Marker Assisted Selection of Brown Planthopper Resistance and Development of Multi-Resistance to Insect and Diseases in Rice (Oryza sativa L.) (DNA 마커를 이용한 벼멸구 저항성 선발 및 복합내병충성 벼 계통 육성)

  • The main objective of this study was to develop the multi-resistance lines to insects(brown planthopper; BPH, rice green leafhopper; GRH) and disease(blast; BL, bacterial blight; BB and rice stripe virus disease;RSV) with good grain quality and plant type by combining conventional breeding and marker assisted selection(MAS) and to eliminate the linkage drag effects between Bph1 gene and culm length, we conducted MAS of Bph1 gene in advanced backcross and double cross progenies. 'Nampyeong', 'Junam' and 'Milyang220' were used as the parent in this study. 'Milyang220' was used as the donor of brown planthopper resistance gene Bph1 with tall culm length. Two backcross progenies were developed using two recipients 'Nampyeong' carrying GRH resistance gene Grh3(t) with good grain appearance and 'Junam' harboring bacterial blight resistance gene Xa3 with short culm length. Two $BC_1$ generations were resulted from the backcrossing of the $F_1$ plants with recurrent parents 'Nampyeong' and 'Junam'. The second rounds of backcrossing($BC_2$) were derived from the cross of selected resistant $BC_1F_1$ plants based on heterozygous genotype of RM28493 linked to Bph1 gene. The double crossed population was constructed from the cross of between each heterozygous $BC_2F_1$ plants at RM28493 locus of '$Nampyeong^*3$ / Milyang220' and '$Junam^*3$ / Milyang220'., The homozygous alleles in Bph1 gene were selected using co-dominant DNA marker RM28493 in double crossed population. Eighty-five lines with multi-resistance to BL, BB, RSV, GRH and BPH were selected by bio-assay and MAS in generation of double crossing. The culm length, head rice ratio and yield of the selected multi resistance lines was ranged from 71 to 88 cm, from 51 to 93%, from 449 to 629 kg/10a. respectively. We can select a promising multi resistance line similar with 'Nampyeong' of major agronomic traits such as culm legnth, head rice ratio and yield. It was designated as Milyang265. Finally this study was developed the multi resistant varieties against to insects and diseases with the good grain quality 'Milyang265' by the advanced backcross and double cross combining MAS and it can be used as genetic resources of multi-resistance to insect and diseases in rice breeding programs.

Development of a Genetic Map of Chili Pepper Using Single Nucleotide Polymorphism Markers Generated from Next Generation Resequencing of Parents (양친의 대량 염기서열 해독을 통해 개발된 SNP 분자표지를 이용한 고추 유전자지도 작성)

  • Molecular markers, as an efficient selection tool, have been and is being used for practical breeding program in chili pepper (Capsicum annuum L.). Recently, a lot of researches on inheritance and genetic analysis for quantitative traits including capsaicinoids, carotenoids, and sugar content in pepper are being performed worldwide. It has been also reported that QTL mapping is a necessary tool to develop molecular markers associated with the quantitative traits. In this study, we suggested a new method to construct a pepper genetic map using SNP (HRM) markers generated from NGS resequencing of female and male parents. Plant materials were C. annuum 'NB1' (female parent), C. chinense 'Jolokia' (male parent), and their $F_2$ population consisting of 94 progenies. Sequences of 4.6 Gbp and 6.2 Gbp were obtained from NGS resequencing of 'NB1' and 'Jolokia', respectively. Totally, 4.29 million SNPs between 'NB1' and 'Jolokia' were detected and the 1.76 million SNPs were clearly identified. Among them, total 145 SNP (HRM) primer pairs covering pepper genetic map were selected, and the 116 SNP (HRM) markers of them were located on this map. Total distance of the map, which consisted of 12 linkage groups and matched with basic chromosome numbers of pepper, was 1,167.9 cM. According to the mapping result, we concluded that our mapping method was suitable to construct a pepper genetic map fast and accurately. In addition, the genetic map could be directly used for QTL analysis of traits different between both parents.

