Comparative Activities of Novel $\beta$-Lactamase Inhibitors, 6-Exomethylene Penamsulfones (CH1240, CH2140) in Experimental Mouse Infection Model

  • The antibacterial activity of novel ${\beta}$-lactamase inhibitors, 6-exomethylene penamsulfones (CH1240, CH2140), has been compared in vivo with that of sulbactam and clavulanic acid against b-lactamase producing strains. In vivo microbiological assessment was used as experimental mouse infection model by gram negative strains. Against Pseudomonas aeruginosa F0013, cefoperazone/CH 1240 was slightly less active than sulbactam. ampicillin/CH 2140 was less effective than sulbactam against escheriachia coli 3457. Especially against Citrobacter diversus 2046E, amoxicillin/CH 2140 was the most potent and amoxicillin/CH 1240 was slightly more active than clavulanic acid. consequently the difference in efficacy between the drug combinations appers to be related to the degree of protection afforded the animals by the b-lactamasse inhibitors. CH1240 and CH2140 are promising new agents and should undergo further investigations.

  • In vitro $\beta$-lactamase inhibitory activity of 6-exomethylenepenam compounds ( 1, 2, 3, 4 and 5) was compared with clavulanic acid, sulbactam and tazobactam. The inhibitory activity of compound 3 was stronger than those of sulbactam and clavulanic acid against Type I and II enzymes and stronger than tazobactam against Type III, IV, TEM enzymes. The inhibitory activity of 5 was stronger than sulbactam and clavulanic acid against Type I and II enzymes and stronger than tazobactam against Type III, and IV enzymes. The in vitro antimicrobial activity of 3, 4 and 5 combined with ampicillin and cefoperazone was compared with the sulbactam against $\beta$-lactamase producing 27 strains. But, synergistic activity of 3 and 5 was inferior to tazobactam.

  • Survey on MIC of antimicrobial drugs and its treatment efficacy in mice were conducted for the strains of Escherichia coli and Salmonella spp isolated from feces of young domestic animals with diarrhea in 1996. A total of 338 strains of E coli and 61 strains of Salmonella spp were examined for the susceptibility to 20 antibiotics and 7 synthetic antimicrobial drugs. The results indicated that the majority of strains were susceptible to amikacin(93.5%), cefoperazone/sulbactam(93.5%), cefotaxim(93.3%), cefomandole(92.8%), cefoperazone(91.6%) and ciprofloxacin(85.1%), in order. Although gentamicin, ciprofloxacin and norfloxacin showed the relatively low MIC distributions, erythromycin, doxycycline, sulfamethoxazole and oxytetracycline revealed the high MIC distributions to most of isolates. The $MIC_{90}$ of antimicrobials for E coli were > $62.5{\mu}g/ml$ in gentamicin, $2.0{\mu}g/ml$ in ciprofloxacin, $1.0{\mu}g/ml$ in norfloxacin, > $500{\mu}g/ml$ in erythromycin, $125{\mu}g/ml$ in doxycycline, > $1000{\mu}g/ml$ in sulfamethoxazole and > $250{\mu}/ml$ in oxytetracycline. In general, the MIC of E coli isolates was higher than that of Salmonella spp isolates. Although variation in synergism or additivity of antibiotic combinations were demonstrated, ampicillin-gentamicin was the most efficacious combination both against E coli and Salmonella spp with the fluctuation of 7.7-77.5%. In the experiment of treatment efficacy in mice, the highest survival ratio(83.3%) after challenge with pathogenic E coli and Salmonella typhimurium was detected in the group treated with gentamicin.

경북지방 환돈에서 분리한 Streptococcus suis의 생화학적 성상 및 약제감수성 (Antimicrobial Susceptibility and Biochemical Characteristics of Streptococcus suis Isolated from Diseased Pigs in Gyeongbuk Province)

