부산지역 소 바이러스성 설사병 바이러스(bovine viral diarrhea virus; BVDV) 감염 실태 조사·연구(2013~2014) (Study on prevalence of antigens to bovine viral diarrhea virus (BVDV) of Cattle in Busan area (2013~2014))

  • Bovine viral diarrhea virus (BVDV) is a very important viral disease virus in cattle, domestic and wild ruminants. The purpose of this study is to investigate the positive rate of bovine viral diarrhea virus antigen by ELISA from Korean native and beef cattle reared in Busan area from March in 2013 to October in 2014. A total of 1,129 bovine blood samples were collected from 140 farms, 1,111 Korean native cattle of 135 farms and 18 beef cattle of 5 farms. Test for antigen was carried out by ELISA method. In general analysis, the positive rate of bovine viral diarrhea virus antigen were 0.7% (8/1,129) cattle and 5.0% (7/140) farm. In regional analysis, the positive rate of BVDV antigen of farm in Kijang-gun, Gangseo-gu, Geumjeong-gu, Saha-gu and Dongnae-gu were 1.4% (2/94), 3.6% (5/37), 0% (0/7), 0% (0/1) and 0% (0/1), respectively, and the positive rate of BVDV antigen of cattle were 0.4% (3/770), 1.5% (5/333), 0% (0/24), 0% (0/1) and 0% (0/1), respectively. The positive rate of BVDV antigen according to sex were 0.6% (6/1,085) female cattle and 4.6% (2/44) male cattle. According to the age of cattle, the positive rate of BVDV antigen in 1 year, 2 years, 3 years and 5 years old were 1.9% (4/215), 0.4% (1/265), 0.9% (2/234) and 1.0% (1/103), respectively, but 4 years (0/198), 6 years (0/55), 7 years (0/24), 8 years (0/14), 9 years (0/10), 10 years (0/7) and 11-15 years (0/3) old were negative, respectively.

  • The survey was performed to provied details about the pattern of bovine brucellosis occurred in Kyunggi province area. The results obtained through the investigations were summarized as follows. Five hundred seventy - three cattle of bovine brucella reactor were occurred in 14 districts among 31 districts in Kyunggi province in 1989-1995. Among them, 370 cattle(64.5% ) were bred in Eastern area(Ichon, Yeju, Kwangju) and 153 cattle(26.7% ) in Southern area(Yongin, Ansung, Peungtack) And the number of farms occurred by bovine brucellosis was 110 ones. When we investigated the occurrance frequency for the 110 farms, the ratio of farms which was brocken out just one time was 67.3a and more than twice was 32.7%. In 573 cattle, 271 cattle were reoccurred in farms which had broken out the bovine brucellosis more than one time. And this survey said the interval of reoccurrance was like this ; within a month 50.2%, within two month 19.2%, within four month 7.4%, within six month 7.4%, within an year 15.1%, within 2 year 7.0%. Brucella abortus was isolated from 38 cattle of the 61 ones cattles, and in type all isolated belong to biotype 1.

  • To investigate epidemological sitution of bovine viral diarrhea infection, serological survey in cattle being raised in Young Dong province were conducted. Bovine sera collected ramdomly from August 1990 to December 1990 were tested for bovine viral diarrhea virus serum neutralizing antibody titers. The results were as follows 1. BVDV SN antibody levels were considerably varies and positive rate was 58(108 heads out of 186) 2. BVDV SN antibodies to breeds of cattle was various and positive rates showed that diary cattle, beef, native cattle(Korean) were 67.52%, 59.38%, 27.00% respectively followed in that order. 3. In the regional prevalence of BVD SN antibodies in cattle, Alpine(92%) was the highest, Young Dong south(59%) middle(44%), and North 30% followed in that order 4. In the age relatated prevalence of BVD SN antibodies, the younger than 6 month old group was the highest 65.7%, and older than 25 month old group was also at 62.2%. Then, 7 to 12 moth old group and 13 to 24 month old group showed to 58.5%, 52.1% respectively. 5. The geometric mean titer (log2) of 108 cattle serum samples showing positive BVD SN antibodies was 4.3. 6. In the geometric mean titer(log2) according to age, younger than 6 month old group (5.2) was the highest, then 7 to 12 month old group 2.8(SD=1.94 standard deviation) was lowliest.

