To investigate the effects of immunization against somatostatin (SRIF) on growth rate, feed efficiency and carcass quality; forth-eight Yorkshire gilts ($age=37.5{\pm}4.3d,\;wt=8.2{\pm}1.6kg$) were randomly assigned to one of the following three treatments (1) control, (2) bovine serum albumin (BSA) and (3) SRIF. Cyclic SRIF was conjugated to BSA as the antigen containing 1 mg of SRIF diluted in 3 ml of saline. The conjugate was injected subsutaneously together with bacterial cell protein (BP) adjuvant on both sides of the neck of each gilt as the initial injection with three subsequent booster injections. Throughout the experiment all pigs were fed ad libitum a corn-soy diet containing 20% protein. Body weight and feed intake were measured on a weekly basis. All pigs in the experiment were slaughtered when they approached 101 kg body weight on the weekly weigh day. After slaughter, carcass parameters were analyzed to assess carcass quality. Results revealed that there were no differences among SRIF, BSA and control treatments for average daily gain, feed efficiency and feed intake during the first 5 wk of the experiment and from 6 wk to slaughter. The results for carcass analysis indicated that active immunization against SRIF had no effect on fat content, lean yield, water content and Canadian carcass index These data, collectively, suggest that the protocol employed in the present investigation for active immunization against SRIF is not an effective method for the enhancement of pig growth and improvement of feed efficiency and carcass quality.
This study aimed to determine the effect of regulating crude protein (CP) levels in total mixed ration (TMR) on the growth performance and carcass characteristics of Hanwoo steers. The regimens were divided into a control group (TMR with 15% CP) and treatment group (TMR with 12.5% CP). The viable cell count and fermentation potential of the prepared experimental feed and the nutrient digestibility were evaluated. Growth performance and carcass characteristics were confirmed by evaluating the weight and intake of Hanwoo steers, and an economic analysis was performed. Only CP digestibility was higher in the control group than in the treatment group (p<0.05). There were no significant differences in the initial and final body weights, daily gain, and feed requirements between the groups (p>0.05). There were no significant differences in carcass weight, yield index, back fat thickness, and loin area (p>0.05), but the appearance rate of quality grade A was higher in the treatment group than in the control group. In addition, there was no difference in the meat quality characteristics, such as marbling, meat color, fat color, texture, and maturity (p>0.05), and the treatment group had a higher rate of 1++ grade than the control group. Economic analysis showed that when 10 cattle were shipped, the control group received a price of 100,041 thousand won, and the treatment group received a price of 102,033 thousand won. When the income for each group was calculated, the production cost of the treatment group was 8% lower than that of the control group; thus, a higher net profit could be expected considering the low feed cost. Therefore, the economic feasibility of steer raising could be improved while maintaining productivity by controlling CP levels in feed. In addition, this study provides useful basic data for precisely adjusting the CP levels in feed for Hanwoo steers.

Autologous peripheral blood stem cell transplantation (PBSCT) has been used as a major treatment strategy for hematological malignancies. The number of CD34 positive cells in the harvested product is a very important factor for achieving successful transplantation. We studied the factors that can predict the number of CD34 positive cells in the harvested product of acute myelocytic leukemia (AML), multiple myeloma (MM) and Non-Hodgkin's lymphoma (NHL) patients after mobilizing them with chemotherapy plus G-CSF. A total of 73 patients (AML 19 patients, MM 28 patients, NHL 26 patients) with hematological malignancies had been mobilized with chemotherapy and granulocyte colony-stimulating growth factor from April, 2000 to February, 2012. Group's characteristics, checkup opinion of pre-peripheral blood on the day of harvest & outcome of PBSC were analyzed and evaluated using SPSS statistics program after grouping patients as below; group 1: CD34 cell counts < $2{\times}10^6/kg$ (n=16); group 2: $2{\times}10^6/kg{\leq}CD34$ cell counts < $6{\times}10^6/kg$ (n=32); group 3: CD34 cell counts ${\geq}6{\times}10^6/kg$ (n=25). We analyzed the clinical characteristics, the peripheral blood (PB) parameters and the number of CD34 positive cells in the PB and their correlation with the yield of CD34 positive cells collected from the mobilized patients. The total number of leukapheresis sessions was 263 (mean: 3.55 session per patient), and the mean number of harvested CD34 positive cells per patient was $7.37{\times}10^6/kg$. The number of CD34 positive cells in product was significantly correlated with the number of platelet and CD34 positive cells in peripheral blood (P<0.05). The number of PB CD34 positive cells was the best significant factor for the quantity of harvested CD34 positive cells on the linear regression analysis (P<0.05). Many factors could influence the mobilization of peripheral blood stem cells. Platelet count and PB CD34 positive cells count were the two variables which remained to be significant in multivariate analysis. Therefore, the number of platelet and CD34 positive cells in peripheral blood on the day of harvest can be used as an accurate predictor for successful peripheral blood stem cell collection.
