Root Yields and Saikosaponin Contents Depending on Planting Time and Cultivated Regions of Bupleurum falcatum L.

  • In order to decide the optimum planting time for Bupleurum falcatum L. which is grown widely in Korea, some experiments were carried out in different regions such as mid-north area (Gangwon-do, Gyeonggi-do and Chungcheongnamdo), mid-south area (Jeollanam-do, Jeollabuk-do, Gyeongsangnam-do and Gyeongsangbuk-do). The mid-south area had the highest total saikosaponin contents of 1.001, which is the medicinal ingredients of Bupleurum falcatum L., in accordance with the different Bupleurum falcatum L. regions. Also this area has the highest extract contents of 24.3%. Average dry root yields in mid-north area (Gangwon-do) were $450{\sim}460$ kg/ha at planting time of March 20th to March 30th, 500 kg/ha at the time of March 30th to April 10th, and $470{\sim}480$ kg/ha at the time of March 10th to March 20th in Chungcheongnam-do. The optimum planting time in the mid-south area (Jeollabuk-do) was March 10th to March 30th with the average dry root yield of $490{\sim}550$ kg/ha. Average dry root yield in Euiseong, Gyeongsangbuk-do area was 470 kg/ha, and the optimum planting time was April 10th, which suggested that the yield increased as the planting time was delayed. At Jeju-do, the volcanic soil of farsouthern area, average dry root yield was 510 kg/ha at the time of March 10th, but the yield from non-volcanic soil was lower than from volcanic soil, which had a dry root yield of 470 kg/ha.

시호 추출물의 화장품 생리활성에 관한 연구 (A Study on the Physiological Activity of Cosmetics in Bupleurum falcatum L. Extract)

  • 시호는 약용작물의 하나로 해열, 진정, 간 장애 억제, 항염, 항암 등 우수한 약리 활성을 지니고 있다. 현재 식품 또는 약용으로 사용되고 있지만 화장품으로서의 사용은 미비하여 화장품 소재로서의 적합성 및 이용 가치를 검토하였다. 시호 추출물을 이용하여 폴리페놀, 플라보노이드, 항산화, 항염 실험을 진행하였다. 폴리페놀 함량은 14.71±0.16 mg·TAE/g, 플라보노이드 함량은 1.42±0.05 mg·QE/g를 나타내었다. 항산화 활성은 농도 의존적으로 우수한 활성을 보였으며, NO 발현 억제능은 1,000 ㎍/mL에서 61.21%의 저해율을 나타내어 항염 활성이 뛰어남을 확인하였다. 본 연구를 통해 시호 추출물의 항산화, 항염 효능이 뛰어나기에 화장품의 천연 소재로서 충분한 가치가 있다고 본다.

Chemical Constituents from the Aerial Parts of Bupleurum falcatum L. and Biological Evidences

  • In this study, phytochemical investigation on the aerial parts of Bupleurum falcatum resulted in the isolation of fourteen compounds including three quinic acid derivatives (1 - 3), five flavonoids (4 - 8), three monoterpene glycosides (9 - 11), and three saikosaponins (12 - 14). Compound 1 was first isolated from nature and unambiguously determined to be 3-O-feruloyl 5-O-caffeoylquinic acid on the basis of the extensive spectroscopic evidence. Biological testing revealed that saikosaponin A (12) and saikosaponin D (13) showed moderate antiproliferative effects on HL-60 and HepG2 cancer cell lines.

  • The study for the quantitative analysis of saikosaponin of Bupleurum falcatum root produced by tissue culture was attempted. The optimal concentration of 2,4-D was 0.1 mg/l for inducing the callus. The induction and growth of the adventitious root from the callus was obtained in the suspension culture containing MS basal medium supplemented with no plant growth regulators. The results of the quantitative analysis of saikosaponins by high performance liquid chromatography (HPLC) showed that the content of saikosaponin a and c was high in the one-year-old root from somatic embryos and that of saikosaponin d was remarkably high in three-months-old adventitious root from callus.

시호(Bupleurum falcatum L.)의 모상근 배양을 통한 Saponin 생산 I. 캘러스, 부정근, 모상근과 재배근의 Saponin 양상과 함량 비교 (Production of Saponin by Hairy Root Culture of Bupleurum falcatum L. I. Comparison of Saponin Content and Pattern in Callus, Adventitous Root, Hairy Root and Cultivated Root)

  • In order to survey possibility to produce saikosaponin from in vitro hairy root culture, culture of callus, adventitous root, and hairy root of Bupleurum falcatum L. were estabilished, and quantitative and qualitative aspects in saikosaponin extracted from these were compared with those of cultivated root. Callus grew well in MS medium containing 0.9 $\mu$M 2, 4-D. In contrast, both of adventitous root and hairy root grew well in hormone-free MS medium. However, hairy root showed more rapid growth with extensive lateral root branches, characteristics of lower content of water and softer than in adventitous root. Among the selected lines of adventitous root and hairy root were observed difference in the growth rate. Mannopine, one of opine synthesized in the transformed tissue with Agrobacterium rhizogenes. A4 were detected in the extract of hairy root lines. Pattern and content of crude saponin from adventitous and hairy root showed no difference, but somewhat difference from those of cultivated root. However, in callus, distinct production-aspect of saponin was not observed.

Variations in Chromosome Numbers and Saikosaponin Contents of Bupleurum falcatum L. from Different Geographical Regions

  • Seven genetic lines of Bupleurum falcatum L. from different geographical regions were analysed for saikosaponin contents and chromosomal numbers. The somatic chromosome numbers of B. falcatum originated from Euisong, Iri, Milyang, Sangnam, Taejon, and Youngchon were 2n=20 while Mishimasaiko showed 2n=26. However, chromosome features were different in plants grown in different geographical regions. Generally, Korean lines had higher saikosaponin contents than Mishimasaiko which is Japanese and Sangnam lines had highest saikosaponin contents compared to other tested lines.

