환경대기 중 프탈레이트 화합물의 농도 측정 (I) - 측정방법 평가와 현장 적용 - (Determination of Phthalates Compounds in the Ambient Atmosphere (I) - Evaluation of a Measurement Method and its Application to a Field Study -)

  • 프탈레이트 화합물은 PVC 수지 등의 가소제로 널리 사용되며, 내분비계장애 및 돌연변이를 유발하는 물질로서 이에 대한 독성은 널리 알려져 있다. 우리나라에서는 소비자용품이나 음용수에서의 프탈레이트 화합물에 대한 측정 사례는 찾아 볼 수 있으나, 대기환경분야에서의 측정사례는 전무한 실정이다. 본 연구에서는 환경대기 중의 프탈레이트 화합물을 측정하기 위해 고용량샘플러로 시료를 채취한 후 속슬렛 장치로 추출하여 GC/MS로 분석하였다. 일부 시료에 대하여 저용량공기채취법과 고용량공기채취법을 동시에 적용하여 두 측정 결과를 비교 평가하였다. 분석대상 항목은 dimethyl phthalate (DMP), diethyl phthalate (DEP), di-n-butyl phthalate (DBP), butyl benzyl phthalate (BBP), di(2-ethylhexyl) phthalate (DEHP) 및 di-n-octyl phthalate (DOP) 등 6개 항목을 선정하였다. 입자상 시료 채취를 위해서는 석영섬유여지를 사용하였으며, 기체상 시료 채취에는 PUF/XAD-2를 사용하였다. 실험실 정도관리 평가 결과 표준시료에 대한 선형성과 재현성은 우수한 결과를 나타내었으며, 정량검출한계는 항목에 따라 대기 중 농도로 환산하여 0.60~17.84 ng/$m^3$ 수준으로 추정되었다. 또한 실제 현장평가를 위해 2007년 1월과 6월에 시화 반월공단 (정왕2동, 원시동)에서 측정한 결과 비교적 분자량이 작고 끓는점이 낮은 DMP, DEP, DBP는 기체상에 많이 존재하며, DBP 보다 분자량이 크고 끓는점이 높은 DEHP와 DOP는 입자상에 주로 존재하는 것으로 나타났다. DEHP는 45.7 ng/$m^3$ ~ 1012.7 ng/$m^3$, DMP는 7.7 ng/$m^3$ ~ 375.1 ng/$m^3$ 수준으로 검출되었다. 전반적으로 볼 때 환경대기 중의 프탈레이트 화합물 측정에는 고용량시료채취법이 저용량법보다 우수한 것으로 나타났다.

Simultaneous Determination of Plasticizers in Food Simulants Using GC/MS

  • Migration levels of plasticizers, di-n-butyl phthalate (DBP), benzyl-butyl phthalate (BBP), di-n-octyl phthalate(DNOP), di-iso-decyl phthalate (DIDP) and di-iso-nonyl phthalate (DINP), di-(2-ethylhexyl) adipate (DEHA), from 46 poly(vinyl chloride) (PVC) wrap films and 54 PVC gaskets into food simulants were determined using gas chromatography/mass spectrometry (GC/MS). The method was validated with limit of detection (LOD) of $0.01{\sim}0.02\;{\mu}g/mL$ for DBP, BBP, DNOP and DEHA, and $2\;{\mu}g/mL$ for DIDP and DINP. The linearity were found to be > 0.99 for all the compounds in concentration range of $0.1{\sim}81.4\;{\mu}g/mL$, and overall recoveries were ranged from 90.4 ~ 99.6%. DBP, BBP, DNOP, DEHA, DIDP and DINP were not detected in food simulants, except 1 wrap sample from which 0.28 and $0.99\;{\mu}g/mL$ of DEHA were detected respectively when tested with 20% ethanol and n-heptane as food simulants. These values were below the regulatory limitation in European Union (EU).

In vivo Effects of Di-n-butyl Phthalate and Di-2-ethylhexyl Phthalate on the Nonspecific Defense Mechanism of the Bagrid Catfish, Pseudobagrus fulvidraco

  • The aim of this study was to investigate in vivo toxicity and effects of two phthalate esters (PEs), di-n-butylphthalate (DBP) and di-2-ethylhexyl phthalate (DEHP), on the immune system of the bagrid catfish, Pseudobagrus fulvidraco. Groups of experimental fish were subjected to daily intraperitoneal injections of 300 or 1000 mg $kg^{-1}$ of DBP or DEHP for 3 days, and the cellularity and functional activity of phagocytes were measured in the spleen and pronephros (head kidney). The number of spleen leukocyte cells increased significantly (p<0.05) in response to low and high doses of DEHP and DBP, respectively; however, the cellularity of the pronephros was more susceptible to higher dose of DEHP than DBP. Nonspecific immunity, as determined by the phagocytic index (PI) and phagocytic capacity (PC), was significantly depressed by DEHP at 1000 $1000mg\;kg^{-1}\;day^{-1}$ in the pronephros at 3 days after injection. Furthermore, significantly (p<0.05) increased levels of serum glutamic oxaloacetate transaminase (GOT) and glutamic pyruvate transaminase (GPT) indicated marked hepatic dysfunction in immunosuppressed fish. Treated fish showed a significant reduction in total serum protein but no significant alteration in lysozyme activity. These results demonstrate the sensitivity of the fish immune response for predicting PE-induced immunotoxicity.

