Identification and Phylogeny of the Human Endogenous Retrovirus HERV-W LTR Family in Human Brain cDNA Library and Xq21.3 Region

  • Human endogenous retroviral long terminal repeats (LTRs) have been found to be coexpressed with sequences of genes located nearby. It has been suggested that the LTR elements have contributed to the structural change or genetic variation of human genome connected to various diseases. The HERV-W family has been identified in the cerebrospinal fluids and brains of individuals with schizophrenia. Using a cDNA library derived from a human brain, the HERV-W LTR elements were examined and five new LTR elements were identified. These elements were examined using a YAC clone panel from the Xq21.3 region linked to psychosis that was replicated on the Y chromosome after the separation of the chimpanzee and human lineages. Fourteen elements of the HERV-W LTR were identified in that region. Those LTR elements showed a high degree of sequence similarity ($91.8-99.5\%$) with previously reported HERV-W LTR. A phylogenetic tree obtained from the neighbor-joining method revealed that new HERV-W LTR elements were closely related to the AXt000960, AF072504, and AF072506 from the GenBank database. The data indicates that several copy numbers of the HERV-W LTR elements exist on the Xq21.3 region and are also expressed in the human brain. These LTR elements need to be further investigated as potential leads to neuropsychiatric diseases.

Study on Changes in Shape of Denatured Area in Skull-mimicking Materials Using Focused Ultrasound Sonication

  • Recently, ultrasound therapy has become a new and effective treatment for many brain diseases. Therefore, skull-mimicking phantoms have been developed to simulate the skull and brain tissue of a human and allow further research into ultrasound therapy. In this study, the suitability of various skull-mimicking materials(HDPE, POM C, Acrylic) for studies of brain-tumor treatments was evaluated using focused ultrasound. The acoustic properties of three synthetic resins were measured. The skull-mimicking materials were then combined with an egg white phantom to observe the differences in the ultrasound beam distortion according to the type of material. High-intensity polyethylene was found to be suitable as a skull-mimicking phantom because it had acoustic properties and a denatured-area shape that was close to those of the skull,. In this study, a skull-mimicking phantom with a multi-layer structure was produced after evaluating several skull-mimicking materials. This made it possible to predict the denaturation in a skull in relation to focused ultrasound. The development of a therapeutic protocol for a range of brain diseases will be useful in the future.

Rabies in a Wildebeest (Connochaetes gnou): A Case Study at Bangabandhu Sheikh Mujib Safari Park, Cox's Bazar, Bangladesh

  • Rabies causes the highest mortality of all viral diseases in the world unless the victim has been protected either by active immunization or post-exposure immunoprophylaxis. Infected stray dogs, raccoons, skunks, foxes and bats are the demonstrated carriers of most cases of rabies. It is difficult to diagnose a rabid animal in the field unless characteristic clinical signs are evident. However, this study used a commercial fast check kit comprised of immunochromatographic test (ICT) strips (ICTS) to diagnose rabies infection in clinically suspected samples obtained from a wildebeest. A 10-year old male wildebeest (approximate weight, 150 kg) died at Bangabandhu Sheikh Mujib Safari (BSMS) Park, Cox's Bazar, Bangladesh with a clinical history of severe excitation and abundant oral secretions. A gross pathological examination revealed no specific lesions indicating any fatal diseases. The entire brain was collected within 6 hours of death, and the brain sample was tested using the ICT strips following the manufacturer's directions. The rabies viral antibody was detected within the brain stem and medulla of the brain tissue of the dead wildebeest.

Invasive Aspergillosis Involving the Lungs and Brain after Short Period of Steroid Injection: A Case Report

  • Invasive pulmonary aspergillosis (IPA) has emerged as a severe infection in patients with immunocompromised hosts. However, recently, several IPA cases, without an apparent predisposition to immunodeficiency, has been reported. A 72-year-old woman was admitted for evaluation of general weakness and poor oral intake. She reported no medical history, except for intraarticular injection of a corticosteroid for joint pain for the duration of two months. A chest radiography revealed multiple cavitary nodules in both lungs. Examination of specimens, obtained by percutaneous needle biopsy, led to a diagnosis of invasive aspergillosis. Brain magnetic resonance imagining revealed numerous peripheral thin enhancing cystic nodules in both cerebral hemispheres. We initiated intravenous administration of amphotercin B. However, the patient died after nine days. Here, we report an invasive aspergillosis case, which involves the lungs and brain after a short period of steroid injection.

A Rare Case of Primary Thymic Adenocarcinoma Mimicking Small Cell Lung Cancer

  • Primary thymic adenocarcinoma is a very rare malignancy of the anterior mediastinum with no standardized treatment. A 36-year-old male patient presented with hoarseness over the past 3 months. A chest computed tomography (CT) scan showed an infiltrative mass to the proximal vessels and aortic arch in left upper mediastinum ($4.1{\times}3.1{\times}5.4cm$). Brain magnetic resonance imaging (MRI) showed focal lesions, suggesting metastasis in the left frontal lobe. A thoracoscopic biopsy of the mediastinal mass confirmed a primary thymic adenocarcinoma forming a glandular structure with atypia of tumor cells. The patient received four cycles of systemic chemotherapy, consisting of etoposide and cisplatin, with concurrent radiotherapy (6,000 cGy/30 fractions) to the mediastinal lesion and the metastatic brain lesion (4,200 cGy/12 fractions). A follow-up chest CT scan and brain MRI showed a decrease in the size of the left upper mediastinal mass and brain lesion. We report a rare case of the primary thymic adenocarcinoma with a literature review.

