  • The potential application of protoplasts useful for studies such as physiology, morphology, genetic engineering, etc has led to the development of suitable methologies for isolation and manupilation of protoplasts from a wide variety of algae (Waaland et al., 1990; Reddy and Fujita, 1991; Chen and Chiang, 1994). Protoplasts technology to seaweeds depends large on the ability to produce viable cells capable of regenerating into whole plantlets (Wakabayashi et al., 1999). Though Capsosiphon fulvescens is one of the important economic seaweeds culturing in Korea, surprisingly protoplasts approach on this species has not been reported so far. Consequently we investigated the various aspects related to the protoplasts of Capsosiphon fulvescens in this study. (omitted)

효소 가수분해를 통한 매생이 유래 β-Glucan 형태의 Oligomer 생산 및 분석 (Production and Characterization of β-Glucan Type Oligomer Produced with Enzymatic Hydrolysis of Capsosiphon fulvescens)

  • ${\beta}$-Glucan type oligomers which have angiotensin I converting enzyme (ACE) inhibitory activity were isolated and characterized from Capsosiphon fulvescens. After C. fulvescens was hydrolysis with Alcalase at $50^{\circ}C$, supernatant was harvested and separated with ultrafiltration membrane (MWCO 2 kDa). Oligomers which were less than 2 kDa of molecular weight were harvested for characterization. The nutrient composition of Alcalase hydrolysate was 89.9% carbohydrate, 4.2% protein and 5.9% sulfate. After ultrafiltration, the nutrient composition of oligomers was changed to 99.88% carbohydrate, 0.07% protein and 0.05% sulfate. The carbohydrate composition of oligomer was glucose (97.2%) and mannose (1.5%). The ACE inhibitory activities of Alcalase hydrolysate and oligomer were 72.1% and 82%, respectively. The molecular weight of oligomer was about 1 kDa. The oligomer was analyzed with FT-IR, $^1H$-NMR and methylation. The oligomers were ${\beta}$-1,3-glucans with ${\beta}$-(1,3)-linked glucose units.

미역과 매생이의 총 페놀함량 및 항산화성 (Total Phenolic Contents and Antioxidant Activities of Undaria pinnatifida and Capsosiphon fulvescens)

  • This study compared the total phenolic contents and antioxidant activities of two seaweed cultivars, Undaria pinnatifida (UP) and Capsosiphon fulvescens (CF), subjected to different drying methods. UP and CF were dried under two different conditions: vacuum drying (VD) at $20^{\circ}C$ and hot-air drying (HD) at $60^{\circ}C$. After drying, the total phenolic content, DPPH, and nitrite scavenging activities of the water extracts were determined. Total phenolic contents were 101.94 mg/100 g for UP and 171.35 mg/100 g for CF upon VD-20, and these values were significantly decreased to 67.59 mg/100 g for UP and 141.48 mg/100 g for CF upon HD-60. UP upon VD-20 and HD-60 had 46.17% and 35.20% DPPH radical scavenging activity, whereas CF upon VD-20 and HD-60 scavenged 57.73% and 35.22%, respectively. UP upon VD-20 and HD-60 had 40.36% and 40.01% nitrite scavenging activity at pH 1.2, whereas CF upon VD-20 and HD-60 scavenged 72.35% and 55.24%, respectively. Nitrite scavenging activities of UP and CF were reduced at pH 3.0.

매생이 추출물이 흰주의 간장조직과 분변 중의 콜레스테롤 함량에 미치는 영향 (Effect of the Green Seaweed Capsosiphon fulvescens Extract on the Liver Tissue and Fecal Cholesterol Content in Rats)

  • We investigated the effect of a green seaweed Capsosiphon fulvescens extract (CFE) on the serum, liver tissue, and fecal cholesterol levels in rats. Male Sprague-Dawley rats (four weeks old) were given on of three diets for four weeks: basal, high cholesterol, and CFE, The total serum and liver tissue cholesterol levels in the CFE group were significantly decreased compared to those in the cholesterol group. The CFE group showed increased amounts of feces, total fecal bile acid and dietary fiber as compared to basal and cholesterol group. Hematoxylin and eosin staining revealed fat droplets in the livers of the rats in the cholesterol group; however, a decreased number of droplets was observed in the rats fed the CFE diet. Our results suggest that CFE supplementation may improve lipid metabolism, by controling serum and liver tissue cholesterol levels, and by increasing the total amounts of bile acid, dietary fiber and cholesterol excretion in feces.

매생이 추출물의 생리 활성과 항산화 활성에 관한 연구 (A Study on Physiological Activity and Antioxidative Activity of Maesangi(Capsosiphon fulvescens) Extract)

  • Physiological activity and antioxidative activity of Maesangi(Capsosiphon fulvescens) extracts with distilled water or 95% ethanol were investigated. For the evaluation of physiological and antioxidative activities, some evaluation assay methods such as measurement of Hunter color value, chlorophyll a/b value, total phenolics, reducing power and thiobarbituric acid(TBA) value of soybean oil were used. Proximate composition and mineral contents of Masaengi were orders of crude protein>crude fiber>moisture>crude ash>crude lipid, and K>Ca>Mg>Na>P>Fe>Zn, respectively. In ethanol extract, the content of total phenolic compounds in Maesangi was determined to half times of that in Dasima(Laminaria). The reducing power of Maesangi-ehtanol extract was about 5 % of vitamin C and was lower than that of Dasima-ethanol extract. The TBA value of Maesangi-ethanol extract on soybean oil oxidation was about 47 % and 68.4 % to control in three and eight days oxidation, respectively. But TBA value difference was not observed significantly with the dosage below 5mL of ethanol extract.

