Anti-diabetic effect and mechanism of Korean red ginseng extract in C57BL/KsJ db/db mice

  • Purpose: Ginseng is a well-known medical plant used in traditional Oriental medicine. Korean red ginseng (KRG) has been known to have potent biological activities such as radical scavenging, vasodilating, anti-tumor and anti-diabetic activities. However, the mechanism of the beneficial effects of KRG on diabetes is yet to be elucidated. The present study was designed to investigate the anti-diabetic effect and mechanism of KRG extract in C57BL/KsJ db/db mice. Methods: The db/db mice were randomly divided into six groups: diabetic control group (DC), red ginseng extract low dose group (RGL, 100 mg/kg), red ginseng extract high dose group (RGH, 200 mg/kg), metformin group (MET, 300 mg/kg), glipizide group (GPZ, 15 mg/kg) and pioglitazone group (PIO, 30 mg/kg), and treated with drugs once per day for 10 weeks. During the experiment, body weight and blood glucose levels were measured once every week. At the end of treatment, we measured Hemoglobin A1c (HbA1c), blood glucose, insulin, triglyceride (TG), adiponectin, leptin, non-esterified fatty acid (NEFA). Morphological analyses of liver, pancreas and white adipose tissue were done by histological observation through hematoxylin-eosin staining. Pancreatic islet insulin and glucagon levels were detected by double-immunofluorescence staining. To elucidate an action of mechanism of KRG, DNA microarray analyses were performed, and western blot and RT-PCR were conducted for validation. Results: Compared to the DC group mice, body weight gain of PIO treated group mice showed 15.2% increase, but the other group mice did not showed significant differences. Compared to the DC group, fasting blood glucose levels were decreased by 19.8% in RGL, 18.3% in RGH, 67.7% in MET, 52.3% in GPZ, 56.9% in PIO-treated group. With decreased plasma glucose levels, the insulin resistance index of the RGL-treated group was reduced by 27.7% compared to the DC group. Insulin resistance values for positive drugs were all markedly decreased by 80.8%, 41.1% and 68.9%, compared to that of DC group. HbA1c levels in RGL, RGH, MET, GPZ and PIO-treated groups were also decreased by 11.0%, 6.4%, 18.9%, 16.1% and 27.9% compared to that of DC group, and these figure revealed a similar trend shown in plasma glucose levels. Plasma TG and NEFA levels were decreased by 18.8% and 16.8%, respectively, and plasma adiponectin and leptin levels were increased by 20.6% and 12.1%, respectively, in the RGL-treated group compared to those in DC group. Histological analysis of the liver of mice treated with KRG revealed a significantly decreased number of lipid droplets compared to the DC group. The control mice exhibited definitive loss and degeneration of islet, whereas mice treated with KRG preserved islet architecture. Compared to the DC group mice, KRG resulted in significant reduction of adipocytes. From the pancreatic islet double-immunofluorescence staining, we observed KRG has increased insulin production, but decreased glucagon production. KRG treatment resulted in stimulation of AMP-activated protein kinase (AMPK) phosphorylation in the db/db mice liver. To elucidate mechanism of action of KRG extract, microarray analysis was conducted in the liver tissue of mice treated with KRG extract, and results suggest that red ginseng affects on hepatic expression of genes responsible for glycolysis, gluconeogenesis and fatty acid oxidation. In summary, multiple administration of KRG showed the hypoglycemic activity and improved glucose tolerance. In addition, KRG increased glucose utilization and improved insulin sensitivity through inhibition of lipogenesis and activation of fatty acid $\beta$-oxidation in the liver tissue. In view of our present data, we may suggest that KRG could provide a solid basis for the development of new anti-diabetic drug.

폐쇄성 수면 무호흡 증후군 치료에서 구개수구개인두성형술의 임상적 유용성 (The Clinical Efficacy of Uvulopalatopharyngoplasty in the Treatment of Obstructive Sleep Apnea Syndrome)

