  • Hepatic fibrosis/cirrhosis is characterized by increased production and deposition of collagen, noncollagenous glycaproteins, and proteoglycans, which mainly compose the extracellular matrix (ECM). Recently, activations of macrophages, myofibroblasts and mast cells are thaught ta be associated with the accumulation of ECM. In order to investigate the kinetics of macrophages, myofibroblasts and mast cells and the relationship between these cells and the accumulation of ECM in carban tetrachloride (CCl$_4$)-injected rat liver, we induced liver cirrhosis of rat by an injection of CCl$_4$ for 14 weeks. • This study examined the carbon dioxide ($CO_2$) pollution inside vehicles under low ventilation condition and evaluated the Air Quality System (AQS) for in-vehicle air quality using two techniques. The low ventilation condition is not recommended in order to keep oxygen-rich condition inside vehicles. Under the low ventilation condition, the in-vehicle $CO_2$ concentrations exceeded 1,000 ppm, the air qualify guidelines in the United States, Western Europe, and Japan, indicating more oxygen deficiency inside vehicles. On the contrary, with the AQS-on condition, the in-vehicle $CO_2$ concentrations were less than 1,000 ppm fer most of the driving time, indicating that the AQS could solve the problem of $CO_2$ accumulation inside vehicles under the low ventilation condition. The AQS test conducted by comparing carbon monoxide (CO) and volatile organic compound (VOC) concentrations inside two vehicles indicated that the AQS effectively decreased the in-vehicle concentrations by 21 to 36%, as compared to medium ventilation condition with the windows closed, the vent opened, and air conditioning on. In addition, The AQS test conducted by comparing the interior and exterior concentrations indicated that the AQS effectively decreased the in-vehicle concentrations by 18 to 31%, as compared to medium ventilation condition.

  • Bacterial cellulose (BC) is widely used in the food industry for products such as nata de coco. The mechanical properties of BC hydrogels, including stiffness and viscoelasticity, are determined by the hydrated fibril network. Generally, Komagataeibacter bacteria produce gluconic acids in a glucose medium, which may affect the pH, structure and mechanical properties of BC. In this work, the effect of pH buffer on the yields of Komagataeibacter hansenii strain ATCC 53582 was studied. The bacterium in a phosphate and phthalate buffer with low ionic strength produced a good BC yield (5.16 and 4.63 g/l respectively), but there was a substantial reduction in pH due to the accumulation of gluconic acid. However, the addition of gluconic acid enhanced the polymer density and mechanical properties of BC hydrogels. The effect was similar to that of the bacteria using glycerol in another carbon metabolism circuit, which provided good pH stability and a higher conversion rate of carbon. This study may broaden the understanding of how carbon sources affect BC biosynthesis.

  • In order to investigate and generalize the effects of carbon and nitrogen sources on the growth of and lipid production in Chlorella sp. 227, several nutritional combinations consisting of different carbon and nitrogen sources and concentrations were given to the media for cultivation of Chlorella sp. 227, respectively. The growth rate and lipid content were affected largely by concentration rather than by sources. The maximum specific growth was negatively affected by low concentrations of carbon and nitrogen. There is a maximum allowable inorganic carbon concentration (less than 500~1,000 mM bicarbonate) in autotrophic culture, but the maximum lipid content per gram dry cell weight (g DCW) was little affected by the concentration of inorganic carbon within the concentration. The lipid content per g DCW was increased when the microalga was cultured with the addition of glucose and bicarbonate (mixotrophic) at a fixed nitrogen concentration and with the lowest nitrogen concentration (0.2 mM), relatively. Considering that lipid contents per g DCW increased in those conditions, it suggests that a high ratio of carbon to nitrogen in culture media promotes lipid accumulation in the cells. Interestingly, a significant increase of the oleic acid amount to total fatty acids was observed in those conditions. These results showed the possibility to induce lipid production of high quality and content per g DCW by modifying the cultivation conditions.

