YSZ based anode supported solid oxide fuel cells (SOFCs) were prepared, and two cells with different electrolyte thicknesses were connected in series for the simulation of a cell-imbalanced fuel cell stack. Pure YSZ cells in a series connection exhibited a rapid degradation when a thick electrolyte cell was operated under a negative voltage. On the other hand, ceria added-YSZ cells in a series connection were stable under similar operating conditions, and the power density and impedance were about the same as those before tests. The improved stability was due to the reduction of internal partial pressure in the electrolyte by locally increasing the electronic conduction. Thus, we propose a new protection method, i.e., the local addition of ceria in the YSZ electrolyte, to extend the lifetime of a cell-imbalanced SOFC stack.
Effects of the cell wall-degrading enzymes derived from Acremonium cellulolyticus and Trichoderma viride on the silage fermentation and in sacco degradation of tropical grasses i.e. rhodesgrass (Chloris gayana Kunth. cv. Callide) and guineagrass (Panicum maximum Jacq. cv. Natsukaze) were investigated in laboratory-scale experiments. These two grasses were either treated with or without the enzymes before ensiling. Untreated rhodesgrass produced acetate fermentation silage (lactate, $13.0\;g\;kg^{-1}$ DM; acetate, $38.7\;g\;kg^{-1}$ DM) with high final pH value and $NH_3$-N content (5.84 and $215\;g\;kg^{-1}$ DM). Addition of enzymes significantly increased (p<0.01) the lactate production (lactate, 45.6; acetate, $34.0\;g\;kg-^{1}$ DM) and decreased (p<0.01) the pH and $NH_3$-N (4.80 and $154\;g\;kg^{-1}$ DM) in the ensiled forages when compared with the control silages. Untreated guineagrass was successfully preserved with a high lactate proportion (lactate, 45.5; acetate, $24.1\;g\;kg^{-1}$ DM), and the addition of enzymes further enhanced the desirable fermentation (lactate, $57.5\;g\;kg^{-1}$ DM; acetate, $19.4\;g\;kg^{-1}$ DM). The content of NDF was lowered (p<0.05) by enzymes in both silages, but the extent appeared greater in the enzyme-treated rhodesgrass (rhodesgrass, $48\;g\;kg^{-1}$ DM; guineagrass, $21\;g\;kg^{-1}$ DM). Changes in the kinetics of in sacco degradation showed that enzyme treatment increased (p<0.01) the rapidly degradable DM (rhodesgrass, 299 vs. $362\;g\;kg^{-1}$ DM; guineagrass, 324 vs. $343\;g\;kg^{-1}$ DM) but did not influence the potential degradation, lag time and degradation rate of DM and NDF in the two silages.

Scandia-stabilized zirconia co-doped with $CeO_2$ is a promising electrolyte for intermediate temperature SOFC, but still shows rapid degradation during a long-term operation. In this study, $CeO_2$ (1 mol%) as a stabilizer is partially substituted with lanthanum oxides ($M_2O_3$, M=Yb, Gd, Sm) to stabilize a cubic phase and thus durability in reducing atmosphere. 0.5M0.5Ce10ScSZ electrolytes were prepared by solid state reaction and sintered at $1450^{\circ}C$ for 10 h to produce dense ceramic specimens. With addition of the lanthanum oxide, 0.5M0.5Ce10ScSZ showed lower degradation rates than 1Ce10ScSZ. Since $Gd_2O_3$ showed the highest ionic conductivity among the co-dopants, an electrolyte-supported cell with 0.5Gd0.5Ce10ScSZ was prepared to compare its long-term performance with that of 1Ce10ScSZ-based cell. Maximum power density of 0.5Gd0.5Ce10ScSZ-based cell was degraded by about 2.3% after 250 h, which was much lower than 1Ce10ScSZ-based cell (4.2%).
As an index of freeze-injury of yeast, the leakage of intracellular substances from yeast cells after freeze-thawing was investigated. It was found that much more ultraviolet-absorbing substances leaked out from non-freeze tolerant yeast (NETY) than from freeze-tolerant yeast. Furthermore, the rate of leakage of cellular substances form NFTY during incubation exceeded that of FTY, indicating that NFTY is more susceptible to freeze-injury than FTY during frozen-storage. An apparent degradation of phospholipid was observed during incubation of perfermented frozen-cells of NFTY, while little change of phospholipid occurred in FTY, These results suggested that the difference in the sensitivity of yeast might be due to the strength of cell membrane in terms of the degradation of phospholipid by enzymes, phospholipases, attached to cell membranes.
