Effects of Exercise on Pasma Glucose and Cholesterol Level in Streptozotocin-induced Diabetic Rats Fed Buckwheat Diet

  • The ability of exercise and buckwheat diet to modify plasma glucose and cholesterol levels in streptozotocin-induced diabetic rats has been studied. Diabetic rats were fed corn starch as a control diet or buckwheat as an experimental diet for 4 weeks. One group of rats were exercise-trained to swim for 60 min/day, 6 days a week. Plasma glucose levels of sedentary rats both on the control diet and on the buckwheat diet were significantly increased from 367.0$\pm$33.6 mg/dl to 545.0$\pm$38.7 mg/dl and from 380.3$\pm$18.9 mg/dl to 540.5$\pm$36.6 mg/dl, respectively. However, this large increase of plasma glucose was not seen in exercised rats on the buckwheat diet (from 345.0$\pm$25.6 mg/dl to 391.4$\pm$34.7 mg/dl). The total plasma cholesterol level was not affected by either diet or exercise. The HDL-cholesterol level tends to increase due to the buckwheat diet or an exercise, but not with a significant increase. Our results suggest that the buckwheat diet is beneficial in lowering the plasma glucose level only when diabetic rats ar exercised.

Effects of Buckwheat on the Insulin Sensitivity, Protein Digestibility and Utility in Diabetic Rats

  • An experiment was performed to determine if buckwheat intake would improve insulin sensitivity in in normal healthy ras and steptozoticin-induced diabetic Sprague-Dauley rats. For four weeks, rats were fed either corn starch as a cotnrol diet or buckwheat as an experimental diet. As a result, the insulin sensitivity and plasma glucose levels in normal rats were not significantly affected by buckwheat fedding. The insulin sensitivity was lower in diabetic rats than in normal rats(p<0.05). Buckwheat tends to decrease the final plasma glucose level and increase insulin sensitivity in diabetic rats, but there was no sifnificant difference. Another five-week experiment was conducted to determine protein digestibility and protein utility in normal healty rats ad streptozotocin-induced diabetic rats on a control diet or buckwheat diet. The diet composition in this experiment was the same as the preceeding experiment. In the cotnrol diet groups, the protein digestibility in diabetic rats was significantly lower than that in normal rats(p<0.05). Buckwheat reduced protein digestibility in both normal and disbetic rats(p<0.05). Interestingly, in buckwheat diet groups, protei digestibility in diabetic rats was similar to that in normal rats. Protein utility was significantly lower indiabetic rats than in normal rats. This phenomenon was observed as early as the first week of the feeding period. However, protein utility was not sifnificanlty altered in both normal and diabetic rats by buckwheat feeding. It follows that decreased protein digestibility and utility in diabetic rts are not further aggravated by buckwheat feeding, suggesting that buckwheat can be a feasible supplement food for the diabetic therapeutic diet.

메밀식이가 제 2형 당뇨환자의 혈당과 혈중지질 대사에 미치는 영향 (Effects of Buckwheat Diet on Serum Glucose and Lipid Metabolism in NIDDM)

  • We have studied the effects of buckwheat diet on serum glucose and lipid metabolism in 9 NIDDM volunteers during 2 weeks. The subjects were given dietary counseling in their own homes at 2-3 day intervals throughout experimental periods and the dietary intake were determined by interview and record methods. The intake of calorie, carbohydrate, protein and fat during the buckwheat diet period were not significantly different compared with control diet and body weight was maintained within 1-2kg. The mean total glycohemoglobin, fructosamine, total cholesterol and LDL-cholesterol levels at the end of buckwheat diet were significantly lower than the end of control diet (P<0.05). Fasting serum glucose, insulin and HDL-chloesterol levels were slightly decreased. The mean triglyceride level was increased but it was not significant. These results indicate that buckwheat diet is an effective therapeutic regimen for the control of metabolic derangements in diabetes mellitus.

메밀급여가 Streptozotocin유발 당뇨쥐의 장기무게 및 당질과 지질대사에 미치는 영향 (Effects of Buckwheat on Organ Weight, Glucose and Lipid Metabolism in Streptozotocin-Induced Diabetic Rats)

  • The present study was undertaken to evaluate the effects of 50% buckwheat diet on the body weight, organ weight, urine albumin, urine glucose, plasma glucose and plasma lipid in normal rats and diabetic rats treated with streptozotocin(STZ). The food intake, body weight, the level of urine glucose in diabetic buckwheat groups were not significantly different with diabetic control group. The level of urine albumin was lower in raw and steam buckwheat group than in the diabetic control group. Compared to the normal control group, liver and kidney weights were heavier in the diabetic groups. Pancreas weight was heavier diabetic buckwheat groups than in normal and diabetic control groups. Fasting plasma glucose level of diabetic buckwheat groups significantly decreased by 18-37% compared with the diabetic control group. Plasma triglyceride level of diabetic buckwheat groups significantly decreased by 34-50% compared with the diabetic control group. Plasma total cholesterol level of diabetic buckwheat groups decreased by 15-27% compared with the diabetic control group. The level of HDL-cholesterol was not affected by buckwheat diet. These results indicate that buckwheat is an effective therapeutic regimen for the control of metabolic derangements in diabetics.

