Bioaccumulation Patterns and Responses of Fleece-flower; Persicaria thunbergii to Cadmium and Lead

  • Application of phytoremediation in the polluted area to remove undesirable materials is a complex and difficult subject without detailed investigation and experimentation. We investigated the accumulation patterns of cadmium and lead in plants naturally grown, the bioavailability of plants to accumulate these toxic metals and the responses of P. thunbergii to cadmium and lead. The soil samples contained detectable lead (<17.5$\mu\textrm{g}$/g), whereas cadmium was not detected in the soils of study area. The whole body of Persicaria thunbergii contained detectable lead (<320.8$\mu\textrm{g}$/g) but cadmium was detected only in the stem (<7.4$\mu\textrm{g}$/g) and root (<10.4$\mu\textrm{g}$/g) of P. thunbergii. Cadmium was not detected in Trapa japonica and Nymphoides peltata, whereas lead was detected in T. japonica (<323.7$\mu\textrm{g}$/g) and N. peltata (<177.5$\mu\textrm{g}$/g). Correlation coefficient between lead content in soil and in these plant samples represented positive correlation. The total content of lead in each plant sample increased in the order of N.peltata$\leq$P.thunbergii

Hepatocyte protection and antioxidant effect of Citri Unshius Pericarpium against cadmium-induced oxidative stress (카드뮴으로 유발된 산화 스트레스에 대한 진피의 간세포 보호 및 항산화 효과)

  • Objective : Citri Unshius Pericarpium is the dried peel of mature fruit of Citrus unshiu Markovich and has been used to treat indigestion, vomiting, and removal of phlegm. This study investigated the hepatoprotective and antioxidant effect of CEE (Ethanol extract of Citri Unshius Pericarpium) in cadmium (CdCl2)-treated HepG2 cells. Methods : Component analysis of Citri Unshius Pericarpium was analyzed by UPLC with C18 column. Cell viability was determined by MTT assay. The enzyme activity of superoxide dismutase (SOD) and the level of reactive oxygen species (ROS) and reduced glutathione (GSH) were analyzed using commercially available kits. Results : Cadmium caused severe HepG2 cell death. Cadmium also increased ROS production, consistent with depletion of GSH and inhibition of the SOD enzyme. However, CEE treatment reduced cell death and relieved oxidative stress caused by cadmium toxicity. CEE lowered ROS levels and improved depletion of GSH levels. CEE also enhanced the enzymatic activity of SOD. In component analysis, hesperidin was the most abundant of the five marker compounds (Narigenin, Narigin, Narirutin, Hesperidin and Hesperidin), which assumes that hesperidin partially contributed to the antioxidant activity of CEE. Conclusion : These results suggested that CEE could be a potential substance to solve heavy metal-related health problems. In particular, inhibition of oxidative stress by CEE can be a way to treat liver damage caused by cadmium.

Method and proficiency test for analysis of toxic metals in livestock products by ICP/MS (ICP/MS를 이용한 축산물의 유해중금속 분석방법 및 숙련도 검증)

  • The analytical method of trace toxic metals in livestock products was confirmed and validated through certified reference material (CRM) and the international proficiency tests. There are some difficulties to determine low levels of toxic metals in livestock products because of interferences due to the matrix. The recoveries of CRM (NIST 1577c) ranged from 73.9 to 119% for lead and from 86.4 to 111% for cadmium in bovine liver. The international proficiency tests were carried out with the milk powder and cocoa powder samples including metals provided by Food Analysis Performance Assessment Scheme (FAPAS$^{(R)}$, UK). The test samples were prepared by microwave digestion using solution of $HNO_3:H_2O_2:H_2O$ (v/v/v = 5 : 2 : 4) and analyzed by ICP/MS. The analytical result of cadmium in milk powder was $121{\mu}g/kg$ with -0.3 of the z-score compared to the assigned value of $131{\mu}g/kg$ by FAPAS$^{(R)}$. The analytical results of lead and cadmium in cocoa powder were $29.2{\mu}g/kg$ and $97.6{\mu}g/kg$, respectively, which satisfied the assigned values of $34.2{\mu}g/kg$ for lead and $126{\mu}g/kg$ for cadmium by FAPAS$^{(R)}$. It is verified that the analytical method is accurate and reliable to determine trace lead and cadmium in livestock products by microwave digestion and ICP/MS.

