  • An experimental study of accelerated weathering on mudstone sample specimens from Haman, Gyeongnam was performed to investigate the variations of physico-mechanical properties of deteriorated rocks due to freeze-thaw weathering. Each complete cycle of freeze and thaw lasted 24 hours, comprising 2 hours of saturating in vacuum chamber, 8 hours of freezing at $-16{\pm}1^{\circ}C$ and 14 hours of thawing at room temperature. Total of 55 cycles of freeze-thaw were completed with measuring the index properties as well as geometries of microfractures. The measured specific gravity and P-wave velocity found to decrease with increasing freeze-thaw cycles. On the other hand, absorption ratio and effective porosity were continuously increased with increasing freeze-thaw cycles. It was found that the index properties of deteriorated sample specimen depend on its initial properties and flaws in rock. The size and density of the traces of the microfracture on slab specimen exhibited abrupt changes after 30 cycles of freeze-thaw weathering. The results obtained in this study show that the box fractal dimension($D_B$) given in this paper has the strong capability of quantifying the size and density of the microfracture.

  • Response surface methodology(RSM) is a very useful statistical techniques for improving and optimizing the product process. By this reason, RSM has been utilized extensively in the industrial world, particularly in the circumstances where several product variables potentially influence some quality characteristic of the product. In order to estimate the optimal condition of product variables, an experiment is being conducted defining appropriate experimental region. However, this experimental region can vary with the experimental circumstances and choice of a researcher. Response surface designs can be classified, according to the shape of the experimental region, into spherical and cuboidal. In the spherical case, the design is either rotatable or very near-rotatable. The central composite design(CCD)s widely used in RSM is an example of 5-level and spherical design. The cuboidal CCDs(CCDs with ${\alpha}=1$) is appropriate when an experimental region is cuboidal but this design dose not satisfy the rotatability as it is not spherical. Practically, a 3-level spherical design is often required in the industrial world where various level of experiments are not available. Box-Behnken design(BBD)s are a most popular 3-level spherical designs for fitting second-order response surfaces and satisfy the rotatability but the experimental region does not vary with the number of variables. The new experimental design with expanded experimental region can be considered if the predicting response at the extremes are interested. This paper proposes a new 3-level spherical RSM which are constructed to expand the experimental region together with number of product variables.

The ovaries were examined 2 times of 10 days interval in 520 posearom and postinsemination subestroHs dairy cows, using rectal palpation and B-mode transrectal ultrasonography. The results of rectal palpation, ultrasonographic examination and measurement of plasma progesterone profiles in 520 subestrous dairy cows were silent brat or error of estrus detection 303 (58.3%), persistent corpus luteum 59 (11.3%), follicular cyst 37 (7.1%), luteal cyst 16 (3.1%), inactive ovary 9 (1.7%), granulosa tumor 1 (0.2%), hydmsalphinx 1 (0.2%), endomehris 81 (15.6%), pyometra 12 (2.3%) and mummified fetus 1 (0.2%), respectively. Accuncy of rectal palpation and ultrasonography for diagiosing ovarian disordeir based on plasma progesterone profiles were silent heat or error of estrus detection 80.5% and 96.7%$\boxUl$ persistent corpus luteum 57.6% and 94.9%, follicular cyst 62.5% and 91.9%1 luteal cyst 62.5% and 87.5%, maclive ovary 55.6% and 88.9% and granulosa cell tumor 100% and 100%, respectively. Acnuucy of rectal palpation for diagnosing uterine disorders based on ultrasonography was pyometra 75.0%1 endometritis 51.9% and mummified fetus 100%, respectively. Cbaracteristic ultrasonographic appearances of ovaries in subestrous dairy cows were as follows; Silent heat or error of estrus detection: anechoic follicle or hypoechoic corpus luteum than ovarian stroma was alternately present on Day 0 (first examination) and Day 10. Follicular cyst: uniformly nonechogenic ovarian structure $\geq $ 25 mm in diameter with a wall < 3 mm was present in ipsilateral on Day 0 and Day 10. Luteal cyst: luteal cyst was similar to follicular cysts but thickness of cystic wall was $\geq $ 3 mm. Inactive ovary : structures within ovaries was not present on Day 0 Bnd Day 10. Characteristic uthssonograpsc appearances of uterus in subestrous dairy cows were as follows; Endometritis: characterized by uterine lumen containing fluid in which 'snowy'echogenic particles art suspended. Pyometra: ultrasonographic appearance of pyometra was diffuse echogenic particles distributed in fluid within the distended uterus, and a thickened uterine wall. These results indicated that ultrasonography was practical far diagnosing reproductive disorders. To diagnosing ovarian disorders, ultrasonography should be carried out 2 times of 10 days interval and rndometritis should be differentiated with uterus of luteal phase in normal cycling cows.