  • 연구배경 : 구개수구개인두성형술(uvulopalatopharyngoplasty : UPPP) 은 폐쇄성 수면 무호흡 증후군 환자에서 가장 보편적으로 시행되고 있는 외과적 치료법이지만 치료 효과에 대하여는 논란의 여지가 많으며, 국내 연구 자료는 매우 부족한 실정이다. 방 법 : UPPP를 시행한 폐쇄성 수면 무호흡 증후군 환자들 중에서 일차 추적 수면다원검사가 가능했던 26명을 대상으로 단기 치료 효과를 관찰하였다. 치료 효과는 두가지 기준에 의해 평가하였다. 첫 번째는 일반적인 평가 기준으로써, UPPP 시행으로 무호흡지수(apnea index : AI) 혹은 무호흡-저호흡지수(apnea-hypopnea index : AHI)가 50% 이상 감소한 경우 혹은 UPPP 시행 후의 AI가 10 미만이거나 AHI가 20 미만인 환자들을 효과가 있는 경우 즉 반응군으로 정의하였고, 두 번째는 저자들이 설정한 평가 기준으로써, UPPP 시행 전의 AI 혹은 AHI와 관계없이 UPPP 시행 후의 AI가 5 미만이거나 AHI가 10 미만인 환자들을 단기적으로 폐쇄성 수면 무호흡 증후군이 치유된 경우 즉 치유군으로 간주하였다. 비만도, AI 및 AHI와 UPPP 시행에 의한 AI 및 AHI 감소율과의 관련성을 관찰하였고, 수면 구조를 비교함으로써 UPPP의 시행으로 수면의 질이 개선될 수 있는지 여부를 분석하였다. UPPP 시행 후 10일 이내에 발생한 문제점을 조기 합병증으로 정의 하였고, 10일 이후에 발생한 부작용은 후기 합병증으로 분류하였다. 결 과 : 26명의 환자 중에서 반웅군은 18명(69.2%) 이었고, 10명(38.5%)은 단기적으로 폐쇄성 수면 무호흡 증후군이 치유된 소견을 보였으며 5명(19.2%)은 UPPP 시행 후에 AI 혹은 AHI가 오히려 증가되었다. UPPP 시행 전의 비만도는 AI 감소용 및 AHI 감소율과 상호 관련성이 없었고, UPPP 시행 전의 AI는 AI 감소율과 상관관계가 없었으며, AHI 역시 AHI 감소율과 관련성이 없었다. 전체 대상 환자, 반웅군 및 비반웅군, 치유군 및 비치유군 각각에서 UPPP 시행 전후 수면 구조의 유의한 변화는 없었다. UPPP 시행에 따른 조기 합병증으로 비교적 심한 수술 부위의 통증, 음식물의 비역류, 기침이나 호흡곤란 등의 호흡기 증상, 수술 부위의 출혈, 연하곤란, 귀의 통증 혹은 봉합 부위 파열 등이 전체 환자(100%)에서 발생하였고, 후기 합병증으로 인후의 건조감, 비역류, 음성의 변화, 연하곤란, 인후의 이물감 혹은 미각변화 등이 22명(84.6%)의 환자에서 관찰되었으나, 합병증은 모두 경미한 것이었다. 결 론 : 폐쇄성 수면 무호흡 증후군 환자의 약 70%가 UPPP의 시행으로 호전을 보이지만, 치료 효과가 충분한 경우는 약 40%에 불과하였다. 치료 효과를 높이기 위해서는 엄격한 기준에 의한 적응증의 결정과 더불어 적절한 수술 방법이 필요할 것으로 생각한다.

조선왕릉 십이지신상(十二支神像)의 도상(圖像) 원류와 전개 과정 (On the Origin and Development of Iconography of the Twelve Zodiac Signs of Royal Tombs of Joseon Dynasty)

  • 조선왕릉은 조선왕조의 역사, 이념, 문화, 미술, 건축, 제례 등이 종합적으로 융합되어 만들어진 결정체이며, 500년 왕조의 역사를 고스란히 보여주는 산물로서 그 상징적 의의가 매우 크다. 왕릉을 구성하고 있는 여러 요소 중에서도 특히 능침 주변에 배치된 석물들은 능침을 수호하는 상징적 기능으로서 중요한 요소였으며, 그 중에서도 봉분 가장 가까이에 표현된 십이지는 그 기능의 가장 중심에 놓여져 있는 대상물이었다. 통일신라시대 능묘에서부터 시작된 십이지신상은 전해지는 규모나 뛰어난 조각수법으로 미루어 능묘 축조에 있어서 중요한 역할과 위치를 차지했었음이 확실하나, 고려시대를 거쳐 조선왕릉에 이르러서는 규모나 형태에서 매우 간소화된 경향을 띠게 되면서 다른 석물들에 비견하여 그 중요성이 간과되어 왔다. 십이지신상은 무덤을 수호하는 역할과 방위의 개념으로서 능묘를 장식했던 중요한 요소 중 하나였으며, 시대별로 다양하게 변화되어 나타나며, 조선왕릉까지 그 전통성이 이어진다는 점에서 역사성과 상징성은 간과할 수 없다. 본고에서는 조선왕릉에 표현된 십이지 도상(圖像)을 크게 3가지로 형식 분류하여 그 원류와 시기별 전개 과정을 고찰해 보았다. 십이지 도상은 사람의 몸과 얼굴을 표현한 인신인수형(人身人首形), 관에 십이지 동물을 표현한 인신인수관형(人身人首冠形), 십이지와 십간 팔괘의 혼합 문자형으로 분류된다. 조선왕릉의 이 3가지 십이지 도상은 그 원류를 중국으로부터 볼 수 있으며, 조선 전기에서 후기에 이르기까지 시대의 흐름에 따라 변화 교체되면서 왕릉을 장식하는 주요한 요소 가운데 하나로서 애용된다. 조선왕릉 십이지신상은 통일신라 이래 이어온 우리나라 능제의 고유성과 전통성을 확인할 수 있는 산 증거로서 그 의미가 크다 하겠다.

