Simultaneous Analysis of Multi-residual pesticides using GC/NPD (GC/NPD를 이용한 다성분 잔류농약의 동시분석)

  • Pesticides were extracted from samples with 70% acetone and methylene chloride in order, and then cleaned up via open-column chromatography apparatus packed with florisil, and finally analyzed simultaneously the organophosphorus pesticides using GC/NPD. Ultra-2 and Ultra-1 fused silica capillary columns were used to separate and identify the products. Recovery of most analytes from soybean sample, taken from pesticide residues well, was greater than(80%) for all except(6) analytes. This method can simultaneously determine multiple pesticides with a high degree of accuracy and precision.

  • The $\alpha$-amylase inhibitor from naked barley was purified by DEAE-cellulose, Concanavalin-A sepharose and superose 6 column chromatography, and confirmed by capillary electrophoresis. The purified $\alpha$-amylase inhibitor showed a single band of 29KD in molecular weight when estimated by the SDS-PAGE. Its purity was increased by 12-fold as compared to its crude extract, and its specific activity was found to be 336.7units/mg. The major amino acids of the $\alpha$-amylase inhibitor from naked barley was appeared to be glutamic acid, asparitic acid and arginine. The inhibitor from naked barley was glycoproteins and carbohydrate content of inhibitor was 1.0%.

  • A connection between pulse-periodical laser radiation power and stability of liquid-metal contacts between powder particles during selective laser sintering (SLS) is determined based on analysis solving the problem of stability of liquid column in the gravity and capillary forces field. On the grounds of obtained relationships the optimization of pulse-periodical laser radiation power and SLS-process duration is realized, that allows to produce voluminous powder porous materials with pre-determined physical and mechanical properties and surface geometry. Results of metallographic investigations of powder porous materials of titanium powder produced with technological regimes calculated by means of obtained relationships are given in the work

The Measurement of Soil Conditioning Effects of Organic Materials (유기물의 토양 개량 효과 측정)

  • Much attention has been given recently to solve the environmental contamination in golf courses Changing to culture practice rather than chemical practice that depends on pesticides and fertilizers is a hot issue in golf courses or grasslands. Organic soil conditioners improve soil-plant envirormental conditions rich in physical properties. In this study, measuring systems to evaluate soil conditioning effects were set up for on-site purpose. After establishing the methodology for evaluating soil conditioner effects, 2 kinds of organic conditioners were rested for examination. The systems for the methodology included a set of simulating equipment for field capacity, an impact type soil column compactor, and an infiltration-percolation system. Test results using the systems showed bulk density and infiltration rate of mixed soil were decreased at highter rates of conditioner, but total porocities were increased. Increased porocities were most capillary pore space which has a positive effect on soil water potential. The systems and methodology in this study seem to have an efficiency to measure the effects of soil conditioner on site purpose.

  • Ginseng saponins were analyzed by thermospray (TSP) LCMS method using ODS column and with acetonitrile/ammonium acetate solution. Optimal condition for TSP Lchfs was found as follows: capillary temperature: 33$0^{\circ}C$ repelled voltage: 200 V, and concentration of ammonium acetate: 0. 05 M. Panaxadiol and panaxatriol type saponins showed characteristic fragment ions. The calibration curve of ginseng saponin showed good linearity with a correlation coefficient of 0.99. Detection limits using selected ion monitoring (SIM) technique were improved by 10~200 times compared to conventional HPLCnnr detection method.

  • Volatile flavor components in powdered Katsuobushi were simultaneous trapped by steam distillation-extraction method, and these were fractionated into the neutral, the phenolic, the acidic and the basic fraction. Volatile flavor components in these fraction were analyzed by the high-resolution GC and GC-MS equipped with a fused silica capillary column. The whole steam volatile concentrate consisted of $48\%$ neutral fraction(NF), $35\%$ phenolic fraction(PF), $12\%$ acidic fraction(AF) and $5\%$ basic fraction(BF). Thirty components such as 8 hydrocarbons, 8 aldehydes, 6 furans, 5 alcohols and 3 ketones were identified from NF. And sixteen components such as phenol, guaiacol, dimethoxy phenol, eugenol in PF, twelve components such as propionic, butanoic, isopentanoic, n-hexanoic, heptanoic, octanoic acid in AF, ten components such as 2,6-dimethylpyrazine, 2-nethylpyridine, 2,4-dimethylthiaBole in BF were identified. NF and PF gave a much higher yield than others and were assumed to be indispensable for the reproduction of aroma of powdered Katsuobushi. It was also identified eight components of volatile carbonyl compounds such as ethanal, propanal, butanal, pentanal by 2,4-DNPH method.