광주(光州)·전남지역내(全南地域內) 소의 바이러스성 질병(疾病)에 관한 혈청학적(血淸學的) 연구(硏究) (Seroepidemiological studies on virus-borne diseases of cattle in Kwangju and Chonnam area)

  • Sera from 304 Holsteins or Korean native cattle were collected from slaughterhouse in Kwangju area to study the infection of major virus-borne diseases. Serum antibody titers against infectious bovine rhinotracheitis virus(IBRV), bovine viral diarrhea virus(BVDV), parainfluenza type-3 virus(PI-3V), bovine ephemeral fever virus(BEFV), bovine Ibaraki virus(BIV), bovine Akabane virus(BAKV), bovine rotavirus(BRV), bovine coronavirus(BCV) were measured by serum neutralization tests. Results which obtained were as follows. Sera from 280 cattle(92.1%) contained antibodies against BRV which rate was the highest among the 8 viruses, and serum antibodies against BCV, BVDV, BIV, BAKV, BEFV, IBRV and PI-3V were detected from 266(87.5%), 149(49%), 108(35.5%), 94(30.9%), 80(26.3%), 32(10.5%) and 24(7.9%) cattle, respectively. Prevalence of seropositives to BVDV, BIV, BAKV, BEFV were higher among Holsteins than among the Korean native cattle(P<0.05). Prevalence of antibody titers against BVDV, BIV and BEFV in Korean native cattle were higher among females than males, while more males contained antibodies to BAKV, IBRV and PI-3V than females in their blood(P<0.05). Seropositives to BVDV, BIV, BAKV, BEFV and IBRV in Holsteins were higher among females than males(P<0.05). In Korean native cattle, serum antibody titers against IBRV and PI-3V ranged from 1:2~1:32 and 1:2~1:64, respectively, while serum antibody titers against the rest 6 viruses ranged from $1:2{\sim}1:{\geq}256$. In Holsteins, serum antibody titers against IBRV and PI-3V ranged from 1:2~1:64 and 1:2~1:32, respectively, while serum antibody titers against the rest 6 viruses ranged from $1:2{\sim}1:{\geq}256$.

전남지방의 소 합포체성 폐렴바이러스(Bovine Respiratory Syncytial Virus) 감염에 관한 혈청학적 연구 (Seroepidemiological Studies on Bovine Respiratory Syncytial Virus of Cattle in Chonnam Province)

  • Sera from 425 Korean native and 203 Holstein cattle were collected from October 1994 to September 1995 from dairy farms and slaughterhouses in Chonnam province to study the exposure rate to bovine respiratory syncytial virus(BRSV). Serum antibody titers against BRSV were measured by neutralization test, and results were as follows: Overall prevalence of seropositive cattle to BRSV were 74.5%, and the exposure rate to BRSV was higher among the Holstein (77.3%) than among the Korean native cattle(73.2%). The serum antibody titers against BRSV ranged from 1:2~$\geq$1:256 in both species. Among Korean native cattle, the most frequent serum antibody titer was 1:4 against BRSV(19.3%), while only 1.4% of seropositive cattle had serum titer of $\geq$1: 256. Among Holstein cattle, 22.7% of examined cattle contained serum titer of 1:8, while 1.5% of seropositive cattle showed $\geq$ 1:256. Antibody titers against BRSV were higher among males than females in both Holstein (82.1% vs. 73.1%) and Korean native (74.5% vs. 69.2%) cattle. Prevalence of seropositive cattle by age in both species were evenly distributed, although the highest number (76.9%) of seropositive were at the age of 3 in Korean native cattle, while 83.5% of seropositive Holstein cattle were of 2 years old. The lowest seropositive rate was observed in cattle of less than 1 year old(25.0%). Seasonal occurrunce of BRSV was the highest in spring season in both Holstein (86.6%) and Korean native (81.0%) cattle (P<0.05).

