A Review on Studies of Marijuana for Alzheimer's Disease - Focusing on CBD, THC

  • Objectives: This study was to discuss the research trend of dementia treatment using cannabis for the purpose of providing the basis of cannabis use for medical purposes in the future. Methods: This study searched publications, which were registered to databases or published by Aug 22, 2019, and targeted the full-text or abstracts of these publications. We selected the final nine studies met all selection criteria. Results: These results implied that the CBD components of cannabis might be useful to treat and prevent AD because CBD components could suppress the main causal factors of AD. Moreover, it was suggested that using CBD and THC together could be more useful than using CBD or THC alone. Conclusion: We hope that there will be a solid foundation to use cannabis for medical use by continuously evaluating the possibility of using cannabis for clinical purposes as a dementia treatment substance and cannabis can be used as a positive tool.

Cannabinol Synthase Gene Based Molecular Markers for Identification of Drug and Fiber Type Cannabis sativa (마약성과 비마약성 대마 품종의 식별을 위한 카나비놀 생합성 유전자 분석법)

  • Cannabis sativa is an important industrial plant utilized to produce fiber, oil, and medicinal ingredients. A chemotype of cannabis is divided into "Drug type" with predominance of tetrahydrocannabinolic acid (THCA) and "Fiber type" with cannabidiolic acid (CBDA). To develop molecular markers for the discrimination of these two types, nucleotide sequences of THCA synthase and CBDA synthase as well as their pseudogenes were retrieved from the recently published cannabis genome in chromosome scale. Gene-specific SNPs were discovered by multiple alignment of these sequences, and 2 dominant marker sets from each gene were designed for selective amplification. Our markers successfully identified "Drug type" and "Fiber type" cannabis plants as well as forensic samples including processed materials. Our molecular markers will provide a fast and efficient system for molecular-based identification of the cannabis plant.

  • With the recent trend towards the legalization and relaxation of regulations surrounding medical cannabis, the diverse benefits of cannabis extracts are being scientifically validated, positioning them as promising next-generation ingredients across various fields. Compounds extracted from cannabis, such as cannabinoid (CB), cannabidiol (CBD), phytocannabinoid (pCB), and hemp seed hexane extracts (HSHE), have been shown to reduce skin inflammation, improve moisture retention exhibit strong antioxidant effects, and improve various skin conditions. In this paper, we review the effects of hemp extracts on the skin and explore their potential role as cosmetic ingredients.

  • 대마는 인류가 이용해 온 가장 오래된 약제 중 하나로 그 원산지는 중앙아시아와 남아시아이다. 식물분류학적으로 대마속 일년생 식물로서 Cannabis sativa, Cannabis indica, Cannabis ruderalis 3종이 있으며, 우리나라에서 재배되고 있는 종은 Cannabis sativa이다. 대마 재배의 역사는 인류의 시작과 그 궤를 같이하며, 동 서양을 막론하고 고대 문명에서 대마에 대한 기록을 쉽게 찾아볼 수 있다. 기록에 의하면 병의 치료나 심리적 치유 및 신에게 제사를 올릴 때 제사장이 사용한 것으로 알려져 있다. 대마의 약효에 대하여는 B.C. 2737년 중국의 신농황제시대의 기록에 관절염과 통증등 의료목적으로 사용했던 최초의 기록이 있으며 본초강목과 동의보감에 저술되어 있다. 우리나라의 대마에 관한 문헌 기록은 삼국지 '위지동이전', 삼국사기 '동성왕편'과 삼국유사에 삼베를 사용한 기록이 있는 것으로 미루어 봤을 때 대마재배의 역사는 삼국시대 이전으로 볼 수 있다. 우리 민족은 생활 속에서 대마를 즐겨 사용하였으며 삼베로 의복과 멍석, 행주 그리고 칠공예품이나 신발등을 만들어 사용하였으며, 죽음에 이르러 삼베옷을 수의로 사용하였다. 대마의 용도는 뿌리, 줄기, 잎, 꽃대 그리고 씨앗까지 다양하게 이용된다. 전통적으로 줄기의 껍질을 이용한 섬유제품이 있으며 실, 의복 및 밧줄등이 있다. 대마 줄기의 속대는 종이, 건축자재, 연료로 사용된다. 씨앗의 경우 식품과 조류의 먹이, 생약으로 이용되고 씨앗의 기름은 연료, 화장품, 맛사지 오일등으로 사용되고 있다. 환각성분이 있어 마리화나 원료로 사용되는 꽃대와 잎은 의약품의 원료로 주목받고 있다. 대마에 관한 최초의 논문은 1843년에 Cannabis indica의 약효에 관한 것으로 보고되었다. 1850년부터 1937년까지 미국의 약전은 대마를 100가지 이상의 질병에 효과가 있는 주요 의약품으로 기재하고 있다. 세계적으로 여러 가지 이유로 대마를 의료 응용과 연구 및 사용을 제한하여 대마에 관한 연구가 침체되었다. 대마의 의학연구는 대마의 약효성분인 칸나비노이드의 발견과 그 구조 및 약효에 관한 연구가 시작되면서 1960년대부터 증가하였으며 2000년 이후에는 칸나비노이드 및 칸나비디올의 다양한 의학적 효과가 밝혀지면서 급격히 증가하고 있다. 대마에 포함된 성분의 의학적 효과가 입증되면서 대마 사용을 합법화한 국가가 증가하면서 대마 산업이 급부상하고 있으며, 의료용뿐만 아니라 기호용, 식품용, 그리고 주류 및 음료시장까지 확대되고 있다. 우리나라도 2019년 3월 질병 치료 목적 대마성분 의약품을 제한적으로 허용하는 마약류 관리에 관한 법률 일부 개정안이 시행되면서 의료용 대마에 관한 연구와 산업화에 관심이 증가하는 추세이다.