Objective: This study evaluated the effect of yeast products on growth performance, visceral organ weights, endogenous enzyme activities, ileal nutrient digestibility and meat yield of broiler chickens fed diets containing autolyzed whole yeast (WY) and yeast cell walls (YCW) at varying levels of inclusion. Methods: Nine dietary treatments consisting of WY or YCW included at 0.5, 1.0, 1.5, or 2.0 g/kg diet and a control diet without yeast supplementation was used in the experiment. Each of the nine treatments was replicated six times with nine birds per replicate. Birds were housed in cages, in climate-controlled rooms and fed starter, grower and finisher diets. Results: There was an improvement (p<0.05) in body weight gain and feed conversion ratio on d 10, 24, and 35 for birds fed 1.0 to 2.0 g/kg WY or YCW diet. Small intestine weight was heavier on d 10 and 24 for birds on higher levels of WY and YCW compared to the control group. On d 10 and 24, there was a significant increase (p<0.05) in tissue protein content and pancreatic enzyme activities (trypsin and chymotrypsin) of birds on 1.5 to 2.0 g/kg WY and YCW diets compared to the control group. Compared to the control group, birds on WY (2.0 g/kg diet) and YCW (at 1.5 and 2.0 g/kg diet) had better (p<0.05) protein digestibility on d 24. On d 35, there was significant improvement (p<0.05) in percentage of carcass, absolute and relative breast weight for broiler chickens fed WY and YCW mostly at 2 g/kg diet compared to birds on the control diet. Conclusion: Supplementation of diets with autolyzed WY and YCW products especially at 1.5 to 2.0 g/kg diet improved broiler chicken performance and meat yield through their positive effects on ileal protein digestibility and pancreatic enzyme activities.
$\beta$-Glucan has been efficiently produced with higher yield by the optimization of liquid cultivation conditions. The optimal composition of medium for batch culture was 5% (w/v) of glucose as a carbon source, 0.5% (w/v) of yeast and 0.5% (w/v) of malt extract as a nitrogen source, 0.1% (w/v) of $KH_2PO_4$ and 0.05% (w/v) $MgSO_4{\cdot}7H_2O$, which had been the base medium for determination of other conditions. The set-up conditions are pH 5.0, $28^{\circ}C$, 1 vvm for aeration and 300 rpm for agitation. In order to minimize the inhibition effect of glucose on the initial growth of mycelia and to maximize the production of extracellular $\beta$-glucan, we have reduced the initial glucose feed to 4% and added 2nd feed at the point of 70 hr from the initial feed. The 2nd feed was composed of glucose 3%, yeast extract 0.1 % and malt extract 0.1 %. It improved the $\beta$-glucan yield upto 5.2 g/L in comparison with 2.8 g/L resulted from batch cultivation. Moreover, the serial treatment of a cell wall lytic enzyme and bromelain to the mycelia was effective for extraction of the cell wall bound $\beta$-glucan. The yield of $\beta$-glucan extraction by the enzyme treatment was 3.5 g/L, which was almost 4 times higher than that by hot-water extraction.
Fermentation characteristics of Jerusalem Artichoke of yeast K.fragilis CBS 1555 were investigated experimentally and quantitatively according to the change of initial sugar concentrations and initial PHs of fermentation broth. Initial sugar concentrations employed were 26, 45, 65, 105, 180, and 215g/1. And initial PHs of fermentation broth were 3, 5.5, 7 and 9. The maximum specific growth rate was observed as 0.4hr-1 at 65g/1 of initial sugar concentration. The maximum specific alcohol production rate was 1.68g/ghr at 105g/1 of initial sugar concentration Cell yield and ethanol yield represent the maximum values such as 0.14 and 0.49 respectively when the initial sugar concentration was 25g/1. The maximum of ethano1 fermentability, 97% was obtained at the initial concentrations, 26 and 45g/1. However, the maximum of total ethanol yield productivity was 2.78g/1hr when the initial concentration was 215g/1. And also the optimum PH was found 5.5 for both specific growth rate and specific alcohol production rate.