텍스트 데이터 시각화를 위한 MVC 프레임워크 (A MVC Framework for Visualizing Text Data)

  • 빅데이터의 중요성에 대한 인식이 확산되고, 관련한 기술이 발전됨에 따라, 최근에는 빅데이터의 처리와 분석의 결과를 어떻게 시각화할 것인지가 매우 관심 받는 주제로 부각되고 있다. 이는 분석된 결과를 보다 명확하고 효과적으로 전달하는 데에 있어서 데이터의 시각화가 매우 효과적인 방법이기 때문이다. 시각화는 분석 시스템과 사용자가 소통하기 위한 하나의 그래픽 사용자 인터페이스(GUI)를 담당하는 역할을 한다. 통상적으로 이러한 GUI 부분은 데이터의 처리나 분석의 결과와 독립될 수록 시스템의 개발과 유지보수가 용이하며, MVC(Model-View-Controller)와 같은 디자인 패턴의 적용을 통해 GUI와 데이터 처리 및 관리 부분 간의 결합도를 최소화하는 것이 중요하다. 한편 빅데이터는 크게 정형 데이터와 비정형 데이터로 구분할 수 있는데 정형 데이터는 시각화가 상대적으로 용이한 반면, 비정형 데이터는 시각화를 구현하기가 복잡하고 다양하다. 그럼에도 불구하고 비정형 데이터에 대한 분석과 활용이 점점 더 확산됨에 따라, 기존의 전통적인 정형 데이터를 위한 시각화 도구들의 한계를 벗어나기 위해 각각의 시스템들의 목적에 따라 고유의 방식으로 시각화 시스템이 구축되는 현실에 직면해 있다. 더욱이나 현재 비정형 데이터 분석의 대상 중 대부분을 차지하고 있는 텍스트 데이터의 경우 언어 분석, 텍스트 마이닝, 소셜 네트워크 분석 등 적용 기술이 매우 다양하여 하나의 시스템에 적용된 시각화 기술을 다른 시스템에 적용하는 것이 용이하지 않다. 이는 현재의 텍스트 분석 결과에 대한 정보 모델이 서로 다른 시스템에 적용될 수 있도록 설계되지 못하는 경우가 많기 때문이다. 본 연구에서는 이러한 문제를 해결하기 위하여 다양한 텍스트 데이터 분석 사례와 시각화 사례들의 공통적 구성 요소들을 식별하여 표준화된 정보 모델인 텍스트 데이터 시각화 모델을 제시하고, 이를 통해 시각화의 GUI 부분과 연결할 수 있는 시스템 모델로서의 시각화 프레임워크인 TexVizu를 제안하고자 한다.

포유류 로드킬 저감을 위한 고속도로 유도울타리 효율성 및 개선방안 연구 - 중앙고속도로 만종~홍천 구간을 사례로 - (The Efficiency and Improvement of the Highway Wild-Life Fences for Decrease of Mammals Road-kill - In Case of Manjong~Hongchun Section on Jungang Highway -)

