Variation of Amylose Content Using dsRNAi Vector by Targeting 3'-UTR Region of GBSSI Gene in Rice (GBSSI 유전자 3'UTR 영역의 발현 억제 dsRNAi 벡터를 이용한 아밀로스함량 조절 벼 개발)

  • The amylose content of starch is a major factor in the texture of cooked cereal grains. Therefore, down-regulation of amylose synthesis is one of the alternative method to improve eating quality of rice. We developed transgenic rice plants designed to suppress granule-bound starch synthase I(GBSSI) gene using RNA interference(RNAi) technology. Transgenic plants with RNAi vector containing the 3'-UTR region of GBSSI showed a lower amylose content in rice endosperm than that of wild-type. The range of amylose content was 5.9~9.0% in the transgenic plants, whereas that of wild-type was 17.7~18.0%. Transgenic rices showed the decrease of short chain and the increase of long chain by analyzing chain length distribution of amylopectin in the endosperm. In the SEM micrographs, we found that compound starch granules in whole grains of the wild-type rice were readily split during fracturing, while the starch granules in RNAi-transgenic lines showed small voluminous, non-angular rounded bodies.

Ventx1.1 as a Direct Repressor of Early Neural Gene zic3 in Xenopus laevis

  • From Xenopus embryo studies, the BMP4/Smad1-targeted gene circuit is a key signaling pathway for specifying the cell fate between the ectoderm and neuro-ectoderm as well as the ventral and dorsal mesoderm. In this context, several BMP4/Smad1 target transcriptional factors have been identified as repressors of the neuro-ectoderm. However, none of these direct target transcription factors in this pathway, including GATA1b, Msx1 and Ventx1.1 have yet been proven as direct repressors of early neuro-ectodermal gene expression. In order to demonstrate that Ventx1.1 is a direct repressor of neuro-ectoderm genes, a genome-wide Xenopus ChIP-Seq of Ventx1.1 was performed. In this study, we demonstrated that Ventx1.1 bound to the Ventx1.1 response cis-acting element 1 and 2 (VRE1 and VRE2) on the promoter for zic3, which is a key early neuro-ectoderm gene, and this Ventx1.1 binding led to repression of zic3 transcription. Site-directed mutagenesis of VRE1 and VRE2 within zic3 promoter completely abolished the repression caused by Ventx1.1. In addition, we found both the positive and negative regulation of zic3 promoter activity by FoxD5b and Xcad2, respectively, and that these occur through the VREs and via modulation of Ventx1.1 levels. Taken together, the results demonstrate that the BMP4/Smad1 target gene, Ventx1.1, is a direct repressor of neuro-ectodermal gene zic3 during early Xenopus embryogenesis.

Selection of Vaccinia Virus-Neutralizing Antibody from a Phage-Display Human-Antibody Library

  • Although smallpox was eradicated in 1980, it is still considered a potential agent of biowarfare and bioterrorism. Smallpox has the potential for high mortality rates along with a major public health impact, eventually causing public panic and social disruption. Passive administration of neutralizing monoclonal antibodies (mAbs) is an effective intervention for various adverse reactions caused by vaccination and the unpredictable nature of emerging and bioterrorist-related infections. Currently, vaccinia immune globulin (VIG) is manufactured from vaccinia vaccine-boosted plasma; however, this production method is not ideal because of its limited availability, low specific activity, and risk of contamination with blood-borne infectious agents. To overcome the limitations of VIG production from human plasma, we isolated two human single-chain variable fragments (scFvs), (SC34 and SC212), bound to vaccinia virus (VACV), from a scFv phage library constructed from the B cells of VACV vaccine-boosted volunteers. The scFvs were converted to human IgG1 (VC34 and VC212). These two anti-VACV mAbs were produced in Chinese Hamster Ovary (CHO) DG44 cells. The binding affinities of VC34 and VC212 were estimated by competition ELISA to $IC_{50}$ values of $2{\mu}g/ml$ (13.33 nM) and $22{\mu}g/ml$ (146.67 nM), respectively. Only the VC212 mAb was proven to neutralize the VACV, as evidenced by the plaque reduction neutralization test (PRNT) result with a $PRNT_{50}$ of ~0.16 mg/ml (${\sim}1.07{\mu}M$). This VC212 could serve as a valuable starting material for further development of VACV-neutralizing human immunoglobulin for a prophylactic measure against post-vaccination complications and for post-exposure treatment against smallpox.