축우의 유행형 (지방병성) 백혈병에 관한 연구 I. 한 유우군의 우백혈병 바이러스 항체와 혈중 임파구의 변동에 관하여 (Studies on enzootic bovine leukosis I. The changes of antibodies against bovine leukemia virus and peripheral blood lymphocyte on Holstein cattle in a dairy farm)

  • To investigate bovine leukosis virus (BLV) infection in the cattle rearing in a dairy farm where a case of bovine lymphosarcoma had been identified several years ago, the 196 Holstein cattle including newborn calves to 12 years of age were tested. The BLV antibody test and peripheral lymphocyte count for bovine leukosis were carried out by the immunodiffusion (ID) test and Bendixen's Kep. These tests were performed 2 to 4 times at the interval of 3 to 5 months. The observed results were as follows: 1. The positive rates of BLV-antibody in the 1st, the 2nd, the 3rd and the 4th tests were 23.3%, 28.1%, 49.0% and 55.7%, respectively. The conversion rates from negative to positive in the 2nd, the 3rd and the 4th tests were 8.9%, 41.4%, and 20.0%, respectively. Results showed that the highest conversion rate was observed at the 3rd test which was conducted after winter. The highest positive rate by ID test were observed in 4 year old cattle in the 1st and 2nd tests, and in 2 year old herd in the 3rd and 4th tests. 2. In hematological test by Bendixen's Key, the positive and suspicious rates in the 1st, the 2nd, the 3rd and the 4th tests were 5.8 and 7.8%, 8.3 and 6.6%, 8.7 and 10.1%, 10.8 and 19.6% respectively. Results showed that the positive and. suspicious rates increased in course of time. 3. 70 to 100% of the positive cattle in hematological test were positive for BLV-antibody test. All of 13 cattle with persistent lymphocytosis (PL) were also positive for BLV-antibody, indicating the high relationship between PL and BLV-antibody. 4. The number of total leukocytes and absolute lymphocytes in the BLV-antibody positive cattle appeared significantly higher than those of the negative cattle. The markedly increased cell counts were observed in the cattle over one year old. 5. The mean of total leukocytes and absolute lymphocytes in the negative cattle for BLV-antibody increased slightly after sero-conversion into positive. 6. In the clinical examinations during experimental periods, none of the 72 positive cattle for BLV-antibody showed any lesions for bovine leukosis.

Characterization and Profiling of Liver microRNAs by RNA-sequencing in Cattle Divergently Selected for Residual Feed Intake

  • MicroRNAs (miRNAs) are short non-coding RNAs that post-transcriptionally regulate expression of mRNAs in many biological pathways. Liver plays an important role in the feed efficiency of animals and high and low efficient cattle demonstrated different gene expression profiles by microarray. Here we report comprehensive miRNAs profiles by next-gen deep sequencing in Angus cattle divergently selected for residual feed intake (RFI) and identify miRNAs related to feed efficiency in beef cattle. Two microRNA libraries were constructed from pooled RNA extracted from livers of low and high RFI cattle, and sequenced by Illumina genome analyser. In total, 23,628,103 high quality short sequence reads were obtained and more than half of these reads were matched to the bovine genome (UMD 3.1). We identified 305 known bovine miRNAs. Bta-miR-143, bta-miR-30, bta-miR-122, bta-miR-378, and bta-let-7 were the top five most abundant miRNAs families expressed in liver, representing more than 63% of expressed miRNAs. We also identified 52 homologous miRNAs and 10 novel putative bovine-specific miRNAs, based on precursor sequence and the secondary structure and utilizing the miRBase (v. 21). We compared the miRNAs profile between high and low RFI animals and ranked the most differentially expressed bovine known miRNAs. Bovine miR-143 was the most abundant miRNA in the bovine liver and comprised 20% of total expressed mapped miRNAs. The most highly expressed miRNA in liver of mice and humans, miR-122, was the third most abundant in our cattle liver samples. We also identified 10 putative novel bovine-specific miRNA candidates. Differentially expressed miRNAs between high and low RFI cattle were identified with 18 miRNAs being up-regulated and 7 other miRNAs down-regulated in low RFI cattle. Our study has identified comprehensive miRNAs expressed in bovine liver. Some of the expressed miRNAs are novel in cattle. The differentially expressed miRNAs between high and low RFI give some insights into liver miRNAs regulating physiological pathways underlying variation in this measure of feed efficiency in bovines.