  • Streptococcus suis는 사람과 돼지에서 다양한 감염을 유발하는 병원체이다. 2004년부터 2009년까지 경북지방 양돈장에서 병성감정 의뢰한 돼지 가검물을 대상으로 S. suis를 분리하여 생화학적 성상, 약제감수성 시험 및 PCR을 이용한 동정을 실시한 결과는 다음과 같다. 40개 농장에서 분리한 61주의 S. suis에 대하여 생화학적 성상시험 결과 모두 S. suis로 동정되었으며, VP 음성, hippurate, esculin, arginine decarboxylase 양성, lactose 분해능이 있는 것으로 나타났다. Trehalose와 starch 분해능은 비교적 높게 나타났으며 생화학적 성상에 따른 phenotype은 11형으로 나누어졌다. 항생제 감수성 시험 결과 대부분의 균주가 amoxicillin/clavulanic acid, ampicillin, cephalothin, cefoperazone, florfenicol에 높은 감수성을 나타내었지만 amikacin, kanamycin, neomycin, streptomycin, erythromycin, clindamycin, tetracycline에는 높은 내성을 나타내었다. 16S-rRNA 특이유전자 부위를 이용한 PCR 결과 공시균주 모두 304 bp에서 증폭산물이 관찰되었다. 이들 결과는 국내에서 사육되고 있는 돼지에서 S. suis 감염을 예방하는데 도움이 될 것으로 기대된다.

국내에서 분리된 Acinetobacter baumannii의 Integron과 연관된 다제내성 분석 (Analysis of Integron-Associated Multi-Drug Resistance of Acinetobacter baumannii Isolated in Korea)

  • 임상분리 균주인 A. baumannii 1625는 imipenem (carbapenem 계열)을 포함한 대부분의 ${\beta}$-lactam계열의 항생제들과 kanamycin, gentamicin, tobramycin 및 제 3세대와 4세대 cephalosporin 계열의 항생제들에 광범위한 내성을 나타내었고, 한국에서는 드문 IMP-1형 metallo-${\beta}$-lactamase (MBL)를 생성하는 균주임이 확인되었다. A. baumannii 1625는 약 2.5 kb 크기의 class 1 integron을 갖고 있었으며, 이 integron내에 aminoglycoside계열 내성 유전자인 accA4, carbapenem 계열 내성 유전자인 $bla_{IMP-1}$, 광범위한 ${\beta}$-lactam 내성 유전자인 $bla_{OXA-2}$ 유전자 cassette들이 차례대로 위치해 있음을 확인하였는데, 이것은 IMP-1형과 OXA-2형 ${\beta}$-lactamase의 유전자를 같은 integron내에 동시에 갖는 새로운 배열 및 구조로서 이전에 국내에서 보고된 바 없는 것이다. 이 2.5 kb 크기의 integron을 항생제 내성이 없는 E. coli에 형질전환 시켰을 때, imipenem, ampicillin, piperacillin, cefazolin, cefoperazone, aztreonam 등의 항생제들에 대하여 8배 이상 증가된 내성정도를 보였다. 이는 A. baumannii 1625의 integron이 다제내성을 부여하는 기능을 하고 있음을 보여준다.

녹농균에 대한 Aminoglycoside계와 Cephalosporin계의 병합작용 (Combined action of Aminoglycoside and Cephalosporin Against Pseudomonas aeruginosa)

  • Thirty-one strains of Pseudomonas aeruginosa were submitted to the synergistic activity test of amikacin(AK) and gentamicin(GM) combined with moxalactam(MX), ceftizoxime(CTZ) or cefoperazone(CFZ). The minimal inhibitory concentrations(MICs) of each drug and drugs combined in various ratios were measured by checkerboard dilution method. The synergism was determined through analysing the MIC distribution curve on isobologram and calculating the fractional inhibitory concentration index(FICI). MICs of GM, AK, MX, CFZ and CTZ against the 31 tested strains were distributed from $12.5{\mu}g/ml$ to $800{\mu}g/ml$, from $0.8{\mu}g/ml$ to $25{\mu}g/ml$, from $3.1{\mu}g/ml$ to $50{\mu}g/ml$, from $3.1{\mu}g/ml$ to $400{\mu}g/ml$, and from $12.5{\mu}g/ml$ to $100{\mu}g/ml$, respectively. The rate synergism of each drug combination by means of FICl was 45.5% in GM-MX, 36.4% in GM-CFZ, 63.6% in GM-CTZ, 48.6% in AK-MX, 35.3% in AK-CFZ, and 35.7% in AK-CTZ combination. Thus, it is suggested that Pseudomonas aeruginosa may effectively be inhibited by various aminoglycoside and cephalosporin combinations.