QTL analysis of agronomic traits in recombinant inbred lines of sunflower under partial irrigation

  • The objective of the present research was to map QTLs associated with agronomic traits such as days from sowing to flowering, plant height, yield and leaf-related traits in a population of recombinant inbred lines (RILs) of sunflower (Helianthus annuus). Two field experiments were conducted with well-irrigated and partially irrigated conditions in randomized complete block design with three replications. A map with 304 AFLP and 191 SSR markers with a mean density of 1 marker per 3.7 cM was used to identify QTLs related to the studied traits. The difference among RILs was significant for all studied traits in both conditions. Three to seven QTLs were found for each studied trait in both conditions. The percentage of phenotypic variance ($R^2$) explained by QTLs ranged from 4 to 49%. Three to six QTLs were found for each yield-related trait in both conditions. The most important QTL for grain yield per plant on linkage group 13 (GYP-P-13-1) under partial-irrigated condition controls 49% of phenotypic variance ($R^2$). The most important QTL for 1,000-grain weight (TGW-P-11-1) was identified on linkage group 11. Favorable alleles for this QTL come from RHA266. The major QTL for days from sowing to flowering (DSF-P-14-1) were observed on linkage group 14 and explained 38% of the phenotypic variance. The positive alleles for this QTL come from RHA266. The major QTL for HD (HD-P-13-1) was also identified on linkage group 13 and explained 37% of the phenotypic variance. Both parents (PAC2 and RHA266) contributed to QTLs controlling leaf-related traits in both conditions. Common QTL for leaf area at flowering (LAF-P-12-1, LAF-W-12-1) was detected in linkage group 12. The results emphasise the importance of the role of linkage groups 2, 10 and 13 for studied traits. Genomic regions on the linkage groups 9 and 12 are specific for QTLs of leaf-related traits in sunflower.

Tea Breeding in Korea (우리나라 차나무 품종육성)

  • When tea trees were introduced to Korea peninsular from China? Historically, Mr. Taeryum, an envoy of Shilla dynasty brought tea seeds from China during Tang dynasty and the seeds were planted at Jiri Mt. by the order of King Heungduk at AD828. During Koryo Dynasty(918 1392), Buddhism spread rapidly all over the country and the tea culture reached its highest stage of prosperity. At the Chosun Dynasty, however, the ceremonial drinking of tea vanished almost completely due to the flourishing Confucian tradition, a kind of substitution of Buddhism. But a few people have supported the traditional tea culture by themselves. Since the independence of Korea soon after the World War II at 1945, Korean War have been exploded at 1950. After economic evolution have been succeeded at 1980, the cultivation area of tea trees has been increased about 2,000ha and the cultural tradition of tea drinking has become popular again at a tea consume quantities amounted to 100g per capita at 2004. The northern limited area of tea plant is lined on the southern part of Korea peninsular. It is very small region compared to China about one million ha and to Japan over 60 thousand ha. It is problem not only the area of tea fields but also the methodology of tea cultivation, for examples without clonal cultivars and mechanical systems. WTO treatments was discussing with Korea, China and Japan government at 2005. Green tea custom is very high at 514% in Korea. If three countries will be agreed the imported tax will be cut off, the Korean tea farmers will be confused because of unstable situation of tea markets. All most of tea farmers should be made the tea fields by seeding not clonal propagation. Because of clonal cultivars have not developed in Korea, there have not been the research institutes for tea plants and manufactures before 1992. Now there are three research institute of tea in Korea; Tea Experiment Station at Bosung of Jeonnam Agricultural Research & Extension Services, Mokpo Experimental Station of National Institute of Crop Science, and Green Tea Cluster Institute of Hadong. Mokpo and Hadong Research Station were established at 2004 and at 2005 but Bosung Station was established at 1992. Seven clonal tea cultivers were selected at Bosung Station; Bohyang, Myngsun, Chanlok, Sunhyang, Mihyang, Jinhyang and Ohsun until 2004. Mokpo Experimental Institute was started the tea provenance testing about 4 provenances: Kangwon-do, Jeonlabuk-do, Jeonlanam-do, and Kyungsangnam-do. Korean new tea cultivers should be selected because Koran wild tea population have been high genetic variation. If tea breeding research will be successful to select new clonal cultivers, the tea farmers of Korea will be stable after WTO treatment with each country.