  • 'Governance' is a subject that is widely discussed these days in the government and the academic world. I think that the new concept, 'governance', is a strategy to develop the democracy of the society in its institutional and functional aspects. When we are going to discuss about governance, public records and their relationships, without understanding the meaning of 'public' in Korea, we cannot expect to discuss the matter precisely. It is said that Korean public service sectors are awash with authoritarianism and unreasonableness, and that they are at the center of seething corruption and bribe scandals. It is the legacy that the regimes adopted in the aftermath of the Japanese colonial rule for 35 years. The colonial legacy included not simply the practice of the Japanese colony, but also people who had collaborated the Japanese. The American military government and Rhee, Sung-Man regime also appointed the same officials to government posts. As was the same case in other areas including economy, press, education, politics, law, etc. In this point of view, "Righting historical wrongs", a controversial issue now in Korea, aims at establishing the right relationships between an individual and the public, and eventually laying the foundation of democracy for future generation, a procedure of achieving good governance. Apparently, Korea has made progress in developing democracy, as well as in reforming the government policy and organization. Many independent commissions are performing the projects instead of the government institutions that mandated to do the job, but has not played their roles. The e-government projects in Korea was launched in 2001 by the former administration. However, the confusion of records-management after the promulgation of the act is the result of the lack of strategy and the inconsistency of the vision. Good record-keeping supports effective, transparent and accountable government. Accountability is a key element of good governance. It is a recognized fact that without information, there is no guidance for decision-making, and accountability. Thus without records, there is no accountability for the decisions of actions. Transparency means that the decisions taken and their enforcement are carried out based on led-out rules and regulations. When both accountability and transparency are non-existent, good governance is bound to fail. Archival institutions have to give an attention to inner-governance because of the new trend of archival practice, namely 'macro-appraisal'. This 'macro-appraisal' is a kind of a functions-based approach. However, macro-appraisal focuses not just on function, but on the three-way interaction of function, structure and citizen, which combined reflect the functioning of the state within civil society, that is to say, its governance. In conclusion, the public and democracy are major challenges in the Korean society. The so-called good governance requires good record management. In this respect, records managers are in the front line of instituting good governance, and achieving better public and democracy for future generation, a procedure of achieving good governance.

Role of the Baseline Heart Rate Variability to the Effect of the Huanglian-Jie-Du Granule in Hwa-Byung Patients: Supplementary Analysis from the Randomized Trial Comparing Huanglian-Jie-Du Granule and Placebo for Hwa-Byung (화병 환자의 심박변이도 차이에 따른 황련해독탕의 효과: 화병에 대한 무작위 대조 비교임상시험 결과의 추가 분석)

  • Background: The purpose of the study was to explore whether the effect of Huanglian-jie-du granule depends on the baseline heart rate variability in patients with Hwa-byung. Methods: We used the supplementary data from a randomized, double-blind, placebo-controlled trial of Huanglian-jie-du granule for Hwa-byung patients. Study drugs were taken orally three times per day for one week. Heart rate variability was measured three times; before the treatment, after the treatment, and month follow-up period. Spearman's rho test was done to explore the role of the baseline heart rate variability to the effect of Huanglian-jie-du granule. Results: Baseline low frequency power of heart rate variability was negatively correlated with the reduction of the insomnia severity index in experimental group (r=-0.493 p=0.02). Also, Huanglian-jie-de granule significantly decreased the low frequency power of heart rate variability in patients with Hwa-byung compared to placebo (mean difference 266 [95% CI: 29~503], p=0.028). In Hwa-byung symptoms, stuffy, pushing up in the chest, and flush of anger were correlated with low frequency power of heart rate variability (p=0.010, p=0.000, and p=0.016, respectively). Conclusions: The effect of the Huanglian-jie-de granule for insomnia seems to be lower when the baseline low frequency power of heart rate variability is higher in Hwa-byung patients. Also, Huanglian-jie-de granule is likely to decrease the sympathetic activity in patients with Hwa-byung. Measurement of heart rate variability may be the useful to understand the state of Hwa-byung patients.

