Results obtained from six secological tests for diagnosing bovine brucellosis-standard plate agglutination test (SPT), standard tube agglutination test(STT), complement fixation test(CFT), Rivanol test (RT), agar gel precipitation test (AGP) and counterimmunoelectrophoresis(CIEP) were compared using 38 sera from brucella reactors and 222 sera from dairy and beef cattle in field. The SPT gave 1.6% apparent false negative reactions and 15.4% apparent false positive reactions when compared with STT which is an official test for bovine brucellosis in this country. The distribution of antibody titers determined by STT showed that 37.5% of 38 reactors had antibody titers ranging from 100 to 200, and the remaining 62.5% had antibody titers of 400 or higher. when 38 reactor judged by STT were tested by CFT and RT, 32 cattle(82.4%) were positive by CFT and 33 cattle (86.8%) were positive by RT, respectively. This results suggest that RT is comparable to CFT in the diagnosis of bovine brucellosis. The results also indicated that both AGP and CIEP were insensitive to detect brucella infection in cattle.
A novel enzyme linked immunosorbent assay (ELISA) is described and compared with other established serologic tests for bovine brucellosis, namely the rose bengal test (RBT), the complement fixation test (CFT), and the tube agglutination test (TAT) approved and used in Korea. A total of 109 bovine serum samples were tested using all the 4 assays and analyzed as to specificity, sensitivity, reproducibility and predictive value. The ELISA showed 100% agreement with the CFT. The least agreement between ELISA was observed with the TAT. The agreement between the ELISA and RBT was not significantly different from that observed between the CFT and the ELISA. It is concluded that the new assay would be a good candidate for routine serologic survey for brucellosis in Korea. A protocol combining the ELISA and the CFT would increase the power for detection of serologically positive individuals and herds.
Reproductive failures, such as abortions were occured among dogs in a large kennel in Chonnam area April in 1994. After an initial positive result by the 2 mercaptoethal rapid slide agglutination test(2ME-RSAT) to Brucella canis on five sera, additional specimens from all dogs in the population were tested. The blood cultures and 3 following serological tests (2ME-RSAT, TAT, AGID) performed on all samples on the basis of surveys, the following results were obtained. 1. Thirty three of 62 dogs were seropositive. 2. Twenty blood samples from 33 dogs were cultured, all of the isolates were identified as B canis. 3. Although there was not siginificant difference in sex, age and breed of the cause brucellosis dectected, abortions was occured late in gestation stage.
Brucellosis is a serious zoonosis, recognized worldwide. It primarily affects animals, which act as reservoirs for human infection as well as being of economic significance to the agri-food industry. Bangladesh has been reported as an endemic area for brucellosis. So a cross sectional study was conducted to determine the seroprevalence and potential risk factors of brucellosis in cattle in Dinajpur and Mymensingh districts of Bangladesh. A total of 182 cattle were examined by Rose Bengal Plate Test (RBPT) between September 2008 and October 2009. Then Positive, doubtful, and negative samples were further confirmed with slow agglutination test (SAT) and both indirect and competitive enzyme linked immunosorbent assay (iELISA and cELISA). A questionnaire was used to collect epidemiological information of the animals. The overall animal-level prevalence was 3.30%. Brucellosis seroprevalence was higher (4.76% by cELISA) in cattle above 48 months than those under 48 months. Female showed higher seroprevalence (10.67%) than male (6.25%). Higher seroprevalence was also found in cattle bred naturally (20.0%) than artificially (8.77%) and cattle that aborted or with previous abortion record (22.22%) showed higher seroprevalence than non-aborted (7.69%). The sensitivity of RBT and SAT was found 100% as compared to cELISA standard test, whereas specificity of RBT (95.35%) was higher than that of SAT (94.32%).

Background: In 2012, the Alaska Section of Epidemiology investigated personnel potentially exposed to a Brucella suis isolate as it transited through three laboratories. Methods: We summarize the first implementation of the United States Centers for Disease Control and Prevention 2013 revised recommendations for monitoring such exposures: (1) risk classification; (2) antimicrobial postexposure prophylaxis; (3) serologic monitoring; and (4) symptom surveillance. Results: Over 30 people were assessed for exposure and subsequently monitored for development of illness. No cases of laboratory-associated brucellosis occurred. Changes were made to gaps in laboratory biosafety practices that had been identified in the investigation. Conclusion: Achieving full compliance for the precise schedule of serologic monitoring was challenging and resource intensive for the laboratory performing testing. More refined exposure assessments could inform decision making for follow-up to maximize likelihood of detecting persons at risk while not overtaxing resources.
This paper describes the epidemiological characteristics of bovine brucellosis in Korea during January 2000~September 2004, which encompasses the period when the incidence of bovine brucellosis increased abruptly. Data from the National Animal Infectious Disease Data Management System were used for this study. A range of epidemiological measures was calculated including annual herd and animal incidence. During the study period, there were 1,183 outbreaks on 638 farms. In beef cattle, annual herd incidence increased from 0.2 (2000) to 11.5 (2004, to September) outbreaks per 10,000 and annual animal incidence varied between 3.4 (2000) and 105.8 (2004, to September) per 100,000, respectively. On 401 (62.9%) infected farms during this period, infection was eradicated without recurrence. Recurrence of infection was significantly higher on farms where abortion was reported (53.3%), compared to farms where it was not (30.0%). On beef cattle farms, infection was introduced most frequently through purchased cattle (46.2%). Based on the results of this study, the establishment and spread of brucellosis in the Korean beef cattle population were mainly due to incomplete or inappropriate treatment of aborted materials and the movement of infected cattle.

