시엽 추출물이 카드뮴에 흡입폭로된 랫드의 독성에 미치는 영향 (Effects of Water Extracts of Persimmon Leaves to Cadmium Toxicity in Rats by Inhalation Exposure)

  • Experimental animals were divided into 5 groups; normal, cadmium control, and 3 experimental groups. Cadmium control and experimental groups were exposed to 1 mg/㎥ of cadmium aerosol in air by inhalation exposure for 6 hours/day, 5 days/week during 4 weeks. Dosages of 20, 40, and 80mg/kg of extracts of persimmon leaves were intraperitoneally injected to experimental groups respectively and several toxicological parameters and induction of metallothionein were measured from the rats that inhaled cadmium aerosol in air. The results of this study were as follows. Cadmium concentration that cadmium control and experimental groups were inhaled was 0.980±0.061 mg/㎥. Mass median diameter of cadmium aerosol for inhalation exposure was 4.93±0.483㎛. Cadmium content of normal group in lung was 0.088㎍/g and the highest cadmium content in lung, 55.492㎍/g was from 80mg/kg dose group. Cadmium concentration of normal group in blood was 0.348㎍/100㎖ and the highest cadmium concentration in blood, 2.642㎍/100㎖ was from cadmium control. Cadmium concentration of normal group in liver was 0.010㎍/g and the highest cadmium concentration in liver, 31.100㎍/g was from 20mg/kg dose group. Cadmium concentration of normal group in kidney was 0.030㎍/g and the highest cadmium concentration in kidney, 2.526㎍/g was from cadmium control. Cadmium concentration of normal group in intestine was O.064㎍/g and the highest cadmium concentration in intestine, 0.300㎍/g was from 80mg/kg dose group. The highest cadmium concentration in urine by week was 6.080㎍/day from 20mg/kg dose group in the fouth week and the highest cadmium concentration in feces by week was 341.731㎍/day from 20mg/kg dose group in the fouth week. Metallothionein concentration of normal group in lung was 5.769㎍/g and the highest in lung, 30.986㎍/g was from 80mg/kg dose group. Metallothionein concentration of normal group in liver was 38.856㎍/g and the highest in liver, 169.378㎍/g was from 40mg/kg dose group. Metallothionein concentration of normal group in kidney was 22.228㎍/g and the highest in kidney, 47.898㎍/g was from 80mg/kg dose group. Metallothionein concentration of normal group in intestine was 2.170㎍/g and the highest in intestine, 13.642㎍/g was from 80mg dose group.

일부 청소년들에서 흡연노출량에 따른 혈중 카드뮴 농도 (Blood Cadmium Concentration According to Exposure of Smoking in Adolescence)

  • Objectives: We surveyed the prevalence of smoking among the adolescent population and analysed the correlation of the two exposure biomarkers-concentration of blood cadmium and urinary cotinine-to the smoking status. Methods: Subjects were 193 middle and high school students in Chungnam province. Blood cadmium and urinary cotinine were compared by sex and smoking status. Smoking status were classified by either the concentration of urinary cotinine or subjective answering as a smoker in the questionnaire. Results: Smoking prevalence was 24.6%, 36.2% and 6.7% among all subjects, male and female subjects, respectively. Average smoking amount was 17.5 and 1.5 cigarettes per month among the male subjects and female subjects, respectively. Mean concentration of urinary cotinine among the male subjects was $135.57{\mu}g/{\ell}$, and that of female subjects was $116.59{\mu}g/$. Direct smokers showed higher concentration of urinary cotinine than those of indirect smokers, and subjects with higher urinary concentration showed higher prevalence of smoking, too. Mean concentration of blood cadmium was $0.0572{\mu}g/d{\ell}$ among the male subjects, and $0.0693{\mu}g/d{\ell}$ among the female subjects. Among the male subjects, both exposure biomarkers showed significant correlation to the smoking status, but among the female subjects urinary cotinine did not show significant correlation to the smoking status. Conclusion: Concentrations of these two biomarkers suggested that this population had significantly high evironmental tobacco smoking(ETS) and efficient stop-smoking programs to reduce ETS should be directed to this population.