  • Objectives : The purpose of this study was to investigate the effects of Injinchunggan-tang (Yinchenchinggan-tang) on DMN-induced liver damage by applying proteomics. Materials and Methods : Sprague-Dawley rats were used in this experiment and were divided into the normal group (normal saline), the control group (DMN) and the sample group (DMN+IJCGT). DMN was injected i.p. once a day three times a week for 3 weeks in the control group. Normal saline instead of DMN was administered to the normal group. In the sample group, Injinchunggan-tang (Yinchenchinggan-tang) extract was orally administered once a day for 10 days after DMN was induced. The livers of each group were processed and analyzed by histology, Western blot, $Oxyblot^{TM}$, CBB and 2-dimensional electrophoresis. Results : In the histological findings of the liver, IJCGT reduced collagen deposition and liver damage in DMN-induced hepatic fibrosis. IJCGT increased MMP-13 protein production assessed by western blot. Protein oxidation induced by DMN treatment was decreased by IJCGT. In the 2-dimensional electrophoresis finding, the level of the increased proteins induced by DMN treatment such as GRP 75, 58kDa glucose regulated protein and heat shock 70kDa protein 5 were decreased by IJCGT. IJCGT was considered to have the protective effects on hepatotoxicity induced by DMN. In the 2-dimensional electrophoresis finding, the level of increased oxidized proteins such as heat shock 70 protein, mitochondrial malonyltransferase, calreticulin precursor, actin, NADP-isocitrate dehydrogenase, ankyrin repeat and SOCS box protein 11 were decreased by IJCGT. IJCGT was considered to have protective effect on the protein production induced by DMN treatment. Conclusion : Injinchunggan-tang (Yinchenchinggan-tang) exerts an inhibitory effect against the fibrosis and protein oxidation induced by DMN treatment in the rat liver. IJCGT was considered to have protective effects on the hepatotoxicity and protein production induced by DMN treatment.

  • A potyvirus was isolated from cultivated Iris plants showing leaf streak mosaic symptom. Reverse transcription and polymerase chain reaction (RT-PCR) product of 1 kb long which encoded partial nuclear inclusion B and N-terminal region of viral coat protein (CP) genes for potyviruses was successfully amplified with a set of potyvirus-specific degenerate primers with viral RNA samples from the infected leaves: The RT-PCR product was cloned into the plasmid vector and its nucleotide sequences were determined. The nucleotide sequence of a CDNA clone revealed that the virus was an isolate of Ornithogalum moseic virus (OrMV) based on BLAST search analysis and was denoted as OrMV Korean isolate (OrMV-Ky). To further characterize the CP gene of the virus, a pair of OrMV-specific primers was designed and used for amplification of the entire CP gene of OrMV-Kr, The virus was easily and reliably detected from virus-infected Iris leaves by using the RT-PCR with the set of virus-specific primers. The RT-PCR product of the CP gene of the virus was cloned and its sequences were determined from selected recombinant CDNA clones. Sequence analysis revealed that the CP of OrMV-Kr consisted of 762 nucleotides, which encoded 253 amino acid residues. The CP of OrMV-Ky has 94.1-98.0% amino acid sequence identities (20 amino acid alterations) with that of other three isolates of OrMV, Two NT rich potential N-glycosylation motif sequences, NCTS and NWTM, and a DAC triple box responsible for aphid transmission were conserved in CPs of all the strains of OrMV. The virus has 58.5-86.2% amino acid sequence identities with that of other 16 potyviruses, indicating OrMV to be a distinct species of the genus. OrMV-Ky was the most related with Pterostylia virus Yin the phylogenetic tree analysis of CP at the amino acid level. This is the first report on the occurrence of OrMV in Iris plants in Korea. Data in this study indicate that OrMV is found in cultivated Iris plants, and may have mixed infection of OrMV and Iris severe mosaic virus in Korea.

  • This study was conducted in order to develop slaughterhouse rumen content (SRC) as a potential feed additive. The moisture content of SRC can reach 80%, and therefore an appropriate dewatering process is required before it can be used. In this study, the effects of heating temperature, heating time, and filler content during the dewatering process on the activity of various enzymes in SRC were investigated. The Box-Behnken experimental design was employed, involving a total of 45 experimental runs, consisting of three variables (heating time, heating temperature, and filler content) with three levels per variable (12, 30 and 48 hr; 60, 75 and $90^{\circ}C$; 12, 22.5 and 33% for heating time, heating temperature, and filler content, respectively). For enzyme activities, xylanase, cellulase, and amylase were examined, and the results were subjected to an analysis of variance. Heating time, heating temperature and filler content had significant effects on the activity of each enzyme (p<0.05). Cellulase and amylase activities decreased (p<0.05) at elevated heating temperatures, whereas xylanase was reasonably stable around $90^{\circ}C$. The activities of all enzymes decreased (p<0.05) with increased heating time. Optimum filler contents for xylanase, cellulase, and amylase activities were 22.5, 12 and 33%, respectively. However, optimum conditions for all variables that simultaneously maximize the activity of all three enzymes could not be ascertained in this study. Nevertheless, the results from the current study can be useful as basic information for the development of SRC as a feed additive enriched with improved major enzymes for livestock feed digestion.