The stability of a solid oxide fuel cell (SOFC) stack is strongly dependent on the magnitude and profile of the internal chemical potential of the solid electrolyte. If the internal partial pressure is too high, the electrolyte can be delaminated from the electrodes. The formation of high internal pressure is attributed to a negative cell voltage, and this phenomenon can occur in a bad cell (with higher resistance) in a stack. This fact implies that the internal chemical potential plays an important role in determining the lifetime of a stack. In the present work, we fabricate planar type anode-supported cells ($25cm^2$) with a bi-layer electrolyte (with locally increased electronic conduction at the anode side) to prevent high internal pressure, and we test the fabricated cells under a negative voltage condition. The results indicate that the addition of electronic conduction in the electrolyte can effectively depress internal pressure and improve the cell stability.
The objective of this study was to examine the impact of three categories of supplements on intake and diet induced difference on degradation of straw substrates. Sixteen crossbred cattle fitted with rumen cannula were randomly divided into 4 equal groups. Animals were fed on wheat straw ad libitum without any supplement except mineral mixture (control; $T_1$) or supplemented with concentrate mixture (CS; $T_2$) or green Lucerne (GLS; $T_3$) or urea-molasses block lick (ULS; $T_4$). Total dry matter intake in $T_2$, $T_3$ and $T_4$ was increased by 70, 54 and 49%, respectively compared to $T_1$ which was only 1.55 kg/100 kg B.Wt. Other than control animals, straw intake was less on $T_3$ than $T_2$ or $T_4$. In Sacco degradation of untreated and urea treated wheat or paddy straw in different treatments indicated that the supplements had a significant (p<0.01) impact on rapidly soluble (A) and insoluble but potentially degradable (B) fractions of straw. Urea treatment increased fraction-A but, provision of supplement improved fraction-B also. Effective degradation (ED) of OM was better on $T_2$. Rate of degradation (C) of OM and CWC was dependent on diet and type of straw but hemicellulose and cellulose were related to latter factor only. ED of cell wall carbohydrates (CWC) was similar in $T_2$ and $T_4$ but higher than $T_3$. CS was more effective in improving the degradation of both untreated and urea treated straw while ULS was effective on the former only. CS had more impact on superior quality straw while contrary was true with ULS. Although GLS improved intake and degradability of untreated and urea treated straws, its bulkiness affected the straw intake compared to other supplements.
Pseudomanas sp. EL-04J was previously isolated from phenol-acclimated activated sludge. This bacterium was capable of degrading phenol and cometabolizing trichloroethylene (TCE). After precultivation in the mineral salts medium containing phenol as a sole carbon source, Pseudomonas EL-04J degraded 90% of TCE $25 \mu\textrm{M}$ within 20 hours. Thus, phenol-induced Pseudomonas sp. EL-04J cells can bdegrade TCE. Followsing a transient lag period, Pseudomonas sp. EL-04J cells degraded TCE at concentrations of at least $250 \mu\textrm{M}$ with no apparent retardation in rate, but the transformance capacity of such cells was limited and depended on the cell concentration. The degradation rate of TCE followed the Michaelis-Menten kinetic model. The maximum degradation ratio ($V_{max}$) and saturation constant ($K_{m}$) were $7nmo {\ell}/min{\cdot}mg$ cell protein and $11 \mu\textrm{M}$, respectively. Cometabolism of TCE by phenol fed experiment was evaluated in $50m {\ell}$ serum vial that contained $10m {\ell}$ of meneral sals medium supplemented with $10 \mu\textrm{M}$ TCE degradation was inhibited in the initial period of 1 mM phenol addition, but after that time Pseudomonas sp. EL-04J cells degraded TCE and showed cell growth.
Tripitaka Koreana were made during Coryo Dynasty from 1236 to 1251 A.D. Buddhist scriptures were engraved on 81.340 wooden plates. Some plates were varnished with Rhus lacquer, but most of them were uncoated. Macroscopically, most of the plates appeared intact due to the storage in a well-ventilated wooden house. Because, they were irregularly used for printings with ink, it can be assumed that they were repeatedly exposed to ink-water and drying processes. The present were made to examine the changes of wood cell structures occurred during long-term aging deterioration processes in these dry archaeological wooden plates. Light, scanning and transmission electron microscopes were employed for this study. Wedge-shaped cracks and delamilations were found from the lumen side toward the compound middle lamellae and they progressed toward primary or secondary walls. A large amount of hypae in vessels and the degradation of vessel-ray pit walls by the fungal hyphae were observed. When compared to the recent wood, the birefringence of wood fibers was considerably lower or completly disappeared, suggesting the degradation of crystalline cellulose in these wood samples. The degradation of the cell wall could be also revealed the calculation of crystallinity with X-ray diffraction and the size of crystalline region was estimated.
