In this study, we examined gangliosides from streptozotocin-induced diabetic rat brain. To obtain the diabetic rat brain, we sacrified the rat three days after injecting the streptozotocin into venus in tail. We measured blood glucose level according to Somogy-Nelson method and measured insulin level using $^{125}$ I-insulin RIA kit. The gangliosides were extracted according to Folch-Suzuki method from the rat brain. We also examined the effect of major lipid components extracted from deer antler on diabetic rat brain. The results showed that the major lipids components lowered both blood glucose and insulin level in normal rat. However only the blood glucose level in diabetic rat was lowered with major lipid components. In diabetic rat brain, gangliosides metabolism were changed. The amount of GMla was increased while GDla, GDlb, and GTlb were not synthesized. Furthermore, undefined ganglioside was found. In major lipid component-treated diabetic rat brain, the ganglioside metabolism proceeded as same as the normal rat. On the contrary, in bovine brain gangliosides-treated diabetic rat brain, the gangliosides metabolism was not recovered to normal one.
Purpose : This research evaluates the effect of the use of absorbable membrane barrier with deproteinized bovine bone (Bio-$Oss^{(R)}$, Switzerland) on bone healing in surgically created critical-sized defects in rat calvaria. Materials and Methods : Two standardized transosseous circular calvarial defects (5 mm in diameter) are made in each calvarium of 30 rats. These rats are divided into negative control group(n=15), positive control group(n=15) and two experimental groups(n=15). In the negative control group, defects are only filled with blood clots. In the positive control group, defects are filled with autogenous bone obtained from calvarium; in the experimental group 1, defects are filled with deproteinized bovine bone; and in the experimental group 2, defects are filled with deproteinized bovine bone with absorbable membrane. At the postoperative 1 week, 3 weeks. and 6 weeks, clinical. histologic and histomorphometric evaluations of the defects are performed. Results : 1. The grafted bone without membrane in the calvarial bone defect was scattered but, the grafted bone with membrane was stable. 2. $BioMesh^{(R)}$ membrane was absorbed beginning at 3 weeks, and was absorbed considerably at 6 weeks while maintaining the structural form of the membrane. 3. The use of membrane blocked soft tissue invasion. 4. In histomorphometric analysis. it showed the greatest amount of new bone formation in the positive control group. The amount of new bone formation was greater in the experimental group 2 than experimental group 1. At 6 weeks. the amount of new bone formation was greater in the positive control group than experimental group l(p<0.005). Conclusion : These results suggest that membrane increase the stability of grafted bone and protects from soft tissue invasion, and the use of the membrane may promote new bone formation in deproteinized bovine bone graft area.
A case of neosporosis is described in a 6-month-old aborted bovine fetus. Grossly, numerous, well-demarcated, 2-3mm, yellowish white foci were scattered in the heart and skeletal muscle. Histopathologic changes were confined to the brain, heart, and skeletal muscle. Multifocal areas of necrotizing encephalitis were observed in the cerebrum as well as cerebellum. Severe multifocal to coalescing nonsuppurative inflammation was observed in the myocardium and skeletal muscle. The tachyzoites of Neospora caninum were demonstrated in the brain, myocardium and skeletal muscle with immunohistochemical method using Neospora caninum-specific antibody. Based on the histopathology and immunohistochemistry, this case was diagnosed as neosporosis in aborted Korean cattle. This is believed to be the first reported case of bovine neosporosis in Korea.
A new inhibitor for peptidylprolyl cis-trans isomerase (PPIase) has been isolated from Ganoderma lucidum and purified to homogeneous state by organic solvent extraction. The purified PPIase inhibitor (GPI) is assumed to be a membrane-associated glycoprotein. GPI inhibits specifically the bovine brain PPIase, a cyclophilin, and has no effect on the FKBP activity. The results of our chemical modification study of GPI indicate the presence of Lys residue(s) at or near its binding site. Like CsA-cyclophilin complex, GPI-bovine brain PPIase complex strongly inhibits the calcineurin activity in vitro, suggesting the possible involvement of GPI in immunomodulating pathway by the formation of PPIase-inhibitor-calcineurin complex.
