Prostaglandin A2-induced Apoptosis is Not Inhibited by Heme Oygenase-1 in U2OS Cells (U2OS 세포에서 prostaglandin A2에 의한 apoptosis는 heme oxygenase-1에 의하여 저해되지 않는다)

  • Prostaglandin $A_2$ ($PGA_2$), one of cyclopentenone PGs, induced both apoptosis and heme oxygenase (HO)-1 expression in U2OS cells. $PGA_2$-induced apoptosis was not perturbed by either over-expression or knock-down of HO-1, whereas $H_2O_2$-induced cell death was inversely modulated by the expression level of HO-1. In addition, N-acetyl-L-cysteine (NAC), a thiol antioxidant, blocked both apoptosis and HO-1 expression induced by $PGA_2$. But, non-thiol antioxidants like butylated hydorxyanisole (BHA) and ascorbic acid did not block either apoptosis or HO-1-induction. Taken together, these results suggest that $PGA_2$ induces both apoptosis and HO-1 expression, which are critically related to the thiol- reactivity of $PGA_2$, but not oxidative stress, and HO-1 expression may be independent or functionally located downstream of apoptosis by $PGA_2$ without contribution to apoptosis progression.

Investigation of Antioxidative Activity and Stability of Ethanol Extracts of Licorice Root (Glycyrrhiza glabra) (감초의 에탄올 추출물의 항산화 활성 및 안정성 조사)

  • This study was carried out to optimize the conditions for the extraction of antioxidative materials from licorice root, Glycyrrhiz glabra. Chipped licorice roots were extracted with several solvents and their antioxidative activities were tested to determine the optimal extraction solvent. Among the solvents tested, 95% ethanol gave the highest free radical scavenging activity, and was therefore chosen as the optimal extracting solvent. The optimum extraction temperature and time were $20^{\circ}C$ and 12 hr, respectively. Next, the free radical scavenging activity of the ethanol extract was compared with that of other known antioxidants such as ${\alpha}-tocopherol$, butylated hydroxytoluene (BHT) and butylated hydroxyanisole (BHA). Ethanol extract of licorice root had greater antioxidative activity than ${\alpha}-tocopherol$ and a similar level to that of the two synthetic antioxidants (BHA and BHT). Moreover, the antioxidative activity of the ethanol extract was inhibited neither by heat treatment at $180^{\circ}C$ for 30 min nor by treatment at extreme pH. These findings suggest that ethanol extract of G. glabra may be useful as a natural antioxidant.

Polyphenol and Anti-oxidant Effects of Kalopanax septemlobus Koidz. Leaf Extracts (음나무(Kalopanax septemlobus Koidz.) 잎 추출물의 폴리페놀 및 항산화효과)

  • The aim of the study was to assess the cosmeceutical activity of Kalopanax septemlobus leaf and it is possible that can be used as a cosmetic ingredient for applicaiton of cosmetic industries. The concentration of total phenolic compound of hot water and 70% EtOH extracts of K. septemlobus leaf showed 104 mg/L and 125 mg/L respectively. In the result of DPPH(1,1- diphenyl-2-picryl -hydrazyl) scavenging radical activity, 70% EtOH extracts of K. septemlobus leaf showed 93.1% and it was similar to BHA(butylated hydroxyanisole) effect at 1,000ppm concentration. Xanthine oxidase inhibition of hot water extracts and 70% EtOH extracts of K. septemlobus leaf were 46.6% and 60.4% at 1,000ppm, respectively. In these results, K. septemlobus leaf has a great potential as a cosmeceutical ingredient with a natural anti-oxidant source.

Comparison of Flavonoid Content and Antioxidant Activities of Peel Extracts from Gardenia jasminoides Ellis by Various Solvents (치자(Gardenia jasminoides Ellis) 과피의 용매별 추출물의 Flavonoid 함량 및 항산화 활성 비교)

  • The purpose of this study was to measure the bioactivity and antioxidant activity of peel from Gardenia jasminoides fructus Ellis (GJE) in Namhae, Korea, following some established methods. CM (Chloroform:Methanol, 2:1, v/v), 70% ethanol, and n-butanol extracts were collected. Flavonoid content and value as a functional food ingredient of GJE peel was investigated through assessing antioxidant [DPPH (1,1'-diphenyl-2-picrylhydrazyl), ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid)], and hydroxyl radical scavenging activities; superoxide dismutase like ability; ferrous ion-chelating capacity; and tannin content by solvent extraction. Solvent extract antioxidant activities significantly increased (p<0.05) at increasing concentrations (0.2, 0.4, 0.6 mg/mL). GJE peel extracts were less active than the positive control [ascorbic acid, BHA (butylated hydroxyanisole), and EDTA (ethylenediaminetetraacetic acid disodium salt dihydrate)]. Based on the results of this study, GJE peel could be used as a natural antioxidant source due to its high antioxidant activity and bioactive compound content.