Study on the anti-inflammatory effects of Cannabis sativa L. seed oil complex (햄프(Cannabis sativa L.)씨드오일 복합물의 항염증 효과에 관한 연구)

  • This study evaluated the potential of hemp seed oil (Cannabis sativa L. seed oil, CSO) and hemp seed oil complex (Cannabis sativa L. seed oil complex, CSOC) as an anti-inflammatory material through comparative analysis. Anti-inflammatory effects of CSO and CSOC were confirmed through lipopolysaccharide (LPS)-induced RAW264.7 model. As a result of confirming the inhibition of lipid oxidation through lipoxygenase inhibitory activity, CSO was not inhibited, but COSC was inhibited by more than 70%. As a result of confirming cytotoxicity through MTT analysis, CSO did not show cytotoxicity, but CSOC showed cytotoxicity at over 200 ㎍/ml. In LPS-induced RAW264.7, the expression of inducible nitric oxide synthase (iNOS) and the production of nitric oxide (NO) were significantly inhibited by CSOC compared to CSO. Additionally, CSOC significantly inhibited the expression of cyclooxygenase (COX)-2 and the production of prostaglandin E2 (PGE2). Through this study, we confirmed that CSOC has superior anti-inflammatory effects than CSO and has the potential to be used as an anti-inflammatory material.

Immunochemical Localization of Tetrahydrocannabinol (THC) in Chemically Fixed Glandular Thrichomes of Cannabis (Cannabaceae)

  • Monoclonal antibody for delta-9-tetrahydrocannabiol (THC Ab), conjugated with protein A-gold, was employed as a probe to detect THC localization in the gland and subjacent cells of chemically fixed bracts of Cannabis. THC was detected in the outer wall of the disc cells, fibrillar matrix, the surface feature of secretory vesicles, and sheath throughout development of the secretory cavity. The probe was absent from vesicles. Label was also present in anticlinal walls of disc cells and walls of dermal and mesophyll cells. Little or no THC Ab was present in disc cells and none were detected in control tissues. This distribution pattern of THC Ab was similar to that in tissues prepared by high pressure cryofixation-cryosubstitution. Consistent association of THC with wall and wall-derived materials suggests that cannnabinoids are synthesized outside the plasma membrane and bound to a wall component, where-upon they are transported to the cavity with wall materials released from the disc cell wall during development of the secretory cavity.

  • More than a year has already passed since the Gyeongbuk Hemp Regulation Free Zone Project was implemented, and it is time to supplement and review the deficiencies in production history management for the domestic hemp industry after the Free Regulatory Zone. In this study, the current status and characteristics of cannabis history management in the Gyeongbuk Hemp Regulation Free Zone project were investigated, and problems with current history management were derived using service design tools such as pilot interviews, shadowing, and Customer journey map. The case of the cannabis production system and the CTS recommended for the legal cannabis market in Oregon, a leading country, was compared and analyzed, and the direction was to secure safety through production cycle history management suitable for growth characteristics, efficient technology application for real-time management, and history information sharing. Through this study, it is expected that it will help improve the current hemp production history management system and properly settle the history management system of domestic hemp industrialization in the future.