배수체 작성에 따른 시호 작물 특성 (Colchicine-Induced Polyploidy and It's Agronomic Characters in Bupleurum falcatum)

  • The effect of colchicine treatment on the agronomic performance and polyploid formation of Bupleurum falcatum using flow cytometry technique was investigated. The roots of 4-leaf stage plants were treated with colchicine (0.5%) for 3, 6, 12, and 24 hours and then transplanted in the field. Agronomic characters (survival rate, plant height, chlorophyll content, bolting rate) were recorded at 4 weeks and 6 months after transplanting while flow cytometry technique was conducted for determination of polyploid formation. Flow cytometry technique revealed polyploid nuclear DNA formation in colchicine treated plants. The highest number of polyploids was obtained at the shortest colchicine treatment time indicating an inverse relationship between colchicine treatment time and polyploid formation. Results also showed that survival and bolting rates were inversely related with the treatment time while plant height and chlorophyll were not significantly affected by the treatment. This study showed a convenient method for determination of colchicine-induced polyploid in B. falcatum and its superior agronomic performance at shorter treatment time.

시호(Bupleurum falcatum)의 생육과 생산성에 미치는 폐활성탄의 효과 (The Effect of Activated Charcoal on Growth and Yield in Bupleurum falcatum)

  • 폐활성탄을 토양개량제로 활용하기위한 기초시험으로서 시호의 포트시험을 실시한 결과 시호의 생장은 폐활성탄(activated charcoal) 10∼30%가 첨가되었을 때 경장과 경태가 크고 분얼수가 많아 생육이 양호하였다. 근의 생장과 비대는 폐활성탄(activated charcoal) 10-30% 첨가되 었을때 주근장과 근직경이 크고, 근중이 무거워서 증수되는 경향이었다. 이와 같은 이유는 활성탄 시용에 의한 토양의 보수력과 보비력이 양호하여 물리성이 향상된 것으로 생각되며 이보다 더 낮은 농도나 높은 농도에서는 효과가 적은 것으로 나타났다. 이상과 같은 결과로 보아 시호를 재배할 경우 폐활성탄(activated charcoal)의 효과가 인정되어 약초의 종류에 따른 농도별 시험이 계속적으로 이루어 져야 할 것으로 사료된다.

약용작물의 경영성과와 효율성 분석 -구릿대와 시호를 중심으로- (An Analysis of Management Performance and Efficiency of Medician Crop Farm - Mainly on Angelica dahurica and Bupleurum falcatum -)

  • 본 연구는 수입산 한약재 수입으로 경영상황이 극한으로 몰린 약용작물 재배농가의 경영개선 방안을 마련하기 위해 구릿대와 시호재배 농가를 중심으로 경영성과 및 경영 효율성에 대한 분석을 실시하였다. 구릿대 재배농가와 시호 재배농가로부터 수집된 자료에 대한 분석결과를 요약하면 다음과 같다. 첫째, 구릿대 재배농가의 10a당 총수입은 평균 1,363천원으로 나타났으며, 소득은 500천원으로 소득율은 36.7%로 분석되었다. 둘째, 시호 재배농가의 10a당 총수입은 평균 1,705천원으로 나타났으며, 소득은 873천원으로 소득율은 44.6%였다. 셋째, 구릿대 재배농가의 기술효율성은 0.790, 순수기술효율성은 0.856, 규모효율성은 0.924로 분석되었다. 넷째, 시호 재배농가의 기술효율성은 0.670, 순수기술효율성은 0.794, 규모효율성은 0.844로 분석되었다. 구릿대와 시호 재배농가모두 기술효율성에 있어서 상대적인 차이가 존재하였으며, 이러한 차이는 규모적 측면보다 순수 기술적 측면이 더 큰 영향을 미치는 것을 확인할 수 있었다. 따라서 경영규모 확대를 통한 효율성 제고보다는 농가들의 기술수준에 적합한 영농교육을 실시하는 것이 효율성을 높이는데 보다 효과적일 것으로 여겨진다.

시호 추출물이 세포성 면역반응에 미치는 영향 (Effect of Bupleurum falcatum extract on cellular immune responses)

  • Bupleurum falcatum has been used for treatment of inflammation, jaundice, influenza and hepatitis as a traditional orient folk medicine. This experiment was carried out to evaluate the effect of B falcatum extract on cellular immune responses in vivo and in vitro. Antigen binding cell(ABC) assay, antibody production, Arthus and delayed-type hypersensitivity(DTH) reaction against sheep erythrocytes(SRBC) were very depressed in B falcatum extract treated group in vivo. The growth of Staphylococcus aureus in brain heart infusion(BHI) broth containing B falcatum extract was remarkably inhibited. Otherwise, that of Salmonella typhyimurium was not significantly increased in vitro. When B falcatum extract pretreated mice were intraperitoneally(IP) injected S typhimurium and S aureus, respectively, the number of bacteria in peritoneal exudates were time dependent declination compared with those of control, and the weight of spleen and the number of macrophage migration into peritoneal cavity have no difference from those of untreated control. B falcatum extract gradually increased phagocytic activities of peritoneal macrophage against Candida albicans time and dose dependently, and was not significant production of migration inhibiotory factor(MIF). But migration abilities of normal leucocytes in B falcatum extract pretreated group were decreased dose dependently. When B falcatum extract was IP administered, these data indicate that B falcatum extract increases level of serum coticosterone. Therefore, B falcatum extract was indirectly mediated in immune system by serum coticosterone having relation to immunosuppression. These results lead to the conclusion that B falcatum extract acts as a trigger or regulator of cellular immune responses in immune system.
