetemcomitans, P. gingivalis, and P. intermedia. HL-60 and J 774A.1 cells showed granular degeneration after treatment with heat-killed and sonic extracts of A. actinomycetemcomitans, P. gingivalis, and P. intermedia Chromatin margination and shrinkage were observed in 774A.1 treated with heat-killed P. intermedia. Cell wall structure and organelles were destroyed and vacuoles were formed in cytoplasm in J774A.1 treated with P. intermedia sonic extract. These results suggest that A actinomycetemcomitans, P gingivalis and P intermedia may have an important role in the formation and progression of pulpal diseases via both modulation of production of IL-$1{\beta}$ and TNF-${\alpha}$ from blood mononuclear cells and cytopathic effects.

Effects of the Synthetic Coprisin Analog Peptide, CopA3 in Pathogenic Microorganisms and Mammalian Cancer Cells

  • A synthetic coprisin analog peptide, 9-mer dimer CopA3 (CopA3) was designed based on a defensin-like peptide, Coprisin, isolated from the bacteria-immunized dung beetle Copris tripartitus. Here, CopA3 was investigated for its antimicrobial activity and cancer cell growth inhibition. CopA3 showed antimicrobial activities against various pathogenic bacteria and yeast fungus with MIC values in 2~32 ${\mu}M$ ranges, and inhibited the cell viabilities of pancreatic and hepatocellular cancer cells, except MIA-Paca2, Hep3B, and HepG2 cells, in a dose-dependent manner. The average $IC_{50}$ values of CopA3 against pancreatic and hepatocellular cancer cells were 61.7 ${\mu}M$ and 67.8 ${\mu}M$, respectively. The results indicate that CopA3 has potential in the treatments of pancreatic and hepatocellular cancers as well as microorganism infection disease.

Roles of Forkhead-box Transcription Factors in Controlling Development, Pathogenicity, and Stress Response in Magnaporthe oryzae

  • Although multiple transcription factors (TFs) have been characterized via mutagenesis to understand their roles in controlling pathogenicity and infection-related development in Magnaporthe oryzae, the causal agent of rice blast, if and how forkhead-box (FOX) TFs contribute to these processes remain to be characterized. Four putative FOX TF genes were identified in the genome of M. oryzae, and phylogenetic analysis suggested that two of them (MoFKH1 and MoHCM1) correspond to Ascomycota-specific members of the FOX TF family while the others (MoFOX1 and MoFOX2) are Pezizomycotina-specific members. Deletion of MoFKH1 (${\Delta}Mofkh1$) resulted in reduced mycelial growth and conidial germination, abnormal septation and stress response, and reduced virulence. Similarly, ${\Delta}Mohcm1$ exhibited reduced mycelial growth and conidial germination. Conidia of ${\Delta}Mofkh1$ and ${\Delta}Mohcm1$ were more sensitive to one or both of the cell cycle inhibitors hydroxyurea and benomyl, suggesting their role in cell cycle control. On the other hand, loss of MoFOX1 (${\Delta}Mofox1$) did not show any noticeable changes in development, pathogenicity, and stress response. Deletion of MoFOX2 was not successful even after repeated attempts. Taken together, these results suggested that MoFKH1 and MoHCM1 are important in fungal development and that MoFKH1 is further implicated in pathogenicity and stress response in M. oryzae.

Prognostic factors for survival of dogs infected with canine parvovirus

  • To determine the prognostic factors for survival of dogs infected with canine parvovirus, clinical and laboratory data of 35 dogs with clinical signs compatible with canine parvoviral enteritis admitted to the Veterinary Medical Teaching Hospital, Seoul National University during the period 1997-1998 were collected. Dogs were grouped by some major covariates, which can be considered as guides to the relative prognosis of dogs in the different subgroups. The Kaplan-Meier survival analysis and Weibull proportional hazard model were used to estimate overall survival, evaluate the comparability between groups, and identify potential prognostic factors. The overall survival rate for all dogs was 45.7% over the study period, and the Kaplan-Meier estimate of one week survival was 0.4989. Gender was the most favorable prognosis ; male dog (median, 6 days) had significantly higher risk of dying than female dog (median, 17 days ; p = 0.0023). In addition to gender, age was significantly associated with survival, with juvenile dogs less than 6-month-old having higher risk (p = 0.0359). Dogs that vaccinated with complete protocol (p = 0.0374) and those of having higher value of mean corpuscular volume (p = 0.0346) were found to be of prognostic importance. The 7 dogs in which white blood cell count of less than 2000 had shorter median survival time (3 days) than the remaining 28 dogs (8 days), but no statistical significance was found between leukopenic and survival. The distribution of packed cell volume and hemoglobin measurement was such that the overall risk of dying in the two groups was comparable. Further studies are needed to more accurately assess these results.