인구집단의 비교를 통한 폐금속광산 지역 주민의 카드뮴 노출수준 및 건강영향평가 (Health Impact Assessment for Cadmium Exposure: Comparison of Residents around Abandoned Mines with the General Population)

  • Objective: We compared the level of blood cadmium exposure with health impacts by using data from a survey of residents near an abandoned mine and a national health survey. Methods: For this study, we used data from 7,046 individuals who participated in the Health Effects Survey of Abandoned Metal Mines (AMS, 2008-2011) and 6,871 individuals who participated in the Korea National Health and Nutrition Examination Survey IV-VI (KNHANES, 2008-2013). To evaluate the health impacts, the quartiles (S1 to S4) were classified according to blood cadmium concentration, and then the odds ratios of S2 to S4 over S1 for exceeding the reference values of renal function, blood pressure, and bone density were compared. Similarly, the odds ratio of AMS over KNHANES was confirmed. Results: In the AMS, adjusted for general characteristics, the geometric mean of blood cadmium concentration was 1.34 ㎍/L, which was statistically significantly higher than that of the KNHANES 1.22 ㎍/L (p<0.001). In the integrated data of AMS and KNHANES, the estimated odds ratio of S4 over S1 for exceeding the reference value was found to be 1.70 for serum creatinine, 1.71 for hypertension, and a T-score 2.02 for the tarsal bone. They were all statistically significant. Conclusion: Residents around abandoned metal mines had a higher blood cadmium concentration than the general population, and the odds for exceeding the reference values were higher for some health indicators. Continuous biomonitoring should be conducted for vulnerable areas such as around abandoned metal mines, and measures to reduce cadmium exposure and management of chronic diseases are needed.

철분부족아동과 정상아동의 혈액, 소변, 머리카락 속의 납, 카드뮴 수준 차이에 관한 연구 (The Difference of Lead and Cadmium Concentration of Blood, Urine and Hair between Children with Suboptimal Iron Status and Normal Children)

  • This study was performed to assess the lead and cadmium status of children residing in urban factory area and to see if there is a difference of lead and cadmium status between the group with normal iron status. The mean lead concentration of male hair was 9.55ppm which is significantly higher than 6.61 ppm that of female. The mean lead concentration of male urine sample was 0.04ppm, 10.3ppm and $48.14\mu{g/L}$, which is higher than 0.08ppm, 6.08ppm and $20.69\mu{g/L}$ of normal group, respectively. In a suboptimal group the proportion of children whose urinary lead is higher than normal$(35\mu{g/L)$was 87.3%, whereas 2.5% for a normal group. The mean cadmium concentration of male hair was 2.58ppm, which is higher than 2.48ppm that of a female. The mean cadmium concentration of erythrocyte, hair and urine was 0.25ppm, 2.65ppm and $38.83\mu{g/L}$, which is higher than 0.20ppm, 2.40ppm and $19.78\mu{g/L}$ of the normal group, respectively. The proportion of children whose urinary cadmium level is higher than the low limit of risk of cadmium intoxication$(40\mu{g/L)}$ was 21.4%, whereas 0% for a mormal group. Urinary lead and cadmium level showed significantly negative correlation with the RBC count, hematocrit, hemoglobin and serum ferritin, whereas they had significantly positive correlation with FEP/Hb(p<0.01). There was no correlation between the IQ and the level of lead and cadmium.