Effect of Various Washing Methods on the Quality of Semi-Dried Pacific Saury Cololabis saira Guamegi (꽁치(Cololabis saira) 과메기의 품질에 세척수가 미치는 영향)

  • We investigated the effects of various washing methods on the quality of semi-dried Pacific saury Cololabis saira, known in Korea as Guamegi. We immersed samples for 5 min in seawater, tap water, chlorinated water (100 mg/kg), ammonia water (100 mg/kg), citric acid (0.1 M), butylated hydroxyanisole (BHA, 0.2 g/kg), or sesame oil (5 g/100 g), and then dried them in the sun for 3 days. The moisture and crude lipid contents after drying were 26.62-32.49 g/100 g and 26.40-33.01 g/100 g, respectively. The moisture content significantly decreased while the crude lipid content increased during drying. The different washing methods did not have a significant effect on the acidity, peroxide values, or levels of thiobarbituric acid or biogenic amine in Guamegi. The lightness of Guamegi during drying significantly decreased, but this decrease was not significantly different among washing methods. The degree of acceptance in a sensory evaluation was higher for Guamegi treated with sesame oil. Our results suggest that these washing methods should not be used to inhibit lipid oxidation, biogenic amine formation, or color changes in semi-dried Pacific saury, because effective components are extracted by the lipids during drying.

  • Objectives : This study was carried out to investigate the inhibitive effect of a combined-herb prescription of Dansam-san (DSS) on formation of foam cells and cytokine. Methods : Experimental formation of foam cells was induced on macrophage RAW 264.7 with ox-LDL. The effect of DSS extract was observed by measuring the changes of CD36, $PPAR-{\Gamma}$, MMP-9, iNOS expression and changes of formation level of foam cells after treating experimentally induced foam cells with DSS extract. Then the antioxidative effect of DSS extract was compared with butylated hydroxyanisole (BHA). Result and Conclusions : Results obtained are as follows: 1. DSS extract showed significant antioxidative effect at 8 mg/ml or more. 2. DSS extract inhibited the formation of foam cells. 3. DSS extract inhibited the creation and revelation of conversion-related material about foam cells. 4. DSS extract prohibited the increase of smooth muscle of vessels.

The In Vitro Antioxidant Properties of Chinese Highland Lichens

  • The antioxidant properties of 46 lichen species, collected from the highly UV-exposed alpine areas of southwestern China, were evaluated for their potential therapeutic utilization. The anti-linoleic acid peroxidation activity, 1,1-diphenyl-2-picryl-hydrazyl (DPPH) scavenging activity, reducing power, and total phenolic contents were all assessed in vitro in the methanol extract of the lichens. A potent reducing power was detected in a number of the lichen extracts, when compared with butylated hydroxyanisole (BHA). In general, it was found that many of the lichens, with antioxidant properties, contained large quantities of phenolic content. Extracts of Peltigera praetextata and Sticta nylanderiana were found to exhibit the most potent activity in all of the antioxidant tests. In particular, extracts of S. nylanderiana displayed a 1.37 times greater anti-linoleic acid peroxidation activity, when compared with the ascorbic acid used as the positive control. S. nylanderiana also possessed the strongest free radical scavenging activity amongst all the tested species, with an inhibition rate of 90.4% at concentration of $330{\mu}g/ml$. Activity-guided bioautographic TLC and HPLC analyses were used to establish which compounds were responsible for the potent antioxidant activities of the S. nylanderiana extract. These analyses revealed lecanoric acid to be primarily responsible for the effective antioxidant properties of S. nylanderiana. Overall, these results have indicated that several highland lichens have the potential of being utilized as novel bioresources for naturally occurring antioxidant therapies.