Incubation of an NADPH-dependent succinic semialdehyde reductase from bovine brain with N-bromosuccinimide (NBS) resulted in a time-dependent loss of enzyme activity. The inactivation followed pseudo-first-order kinetics with the second-order rate constant of $6.8\times{10}^3$$M^-1$$min^{-1}$. The inactivation was prevented by preincubation of the enzyme with substrate succinic semialdehyde, but not with coenzyme NADPH. There was a linear relation-ship between oxindole formation and the loss of enzyme activity. Spectro-photometric studies indicated that about one oxindole group per molecule of the enzyme was formed following complete loss of enzymatic activity. It is suggested that the catalytic function of succinic semialdehyde reductase is modulated by binding of NBS to a specific tryptophan residue at or near the substrate binding site of the enzyme.
In the present study, the approach of high accuracy mass measurements for phospholipid identifications was evaluated using a 7 T ESI-FTMS/linear ion trap MS/MS. Experiments were carried out for porcine brain, bovine liver, and soybean total lipid extracts in both positive and negative ion modes. In total, 59, 55, and 18 phospholipid species were characterized in the positive ion mode for porcine brain, bovine liver, and soybean lipid extracts, respectively. Assigned lipid classes were PC, PE, PEt, PS, and SM. In the negative ion mode, PG, PS, PA, PE, and PI classes were observed. In the negative ion mode, for porcine brain, bovine liver, and soybean lipid extracts, 28, 34, and 29 species were characterized, respectively. Comparison of our results with those obtained by other groups using derivatization-LC-APCI MS and nano-RP-LC-MS/MS showed that our approach can characterize PC species as effectively as those methods could. In conclusion, we demonstrated that high accuracy mass measurements of total lipid extracts using a high resolution FTMS, particularly, 7T FTMS, plus ion-trap MS/MS are very useful in profiling lipid compositions in biological samples.
세포막 정보전달 경로에서 수용체에 전달된 정보를 세포내로 전달하는데 조절 단백질로 알려진 GTP 결합단백질을 소외 뇌조직으로부터 정제하고 그 분자량등을 관찰하였다. 분쇄한 소의 뇌조직으로부터 세포막을 분리 해내고 1% cholate를 이용하여 세포막 단백질을 얻었으며 DEAE-Sephacel column chromatography를 시행하였다. 여기서 얻은 GTP 결합단백질은 다시 Ultrogel AcA 34 column chromatography, heptylamine-Sepharose column chromatography 순으로 실시하여 $GTP{\gamma}S$와 결합하는 단백질 분획을 모았고 활성도와 일치하는 분획을 얻었다. 전기영동으로 관찰한 결과 $Go{\alpha}$가 분자량 39,000 dalton. $G{\beta}$가 36,000 dalton인 band를 확인하였고 나머지 다른 단백질도 함께 관찰되어 heptylamine-Sepharose 분획을 다시 DEAE-Sephacel column에 적용하여 순수한 band를 구하였다. GTP 결합단백질의 활성화는 GTP가 결합될 때 ${\alpha}$부분과 결합하고 ${\beta}{\gamma}$는 떨어져 나간다. 그러므로 heptylamine-Sepharose column분획의 활성도에서 $Go{\alpha}$의 band 분획과 곡선의 활성도가 일치하고 ${\beta}$는 곡선이 하향하는 분획에서 전기영동상에 관찰되었다.
The synthesis and methylation in vivo of myleline basic protein(MBP) during the mouse brain devlopment was found to be the highest in youngest brain and declined progressively in mature brains. The relative rate of protein synthesis and methylation was a maximal ration in the youngest brain, This high ratio was wdll correlated with the higher protein methylase I (PM I) activity in younger brains. The jimpy mouse is the most severely affected dysmyelinating mutant and is characterized by failure to incorporate MBP into myelin. sheath. The MBP-specific PM I activity in 15-, 18-, and 21-days old hemizygous jimpy mice(jp/y)brains decreased by 20, 50 and 75%, respectively. Myelin fraction with different degrees of compaction were isolated from bovine brain, the most compact myelin fraction exhibited higher methylaccepting activity than the less compact dense fractions.