Determination of Genetic Diversity among Korean Hanwoo Cattle Based on Physical Characteristics

  • This study was conducted to establish genetic criteria for phenotypic characteristics of Hanwoo cattle based on allele frequencies and genetic variance analysis using microsatellite markers. Analysis of the genetic diversity among 399 Hanwoo cattle classified according to nose pigmentation and coat color was carried out using 22 microsatellite markers. The results revealed that the INRA035 locus was associated with the highest $F_{is}$ (0.536). Given that the $F_{is}$ value for the Hanwoo INRA035 population ranged from 0.533 (white) to 1.000 (white spotted), this finding was consistent with the loci being fixed in Hanwoo cattle. Expected heterozygosities of the Hanwoo groups classified by coat colors and degree of nose pigmentation ranged from $0.689{\pm}0.023$ (Holstein) to $0.743{\pm}0.021$ (nose pigmentation level of d). Normal Hanwoo and animals with a mixed white coat showed the closest relationship because the lowest $D_A$ value was observed between these groups. However, a pair-wise differentiation test of $F_{st}$ showed no significant difference among the Hanwoo groups classified by coat color and degree of nose pigmentation (p<0.01). Moreover, results of the neighbor-joining tree based on a $D_A$ genetic distance matrix within 399 Hanwoo individuals and principal component analyses confirmed that different groups of cattle with mixed coat color and nose pigmentation formed other specific groups representing Hanwoo genetic and phenotypic characteristics. The results of this study support a relaxation of policies regulating bull selection or animal registration in an effort to minimize financial loss, and could provide basic information that can be used for establishing criteria to classify Hanwoo phenotypes.

Investigation into Whether Agronomic Traits Are Fixed for the Breeding of Drought Tolerance and BPH-resistant Crosses (내건성 및 BPH 내성 계통의 육성을 위한 농업형질 고정여부 조사)

  • Late in the 1960s, South Korea established cultivation technology and began breeding new "Tongil" type rice cultivars by crossing indica and japonica. To date, this cultivation technique has been used to produce a wide variety of cultivars to meet consumer preferences and adapt to extreme weather conditions. Once major genetic traits are no longer segregated through advanced generations, varieties obtained from a cross between different crops may become a new variety. In this study, we confirmed the agronomic traits of F4, F5, and F6 by advanced generation a population in which drought tolerance and brown planthopper (BPH) resistance were crossed. HV23, Ilmi/HV23, and Drimi2ho/HV23 were used. HV23 was drought-tolerant, and Drimi2ho was resistant to BPH. As a result, it was possible to consider that the agronomic traits were fixed because none of them showed a significant difference from the others. While it takes more than 10 years for the pedigree method of breeding, this study confirmed that the agronomic traits were fixed in 4-6 years. In the future, we will investigate the homology of the CaMsrB2 gene and the Bph1 gene, to confirm that both genes are closely related to each other, and analyze the stable inheritance of the introduced gene for multiple successive generations.

Introgression of Oryza minuta into Rice, Oryza sativa (벼 Oryza sativa x O. minuta 여교배 계통에서 이입 염색체단편 검정)

  • An introgression line, WH79006 was produced from a single plant from $BC_5F_3$ families from a cross between Hwaseongbyeo used as a recurrent parent and O. minuta (BBCC, Ace. No. 101154) as a donor parent, which was subsequently self-pollinated for three generations. WH79006 resembled the O. sativa parent, Hwaseongbyeo. However it differed from Hwaseongbyeo in several traits including days to heading, culm length, grain size, spikelets per panicle and fertility. These differences in the traits between WH79006 and Hwaseongbyeo can be attributed to the O. minuta introgressions. To detect the introgressions, 294 SSR markers of known chromosomal position have been used. At least, 28 introgressed chromosomal segments have been identified using SSR markers and they map to all chromosomes except chromosome 2. The size of the introgressed segments ranged from 4 to 35cM. A QTL related to culm length was detected using 75 $F_2$ plants from the Hwaseongbyeo/WH79006 cross. This QTL, cl6 located on chromosome 6 explained $9.6\%$ of the total phenotypic variation in the population. This QTL has not been detected in the previous QTL studies between Oryza sativa cultivars, indicating potentially novel alleles from O. minutan.