  • This study introduced a carbon-free electrode for $Li-O_2$ cells with the aim of suppressing the side reactions activated by carbon material. Micro-particles of poly(3,4-ethylenedioxythiophene) (PEDOT), a conducting polymer, were used as the base material for the air electrode of $Li-O_2$cells. The PEDOT micro-particles were treated with $H_2SO_4$ to improve their electronic conductivity, and LiBr and CsBr were used as the redox mediators to facilitate the dissociation of there action products in the electrode and reduce the over-potential of the $Li-O_2$ cells. The capacity of the electrode employing PEDOT micro-particles was significantly enhanced via $H_2SO_4$ treatment, which is attributed to the increased electronic conductivity. The considerable capacity enhancement and relatively low over-potential of the electrode employing $H_2SO_4$-treated PEDOT micro-particles indicate that the treated PEDOT micro-particles can act as reaction sites and provide storage space for the reaction products. The cyclic performance of the electrode employing $H_2SO_4$-treated PEDOT micro-particles was superior to that of a carbon electrode. The results of the Fourier-transform infrared spectroscopic analysis showed that the accumulation of residual reaction products during cycling was significantly reduced by introducing the carbon-free electrode based on $H_2SO_4$-treated PEDOT micro-particles, compared with that of the carbon electrode. The cycle life was improved owing to the effect of the redox mediators. The refore, the use of the carbon -free electrode combined with redox mediators could realize excellent cyclic performance and low over-potential simultaneously.

  • A combined process consisted of a biofilm reactor and a continuously stirred-tank reactor (CSTR) was investigated for highly loaded ammonium wastewater treatment via nitrite accumulation. To enhance ammonium oxidizing bacteria over nitrite oxidizing bacteria on the surface of carriers, the biofilm reactor was operated at temperature of $35^{\circ}C$ for more than three months but the influent ammonium (500 mg-N/L) was partially oxidized to nitrite (240 mg-N/L). As pH was increased from 7.5 to 8.0, nitrite accumulation was fully achieved due to the inhibition of nitrite oxidizing bacteria under high free ammonia concentration. The biofilm reactor performance was severely deteriorated at the hydraulic retention time of 12 hr, at which incomplete nitrification of ammonia was observed. Various solubilization methods were applied to sewage sludge for enhancing its biodegradability and the combined method, alkaline followed by ultrasonic, gave the highest solubilization efficiency (58%); the solubilized solution was used as the external carbon source for denitrification reaction in CSTR. FISH analysis showed that the dominant microorganisms on the carriers were ammonium oxidizing bacteria such as Nitrosomonas spp. and Nitrospirar spp. but low amounts of nitrite oxidizing bacteria as Nitrobacter spp. was also detected.

  • Glycogen plays important roles in bacteria. Its structure and storage capability have received more attention recently because of the potential correlations with environmental durability and pathogenicity. However, the low level of intracellular glycogen makes extraction and structure characterization difficult, inhibiting functional studies. Bacteria grown in regular media such as lysogeny broth and tryptic soy broth do no accumulate large amounts of glycogen. Comparative analyses of bacterial media reported in literature for glycogen-related studies revealed that there was no consistency in the recipes reported. Escherichia coli $DH5{\alpha}$ is a convenient model organism for gene manipulation studies with respect to glycogen. Additionally, M9 minimal salts medium is widely used to improve glycogen accumulation, although its composition varies. In this study, we optimized the M9 medium by adjusting the concentrations of itrogen source, tryptone, carbon source, and glucose, in order to achieve a balance between the growth rate and glycogen accumulation. Our result showed that $1{\times}M9$ minimal salts medium containing 0.4% tryptone and 0.8% glucose was a well-balanced nutrient source for enhancing the growth and glycogen storage in bacteria. This result will help future investigations related to bacterial physiology in terms of glycogen function.

  • The effects of environmental factors on the extracellular release of organic carbon by Scenedesmus qudricauda were studied. The PER (percentage extracellular release) was greater at high temperature and at high concentration of nitrogen and phosphate. The PER variation according to the change of M/P ratio showed high values at each extreme N/P ratio. This result suggested that the limitation of nitrogen or phosphorous resulted in the accumulation of carbohydrates as photosynthetic products, and the products in high concentration were excreted through algal cell membrane.