미성숙 수컷 랫드에서 Hershberger 시험에 의한 Di(n-butyl) Phthalate의 항안드로젠 효과 (The Antiandrogenic Effects of Di(n-butyl) Phthalate in Immature Male Rats: Establishment of Hershberger Assay for Endocrine Disruptors)

  • Hershberger assay is known as one of the in vivo-short-term scrrning assays for endocrine disrupting chemicals (EDCs), but this method is not a validated test system. In the present study, the establishment of Hershberger assay to detect EDCs was tried using a model substance, di(n-butyl)phthalate (DBP), a plasticizer for plastics. Thirty-six immature male rats were randomly assigned to six groups: DBP 0, 40, 200, and 1000mg/kg, a positive control (flutamide 20 mg/kg), and a combination group(DBP 1000mg/kg and testosterone 50 ug/kg). DBP and flutamide were administered by gavage to male rats from day 21 to 40 post partum. Testosterone was subcutaneously injected during the same period. We evaluated body weigth gain, weights of ventral prostate, seminal vesicle, and levator ani and bulvocavernous muscle, and serum concentrations of testosterone and lutenizing hormone in male rats. The weights of seminal vesicle and levator ani and bulvocavernous muscle of males receiving 1000mg/kg of DBP was significantly lower than controls. There was no effect of DBP-treatment on body weight gain, prostate weight, and hormone concentrations. In the positive control group, the weights of seminal vesicle and levator ani and bulvocavernous muscle of males receiving 20mg/kg of flutamide were significantly lower than controls. In the combination group, there was no effect of co-treatment of DBP and testosterone on all parameters effect against DBP. This method was found to be a useful short-term screening assay system for EDCs.

초등학생의 식습관과 요 중 프탈레이트 대사체 농도와의 관계 (Relationship between Dietary Habits and Urinary Phthalate Mtabolite Concentrations in Elementary School Children)

  • Objectives: The purpose of this study was to examine the relationship between dietary habits and concentrations of urinary phthalate metabolite in elementary school children. Methods: This study was conducted in Seoul, South Korea. We collected urine samples from 156 children from a single school in September 2016. Information on dietary habits was obtained from the parents of the children. Five types of urinary phthalate metabolites were analyzed using a high-performance liquid chromatography tandem mass spectrometer. Multiple linear regression analysis was used to determine the factors affecting the concentrations of urinary phthalate metabolite. Results: Girls had a significantly lower concentration of urinary mono-n-butyl phthalate (MnBP) (p=0.006) than did boys. When the consumption of bean processed food and wrapped delivery food were more than once a week, urinary MnBP (p=0.021) and monobenzyl phthalate (MBzP) (p=0.032) concentrations were the highest, respectively. Conclusion: Several urinary phthalate metabolite concentrations were associated with demographic and dietary habits. Particularly, urinary MnBP and MBzP were associated with several processed foods. The findings of this study might be useful as basic data for establishing and educating on environmental health policy.

Perinatal Phthalate/Adipate Esters Exposure in Rats: Effects on Maternal Body Weight Changes and Developmental Landmarks in Offspring Rats

  • Di-n-butyl phthalate (DBP), diisononyl phthalate (DINP) and di-(2-ethylhexyl) adipate (DEHA) are ubiquitously distributed chemicals that are widely used as plasticizers and also found at low levels in foods. The aims of this study were to determine whether perinatal exposure to DBP, DINP and DEHA could alter normal patterns of neonatal development. Dams were provided with pulverized soy-free diet containing 20, 200, 2,000 and 10,000 ppm of DBP, 40, 400, 4.000 and 20,000 ppm of DINP, or 480, 2,400 and 12,000 ppm of DEHA from gestational day 15 to postnatal day 21. Exposure to the high doses of DBP, DINP and DEHA during gestational period significantly decreased food consumption and body weight gain of dams. These chemicals reduced neonatal body weight as well as that of the after maturation. Also, exposure to DINP of all the doses used and the higher doses (2,400 and 12,000 ppm) of DEHA decreased AGD at PND 1 in male neonates, though that to DBP did not affect AGD in males. In female neonates, an increase in AGD was observed in DBP- and DINP-exposed animals at the highest doses. Moreover, these chemicals affected survival rate of pups at PND 5, and delayed onset of eye opening in all chemica1-exposed groups at PND 17. These results suggest that perinatal exposure to these chemicals may affect the normal development and/or growth of offspring.