동물세포의 부착에 관여하는 살모넬라 유전자의 특성 연구 (Characterization of Salmonella Adhesins Required for Colonization of Animals)

  • 살모넬라는 질병을 일으키기 위해서 장내의 상피세포에 먼저 부착하여 집락을 이루게 된다. 이에 따라 살모넬라는 부착에 관여하는 세포내 몇 몇 소기관들을 합성한다. 이러한 소기관에는 type 1 fimbriae, plasmid-encoded fimbriae, long polar fimbriae 그리고 thin aggregative fimbriae 등이 있다. 본 논문에서는 type 1 fimbriae, thin aggregative fimbriae, LP fimbriae, 그리고 PE fimbriae 각각을 결손시킨 변이주와 4종류를 모두 결손시킨 변이주를 만들 수 있었다. 변이주들에 대해서 세포배양 부착성 실험을 한 결과, 각각을 결손시킨 변이주들은 몇몇 mammalian 세포주에서 야생형 살모넬라와 동일한 양상으로 부착성을 나타내었고 마우스 실험에서 야생형과 거의 마찬가지로 독성을 가지고 있었다. 반면, 4종류의 fimbriae가 모두 결손된 균주는 닭에서는 독성을 나타내었으나 마우스에서는 매우 약독화되어있음을 확인할 수 있었다. 이상의 결과로부터 오는 차이점은 닭과 비교하여 마우스에서 살모넬라의 부착을 매개하는 표면 구조에 연관성이 있음을 제시하였다.

결핵균 감염에 의한 THP-1 세포에서의 Prothymosin alpha 유전자 발현증가 (Up-regulation of Prothymosin alpha in THP-1 Cells Infected with Mycobacterium tuberculosis)

  • Mycobacterium tuberculosis is capable of growing and survival within macrophage. The purpose of this study was to identify the genes regulated by infection of mycobacteria in human monocytic THP-1 cells. We used the differential display reverse transcriptase polymerase chain reaction (DD RT-PCR) and nothern blot analysis to confirm the differentially expressed genes from THP-1 cells infected with live Mycobacterium tuberculosis H37Rv, heat-killed Mycobacterium tuberculosis H37Rv and live Mycobacterium bovis BCG. Among many up or down-regulated clones, 27 clones were sequenced and compared with known genes on GenBank. Thirteen of over-expressed clones from THP-1 cells infected with live Mycobacterium tuberculosis H37Rv were identical to human prothymosin alpha, eight were novel clones and six clones showed homology with Human ferritin H chain, Esherichia coli bgl, Mouse RNA-dependent EIF-2 alpha kinase, E. coli htrL, Hyaluronan receptor and T cell receptor. Our result suggests that Mycobacterium tuberculosis might regulate prothymosin alpha gene transcription in monocytic THP-1 cell.

핵다면체 바이러스의 감염증상과 전자현미경적 연구 (Infection Symptom and Electron Microscopic Visualization of Nuclear Polyhedrosis Virus)

  • 핵다면체 바이러스는 S. frugiferda 세포주에 감염되었다. 감염 12시간 후에 세포는 운동성을 잃고 세포의 핵은 팽창되었다. 감염 24시간 후에 세포는 비정상적인 형태로 되었으며, 작은 PIB가 나타나기 시작하였다. 감염 48시간 후에는 전체 세포에 PIB가 형성되었고, 감염 76시간 후에는 핵속에 있는 PIB는 일부가 세포밖으로 방출되었다. 전자현미경을 통해 감염후 13시간이 지난후 NPV를 관찰한 결과 세포의 핵속에서는 virogenic stroma가 형성되었으며, 그 부분에서 nucleocapsids가 형성되었다. 감염 48시간 후에는 많은 nucleocapsid들은 bundle을 형성하고, PIB에 봉입되었다. PIB는 대부분 4면체이며, 크기는 $3{\sim}10{\mu}m$ 정도이었다. Virion은 막대형으로 nucleocapsid는 $30{\sim}40{\times}300{\sim}400nm$이었다.

Intestinal histopathology and in situ postures of Gymnophalloides seoi in experimentally infected mice

  • The intestinal histopathology and in situ postures of Gymnophalloides seoi (Digenea: Gymnophallidae) were studied using C3H/HeN and C57BL/6 mice as experimental hosts; the effects of immunosuppression were also observed. The metacercariae isolated from naturally infected oysters, 300 or 1,000 in number, were infected orally to each mouse, and the mice were killed at days 3-21 post-infection (PI). In immunocompetent (IC) mice, only a small number of flukes were found in the mucosa of the duodenum and jejunum during days 3-7 PI, with their large oral suckers pinching and sucking the root of villi. The intestinal mucosa showed mild villous atrophy crypt hyperplasia, and inflammations in the villous stroma and crypt, with remarkable goblet cell hyperplasia. These mucosal changes were almost restored after days 14-21 PI. In immunosuppressed (IS) mice. displacement as well as complete loss of villi adjacent to the flukes was frequently encountered, otherwise the histopathology was generally mild, with minimal goblet cell hyperplasia. In these mice, numerous flukes were found, and it seemed that they were actively moving and rotating in situ. Several flukes were found to have invaded into the submucosa, almost facing the serosa. These results indicate that in IC mice the intestinal histopathology caused by G. seoi is generally mild, and the flukes do not penetrate beyond the mucosa, however, in IS mice. the flukes can cause severe destruction of neighboring villi. and some of them invade into the submucosa.