마우스 뇌조직에서 지질과산화를 막아주는 것으로 알려진 청겨자채에 대해서 50%MeOH 추출물(2mg/mL)을 사용하여 소 뇌조직의 산화적스트레스를 유발한 ascorbate/Fe$^{3+}$ system에서 항산화 활성을 조사하였다. 청겨자채 추출물의 용매순차추출 후 TBARS 활성을 각각 용매 분획(100)$\mu\textrm{g}$/mL)별로 확인한 결과 각각의 용매 분획 중 n-BiOH 분획 (49%)이 가장 높았고, EtOAc(40%), H$_{2}$O(9%) 및 n-hexane (0%) 순이었다. n-BuOH 분획을 silica gel column chroma-tography를 반복하였으며, CE, TLC를 통해서 sinapic acid와 ferulic acid를 확인하였다. CE를 통하여 그 함량을 정량한 결과, 건조중량(g)당 sinapic acid는 24.3 ppm 이었고 ferulic acid는 42.8 ppm으로 나타났다. 최종 정제활성물질을 $^{1}$J-NMR(CDCL$_{3}$)와 $^{13}$C-NMR(CDCL$_{3}$)로 확인하였으며 sinapic acid methyl ester, ferulic acid acid와 ferulic acid와 RF치가 일치하는지 여부를 TLC상에서 비교하였다. 동일한 50$\mu\textrm{g}$/mL 농도에서 sinapic acid methyl ester는 40%, ferulic acid methyl ester 는 35%의 항산화 활성을 나타내었으며 이것은 시판되는 표준물질은 sinapic acid(22%)와 ferulic acid (18%)에 비해 약간 높게 나타났다. 이것은이 화합물에 라디칼을 소거하는 작용기와 관계가 있는 것으로 여겨진다. 청겨자채 추출물 중에도 항산화 활성이 비교적 널리 분포된 것으로 보아 이 성분들은 체내에서 단독으로 또는 상호작용으로 항산화 작용을 나타낼 것으로 예상된다.
Introduction: Although several types of calcium-phosphate coumpound have been frequently applied to osseous defects at maxillofacial area for many years, there is a controversy about its efficiency on bone conductivity comprared to xenograft bone substitute. Alloplastic carbonapatite has been introduced to improve disadvantages of hydroxyapatite and to mimic natural bone containing carbon elements. However, a preclinical study about its efficiency of osteoconductivity has not been reported. This study was performed to evaluate the early osteoconductive potential of synthetic carbonapatite with multiple pores relative to anorganic bovine xenograft. Materials and methods: Total 5 beagle dogs were used for maxillary augmentation model. The control (anorganic bovine xenograft) and experimental groups (synthetic carbonapatite) were randomly distributed in the mouth split design. After bone graft, all animals were sacrificed 4 weeks after surgery. Histological specimens with Masson Trichrome staining were made and histomorphometrically analysed with image analyser. The statistical analysis was performed using paired t-test. Results: In both groups, all animals had no complications. The experimental group showed relatively much new bone formation around and along the bone substitutes, whereas it was clearly reduced in the control group. The ratios of new bone area to total area, to material area and to the residual area excluding materials were higher in the experimental group ($0.13{\pm}0.03,\;0.40{\pm}0.13,\;0.20{\pm}0.06$ respectively) than in the control group ($0.01{\pm}0.01,\;0.03{\pm}0.02,\;0.03{\pm}0.03$, respectively). And the differences between both groups were statistically significant (p<0.001, <0.01, <0.01, respectively), while the ratio of material area to total area in two groups was not significant. Conclusion: Carbonapatite showed a high osteoconductivity in the early stage of bone healing compared to bovine derived anorganic bone substitute. This study suggests that this bone materials can be applied as a reliable bone substitute in the clinical treatment.
