In this study, we investigated a calcium phosphate-calcium sulfate injectable bone substitute (IBS) with organic reinforcement of chitosan, citric acid and hydroxypropyl-methyl-cellulose (HPMC). The powder component of IBS consisted of tetra calcium phosphate (TTCP), dicalcium phosphate dihydrate (DCPD) and calcium sulfate dihydrate (CSD). The liquid component was a solution of citric acid and chitosan. The effect of HPMC in terms of setting time, compressive strength and apatite forming ability on this IBS was investigated. The mass content of HPMC in liquid phase was varied in array of 0%, 2%, 3% and 4%. The setting times obtained between 20 and 45 minutes. Compressive strength was achieved over 20 MPa after incubation at 370C and in 100% humidity for 28 days. Porosities were evaluated in relation with compressive strength. Elastic moduli of the 28 days after-incubation IBS were obtained around 4GPa

This study was to investigate the caponization effects on bone characteristics in male chickens, and the optimum testosterone implantation dosage on bone characteristics improvement. Healthy Single Comb White Leghorn cockerels were caponized at 12-wk-old and selected at 16-wk-old for a 10-wk feeding experiment. Fifteen intact male and caponized male chickens (capon) respectively were assigned to trial 1. Ten sham-operated chickens and 40 capons (randomly allocated into four treatments) were implanted with cholesterol (1.62 mm i.d., 3.16 mm o.d., $9.24{\pm}0.36$ mg), low (1 mm i.d., 3 mm o.d., $5.88{\pm}0.23$ mg), medium (1.62 mm i.d., 3.16 mm o.d., $9.81{\pm}0.17$ mg) or high dose (2 mm i.d., 4 mm o.d., $16.7{\pm}0.24$ mg) of testosterone in trial 2. The results from trial 1 showed that the tibia length, relative tibia weight, breaking strength, bending moment and stress in intact males were higher than capons (p<0.05). The blood phosphorus concentration in capons was higher than the intact male chickens (p<0.05). Caponization also resulted in more antrums and osteoclasts within periosteum and cortical bone from histological observation. In trial 2, the adverse impact of caponization on the bone breaking strength, bending moment and stress could be alleviated through medium dose testosterone implantation. It appears that caponization reduced androgen secretion hence influenced the biomechanical characteristics of bone (tibia) and these adverse effects could be alleviated through appropriate dose of testosterone implantation.
Osteoporosis is a major worldwide public health problem that poses a great economic burden to society. Puerariae Radix, the dried root of Pueraria lobata (Wild.) Ohwi, has been widely used in Asia. This study investigated the effects of Puerariae Radix (PR) on bone loss in ovariectomized (OVX) mice. C3H/HeN mice (10 weeks old) were divided into sham and OVX groups. The OVX mice were treated with vehicle, $17{\beta}-estradiol$ ($E_2$), PR (oral administration, 250 mg/kg/day) or PR (intraperitoneal administration, 50 mg/kg/every other day) for 6 weeks. Grip strength, uterus weight, serum alkaline phosphatase (ALP), estradiol concentration and osteoclast surface levels were measured. Tibiae were analyzed using microcomputed tomography. There were no significant differences in the degree of grip strength, body weight and uterine weight between OVX group and PR-treated group. As compared with the OVX group, the serum estradiol levels were significantly increased in the PR-treated group. PR (i.p.) significantly preserved trabecular bone volume, trabecular bone number, structure model index and bone mineral density of proximal tibiae metaphysic. The administration of PR lowered serum ALP and osteoclast surface levels in OVX mice, suggesting that PR can reduce the bone turnover rate in mice. The results indicate that the supply of PR can prevent OVX-induced bone loss in mice.
Objectives: To select the optimal ranges showing obvious synergic anti-osteoporotic potential after adjust mixed formula consisted of Morindae Radix (MR) and Cistanchis Herba (CH) as compared with those of each single formula or risedronate sodium (RES) using bilateral ovariectomized (OVX) female mice. Methods: Fourteen groups, total eight sham or 104 OVX mice were selected based on the body weights at 34 days after OVX surgery. After that, 9 types mixed compositions, single formula of MR and CH, and RES were orally administered for 35 days. And we measured changes in body weight and gain, femur weight, bone mineral density (BMD), bone strength (failure load) and mineral content - calcium (Ca) and inorganic phosphorus (IP), osteocalcin contents and bone specific alkaline phosphatase (bALP) activities of all mice. Results: The OVX-induced estrogen-deficient osteoporotic signs were significantly inhibited by 35 days of continuous oral treatment of all treated mice as compared with OVX control mice. Especially, MR:CH 1:3 and 1:1 mixed formula treated mice showed significantly more favorable inhibitory activities against estrogen-deficient osteoporosis symptoms as compared to those of each single formula of MR and CH. Although RES also ameliorated the decreases of the femur BMD, strength and trabecular bone architectures through the inhibited the increases of bone turnover, but they did not critically influenced on the bone formations. Conclusions: The results suggest that MR:CH 1:3 mixed formula showed somewhat lower anti-resorptive effects as compared to those of RES, but they also showed bone formation effects. therefore, it is expected that MR:CH 1:3 mixture will be promising as a potent protective agents for relieving the osteoporosis in menopausal women.