Simultaneous Determination of Mercury, Bromine, Arsenic and Cadmium in Biological Materials by Neutron Activation Analysis

  • A method for the simultaneous determination of mercury, bromine, arsenic and cadmium in biological samples is described. Following neutron activation and a simple distillation of volatile compounds, mercury and bromine were determined by gamma-ray spectrometry. Arsenic and cadmium were further separated by cation exchange separation and determined similarly. Determination limits for mercury, bromine, arsenic and cadmium were 0.001$\mu\textrm{g}$, 0.003$\mu\textrm{g}$, 0.001$\mu\textrm{g}$ and 0.02$\mu\textrm{g}$, respectively. The method has been applied to the determination of mercury, bromine, arsenic and cadmium in rice and fish samples. Analysis of a standard kale powder yielded the values of 0.046$\mu\textrm{g}$/g for mercury, 24.5$\mu\textrm{g}$/g for bromine 0.17 $\mu\textrm{g}$/g for arsenic and 0.50$\mu\textrm{g}$/g for cadmium.

Synthesis and Characterization of ${\omega}-Sulfonated$ Polystyrene-stabilized Cadmium Sulfide Nanoclusters

  • We report an important and useful method for preparing ${\omega}-sulfonated$ polystyrene-stabilized cadmium sulfide (CdS) nanoclusters. The ${\omega}-sulfonated$ polystyrene $(M_n\;=\;5000\;g/mol)$ was prepared successfully through chain-end sulfonation of poly(styryl)lithium using 1,3-propanesultone; the resulting polymer was used successfully as a polymeric stabilizing agent for the preparation of semiconductor CdS nanoclusters by reduction of cadmium acetate in a mixture of toluene and methanol (9:1, v/v). The nanoclusters that formed were characterized by a combination of transmission electron microscopy, X-ray diffraction, and UVN is spectroscopic analysis. The ${\omega}-sulfonated$ polystyrene-stabilized CdS nanoclusters synthesized in this study exhibited the cubic phase (zinc-blende phase) structure in the range of 2-8 nm.

The analysis of metallothionein of acute cadmium poisoning - animal study - (카드뮴중독자 검색을 위한 메탈로치오네인 분석 -동물실험을 중심으로-)

  • The purpose of this study was to investigate the metallothionein of acute cadmium poisoning mice as a Cadmium index. Forty male ICR mice were injected with cadmium chloride solution from 1/8LD50 to 1/2LD50 dose. At 24 hours after exposed Cd, I examined Cd and metallothionein (MT) intissues(liver and kidney) and fluids(whole blood and urine) and also measured low molecular proteins, N-acetyl-D-glucosaminidase(NAG) and 2- microglobuline (2-MG) in urine. The concentration of Cd and MT of liver kidney whole blood and urine were increased with dose dependent manner. Urinary Cd and urinary MT and very good significance (p<0.01) and urinary MT had good significance with kidney Cd and NAG but not 2-MG Conclusionally MT in urine was very correlated with kidney Cd and urine Cd, So MT maybe useful as a Cd poisoning index.

  • A cadimium-resistant strain isolated from Anyang stream, Azomonas agilis PY101 exhibited strong resistance to 1000 ppm of cadmium ion $(Cd^{2+})$. A agilis PY101 also exhibited resistance to various antibiotics, such as amoxicillin, amplicillin, bacitracin, cefazolin, erythromycin, penicillin, tetracycline, and vancomycin. In the presence of $Cd^{2+}$, the growth of A. agilis PY101 started after an extended lag phase and produced a green-fluorescent pigment induced by cadmium. The dramatic decrease (approximately 400ppm) of concentration of $cd^{2+}$ in the culture medium during the growth phase of A. agilis PY101 was confirmed by the inductively coupled plasma-atomic emission spectrophotometer. Transmission electron microscopic analysis revealed that A. agilis PY 101 actively accumulated $Cd^{2+}$ in the cytoplasm.

  • In this study, we investigated the effect of cadmium on genes expression related to zinc homeostasis in HT22 hippocampal neuron cell line by RT -PCR and western blotting technics. In the time-course effect, cadmium up-regulated the relative levels of MT -I and MT -II to~b-actin at 4 hr after treatment. These effects were consistent with MT -I/II protein contents by western blot analysis. But MT -III, a specific MT isoform in brain, was not affected by cadmium. (omitted)

  • Cadmium is an environmental pollutant and exhibits nephrotoxicity, hepatotoxicity and immunotoxicity. Recently, cadmium was found to induce DNA fragmentation, a biochemical hallmark of apoptosis, in cultured renal cells, hepatocytes and neuroblastoma cell. Therefore, the various toxicities of cadmium are thought to be caused by the induction of apoptosis. Lipids-derived pro-apoptotic ceramide has emerged as an important intracellular signaling molecule that mediates diverse cellular effects, of which programmed cell death, or apoptosis, has attracted significant interest.