  • 본 연구는 고속도로 야생동물 유도울타리 설치효과를 분석하여 적정한 유도울타리 설치 개선 및 관리방안을 제시하고자 중앙고속도로 만종분기점~홍천나들목구간을 대상으로 수행하였다. 고속도로에 설치된 유도울타리는 포유류의 도로침입 방지효과가 있었으며, 방음벽, 낙석방지울타리, 2단 가드레일과 분리방호벽도 로드킬 예방에 효과적인 시설이었다. 유도울타리의 로드킬 저감 효과는 출입문이 설치된 높이 1.5m의 울타리보다 출입문이 설치되지 않은 높이 1.0m 구간이 로드킬 감소효과가 높아 출입문의 유무가 로드킬 저감에 중요한 요인으로 판단되었다. 또한 도로시설물의 연결지점에서 로드킬 저감 효과가 컸으며 출입문이 있더라도 관리가 잘되는 지역도 로드킬 감소효과가 높았다. 유도울타리는 설치연장이 증가함에 따라 로드킬 감소율이 높았다. 유도울타리 끝 부분과 절토부 주변은 로드킬에 취약하여 이에 대한 개선이 요구되었다. 울타리 유지관리용 출입문은 설치간격이 가까울수록 로드킬 발생량이 높아 출입문 관리가 로드킬 저감에 효과가 많은 것으로 판단되었다. 고속도로 유도울타리 설치 개선방안은 암반의 급경사 지역 외에 모두 설치되어야 하고, 도로상의 낙석방지책, 2단 가드레일, 방호벽 등은 적정 시설개선을 통해 유도울타리로 연결 설치되어야 한다. 출입문을 설치할 경우 자동개폐식 또는 중력개폐식으로 설치하여 개방을 방지하고 야생동물 탈출로가 조성이 되어야 한다. 겸용생물이동통로는 토양층 복원이나 식생공간을 조성하여 생물이동통로화 하여야하며 로드킬로 포식자에 의한 2차 로드킬을 막기 위해 사고난 사체는 즉시 도로외부로 처리가 되어야 한다. 울타리는 최소 사고예방 목표지점으로부터 양방향 500m 이상 즉 1,000m 이상 설치되어야 하며 가급적 다음 도로시설인 교량, 통로박스 등까지 연결하여 야생동물을 생물이동통로로 안전하게 유도하여야 한다.

부산지역 일반주택에서의 라돈농도측정 (Radon concentration measurement at general house in Pusan area)

  • 1980년대 초까지 우리들은 라돈이 우리의 건강을 해친다는 생각을 하지 못하고 살아 왔다. 그러나 과학자들은 오래 전부터 우리가 사는 실내에 라돈 방사능의 위험이 도사리고 있다는 사실을 알게 되었다. 특히 우리나라에서는 라돈에 대한 위해와 인체에 미치는 영향에 대한 관심이 저조하다. 최근 들어 라돈 오염에 대한 의식을 가지고 서울 지하철의 일부 역, 학교 시설의 실내 공기, 주택 내 공기 중 라돈 문제의 중요성과 위험성에 대해 알리고 측정, 관리하는 관심을 가지게 되었다. 일반적으로 건물의 지반에서 방출된 라돈가스가 건물 바닥 갈라진 틈새 등을 통해 실내로 들어옴으로써 라돈이나 라돈낭핵종의 실내 공기 중 농도는 증가하게 된다. 따라서, 균열된 건물 바닥의 틈, 지하로부터 실내로 들어오는 상하수 파이프와 지반 사이에 틈새가 많을 수록 실내의 라돈 농도는 높아진다. 이와 같이, 라돈은 지각 뿐만 아니라 건축 자재물 상수, 취사용 천연가스 등을 통해서도 실내로 들어오지만 라돈의 85%이상은 지각으로부터 방출된 것이다. 폐암의 한 원인으로 지목 받는 라돈과 라돈 낭핵종에 의한 건강상의 위해는 토양 중 우라늄의 함량이 높은 지역과 광산의 갱내, 동굴, 주택과 같이 밀폐된 공간에서 특히 높아진다. 라돈 농도의 안전한 준위란 알 수 없으며 크든 작든 간에 항상 위험이 존재한다. 그러므로 중요한 것은 주택 및 건물 내에서 라돈의 농도를 낮춤으로써 폐암의 위험을 감소시키는 것이다. 따라서 일반 주택 라돈 농도 측정이 필요한 것으로 생각되어, 신틸레이터 라돈 모니터를 이용하여 월별로 라돈 농도를 측정하였다. 연구결과는 지상보다는 지하가 1년 내내 높게 나타났으며, 여름보다는 겨울이 높게 나타났다. 특히 미국 환경 보호청이 권고하는 주택 내 4 pCi를 넘는 달은 지하 내에서만 나타났으며, 12개월 중 4개월로 나타나 라돈 피폭 심각성을 알게 되었다. 그러므로 라돈에 관한 기준치의 설정과 규제 및 저감 대책의 마련이 시급하다는 생각이 들며, 라돈 농도 측정한 결과를 알리고자 한다.