Key Structural Features of PigCD45RO as an Essential Regulator of T-cell Antigen Receptor Signaling (T-세포 항원 수용체 매개 신호전달 조절자로서 돼지 CD45RO 구조특성)

  • Pig CD45, the leukocyte common antigen, is encoded by the PTPRC gene and CD45 is a T cell-type specific tyrosine phosphatase with alternative splicing of its exons. The CD45 is a coordinated regulator of T cell antigen receptor (TCR) signal transduction achieved by dephosphorylating the phosphotyrosine of its substances, including $CD3{\zeta}$ chain of TCR, Lck, Fyn, and Zap-70 kinase. A dysregulation of CD45 is associated with a multitude of immune disease and has been a target for immuno-drug discovery. To characterize its key structural features with the effects of regulating TCR signaling, this study predicted the unknown structure of pig CD45RO (the smallest isoform) and the complex structure bound to the ITAM (REEpYDV) of $CD3{\zeta}$ chain via homology modeling and docking the peptide, based on the known human CD45 structures. These features were integrated into the structural plasticity of extracellular domains and functional KNRY and PTP signature motifs (the role of a narrow entrance into ITAM binding site) of the tyrosine phosphatase domains in a cytoplasmic region from pig CD45RO. This contributes to the selective recognition of phosphotyrosine from its substrates by adjusting the structural stability and binding affinity of the complex. The characterized features of pigCD45RO can be applied in virtual screening of the T-cell specific immunomodulator.

Arg-Leu-Tyr-Glu Suppresses Retinal Endothelial Permeability and Choroidal Neovascularization by Inhibiting the VEGF Receptor 2 Signaling Pathway

  • Vascular endothelial growth factor (VEGF) plays a pivotal role in pathologic ocular neovascularization and vascular leakage via activation of VEGF receptor 2 (VEGFR2). This study was undertaken to evaluate the therapeutic mechanisms and effects of the tetrapeptide Arg-Leu-Tyr-Glu (RLYE), a VEGFR2 inhibitor, in the development of vascular permeability and choroidal neovascularization (CNV). In cultured human retinal microvascular endothelial cells (HRMECs), treatment with RLYE blocked VEGF-A-induced phosphorylation of VEGFR2, Akt, ERK, and endothelial nitric oxide synthase (eNOS), leading to suppression of VEGF-A-mediated hyper-production of NO. Treatment with RLYE also inhibited VEGF-A-stimulated angiogenic processes (migration, proliferation, and tube formation) and the hyperpermeability of HRMECs, in addition to attenuating VEGF-A-induced angiogenesis and vascular permeability in mice. The anti-vascular permeability activity of RLYE was correlated with enhanced stability and positioning of the junction proteins VE-cadherin, ${\beta}$-catenin, claudin-5, and ZO-1, critical components of the cortical actin ring structure and retinal endothelial barrier, at the boundary between HRMECs stimulated with VEGF-A. Furthermore, intravitreally injected RLYE bound to retinal microvascular endothelium and inhibited laser-induced CNV in mice. These findings suggest that RLYE has potential as a therapeutic drug for the treatment of CNV by preventing VEGFR2-mediated vascular leakage and angiogenesis.