  • Members of the TGA family of basic domain/leucine zipper transcription factors regulate defense genes through physical interaction with NON-EXPRESSOR OF PR1 (NPR1). Of the seven TGA family members, TGA4/octopine synthase (ocs)-element-binding factor 4 (OBF4) is the least understood. Here we present evidence for a novel function of OBF4 as a regulator of flowering. We identified CONSTANS (CO), a positive regulator of floral induction, as an OBF4-interacting protein, in a yeast two-hybrid library screen. OBF4 interacts with the B-box region of CO. The abundance of OBF4 mRNA cycles with a 24 h rhythm under both long-day (LD) and short-day (SD) conditions, with significantly higher levels during the night than during the day. Electrophoretic mobility shift assays revealed that OBF4 binds to the promoter of the FLOWERING LOCUS T (FT) gene, a direct target of CO. We also found that, like CO and FT, an OBF4:GUS construct was prominently expressed in the vascular tissues of leaf, indicating that OBF4 can regulate FT expression through the formation of a protein complex with CO. Taken together, our results suggest that OBF4 may act as a link between defense responses and flowering.

  • Soil moisture is one of the important components in hydrological processes and also controls the subsurface flow mechanism at a hillslope scale. In this study, time series of soil moisture were measured at a hillslope located in Gwangneung National Arboretum, Korea using a multiplex Time Domain Reflectometry(TDR) system measuring soil moisture with bi-hour interval. The Box-Jenkins transfer function and noise model was used to estimate spatial distributions of soil moisture histories between May and September, 2007. Rainfall was used as an input parameter and soil moisture at 10 cm depth was used as an output parameter in the model. The modeling process consisted of a series of procedures(e.g., data pretreatment, model identification, parameter estimation, and diagnostic checking of selected models), and the relationship between soil moisture and rainfall was assessed. The results indicated that the patterns of soil moisture at different locations and slopes along the hillslope were similar with those of rainfall during the measurment period. However, the spatial distribution of soil moisture was not associated with the slope of the monitored location. This implies that the variability of the soil moisture was determined more by rainfall than by the slope of the site. Due to the influence of vegetation activity on soil moisture flow in spring, the soil moisture prediction in spring showed higher variability and complexity than that in early autumn did. This indicates that vegetation activity is an important factor explaining the patterns of soil moisture for an upland forested hillslope.

  • The ship sailing among waves are suffered the various wave loads that comes from its motion throughout its life. Because there are dynamic, the analysis of ship structure must be considered as the dynamic problem precisely. In the rationally-based design, the dynamic structural analysis is carried out using dynamic wave loads provided from the results of the ship mouton calculation as the rigid body. This method is based on the linear theory assumed low wave height and small amplitude of motion. But at the rough sea condition, high wave height, relatively ship's depth, is induced the large ship motion, so the ship section configulation below water line is rapidly changed at each time. This results in non-linear problem. Considering above situation in this paper, the strength analysis method is introduced for the hull glider among waves considering non-linear hydrodynamic forces. This paper considers that the overall or primary level of the ship structural dynamic loading and dynamic response provided from the non-linear wave forces, and bottom and bow flare impact forces estimated by momentum slamming theory, in which the ship is idealized as a hollow thin-walled box beam using thin-walled beam theory and the finite element method. This method is applied to 40,000 Ton Double-Skin Tanker and attention is paid to the influence of the response of ship speed, wave length and wave height compared with linear strip theory.

  • This study was investigated the changes of quality stability and physicochemical characteristics of the Korean red ginsengs stored for a long times over 20 years. The Korean red ginsengs were stored for 4 to 22 years in canned packaging with polypropylene film and wooden box at room temperatures. The unusal phenomena such as discoloration and pin hole in packaging were not observed. General bacteria showed the vlaues of below 100 CFU/g, coliform groups and molds were not found in any samples stored for 22 year. Any samples also were not detected in mycotoxins. The contents of moisture, ash and crude saponin were the levels of 10.6~11.1%, 3.8~4.2% and 4.1~4.7% during the whole storage periods, respectively. The contents of maltol, which has been known as characteristic flavour and antioxidant of Korean red ginseng, showed remarkably increasing tendency from 0.10 mg/g for 4 years to 2.53 mg/g for 22 years during the storage. The contents of AFG (arginyl-fructosyl-glucose), arginine and free sugar were slightly decreased. Acidic polysaccharide and ginsenoside were not changed significantly during the storage periods. The contents of acidic polysaccharide and total ginsenosides were the 75.1~76.3 mg/g and 15.1~16.6 mg/g, respectively. The sums of ginsenoside-Rg1,-Rb1 and -Rg3s were the ranges of 9.3~9.9 mg/g and PD (ginsenoside-Rb1, -Rb2,-Rc,-Rd,-Rg3s,-Rg3r)/PT (ginsenoside-Rg1,-Rg2,-Re,-Rf,-Rh1) saponin ratios were the levels of 1.4~1.5. These results suggest that Korean red ginsengs stored for long periods show relatively stable quaility stabilities and not significantly changed the contents of ginsenoside and polysaccharide during the storage up to 22 years.