This experiment was conducted to obtain the have higher contents of pharmaceutical constituents as well as higher yield from colchicine induced diploid and tetraploid extracts of Platycodon grandiflorum. In order to determine the biological activity, this study was focused to evaluate the cytotoxicity, antimicrobial on the bronthus disease bacteria, antioxidant enzyme activity of diploid and tetraploid extracts in P. grandiflorum. The activities of antioxidant enzyme according to different solvent extracts were measured as superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX). The cytotoxicity of methanol extracts of P. grandiflorum showed significant differences between tetraploid and diploid. That is, the cytotoxic effect against human cancer cell was higher in tetraploid than in diploid. At all extracts concentration, tetraploid samples showed high toxicity and the $IC_{50}$ (concentration causing 50% cell death) value showed the highest on HCT-116 cell ($105.91{\mu}g/mL$), and exhibited significant activity against the Hep 3B cell ($140.67{\mu}g/mL$), SNU-1066 cell ($154.01{\mu}g/mL$), Hela cell ($158.37{\mu}g/mL$), SNU-601 cell ($182.67{\mu}g/mL$), Calu-6 cell ($190.42{\mu}g/mL$), MCF-7 cell ($510.19{\mu}g/mL$). Antimicrobial activities of diploid P. grandiflorum were relatively low compared to tetraploid P. grandiflorum on most of the bacterial strains. In tetraploid P. grandiflorum, K. pneumoniae showed the clear zone formation (18~19 mm) of growth inhibition, followed by the clear zone formation of 13~15 mm on C. diphtheria and S. pyogenes. The antimicrobial activities in diploid P. grandiflorum were the highest on K. pneumonia (14~15 mm), and showed the clear zone formation of 11~12 mm on C. diphtheria and 12~13 mm on S. pyogenes. The antimicrobial activity is thought to look different depending on the bacterial strains and the polyploidy of P. grandiflorum. The root extract of P. grandiflorum had the highest (97.2%) SOD enzyme activity in ethyl acetate partition layer of tetraploid while water partition layer of diploid showed the lowest (48.6%) SOD enzyme activity. The activity of CAT showed higher values in the root of tetraploid than in the diploid of P. grandiflorum in all partition layers except butyl alcohol. The activities of APX and POD showed higher values in the root of tetraploid than in the diploid of P. grandiflorum in all fraction solvents except water layer. These results indicate that the tetraploid P. grandiflorum can be used as a source for developing cytotoxic agent and antimicrobials which can act against bronchus diseases bacterial strains.
Korean Lentinus edodes SR-1 was cultured to multiply the mycelia in the complete broth medium (C/N=13.1) for mushroom, and protein-bound polysaccharides were extracted from the cultured broth containing mycelia (The whole cultured broth was used to increase the yields: 80% of protein-bound polysaccharides were existed at the cell wall of mycelia and 20% of those were secreted extracellularily in this culture). Protein-bound polysaccharides in the cultured broth containing mycelia were extracted by using three different methods: 1) Extraction with hot water, 2) Disintegration of cell wall by glass bead mill treatment before extraction with hot water, and 3) Cellulase treatment before extraction with hot water. The highest yield was obtained (930 mg polysaccharides/100 mL culture broth) when protein-bound polysaccharides were extracted with 2) method. The extracted crude protein-bound polysaccharides were purified using protease, DEAE-cellulose and Sephadex G-100. The growth inhibition activity for $P_{388}$, mouse leukemic cell, increased (53.7, 62.2, 93.7% and 97.4%) as the purification level increased. Protein-bound polysaccharides contained 46.1% of polysaccharides, 7.3% of protein, and trace amounts of minerals. Polysaccharides contained glucose, galactose, xylose and mannose. The content of proline and glycine were high, however, methionine and leucine were not found. The major minerals were Na, K, Zn, and Ca.
The research is concerned with optimization of growth medium composition in an attempt to improve the product yield of Bacillus licheniformis amylase (BLMA) in recombinant E. coli containing the BLMA gene. BLMA has the catalytic activity of producing branched oligosaccharides from starch. The medium optimization was performed in flask cultures based on the Box and Wilson method. The optimized medium is composed of tryptone 18.0 g/l, yeast extract 22.4 g/l, NaCl 5.3 g/l and glucose 2.1 g/l. In a jar fermenter culture with the conventional LB medium, the recombinant E. coli yielded 1.39 g/l of final dry cell mass and 5.11 U/ml of enzyme activity. In the optimized medium, however, the final cell mass was increased to 6.01 g/l and the enzyme activity to 23.2 U/ml. Medium optimization improved cell mass by 4.3 times and enzyme activity by 4.5 times. Such an increase in enzyme activity is mainly due to an enhancement of cell mass.

Abiotic stress is a major problem in global agriculture as it negatively affects crop growth, yield, and quality. Wheat (Triticum aestivum) is the world's second-highest-producing food resource, so the importance of mitigating damage caused by abiotic stress has been emerging. In this study, we performed GWAS to search for SNPs associated with salt tolerance and drought tolerance. NaCl (200 mM) treatment was performed at the seedling stage using 613 wheat varieties in Korean wheat core collection. Root length, root surface area, root average diameter, and root volume were measured. Drought stress was applied at the seedling stage, and the above phenotypes were measured. GW AS was performed for each phenotype data using the MLM, MLMM, and FarmCPU models. The best salt-tolerant wheat varieties were 'MK2402', 'Gyeongnam Geochang-1985-3698', and 'Milyang 13', showing superior root growth. The significant SNP AX-94704125 (BA00756838) were identified in all models. The genes closely located to the significant SNP were searched within ± 250 kb of the corresponding SNP. A total of 11 genes were identified within the region. NB-ARC involved in the defense response, FKSI involved in cell wall biosynthesis, and putative BP Ml involved in abiotic stress responses were discovered in the 11 genes. The best drought-tolerant wheat varieties were 'PI 534284', 'Moro of Sind', and 'CM92354-33M-0Y-0M-6Y-0B-0BGD', showing superior root growth. This study discovered SNPs associated with salt tolerance in Korean wheat core collection through GWAS. GWAS of drought tolerance is now proceeding, and the GWAS results will be represented on a poster. The SNPs identified by GWAS can be useful for studying molecular mechanisms of salt tolerance and drought tolerance in wheat.