  • 최근의 연구들은 알츠하이머병의 병리가 단순히 아밀로이드와 타우단백질의 축적만이 아닌, 혈액뇌장벽(blood brain barrier; 이하 BBB) 이상과 같은 미세혈관병리와 밀접한 관련이 있음을 밝히고 있다. BBB 투과도 변화는 아밀로이드 및 타우 단백질 축적뿐 아니라 신경염증과 신경 퇴행성변화를 일으킴으로써 결국 임상 치매를 야기한다. 최근에는 알츠하이머 및 관련 질환의 BBB 이상을 보기 위한 MR 영상의 이용이 증가하고 있다. 이 종설에서는 알츠하이머병에서 BBB와 관련된 병리를 소개하고, BBB 투과도 변화에 대한 MR 영상 연구들을 소개할 것이다. 또한 BBB 투과도 측정을 위한 MR 방법론 개요와 문제점들을 제시할 것이다.

  • The human gastrointestinal (GI) tract houses a diverse microbial community, known as the gut microbiome comprising bacteria, viruses, fungi, and protozoa. The gut microbiome plays a crucial role in maintaining the body's equilibrium and has recently been discovered to influence the functioning of the central nervous system (CNS). The communication between the nervous system and the GI tract occurs through a two-way network called the gut-brain axis. The nervous system and the GI tract can modulate each other through activated neuronal cells, the immune system, and metabolites produced by the gut microbiome. Extensive research both in preclinical and clinical realms, has highlighted the complex relationship between the gut and diseases associated with the CNS, such as Alzheimer's disease, Parkinson's disease, and amyotrophic lateral sclerosis. This review aims to delineate receptor and target enzymes linked with gut microbiota metabolites and explore their specific roles within the brain, particularly their impact on CNS-related diseases.

Pekingese에서의 Neuronal Vacuolation (Neuronal Vacuolation in a Pekingese)

  • A 6-month-old female Pekingese was euthanized due to poor progrosis after 1 month history of neurologic signs that include depression, ataxia, urination and defecation difficulty. At necropsy, no significant gross abnormalities were noted Histologically, neuronal vacuolation was noted in the brain, primarily cerebellum and occasionally in the brain stem area. Neuronal necrosis and secondary axonal swelling were also observed. Differential diagnoses were able to rule out other diseases which can induce neuronal vacuolation such as lysosomal storage disease, prion infection, and postvaccinal change.

중추신경계 질환의 진단과 치료를 위한 엑소좀의 활용 (Application of Exosome for Diagnosis and Treatment of Diseases in the Central Nervous System)

  • 엑소좀은 단백질, mRNA 및 miRNA를 포함하고 모든 유형의 세포에서 분비되는 세포 외 소포의 일종이다. 방출된 엑소좀은 인접하거나 멀리 있는 다른 세포에 의해 선택적으로 흡수되어 그 내용물을 방출하고 표적 세포를 재프로그래밍한다. 엑소좀은 세포에 의해 생성되는 작은 천연 소포이므로 무독성과 비면역원성의 특징이 있는 것으로 받아들여지고 있다. 최근에는 엑소좀이 중추신경계에 대한 약물 전달체로 과학적 관심을 받고 있다. 중추신경계에는 약물의 침투를 어렵게 하는 혈뇌장벽이 있고 이는 퇴행성신경질환의 치료제 개발에 큰 걸림돌이 되어왔다. 그러나 축적된 연구결과들을 볼 때, 엑소좀이 주로 트랜스사이토시스를 통해 혈뇌장벽을 통과할 수 있음이 제시되었다. 이러한 결과를 종합하면, 엑소좀은 혈뇌장벽을 넘어 뇌 실질조직에 약물을 전달할 수 있는 새로운 전달 수단이 될 것으로 기대된다. 또한 세포의 종류와 질병상태에 따라 분비되는 엑소좀의 종류가 다르기 때문에 엑소좀은 중추신경계 질환의 진단을 위한 바이오마커로도 활용될 수 있다. 본 총설 논문에서는 중추신경계 질환에 대한 바이오마커 및 치료 옵션으로서의 임상시험을 포함한 엑소좀에 대한 최근 연구동향을 정리하였다.

뇌파 기반 뇌-컴퓨터 인터페이스 기술의 소개 (Introduction to EEG-Based Brain-Computer Interface (BCI) Technology)

  • There are a great numbers of disabled individuals who cannot freely move or control specific parts of their body because of serious neurological diseases such as spinal cord injury, amyotrophic lateral sclerosis, brainstem stroke, and so on. Brain-computer interfaces (BCIs) can help them to drive and control external devices using only their brain activity, without the need for physical body movements. Over the past 30 years, several Bel research programs have arisen and tried to develop new communication and control technology for those who are completely paralyzed. Thanks to the rapid development of computer science and neuroimaging technology, new understandings of brain functions, and most importantly many researchers' efforts, Bel is now becoming 'practical' to some extent. The present review article summarizes the current state of electroencephalogram (EEG)-based Bel, which have been being studied most widely, with specific emphasis on its basic concepts, system developments, and prospects for the future.