Antimicrobial and Antioxidant Activities of Ethanol Leaf Extract of Dendropanax morbiferus Lev. (황칠나무(Dendropanax morbiferus Lev.) 잎 에탄올 추출물의 항균 및 항산화활성)

  • In the study reported here, the antimicrobial and antioxidant activities of the ethanol leaf extract of Dendropanax morbiferus Lev. from Jeju Island was investigated. Of the 14 strains of 12 species of microorganisms tested, the extract exhibited antimicrobial activity observed against seven Gram-positive bacteria of four species, but not against six Gram-negative bacteria and a yeast strain. Using the disc diffusion method, the diameter of the inhibition zone increased with application of the extract with every strain and the highest growth inhibition was exhibited with Staphylococcus aureus KCTC 1916 at 5 mg/ml. The minimal inhibitory concentration of the extract (MIC) by turbidity was 2.5 mg/ml against Bacillus cereus KACC 12672 and 15 mg/ml when with Enterococcus faecalis KCTC 3206. The minimum bacterial concentration (MBC) values defined as being $${\geq_-}99.9%$$ reduction in viable cells against the tested strains was higher than the MIC values. Time killing curves using the optimum MIC were performed on seven strains incubated for 48 hr. The growth of B. cereus KACC 12672 was detected after 12 hr and no significant growth was found in the others strains after 48 hr (p<0.05). The 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity of the ethanol leaf extract was similar to that of butylated hydroxyanisole (BHA) at the same concentration. These results indicate that leaf extract of D. morbiferus Lev. can be utilized as a natural preservative and an antioxidant.

In Vitro Antioxidant Activity of Alnus firma Extracts (사방오리(Alnus firma) 추출물의 in vitro 항산화 활성)

  • This study evaluated the antioxidant activity of the extract and fractions of Alnus firma. Alnus firma had the highest total phenolic content ($452.80{\pm}7.01{\mu}g$ gallic acid equivalents/mg) in a methanol (MeOH) fraction and the highest total flavonoid content ($112.29{\pm}11.14{\mu}g$ rutin equivalents/mg) and antioxidant capacity ($936.23{\pm}0.07{\mu}g$ ${\alpha}$-tocopherol equivalents/mg) in an ethylacetate (EA) fraction. The antioxidant activities of various solvent extract fractions of Alnus firma were evaluated using various antioxidant assays, including ${\beta}$-carotene-linoleate assay, reducing power assay, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, metal chelating activity assay, superoxide anion radical scavenging assay, and lipid peroxidation inhibition assay using the ferric thiocyanate method. These activities were compared with those of ascorbic acid, butylated hydroxyanisole (BHA), gallic acid (GA), butylated hydroxytoluene (BHT), and ${\alpha}$-tocopherol. First, at a $250{\mu}g/ml$ concentration, the EA and MeOH fractions of A. firma showed 92.43% and 89.20% DPPH radical scavenging activity, respectively. Second, $50{\mu}g/ml$ of the EA fraction exhibited 72.49% superoxide anion radical scavenging activity, a little greater than the same dose of GA (60.88%). Finally, 0.5 and 1 mg/ml of the EA fraction showed 73.45% and 73.29% inhibition of peroxidation in the ${\beta}$-carotene-linoleic acid system, respectively. The decreasing order of reducing power was EA fraction > n-butanol (BuOH) fraction > dichloromethane (DCM) fraction > n-hexane (HX) fraction. The results obtained in the present study indicated that Alnus firma can be used as an easily accessible potential source of natural antioxidants.

Antioxidant Activity of Chestnut (Castanea crenata S.et Z.) bur Fermented by Lactobacillus casei (유산균 발효에 의한 밤송이추출물의 항산화 효과)

  • The aim of this study was to show the antioxidant properties of chestnut (Castanea crenata) bur extracts fermented by Lactobacillus casei. The chestnut has been used as a cosmetic material in Korea for many years. This study showed that reactive oxygen species (ROS) were inhibited by the fermentation materials of chestnut bur extracts. The antioxidant activities were analyzed and expressed as EDA, ABTS, reducing power, SOD-like activity, hydrogen peroxide scavenging activity, superoxide anion radical scavenging activity, and nitric oxide scavenging ability. The antioxidant activities of fermentation materials from L. casei of chestnut bur extracts were higher than those of butylated hydroxyanisole (BHA), epigallocatechin gallate (EGCG), and ascorbic acid (AA). Therefore, we expect that fermentation materials from L. casei of chestnut bur extracts are valuable resources as natural antioxidants and functional cosmetics ingredients.