The study was carried out to evaluate the diagnostic efficacy of the tube agglutination test (TAT), enzyme-linked immunosorbent assay (ELISA) and the 2-Mercaptoethanol agglutination test (2-MAT) to detect human brucellosis patients in Korea. We examined 87 serum samples of people in the rural farm areas where bovine brucellosis had been reported. People in this study were divided into seven groups- farmers and their families, veterinarians, veterinary quarantine workers, livestock health control officers, artificial inseminators, livestock traders and healthy control individuals. Among 87 people, 65 were males and 22 were females ranging in age from 13 to 72 years. Of 87 serum samples, ELISA detected 21.84%, TAT detected 11.50% and 2-MAT detected 8.05% Brucella positive sera. Brucella specific IgG ELISA antibody titer was recorder higher in the individuals between the ages of 50 and 65 years. The highest prevalence rate of brucellosis(29.4%) was recorded in the cattle farmers and their family members followed by quarantine veterinary office workers (25%) and practicing veterinarians 01.1%). The majority of the Brucella sero-positive individuals in this study had a history of direct contact with animals.
Objectives : We conducted an investigation on 14 cases of brucellosis in Gyeongsangbuk-do during 2003-2004 to understand the source of infection and the transmission routes of brucellosis. Methods : The authors visited the each of the health centers and we examined the patients, their written epidemiologic questionnaire and the occurrence of bovine brucellosis. We visited the patients' living and work areas, and we examined their occupations, the date they developed symptoms, the progress of their symptoms, whether or not they were treated, their current status, whether or not they consumed raw milk and raw meat, and if their work was related to cattle breeding and the related details. We reviewed the results of the blood tests and medical records and we examined the cattle's barn. Results : There were 3 patients in 2003 and 11 patients in 2004. All of their brucella antibody titer exceeded 1:160. The patients' symptoms were fever, myalgia, malaise, chills and an influenza-like illness, but the clinical signs were absent on the medical records. Brucella abortus were cultured from 3 of the patients' blood samples. Conclusions : When the authors discovered the transmission routes, they were divided into 4 different sorts. The first route was related to cattle birth such that patients touched the calves or placentas that were infected with the Brucella species. The second route was related to performing artificial insemination on the cattle and the semen that was used for artificial insemination. The third route was due to the ingestion of raw meat and milk. The last route was due to sexual intercourse between the patients.
The present study was carried out to investigate the prevalence of brucellosis in Kyungbuk area for the 3 years from 1966 to 1998. Collective milk samples were routinely screened to detect positive farms by using the milk ring test(MRT), and serum agglutination test was performed to detect sero-positive individuals in the MRT positive farms. Attempt were made to isolate the causative organismas from slaughtered sero-positive reactors and some biochemical and polymerase chain reation characters of the isolates were also made to identify the organisms. Seroprevalence to brucellosis in peoples who are close contact with infected dairy herds was also investigated. Brucellosis of dairy cattle was rare before 1997, but has been broken more frequently since early 1998. By the MRT for dairy herds, positive rate was gradually increased every year : 0.6% in 1996, 1.5% in 1997, 3.9% in 1998. Among 262 MRT-positive herds, only 21 herds(8.0%) showed positive brucellosis in serological test. The isolation rates of Brucella sp from tested materials were 51.2% in supramammary glands, 39.5% in milks, and 50.0% in pulmonary Iymphnode, respectively. Isolated strain and biotype were Brucella(B) arbortus biotype 1 in 26 heads, and were B suis biotype 1 in 2 heads. Isolated strain and vaccine strain were very similar in their colony morphology and staining. In drug susceptibility, isolated stains(B abortus) and vaccine strain(B abortus RB-51) were sensitive to ampicillin, gentamycin, kanamycin, neomycin, penicillin, streptomycin, and to tetracycline, but resistant to erythromycin. In the PCR, field strains reacted to BA and IS711 primers, and vaccine strain reacted to BA, IS711, and RB5l primers. In the plate agglutination test of 96 sera of human contacted with animals, serum antibody titer detected 1 : 100 in one person, 1 : 200 in one, and below 1 : 25 in the others.
In Korea, burial is the most common method of disposing animal carcasses culled due to brucellosis infection. However, burial has many disadvantages such as shortage of appropriate burial sites, possibile pollution of ground water supply, and negative view of the public. In this study, we have reviewed 3 legal methods for disposing bovine carcasses, which are burial, incineration, and rendering. We also described the overall process, advantages and disadvantages, and required costs for each method. About 75% of bovine brucellosis outbreak farms had less than 3 reactors, and in our study, rendering required the least amount of cost for farms with a small number of reactors (1-3 heads). Our findings suggest that the use of rendering should be encouraged for farms with bovine brucellosis and other methods considered only if rendering is inappropriate.
