Effect of Collagen Concentration on the Viability and Metabolic Function of Encapsulated Hepatocytes

  • Chitosan/alginate capsules were formed by electrostatic interactions and had appropriated mechanical strength, permeability to albumin, and stability to hepatocytes. Rat hepatocytes were isolated and immobilized in chitosan/alginate capsules. During the encapsulation process with hepatocyte, 10% of viability was decreased mainly due to the low pH of the chitosan solution. Among various capsule fabrication methods, the chitosan-alginate capsule showed the highest mechanical strength. Addition of collagen in the capsule with hepatocytes enhanced hepatic metabolism as well as the cell viability for 2 weeks of culture. The hepatocyte in the capsule without collagen decreased the viability to 10% for 2-week cultures.

  • To separate chitosanoligosaccharides easily by size exclusion, an coencapsulating technology of substrate and enzyme was developed. Chitosan and chitosanase were enveloped in this membrane and the product released to medium by size exclusion. The lower limit of the alginate concentration and the agitation speed were 0.5% and 40 rpm, respectively. Membrane thickness and capsules diameter were $10{\mu}m$ and approx. 3.0mm, 1.5mm, respectively. The molecular weight difference by concentration and cps of alginate were of little significance. And also, the molecular weight of distribution according to enzyme concentration was low concentration of enzyme produced high molecular weight of oligosaccharides. At 1.5mm size of capsule, product diffusion rate to outer part was higher than other capsules. The molecular weight distribution of the released oligosaccharides ranged from 1000 to 6000 Da.

사자 부신의 형태학적 관찰 (Morphological study on the adrenal gland of the lion(Panthera leo))

  • The morphological and immunohistochemical characteristics of the adrenal gland of the lion(Panthera leo) have been studied. In the light microscopic level, the adrenal gland was surrounded by thick capsules arid weak development of the zona glomerulosa was detected. Some medullary tissues were directly connected with capsules, however the adrenal gland in the lion 'demonstrated somewhat pseudolobulated appearance. Weakly or strongly stained chromogranin-immunoreactive cells were detected in medullary regions of the adrenal gland of the lion by immunohistochemistry, and weakly stained chromogranin-immunoreactive cells were detected more frequent than strongly stained ones.

알긴산소다를 이용한 멸치어유의 미세캡슐화 (Microencapsulation of Anchovy Oil by Sodium Alginate)

  • Microencapsulation of anchovy oil as a core material in sodium alginate as a wall material was inves tigated. Microencapsulation was accomplished by injecting an oil/water emulsion, consisting of a mixture of liquefied sodium alginate and emulsifier, under high pressure through an orifice submerged in a calcium lactate solution. Microcapsules suspended in a dispersion fluid were observed under a fluorescence mi croscope to verify the presence of the capsules and to note coalescence or degradation of the capsules. Optimum conditions for microencapsulation of anchovy oil were obtained when 1.0% aqueous solution of sodium alginate contained 3% of a 1:1 ratio of ESPR 25(polyglycerine+polylinoleate) and TW 20(sorbitan laurate+ethylene oxide) as an emulsifier in terms of capsule size and size distribution, and emulsion stability. The airless sprayer produced microcapsules with a diameter between 15.9 and 73.9 m with different concentration of a wall material. The optimum mixing ratio of wall material to core material was 90:10(wt/wt). 0.2% calcium lactate was appropriate as a dispersion fluid.

  • The purpose of the present study was to evaluate the bioequivalence of two Erdosteine capsules, $Erdos^{TM}$(Dae Woong Pharmaceutical Co., Ltd.) and $Ersteine^{TM}$(Dae Won Pharmaceutical Co., Ltd.), according to the guidelines of Korea Food and Drug Administration (KFDA). Twenty-four normal male volunteers,23.88 ${\pm}$2.89 year in age and 68.50 ${\pm}$ 7.71 kg in body weight, were divided into two groups and radkomied 2${\times}$2 cross-over study was empolyed. Ater three capsules containing 300 mg of erdosteine per capsule were orally administered, blood was taken at predetermined time intervals and concentrations of erdostene I in plasma were determined using HPLC.   • PLA 3D printed capsule of FDM method has advantages of mass production and low cost. However, it has a different strength depending on the direction in witch it is laminated. In this paper, structural design of several capsules, FEM analysis, and Compressive strength tests were conducted. As a result, the proposed capsule has a strong load of up to 217.9% compared to general capsule without a reinforcing structure.

  • The microfluidic controlled droplet manufacturing system is one of the most powerful methods for capsule manufacturing. The microfluidic control method can control the type and size of the capsule by changing the size and configuration of the channel. In addition, by increasing the number of channels, capsules of uniform size can be mass-produced. In this paper, a capsule manufacturing system including flow-focusing and T junction method was designed. In addition, the effectiveness of this system was verified by manufacturing multi-emulsion capsules with a size of 2.2 to 3 mm.

Polydopamine-coated chitosan hydrogels for enzyme immobilization

  • To address inherent weaknesses such as low mechanical strength and limited enzyme loading capacity in conventional chitosan or alginate beads, an additional step involving the exchange of anionic surfactants with hydroxide ions was employed to prepare porous chitosan hydrogel capsules for enzyme immobilization. Consequently, excellent thermal stability and long-term storage stability were confirmed. Furthermore, coating the porous chitosan hydrogel capsules with polydopamine not only improved mechanical stability but also exhibited remarkable enzyme immobilization efficiency (97.6% for M1-D0.5). Additionally, it was demonstrated that the scope of application for chitosan hydrogel beads, prepared using conventional methods, could be further expanded by introducing an additional step of polydopamine coating. The enzyme immobilization matrix developed in this study can be selectively applied to suit specific purposes and is expected to be utilized as a support for the adsorption or covalent binding of various substances.