Breeding of New Synthetic Egg Production Line in Domestic Chicken by Intlroducing Sex Linked Gene. I. Production of the Autosexing Breed (성 감별 유전자를 도입한 다산계계통 신품종 육종에 관한 연구 I. 반생유전계통 조성)

  • This study was carried out to build up new synthetic egg Production lines which had sex linked gene for feather color sexing and had also superior combining ability for producing the best commercial chicks. In order to make autosexing layer line, the commercial layers which had Z$^{s}$ Z$^{s}$ and Z$^{s}$ W were mated. Among progeny, the chicks which had homozygote of silver gene and non-silver gene were selected for making dam and sire lines. Afterwards the closed flock breeding method was utilized to improve general performances of the each line. The performances of egg production in synthetic line were 161 day for age at sexual maturity, 219 eggs for total egg number to 60 weeks of age, 84% for hen-day egg production and 619 for average egg weight. There was no difference in egg production between new synthetic lines and imported breeds. In the analysis of genetic trends, the estimates of genetic parameter in the autosexing lines were similar to those of the general population of layer breeders. This results indicated the consistency of genetic variation from this selection.

Effect of errors in pedigree on the accuracy of estimated breeding value for carcass traits in Korean Hanwoo cattle

  • Objective: This study evaluated the effect of pedigree errors (PEs) on the accuracy of estimated breeding value (EBV) and genetic gain for carcass traits in Korean Hanwoo cattle. Methods: The raw data set was based on the pedigree records of Korean Hanwoo cattle. The animals' information was obtained using Hanwoo registration records from Korean animal improvement association database. The record comprised of 46,704 animals, where the number of the sires used was 1,298 and the dams were 38,366 animals. The traits considered were carcass weight (CWT), eye muscle area (EMA), back fat thickness (BFT), and marbling score (MS). Errors were introduced in the pedigree dataset through randomly assigning sires to all progenies. The error rates substituted were 5%, 10%, 20%, 30%, 40%, 50%, 60%, 70%, and 80%, respectively. A simulation was performed to produce a population of 1,650 animals from the pedigree data. A restricted maximum likelihood based animal model was applied to estimate the EBV, accuracy of the EBV, expected genetic gain, variance components, and heritability (h2) estimates for carcass traits. Correlation of the simulated data under PEs was also estimated using Pearson's method. Results: The results showed that the carcass traits per slaughter year were not consistent. The average CWT, EMA, BFT, and MS were 342.60 kg, 78.76 ㎠, 8.63 mm, and 3.31, respectively. When errors were introduced in the pedigree, the accuracy of EBV, genetic gain and h2 of carcass traits was reduced in this study. In addition, the correlation of the simulation was slightly affected under PEs. Conclusion: This study reveals the effect of PEs on the accuracy of EBV and genetic parameters for carcass traits, which provides valuable information for further study in Korean Hanwoo cattle.

Estimation of heritability and genetic correlation of body weight gain and growth curve parameters in Korean native chicken

  • Objective: This study estimated the genetic parameters for body weight gain and growth curve parameter traits in Korean native chicken (KNC). Methods: A total of 585 $F_1$ chickens were used along with 88 of their $F_0$ birds. Body weights were measured every 2 weeks from hatching to 20 weeks of age to measure weight gain at 2-week intervals. For each individual, a logistic growth curve model was fitted to the longitudinal growth dataset to obtain three growth curve parameters (${\alpha}$, asymptotic final body weight; ${\beta}$, inflection point; and ${\gamma}$, constant scale that was proportional to the overall growth rate). Genetic parameters were estimated based on the linear-mixed model using a restricted maximum likelihood method. Results: Heritability estimates of body weight gain traits were low to high (0.057 to 0.458). Heritability estimates for ${\alpha}$, ${\beta}$, and ${\gamma}$ were $0.211{\pm}0.08$, $0.249{\pm}0.09$, and $0.095{\pm}0.06$, respectively. Both genetic and phenotypic correlations between weight gain traits ranged from -0.527 to 0.993. Genetic and phenotypic correlation between the growth curve parameters and weight gain traits ranged from -0.968 to 0.987. Conclusion: Based on the results of this study population, we suggest that the KNC could be used for selective breeding between 6 and 8 weeks of age to enhance the overall genetic improvement of growth traits. After validation of these results in independent studies, these findings will be useful for further optimization of breeding programs for KNC.