Development of Bovine Specific Leptin Radioimmunoassay and Relationship of Plasma Leptin with Vitamin A and Age of Wagyu

  • Leptin is produced by adipocytes and its role in the regulation of lipid metabolism, feed intake, productive and reproductive performance of domestic animal species has been greatly stressed and extensively investigated in recent years. This study was conducted to develop a radioimmunoassay (RIA) for the estimation of plasma bovine leptin and to determine plasma leptin concentration in fattening Japanese Black cattle (Wagyu) and its crossbreds at commercial farms. Relationships of plasma leptin with plasma vitamin A and age of crossbred cattle were also determined. Recombinant bovine leptin (rbleptin) was produced by the E. coli overexpressed leptin as a GST (glutathione S-transferase)-fusion protein. Then antiserum against bovine leptin was obtained by its immunization in rabbits. Using this antiserum, a bovine specific RIA was developed and plasma leptin level was determined in 120 crossbred fattening cattle (WagyuHolstein, 50:50) at commercial farms. The plasma leptin level increased with the age of cattle and its level was greater in the crossbred heifers than in the steers. Plasma vitamin A level was negatively correlated with plasma leptin level in crossbred heifers and steers. This relationship was stronger in heifers than in steers. Plasma leptin was gradually increased with advancing age in fattening Wagyu cattle. In conclusion, development of a bovine specific RIA to estimate plasma leptin will contribute to better understanding of the role of leptin in cattle.

Identification of Bovine Lymphocyte Antigen DRB3.2 Alleles in Iranian Golpayegani Cattle by DNA Test

  • The bovine lymphocyte antigen (BoLA)-DRB3 gene encodes cell surface glycoproteins that initiate immune responses by presenting processed antigenic peptides to CD4 T helper cells. DRB3 is the most polymorphic bovine MHC class II gene which encodes the peptide-binding groove. Since different alleles favour the binding of different peptides, DRB3 has been extensively evaluated as a candidate marker for associations with various bovine diseases and immunological traits. For that reason, the genetic diversity of the bovine class II DRB3 locus was investigated by polymerase chain reaction-restriction fragment length polymorphism method (PCR-RFLP). This study describes genetic variability in the BoLA-DRB3 in Iranian Golpayegani Cattle. Iranian Golpayegani Cows (n = 50) were genotyped for bovine lymphocyte antigen (BoLA)-DRB3.2 allele by polymerase chain reaction and restriction fragment length polymorphism method. Bovine DNA was isolated from aliquots of whole blood. A two-step polymerase chain reaction followed by digestion with restriction endonucleases RsaI, HaeIII and BstYI was conducted on the DNA from Iranian Golpayegani Cattle. In the Iranian Golpayegani herd studied, we identified 19 alleles.DRB3.2${\times}$16 had the highest allelic frequency (14%), followed by DRB3.2${\times}$7 (11%). Six alleles (DRB3.2${\times}$25, ${\times}$24, ${\times}$22, ${\times}$20, ${\times}$15, ${\times}$3) had frequencies = 2%. Although additional studies are required to confirm the present findings, our results indicate that exon 2 of the BoLA-DRB3 gene is highly polymorphic in Iranian Golpayegani Cattle.