매생이 분말을 첨가한 스폰지 케이크의 품질특성 (Quality Characteristics of Sponge Cake added with Mesangi(Capsosiphon Fulvescens) Powder)

  • This study was carried out to investigate the quality characteristics of sponge cake with mesangi(Capsosiphon fulvescens) powder added to give a control 0(control), 2, 4, 6 and 8% by weight per wheat flour. The crude protein was 33.37% for mesangi on a dry basis. Batter specific gravity, viscosity and baking loss increased with increasing mesangi powder content, whereas the specific loaf volume tended to decrease. Values of L and B decreased with increasing mesangi powder content while a value was minimized for the 4% mesangi powder. Hardness, cohesiveness, springiness, gumminess, and brittleness of sponge cakes with added mesangi powder content. The sensory tests indicated that aircell uniformity, firmness, sweetness and overall preference were not affected by the addition of 2 and 4% mesangi powder compared to control. Therefore, the addition of 4% mesangi powder to sponge cake was suggested as bing suitable to improve the quality.

Moisture Sorption and Thermodynamic Properties of Vacuum-Dried Capsosiphon fulvescens Powder

  • The moisture sorption isotherms of vacuum-dried edible green alga (Capsosiphon fulvescens) powders were determined at 25, 35, and $45^{\circ}C$ and water activity ($a_w$) in the range of 0.11~0.94. An inversion effect of temperature was found at high water activity (>0.75). Various mathematical models were fitted to the experimental data, and Brunauer, Emmett, and Teller model was found to be the most suitable model describing the relationship between equilibrium moisture content and water activity (<0.45). Henderson model could also provide excellent agreement between the experimental and predicted values despite of the intersection point. Net isosteric heat of adsorption decreased from 15.77 to 9.08 kJ/mol with an increase in equilibrium moisture content from 0.055 to $0.090kg\;H_2O/kg$ solids. The isokinetic temperature ($T_{\beta}$) was 434.79 K, at which all the adsorption reactions took place at the same rate. The enthalpy-entropy compensation suggested that the mechanism of the adsorption process was shown to be enthalpy-driven.

매생이(莓山苔) 추출물이 NC/Nga 마우스의 접촉성 피부염에 미치는 효과 (Effects of Capsosiphon Fulvescens Extract on Contact Dermatitis in NC/Nga Mice)

  • Objectives : The aim of this study is to report the effect of Capsosiphon fulvescens(Maesangi) extract(MSE) on contact dermatitis(CD). Methods : CD induced by Dermatophagoides farinae body(Dfb) in NC/Nga mice. 4% sodium dodecyl sulfate(SDS) $150{\mu}l$ and Dfb(10mg/ml) ointment 100mg were topically treated twice a week for four weeks. MSE 200 mg/kg was topically treated every 4weeks. Every week, we examined the skin lesions, weight of spleen and lymph node, epidermal thickness, production of Tumor necrosis factor-alpha($TNF-{\alpha}$) and interleukin-6(IL-6). Results : MSE reduced features of Dfb-induced skin lesions, weight of spleen and production of $TNF-{\alpha}$. MSE significantly reduced weight of lymph node, epidermal thickness, number of mast cells and production of IL-6. Conclusions : MSE may have a potential therapeutic effects for CD by suppressing allergic inflammation.

  • 전남 지방에서 생산되는 매생이를 채취하여 이화학적 성분을 조사하였다. 일반성분은 건물기준 탄수화물이 53.65%로 가장 많이 함유하고 있었고, 조단백질 31.76%, 회분 13.58%와 조 지방이 1.01%로 나타났다. 매생이에 주로 함유되어 있는 무기성분으로는 Na>Mg>Ca>K>P 순으로 나타났으며 Se도 검출되었다. 유리당은 총 5종의 유리당(fructose, glucose, sucrose, maltose, galactose)이 분석되었으며 유기산은 l-malic acid, citric acid, tartaric acid, D-malic acid 등 4종이 검출되었다. 매생이의 구성 아미노산은 전체적으로 고루 함유되어 있었고 그 중 glutamic acid, aspartic acid와 alanine이 전체의 36.2%를 차지하였다. 매생이에 함유된 주요 지방산으로는 palmitic acid, linolenoc acid, stearidonic acid 순이었으며 고도 불포화지방산이 전체의 58.37%를 차지하고 있었다.

  • The green marine algae, Capsosiphon fulvescens has been cultivated in south coast of southern Korea for many years on a commercial scale. This species is very popular in Korean as a food supplement because of its attractive flavor and flexcible taste. It is, therefore, necessary to isolate and utilize qualified germplasms for mass production of this economic seaweed. Several reports have been published on phycological applications of RAPDs including the characterization of interspecific genetic variation, the identification of isolates and hybrids, and the study of phylogenetic relationships. However few authors have used RAPDs to assess the genetic variability among populations of a seaweed species(van Oppen et al., 1994; Alberto et al., 1997). The present study was undertaken for characterizing the identities of Capsosiphon fulvescens populations cultivating in Korea through the analysis of PCR based random amplified polymorphic DNAs (Welsh and MacClelland, 1990; Willams et al., 1990) and chemical composition aimed to isolate the useful strains for aquaculture.