Time-dependent Changes of Cadmium and Metallothionein after Short-term Exposure to Cadmium in Rats

  • The time-dependent changes in cadmium (Cd) concentration were studied in Female Sprague-Dawley (SD) rats during and after Cd exposure via drinking water (10 and 50 ppm) for 30 days. The cadmium concentration in muscle, liver, kidney, blood plasma, and urine, and the metallothionein concentration in blood plasma were determined every 10 days during exposure and every 7 days after exposure for 3 weeks. The muscle Cd concentration did not change during, and neither after, exposure. The liver Cd concentration increased from 1.4 to 3.3 (at 10 ppm) and from 6.1 to 10.1 folds (at 50 ppm) during exposure and remained higher than those of controls in both groups even during post-exposure period. The kidney Cd concentrations were 2.3 to 5.1 (at 10 ppm) and 4.9-14.0 folds (at 50 ppm) higher than those of controls during exposure and also remained elevated during the post-exposure period. Plasma Cd concentrations were not significantly different from those of controls in both groups. Urine Cd concentrations were more than 2 folds (at 10 ppm) and 6.5 to 12.6 folds (at 50 ppm) higher than those of controls but rapidly decreased over the 7 days of withdrawal. Blood plasma metallothionein concentrations were more than 2.4 folds (at 10 ppm) and 3.1 to 7.4 folds (at 50 ppm), and they remained elevated till 7 days (10 ppm) and 14 days (at 50 ppm) after exposure. Our data support that Cd in urine could be a useful biomarker during Cd exposure period and metallothionein in blood plasma could be as a supportive biological marker for during and post Cd exposure.

  • 납땜공 22명(名)과 대조군으로서 의과대학생 28(명)名을 대상(對象)으로 혈액(血液)을 $5{\sim}10ml$ 채취하여 Cd, Pb 함량(含量)을 측정(測定)한 후 대조군에 대한 납땜공의 혈중(血中) 함량(含量)의 통계치(統計値)를 비교한 결과는 다음과 같다. 1. 혈중 Cd과 Pb의 분포양상은 대조군과 납땜공 모두 비정규분포를 보이고 있으며, 대조군보다 납땜공에서 좌비대칭(左非對稱)으로 심하게 치우치면서 예리한 봉(峰)을 보이고 있다. 2. 혈중 Cd과 Pb함량의 개체간 변이는 대조군과 납땜공 모두 심하였으며, 특히 Pb의 경우 대조군보다 남땜공에 서 심하였다. 3. 대조군에 대한 납땜공의 혈중 Cd과 Pb 함량의 평균치는 Cd과 Pb의 경우 각각 통계적으로 유의했다(Cd; p<0.01, Pb; p<0.01). 4. 혈중 Cd 함량의 상대비는 산술평균으로 1.36배(倍), 기하평균으로 1.39배(倍)였다. 헐중 Pb 함량의 상대비는 산술평균으로 1.53배(倍), 기하평균으로 1.30배(倍)였다. 5. 혈중 Cd과 Pb 함량사이의 상관관계는 대조군과 납땜공에서 각각(各各) 통계적으로 유의했다(controls; p<0.1, solderers; p<0.1).

송엽 추출물을 이용한 카드뮴 독성의 해독에 관한 연구 (A Study on Antitoxic Effects of Pine Leaf Extracts against Cadmium Toxicity in Rats)

  • This study was conducted to investigate the antitoxic effects of Pine Leaf extracts against cadmium toxicity. The experimental rats were divided into 5 groups, such as control group cadmium alone treatment group and simultaneous treatment groups of cadmium and three doses of Pine Leaf extracts. Each group was administered with different dose of Pine Leaf extracts such as 0.5 mg, 2.5 mg, 5.0 mg/kg wet weight in pallets for 12 weeks. Cadmium Chloride($CdCl_2$) was administered by 4 mg/kg body weight. The results were summarized as follows: The simultaneous administration of cadmium and Pine Leaf significantly more decreased cadmium concentration in liver tissues compared to the administration of cadmium only. When blood were measured, no significantly difference in haemoglobin, haematocrit, erythrocyte values compared to the administration of cadmium only, but RBC significantly more increased. The simultaneous administration of cadmium and Pine Leaf more no sigmificantly difference metallothionein concentration in liver than the administration of cadmium only. There were showed the histopathological slight changes in the liver and kidney tissues of rats.