  • A rapid gas chromatographic (GC) profiling method for the simultaneous analysis of volatile and nonvolatile organic acids was applied to alcoholic beverages (white wine, red wine, brandy, and beer). It involves the solid-phase extraction of organic acids using Chromosorb P as the sorbent and diethyl ether as the eluent with subsequent triethylamine treatment. The resulting triethylammonium salts of acids were directly converted to volatile tert.-butyldimethylsilyl derivatives, which were analyzed by dual-capillary column GC and GC-mass spectrometry. From the alcoholic beverages studied, more than 29 organic acids were detected. When the simplified retention infer (RI) spectra of organic acids, and the direct comparisor method between alcoholic beverages and a test sample were attempted to identify a test sample, it was quickly recognized to be a red wine with the 998 ppt match quality value.

Volatile Flavor Components in Chinese Quince Fruits, Chaenomeles sinensis koehne (모과의 휘발성 Flavor 성분에 관한 연구)

  • Volatile flavor components in the Chinese quince fruits were trapped by simultaneous steam distillation-extraction method, and these were fractionated into the neutral, the basic, the phenolic and the acidic fraction. In the identification of carboxylic acids, the acidic fraction was methylated with diazomethane. Volatile flavor components in these fractions were analyzed by the high-resolution GC and GC-MS equipped with a fused silica capillary column. The total of one hundred and forty-five compounds from the steam volatile concentrate of the Chinese quince fruits were identified: they were 3 aliphatic hydrocarbons, 1 cyclic hydrocarbon, 4 aromatic hydrocarbons, 9 terpene hydrocarbons, 17 alcohols, 3 terpene alcohols, 6 phenols, 21 aldehydes, 7 ketones, 28 esters, 27 acids, 3 furans, 2 thiazoles, 2 acetals, 3 lactones and 9 miscellaneous ones. The greater part of the components except for carboxylic acids were identified from the neutral fraction. The neutral fraction gave a much higher yield than others and was assumed to be indispensable for the reproduction of the aroma of the Chinese quince fruits in a sensory evaluation. According to the results of the GC-sniff evaluation, 1-hexanal, cis-3-hexenal, trans-2-hexenal, 2-methyl-2-hepten-6-one, 1-hexanol, cis-3-hexenol, trans, trans-2, 4-hexadienal and trans-2-hexenol were considered to be the key compounds of grassy odor. On the other hand, esters seemed to be the main constituents of a fruity aroma in the Chinese quince fruits.

Development of Portable Preconcentration-Gas Chromatography System for Fast Analysis of Trace Benzene, Toluene and Xylene in Air (대기 중 극미량의 벤젠, 톨루엔 및 자일렌의 신속한 분석을 위한 휴대용 농축-기체 크로마토크래피 시스템 개발)

  • An automated on-line portable preconcentration-short column gas chromatograph was developed, which used preconcentrator using adsorption tube with Tenax-GR and Curie-point heating. The developed system operated with 3 steps of processing, preconcentration, thermal desorption, and analysis and cleaning, and could continued operating within 1~2 min cycle. The recoveries of preconcentrator for toluene was ranged between $94.7{\pm}6.6%$ and $103.8{\pm}3.1%$ with less than 7% of RSD. For benzene, toluene and xylene(BTX) standard gas test, IDL was 41, 49, $472ng/m^3$ benzene, toluene and o-xylene, respectively. The BTX mixture was analyzed within 30 sec with baseline separation by the system equipped with 4 m long capillary column. The deficiency of separation power caused by short column was solved by the control of sample injection volume and inlet/outlet pressure ratio. The automated portable preconcentration-short column gas chromatograph system was found to be useful for the continuous air monitoring of BTX at ppb levels in ambient air.

Study on Antimicrobial Activity and Analysis of Essential Oil Components of Cinnamomum cassia and Prunellae Herba (육계 및 하고초의 향기성분 분석과 항균 활성 연구)

  • The essential oil obtained by steam distillation from medicinal plants of Cinnamomum cassia and Prunellae Herba. Analysis of essential oils were performed on GC/MS selective detector. Separations were performed fused silica capillary column. The carrier gas was ultra pure helium with a flow of 1 $m{\ell}/min$ and the splitless injector temperature was set as $280^{\circ}C$. The column temperature program was as follows: initial temperature of $70^{\circ}C$ for 4 min, and increased by $2^{\circ}C/min$ 70 to $100^{\circ}C$ (held 2 min), After that the temperature was varied from 100 to $200^{\circ}C$ at $5^{\circ}C/min$ (held 20 min), increase to $280^{\circ}C$ (held 5 min) at $10^{\circ}C/min$, in a total run time of 73 min. Ten volatile flavor components were identified from C. cassia and ten volatile flavor components were identified from Prunellae Herba. Strong inhibition of growth of Vibrio parahaemolyticus was obtained with all doses of C. cassia tested. Moreover, antimicrobial activity of C. cassia occurred in a dose dependant manner.