Material Properties and Conservation of Imjin Jangcho (Drafts of Imjin War Reports of Admiral Yi Sun-sin) of the 76th National Treasure of South Korea (국보 제76호 임진장초(壬辰狀草)의 지질분석과 보존처리)

  • The $76^{th}$ National Treasure is consisted of seven volumes of Nanjung Ilgi (War Diary of Admiral Yi Sun-sin; each titled by the author based on the zodiac name of the year covered, i.e. Imjin Ilgi, Gyesa Ilgi, Gabo Ilgi, Byeongsin Ilgi, Jeongyu Ilgi, Sok Jeongyu Ilgi, Musul Ilgi), Seogancheob (Letters of Admiral Yi) and Imjin Jangcho (Drafts of Imjin War Reports of Admiral Yi). These are currently in the custody of Hyeonchungsa shrine, the state-managed memorial shrine of Admiral Yi, and has been added to UNESCO's Memory of the World Register in June 2013 in the appreciation of its historical and academic importance as a commander's hand-written battlefield accounts during Japanese invasions of Korea. Imjin Jangcho, among these nine volumes, is a record of battles, including observations on the enemy fleets, suggestions on naval matters, battle expenditure, and methods of battle preparation, transcribed by someone else according to Admiral Yi's instructions. It is Admiral Yi's honest detailobservations on the war situation during Japanese invasions of Korea from 1592 as a Naval Commander of Left Jeolla Province, till 1594 (Year 27 of King Seonjo's reign) as a Commander-in-Chief of the Naval Forces of the South. It is a draft of the war report to be submitted to the King in a single volume as a folio (each sheet folded once to produce one leaf) bound in a side-stitched binding with five holes and inner binding of twisted paper. This study explores the condition and the treatment performed on Imjin Jangcho which could have an implication for the conservation of cultural assets on paper of late Joseon Dynasty.

Collapse Type and Processes of the Geumosan Caldera in the Southern Gumi, Korea (구미 남부 금오산 칼데라의 함몰 유형과 과정)

  • The Gumi basin, situated in the mid-southeastern Yeongnam Massif, has the Cretaceous stratigraphy that is divided into Gumi Formation, andesitic rocks (Yeongamsan Tuff, Busangni Andesite), rhyolitic rocks (Obongni Tuff, Doseongul Rhyolite, Geumosan Tuff) and Intrusives (ring dikes, other dikes) in ascending order. The Geumosan Tuff is composed mostly of many ash-flow tuffs which are associated with Geumosan caldera along with the ring dikes. The caldera is outlined by ring faults and dikes and has about 3.5 × 5.6 km in diameters. The intracaldera volcanics show a downsag structure that is dipped inward in their flow and welding foliations. The caldera block represent an asymmetric subsidence, which drops 350 m in the northern margin and 600 m in the southern one. Based on these data, the Geumosan caldera is geometrically classified as an asymmetric piston subsidence caldera that suggests a single caldera cycle. The caldera reflects the piston subsidence of the caldera block bounded by the outward-dipping ring faults following a voluminous eruption of magma from the chamber. The downsag in the caldera block refers to the downsagging during the initial subsidence at the same time as the full development of the bound fault. In the ring fissures following the sagging, magma was injected due to the overpressure of magma chamber caused by subsidence.

  • This study regards the need for jobs to overcome the poverty of the elderly in an aged society, and as well as researches ways to participate in active economic activities in old age. The results of the study are as follows. First, the legal system needs to be improved. In addition to institutional support for the expansion of jobs for the elderly, improvement of awareness of the elderly and reinforcement of infrastructure such as a delivery system are required, and a legal basis for this is needed. To this end, occupations targeting the elderly must be selected first, and legal procedures must be prepared by the government. Second, there must be social consideration and support for the elderly. Extension of the retirement age plays an important role in enhancing employment for the elderly. Along with the improvement of the legal system, social consideration is required. Therefore, since the elderly policy without social consensus is bound to fail, various support methods that can lead to social support must be devised. Third, the elderly must have a sense of poverty and willingness to rehabilitate themselves. Elderly people should not only ask for their children and social support. The elderly must be able to find a way to become the economic agent themselves. To do this, a basic awareness of the elderly poverty consciousness is needed, and a change of awareness to escape the poverty of the elderly is necessary. Therefore, the elderly need to be self-reliant, and they must choose jobs according to their own abilities. In conclusion, the elderly job program is a part of income increase that solves the poverty of the elderly in the aged society, and the elderly are required to actively participate in economic activities.