인공동결공법 적용 후 융해에 따른 해성 점토지반의 역학적 특성 평가 (Evaluation of mechanical characteristics of marine clay by thawing after artificial ground freezing method)

  • 인공동결공법(artificial ground freezing method)은 연약지반 및 도심지에서의 지하구조물 시공에 적합한 차수 및 지반보강 공법이다. 인공동결공법은 동결관(freezing pipe)을 지중에 매설한 후 냉매(refregerant)를 순환시켜 대상 지반에 차수벽 및 지지체의 역할을 수행하는 동결벽체(frozen wall)를 형성한다. 그러나 간극수의 동결에 따른 간극수의 부피팽창은 지반의 변형을 야기시킬 수 있고, 시공완료 후 동결토의 융해에 따른 지반의 소성변형 및 입자의 재배치 등은 지반의 역학적 특성을 변화시킨다. 본 논문에서는 인공동결공법에 따른 해성 점토지반(marine clay)의 동결속도를 평가하기 위하여 인공동결공법 현장실증시험을 수행하였다. 현장실증시험은 지중에 3.2 m 깊이로 매설된 동결관 1공 내로 초저온 냉매인 액화질소를 순환시키는 방법으로 수행되었다. 또한, 원지반과 인공동결공법에 의해 동결/융해된 지반에 대한 피에조 콘 관입시험(piezo cone penetration test, CPTu) 및 공내재하시험(lateral load test, LLT)을 수행함으로써 동결/융해(freezing-thawing)에 따른 해성 점토지반의 강도 및 강성 특성의 변화를 평가하였다. 시험결과, 부피가 약 $2.12m^3$인 원기둥 모양의 동결체를 형성하는데 총 3.5일이 동안 약 11.9 ton의 액화질소가 소요되었다. 동결/융해에 따른 지반의 강도 및 강성 저하는 각각 48.5%, 22.7%로 산정되었다.

PL의 브랜드확장이 소비자태도에 미치는 영향에 관한 연구 : 모브랜드 적합도 인식 차이의 조절효과를 중심으로 (The Effect of Brand Extension of Private Label on Consumer Attitude - a focus on the moderating effect of the perceived fit difference between parent brands and an extended brand -)

  • 브랜드확장은 다양한 마케팅 영역 중에서도 전통적으로 활발하게 연구가 진행되어 왔던 영역으로서, 본 연구는 최근 그 중요성이나 활용도가 급증하고 있는 PL(Private Label)제품에 대해 브랜드확장의 개념을 활용하여 차별적으로 접근하고자 하였다. 최근 PL제품에 관한 마케팅연구가 활발하게 진행되고 있으나, 대부분 기존 틀에서 크게 벗어나지 못한 채 단순한 적용에 그치고 있으며, 특히 브랜드확장에 관련된 연구들에서도 PL시장의 특성을 제대로 반영하고 있다고 볼 수 없다. 특히 PL제품의 확장에 있어서는 두 가지 모브랜드가 존재할 수 있는데, 이에 대한 연구는 부재한 상황이다. 이에 본 연구에서는 확장 PL제품의 태도에 영향을 미치는 변수로서 두 가지 모브랜드인 유통업체와 기존 PL제품에 대한 태도를 제시하였다. 또한 개별 모브랜드가 PL제품의 태도에 미치는 영향은 개별 모브랜드와 확장 PL제품간 유사성에 의해 상이할 것이라고 제안하였으며, 유통업체와 기존 PL제품에 대한 태도에 영향을 미치는 변수로서 신뢰와 만족을 제시하였다. 분석결과 유통업체와 기존 PL제품에 대한 태도 모두 확장 PL제품의 태도에 유의한 영향을 미쳤으며 동시에 적합도 정도에 따라 그 영향력이 상이함도 실제 데이터를 통해 검증하였다. 즉 확장 PL제품의 태도는 모브랜드의 적합도가 보다 강하게 형성된 모브랜드의 영향을 더 크게 받는 것을 확인할 수 있었다. 이를 토대로 향후 PL제품을 확장할 경우 소비자가 보다 긍정적인 태도를 갖고 있는 모브랜드에 기초하여 해당 모브랜드와의 연상이 강하게 나타날 수 마케팅 전략을 구사할 필요가 있을 것이다.

Sensory Information Processing

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times.