Background: Osteosarcopenia is a common condition characterized by the loss of both bone and muscle mass, which can lead to an increased risk of fractures and disability in older adults. The study aimed to elucidate the response of various mouse strains to treatment with Rg3, one of the leading ginsenosides, on musculoskeletal traits and immune function, and their correlation. Methods: Six Collaborative Cross (CC) founder strains induced muscle atrophy and bone loss with dexamethasone (15 mg/kg) treatment for 1 month, and half of the mice for each strain were orally administered Rg3 (20 mg/kg). Different responses were observed depending on genetic background and Rg3 treatment. Results: Rg3 significantly increased grip strength, running performance, and expression of muscle and bone health-related genes in a two-way analysis of variance considering the genetic backgrounds and Rg3 treatment. Significant improvements in grip strength, running performance, bone area, and muscle mass, and the increased gene expression were observed in specific strains of PWK/PhJ. For traits related to muscle, bone, and immune functions, significant correlations between traits were confirmed following Rg3 administration compared with control mice. The phenotyping analysis was compiled into a public web resource called Rg3-OsteoSarco. Conclusion: This highlights the complex interplay between genetic determinants, pathogenesis of muscle atrophy and bone loss, and phytochemical bioactivity and the need to move away from single inbred mouse models to improve their translatability to genetically diverse humans. Rg3-OsteoSarco highlights the use of CC founder strains as a valuable tool in the field of personalized nutrition.
Using the favorable resorption behavior of amorphous Calcium phosphate (CaP) we fabricated a gelatin basednano fibrous mat by electrospinning for using as a fast healing patch for minorbone defects. Bone is predominantly formed by an inorganic phase of nano-crystalline HAp materials and nano fibrous protein material of collagen. The osteoblast cells, which are the bone formation cells and are key to the new bone formation, receive these materials to form new bone. Taking these considerations we make a new nano fibrous mat of amorphous CaP and gelatin, which is derived from collagen itself. A polymer carrier of poly caprolactone(PCL) was used in the system to stabilize the materials in biological condition. The electrospinning conditions were optimized for smooth mat without any droplet formation. The fabricated mat was characterized for its morphologyby SEM. Mechanical properties like tensile strength was evaluated. To investigate the bio-compatibility we performed the MTT assay and investigated its resorption behavior and apatite formation behavior by SBF immersion.
The microarchitecture of trabecular bone plays a significant role in mechanical strength due to its load-bearing capability. However, the complexity of trabecular microarchitecture hinders the evaluation of its morphological characteristics. We therefore propose a new classification method based on static multiscale theory and dynamic finite element method (FEM) analysis to visualize a three-dimensional (3D) trabecular network for investigating the influence of trabecular microarchitecture on load-bearing capability. This method is applied to human vertebral trabecular bone images obtained by micro-computed tomography (micro-CT) through which primary trabecular bone is successfully visualized and extracted from a highly complicated microarchitecture. The morphological features were then analyzed by viewing the percolation of load pathways in the primary trabecular bone by using the stress wave propagation method analyzed under impact loading. We demonstrate that the present method is effective for describing the morphology of trabecular bone and has the potential for morphometric measurement applications.
In recent years, quantitative ultrasound (QUS) technologies have played a growing role in the diagnosis of osteoporosis. Most of the commercial bone somometers measure speed of sound (SOS) and/or broadband ultrasonic attenuation (EUA) at peripheral skeletal sites. However, the QUS parameters are purely empirical measures that have not yet been firmly linked to physical parameters such as bone strength or porosity. In the present study, the theoretical models for wave propagation in cancellous bone, such as the Biot model, the stratified model, and the modified Biot-Attenborough (MBA) model, were applied to predict the dependence of phase velocity on porosity in cancellous bone. The optimum values for the input parameters of the three models in cancellous bone were determined by comparing the predictions with the previously published measurements in human cancellous bone in vitro. This modeling effort is relevant to the use of QUS in the diagnosis of osteoporosis because SOS is negatively correlated to the fracture risk of bone, and also advances our understanding of the relationship between phase velocity and porosity in cancellous bone.
The influx of Western culture and the food culture have altered the tableware of Korea, increasing the amount of using tableware. The representative tableware is known as bone china. Considering not only the amount of bone china waste generated from broken tableware at homes and but also faulty bone china products at factories, the bone china waste disposed in simple landfills is to be a serious environmental problem to solve. In this study, following "the research on development of ceramics body using waste celadon" of which the possibility was confirmed, development of ceramics body is conducted by recycling waste bone china. From the result of the study, it was understood that maximum addition of waste bone china was 20% of the whole material on the basis of measurement of plasticity. So 20% of pulverized bone china was added and the characteristics were evaluated, and the result was that porosity, water absorptivity and plasticity were 0.756%, 0.339% and 24.13%, respectively. Bending strength was $672\;kgf/cm^2$, which showed a good sintering condition. Therefore, it was considered that waste bone china could be utilized for development of new porcelain material.