ISOLATION OF Campylobacter jejuni AND C. coli FROM DOMESTIC AND EXPERIMENTAL ANIMALS AND THEIR DRUG SUSCEPTIBILITY

  • A total of 526 domestic and experimental animals in Miyagi prefecture, Japan were investigated for fecal carriage of Campylobacter jejuni and Campylobacter coli. C. jejuni was detected in chickens (8.2%), dogs (6.3%), pigs (4.3%), cattle (1.8%) and hamsters (1.4%). C. coli was only detected from pigs (20.7%). Drug susceptibility test was performed on 5 strains of C. jejuni isolated from chickens and 13 strains of C. coli isolated from pigs to tylosin (TS), thianphenicol (TP), carbadox (CDX), chroltetracyclin (CTC), vancomycin (VCM), cefoperazone (CPZ), latamoxef (LMOX), GM were highly effective and CTC, CP and PL were moderately effective against both C. jejuni and C. coli. TS and TPH were moderately effective against C. jejuni; however, they were less effective to C. coli. One strain of C. jejuni against CTC considered to be drug resistant. The results suggest that C. jejuni and C. coli can be controlled by several drugs effectively, although a drug resistant strain exists.

  • The identification step of raw drug materials is an indispensible procedure in the GMP manufacturing process within the pharmaceutical industry. However, wet chemistry methods for identification of drug materials, used by the various Pharmacopeia are time-consuming and expensive steps. In this paper, near-infrared spectroscopy (NIRS) has been developed for identifying eleven drug substances including calcium pantothenate, cefaclor, cefoperazone, cephradine, dextromethorphan, ehtambutol, nicotinamide, pyrozinamide, tramadol, vitamin C, and vitamin E. Also the aim of ths work is to consturct a new algorithm for calibration model using soft independence modeling of class analogy (SIMCA) with Malinowskis Indicator Function (IND), which is used for finding the number of principal components of each class of the SIMACA model. The use of NIR technique with pattern recognition to qualify raw materials can make it possible to monitor process in real time as well as to control all procedures in the pharmaceutical industry. As the result, the samples identified of 183 different batches from 11 different compounds were separated clearly by SIMCA with 2nd derivative spectra in the NIR region of 1100∼2400 nm.

이질균속의 항균제 내성 (Antimicrobial Drug Resistance in Shigella cultures Isolated in Korea (1983))

  • One hundred and forty strains of Shigella cultures isolated from the twelve hygiene laboratories of cities and provincial level and general hospital laboratories in 1983, and were tested for their resistance to 13 antimicrobial drugs and their R-Plasmid transfer. One hundred and forty (100%) of isolates were susceptible to amikacin, gentamicin, tobramycin, a total of 94.3% of all shigella isolates were resistant to 1 or more of the 13 antimicrobial agents tested. The most commonly found resistance was to chloramphenicol (94%), followed by streptomycin (93%), tetracycline (92%), piperacillin (90%), ampicillin (83%), cefoperazone (42%), nalidixic acid (14%), cephalothin (17%), rifampicin (22%), and kanamycin (6%). Sixty percent of strains among 140 were resistant to ampicillin, chloramphenicol, streptomycin, tetracycline at same time. The transfer of drug resistance by conjugation was tested and 94 strains (94.3%) which were resistant to one or more drugs were found to transfer their drug resistance to E. coli.

2005년 한국에서 시판된 먹는 샘물의 미생물 오염 (Assessment of Bacterial Contamination of Bottled Water in Korea, 2005)

  • In recent years, there has been an increase in consumer demand for bottled waters. There is a perception that consumption of natural mineral water represents a healthy life style and that these products are relatively safe. In this study, the microbiological quality of 39 samples of bottled water, purchased from retail store in Korea, was investigated during the 2005. Applying pour plate method, the 1 mL of water samples were analyzed for the presence and enumeration of total general bacteria and Pseudomonas spp.. Nineteen samples representing 9 brands of bottled water contained general bacteria ($1.54{\times}10^2$ CFU/mL). In addition four samples contained Pseudomonas spp. and Camamonas acidovorans. The susceptibility of the strains tested against 25 antimicrobial agents, Pseudomonas fluorescens were resistant to Lincomycin, Amoxacilin/Clavulanic acid and Cefazolin (> $100{\mu}g/mL$). Also Comamonas acidovorans were intermediate to Cephalothin and resistant to Cefoperazone.