국민건강영양조사를 이용한 성인의 혈중 중금속 농도와 생선 섭취 및 심혈관 질환과의 관련성-국민건강영양조사 (2008~2010년) (The association of heavy metals in blood, fish consumption frequency, and risk of cardiovascular diseases among Korean adults: The Korean National Health and Nutrition Examination Survey (2008-2010))

  • Limited studies are available concerning the effect of heavy metal exposure on cardiovascular diseases. As environmental pollution increases, food contamination, including heavy metal contamination of fish, also increases. However, researches based on the intake of heavy metals, cardiovascular disease, and fish intakes are inconclusive. We assessed an association of heavy metal exposure with cardiovascular disease and fish intake in a nationally representative group of general Korean adults. We used data from the combined 2008-2010 Korean National Health and Nutrition Examination Survey (KNHANES), and analyzed the data of 5,139 Koreans who participated in KNHANES. All participants were older than 20 years, and were diagnosed with stroke, ischemic heart disease, or hypertension. The mean blood cadmium, lead concentration, and mercury concentration of subjects were $1.07{\pm}0.01{\mu}g/L$, $2.49{\pm}0.02{\mu}g/dL$, and $5.19{\pm}0.08{\mu}g/L$, respectively. We used the survey logistic regression model to account for the complex sample design of the cardiovascular disease risk in order to estimate the odds ratios (OR). After adjusting for age, education, income, alcohol, smoking, and BMI, the increase of serum cadmium in blood was associated with the increase in the prevalence of hypertension. Further, the increase in blood cadmium concentration was associated with the increase of both systolic blood pressure (SBP) and diastolic blood pressure (DBP). Although higher fish intakes were significantly associated with higher blood mercury concentration (p for trend < 0.0001), fish intakes did not affect either blood cadmium or lead concentration. Comparing the highest fish intake group with the lowest intake group, the OR of hypertension was 0.31 (95% CI: 0.19-0.59) in the crude model for total fish. However, these associations were no longer significant after the adjustment for potential confounding factors. In conclusion, cadmium in blood was associated with an increased risk of hypertension in the general Korean adult population. However, we found no evidence of a clear relationship between cardiovascular disease and frequency of fish consumption.

치매환자 여부와 혈중 납, 카드뮴, 수은 및 망간 농도와의 상관성 (Correlation between Dementia and Lead, Cadmium, Mercury, and Manganese Concentrations in the Blood)

  • Objectives: The causes of dementia have been reported in various ways, but there has been little research on the interrelationship between heavy metals and dementia, and the results also show little consistency. Therefore, it is imperative to compare the levels of heavy metal exposure between the dementia-suffering group and a control group to confirm the correlation between the level of heavy metal exposure and the likelihood of dementia. Methods: In order to assess the dementia level of the elderly, the Global Deterioration Scale (GDS) and Mini Mental State Examination (MMSE) were applied. To analyze the concentration of heavy metals in the blood, blood was collected from the veins of study subjects and measured using Inductively Coupled Plasma-mass spectrometry (ICP-MS). Results: There was a statistically significant correlation between lead and manganese concentrations in the blood and the MMSE and GDS. It was found that there was a statistically significant correlation between cadmium concentration in the blood and the GDS, but the MMSE was less relevant. It was found that the blood mercury concentration and the MMSE and GDS were less relevant. The lead concentration in the blood was 0.95±0.74 ㎍/dL in the dementia patient group and 0.33±0.22 ㎍/dL in the normal group, while cadmium was 0.69±0.37 ㎍/L in the dementia group and 0.18±0.10 ㎍/L in the normal group. Mercury was 0.81±0.31 ㎍/L in the dementia group and 1.16±0.80 ㎍/L in the normal group. Manganese was 6.83±2.01 ㎍/L in the dementia group and 4.78±1.59 ㎍/L in the normal group. All of these show statistically significant differences. Conclusions: As the concentration of lead, cadmium and manganese in the blood increases, the MMSE scores and GDS scores were found to worsen, and it was confirmed that there is a correlation between heavy metal exposure and cognitive degradation.