Comparative Morphology of Plastids on Vegitative Tissue of Cannabis sativa L. (대마(Cannabis sativa L.) 영양조직의 색소체 비교)

  • Various plastids in leaf and bracteal tissues of Cannabis sativa L. were examined by electron microscopy. Young chloroplasts without starch grain in mesophyll cells were ellipsoidal, and osmiophilic globules within them were common in stroma. During the plastid differentiation, the mature chloroplasts in mesophyll were changed in shape depending on the numbers and sizes of starch grain in stroma. Electron-dense granular substances were occurred along the outer membrane of chloroplasts in mesophyll. Typical plastids with reticulate body were present in the glandular trichomes. Electron-grey material appeared along the surface of a plastid. A light area in reticulate body is considered to represent junction point of thylakoids.

Efficacy of cannabis-based medications compared to placebo for the treatment of chronic neuropathic pain: a systematic review with meta-analysis

  • Background: Chronic neuropathic pain (NP) presents therapeutic challenges. Interest in the use of cannabis-based medications has outpaced the knowledge of its efficacy and safety in treating NP. The objective of this review was to evaluate the effectiveness of cannabis-based medications in individuals with chronic NP. Methods: Randomized placebo-controlled trials using tetrahydrocannabinol (THC), cannabidiol (CBD), cannabidivarin (CBDV), or synthetic cannabinoids for NP treatment were included. The MEDLINE, Cochrane Library, EMBASE, and Web of Science databases were examined. The primary outcome was the NP intensity. The risk of bias analysis was based on the Cochrane handbook. Results: The search of databases up to 2/1/2021 yielded 379 records with 17 RCTs included (861 patients with NP). Meta-analysis showed that there was a significant reduction in pain intensity for THC/CBD by -6.624 units (P < .001), THC by -8.681 units (P < .001), and dronabinol by -6.0 units (P = .008) compared to placebo on a 0-100 scale. CBD, CBDV, and CT-3 showed no significant differences. Patients taking THC/CBD were 1.756 times more likely to achieve a 30% reduction in pain (P = .008) and 1.422 times more likely to achieve a 50% reduction (P = .37) than placebo. Patients receiving THC had a 21% higher improvement in pain intensity (P = .005) and were 1.855 times more likely to achieve a 30% reduction in pain than placebo (P < .001). Conclusion: Although THC and THC/CBD interventions provided a significant improvement in pain intensity and were more likely to provide a 30% reduction in pain, the evidence was of moderate-to-low quality. Further research is needed for CBD, dronabinol, CT-3, and CBDV.

Characterization of Three Fusarium spp. Causing Wilt Disease of Cannabis sativa L. in Korea

  • In July 2021, wilting symptoms were observed in adult and seedling hemp (Cannabis sativa L. cv. Cherry Blossom) plants grown in a greenhouse. As the disease progressed, yellowing and wilting symptoms on the leaves developed, resulting in whole plant death. In seedling plants, typical damping-off symptoms were observed. To identify the pathogen, the roots of diseased plants were sampled, surface sterilized, and cultured on potato dextrose agar (PDA) media. From the culture, 4 different fungal isolates were recovered and purely cultured. Each fungal isolate showed distinct growth shapes and color development on malt extract agar, oatmeal agar, sabouraud dextrose agar, and PDA media. Microscopic observation and molecular identification using ribosomal DNA internal transcribed spacer sequencing identified them as 3 Fusarium spp. and 1 Thielaviopsis paradoxa. Additional sequencing of elongation factor 1-alpha and b-tubulin regions of 3 Fusarium spp. revealed that 2 of them are Fusarium solani, and the other one is Fusarium proliferatum. To examine which isolate can act as a causal agent of wilt disease of hemp, each isolate was tested for their pathogenicity. In the pathogenicity test, F. solani AMCF1 and AMCF2, and F. proliferatum AMCF3, but not T. paradoxa AMCF4, were able to cause wilting disease in hemp seedlings. Therefore, we report that F. solani AMCF1 and AMCF2, and F. proliferatum AMCF3 as causal agents of Fusarium wilt of hemp plants. To our knowledge, this is the first report of the wilt disease of C. sativa L. caused by Fusarium spp. in Korea.