  • 메밀보충급여가 백서의 혈압 및 혈당에 미치는 효과를 검토 하고자 AIN-76 diet와 AIN-76 diet의 탄수화물원을 박피 메밀분으로 교체한 식이로 4주간 사육하여 성장율, 혈당, 인슐린 분비능, 혈압 등의 변화를 측정하고 고찰한 결과는 다음과 같다. 동일실험 기간동안 체중증가량 및 식이섭취량은 대조군과 메밀투여군 사이에 큰 차이를 보이지 않았다. 메밀보충 급여군은 혈당강하작용이 있음이 관찰되었으며, 혈압은 유의성이 없으나 수치적으로 어느 정도 감소시키는 경향을 보였다. 또한 메밀보충급여에 의해 공복시 혈청 인슐린 수준은 낮았으며 혈당을 투여한 후 혈중의 당이 증가하면 분비되는 인슐린 양도 빠르게 증가하는 경향을 나타내었으나 유의성은 없었다.

  • Wistar Strain male rats were divided into four different diet groups and the control group was fed on seven percent casein diet (C), the second with a mixture of 80% of rice and 20% of buckwheat (RW), the third with rice only (R), and the fourth with a mixture of rice and buckwheat of equal nitrogen level (RWS). Each diet group consisted of six rats was fed for three weeks by the adlibitum feeding method. The results of the experiment, i, e, the growth gain and change in the level of branch-amino acids in the plasma determined by the micro-bioassay method were as follows. 1. The group C showed the largest growth gain and the rest did in the order of RW group, R group, and RWS group. 2. It was shown that rats fed on diet of higher protein score tend to have higher level of free branch-amino acids in the plasma. 3. Thus, the protein score may be estimated based on the level of the free branch-amino acids in the plasma.

Hypocholesterolemic Effect of Tartary Buckwheat (F. tataricum Gaertn.) Extract from High Fat Diet Mice

  • To investigate the efficacy of extract of Tartary buckwheat (F. tataricum Gaertn.) in high fat diet (HFD) in mice, the F. tataricum Gaertn. extract (FTE) was orally administered to mice with a HFD at 300 mg/kg/day for 4 weeks. Our results show that FTE significantly inhibited fat accumulation. Moreover, FTE markedly reduced the final body weight with a decrease in epididymal adipose tissue mass and adipocyte size compared with the untreated HFD-induced group. Additionally, FTE ameliorated serum total cholesterol, triglyceride, and low-density lipoprotein cholesterol levels. The results show that Tartary buckwheat possesses hypocholesterolemic effect through downregulating lipid metabolism. Further studies are required in this area to strengthen the anti-obesity effects of FTE with active component, and it can be used a pro-drug instead of whole extract.

  • 흰주의 체중은 실험기간동안 증가하였으며, 식이 별로는 대조군이 가장 높은 체중 증가를 보였다. 콜레스테롤의 첨가는 흰쥐의 정상적인 성장을 억제하였으며, 콜레스테롤 첨가군에서는 콜레스테롤 대조군의 체중증가량이 유의하게 가장 낮았다. Rutin과 메밀 채소를 식이에 첨가하였을 때 흰쥐의 콜레스테롤에 의한 성장억제 작용을 효과적으로 감소시키는 것으로 나타났으며, 첨가량의 증가에 따라 혈청의 총 콜레스테롤 함량과 과산화지질 함량은 낮아지는 반면에 HDL-cholesterol 함량은 높아지는 경향을 보였다.

  • Wistar계(系) 흰쥐숫놈을 casein 대조군(大照群), 백미사료군(白米飼料群), 백미(白米) 80%에 교맥분(蕎麥粉) 20%를 가(加)한 사료군(飼料群), 백미(白米)와 교맥분(蕎麥粉)의 질소(窒素) level을 같게한 사료군(飼料群)으로 하여 각군(各群) 6마리씩을 ad libitum방법(方法)으로 3주간(週間) 사육(飼育)하였을 때의 성장결과(成長結果)는 아래와 같다. 백미(白米) 80%에 교맥분(蕎麥粉) 20%를 혼합(混合)한 사료군(飼料群)은 다른 시험군(試驗群) 보다 체중증가율(體重增加率)이 가장 좋았다. 이것은 쌀에 잡곡(雜穀)을 적당한량(量) 혼합(混合)함으로써 단백가(蛋白價)를 개선(改善)할 수 있다는 이론(理論)을 동물사육실험(動物飼育實驗)으로서도 잘 입증(立證)해 줌을 볼 수 있었다.

  • STZ 유발 당뇨쥐에 날메밀, 볶은메밀, 찐메밀을 식이의 50%가 되도록 각각 혼합한 메밀식 이를 급여하여 2주간 사육시킨 후 변의 단백질 함량, 췌장무게, 췌장의 $\alpha-amylase,$ chymotrypsin, lipase환성 및 변의 $\alpha-amylase,$ chymotrypsin, trypsin 활성을 각각 측정하였다. 변중 단백질 함량은 당뇨대조군에 비하여 당뇨날메밀군, 당뇨볶은메밀군, 당뇨찐메밀군에서 각각 99%, 91%, 103% 유의적으로 증가하였으며 , 췌장 무게는 당뇨대조군과 비교하여 당뇨날메밀군에서 24% 유의적으로 증가하였다. 췌장의 $\alpha-amylase$ 및 lipase 환성은 당뇨대조군과 당뇨메밀군들 사이에 유의적인 차이가 없었으나 chymotrypsin 활성은 당뇨찐메밀군에서 45% 유의적으로 감소하였다. 변의 $\alpha-amylase활성은$ 당뇨대조군과 당뇨메밀군들 사이에 유의적인 차이가 없었으나, chymotrypsin 활성은 모든 당뇨메밀군들에서, trypsin 활성은 당뇨볶은메밀군에서 유의적으로 증가하였다.
