Effects of Fruit Extract Drink on Alcohol Metabolic Enzymes in Ethanol-treated Rats

  • Alcoholism is a significant global health problem. Alcohol dehydrogenase and aldehyde dehydrogenase play important roles in the metabolism of alcohol and aldehyde. In this study, we aimed to investigate the eliminatory effects of a fruit extract drink on alcohol metabolism in drunken Sprague-Dawley (SD) rats. Male SD rats were given a fruit extract drink or a commercial product (10 mL/kg) 30 min prior to 40% (5 g/kg) ethanol ingestion. To assay the effect of the fruit extract drink on blood ethanol concentration, blood samples were taken from the saphenous vein at 3 and 5 h after ethanol ingestion. The blood concentrations of alcohol, alcohol dehydrogenase, and aldehyde dehydrogenase were significantly lower in the fruit extract drink group than in the control group, in a time-dependent manner. However, the alanine aminotransferase and aspartate aminotransferase activities of all experimental groups were unaltered compared to those of the control group. These results suggested that fruit extract drink intake can have a positive effect on the reduction of alcohol, alcohol dehydrogenase, and aldehyde dehydrogenase concentrations in the blood and may alleviate acute ethanol-induced hepatotoxicity by altering alcohol metabolic enzyme activities.

  • This study was conducted to evaluate the hangover relieving effect of Sanghwang mushroom mycelium extract (SME). The extract showed 1,1-diphenyl-2-picrylhydrazyl radical scavenging effect in a concentration-dependent manner and high antioxidant capacity ($56.67{\pm}1.77%$) when administered at $120{\mu}g/mL$. In addition, SME significantly increased (p < 0.005) the aldehyde dehydronase (ALDH) activity ($126.03{\pm}9.11%$) when applied at 8 or $16{\mu}L/mL$. A locomotor activity test showed that the alcohol-water treated group showed significantly decreased motor activity at 90 min post-administration. However, the alcohol-SME treated group showed a 20-fold higher motor activity than that observed in the alcohol-water treated group at 90 min post-administration. Blood was harvested from each mouse at 90 min post-administration, and both alcohol and aldehyde concentrations were measured. The alcohol-SME treated group showed significantly lower (p < 0.5) alcohol ($120.13{\pm}12.83{\mu}g/mL$) and aldehyde ($7.26{\pm}1.22{\mu}g/mL$) concentrations than the values observed in the alcohol-water treated group. These results suggest that the hangover relieving effect of SME results from increased ALDH activity, which reduces the aldehyde concentration in the blood.

  • Alcohol and acetaldehyde concentrations were measured in the blood and brain of rats which were treated with 20% alcohol (control group) or co-administered 20% alcohol with ginseng extract residue roasted (test group). There was no change in blood alcohol concentration between control and test group. However, the brain alcohol concentration was lowered in the test group which was treated for seven days. The concentration of aldehyde in the brain and blood was lowered in the test group. The activities of monoamine oxidase b in various regions of brain were recovered to normal group in the test groups. However, the Quantities of naloxone binding receptors were not changed by ginseng extract residue roasted.

  • Alcohol is the most widely psychoactive drug and has known in almost all civilization since ancient time. Recently increase consuming alcoholic beverages, alcohol is on of the major public health problems in the world. Alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) play important roles in the metabolism of alcohols and aldehydes. The drink consists of medicinal herbs, Puerariae Radix, Phyllostachyos Folium, Citri Pericarpium, Polygonati Rhizoma, Rehmanniae Rhizoma (Vinegar), which have been widely used in oriental medicine. This study was designed to investigate effects of medicinal herbal drink (MHD) on alcohol metabolism in drunken SD rats subjects. In experiment, rats were treated to ethanol (EtOH, 3 g/kg, PO) at 60 min. after saline (CON) or MHD (1 ml/kg, PO) administration. The blood alcohol concentration (BAC), blood acetaldehyde concentration (BALC) activities of ADH, ALDH, AST and ALT were significantly decreased in MHD group than in control group as a time-dependent manner. And drinking water volume in MHD group with duplicate treatment, were significantly decreased than in CON group. These results suggested that MHD intake could give an influence upon the reduction in BAC and BALC may alleviate acute ethanol-induced hepatotoxicity by altering alcohol metabolic enzyme activities.

Effects of Phellinus spp. Extract on Alcohol Metabolic Enzymes in Alcohol-treated Rats

  • Alcoholism is a significant health problem in the world. The liver is the first and primary target organ for alcohol metabolism. Alcohol dehydrogenase and aldehyde dehydrogenase play important roles in the metabolism of alcohol and aldehyde. In this study, I aimed to investigate the eliminatory effects of a Phellinus spp. extract on alcohol metabolism in drunken Sprague-Dawley (SD) rats. Male SD rats were given Phellinus spp. extract at 30 min after 40% (5 g/kg) alcohol ingestion. To assay the effect of Phellinus spp. extract on blood alcohol concentration, blood samples were taken from the tail vein at 1, 3 and 5 h after alcohol ingestion. The concentrations of alcohol, alcohol dehydrogenase, and aldehyde dehydrogenase in Phellinus spp. extract treated rat were significantly lower than that of the control with a time-dependent manner. In addition, the alanine aminotransferase and aspartate aminotransferase activities of Phellinus spp. extract-treated groups were altered compared to those of the control group. These results suggest that Phellinus spp. extract intake can have a positive effect on the reduction of alcohol, alcohol dehydrogenase, and aldehyde dehydrogenase concentrations in the blood and may alleviate acute alcohol-induced hepatotoxicity by altering alcohol metabolic enzyme activities. Phellinus spp. extract is thus a good nutraceutical candidate.

약용식물 추출물의 숙취 해소 효과에 관한 연구 (Effect of medicinal plant extract for hangover relief)

  • The present study was performed to evaluate the effect of medicinal plant extract on relieving hangovers in mice administered alcohol. The animals were divided into three groups. Each group was treated with fermented plant extract, non-fermented plant extract, or water 30 min after consuming ethanol (2 mL/kg). A locomotor activity test showed that all groups had decreased motor activity until 40 min after plant extract administration. The mice treated with water had lower motor activity until 100 min post-administration. However, the group treated with non-fermented plant extract showed increased motor activity 40 min post-administration, and the higher activity level was maintained until 120 min post-administration. The animals treated with fermented plant extract had a level of motor activity between those of the groups treated with water or non-fermented plant extract. Blood was collected from each mouse 120 min post-administration and aldehyde concentration was measured. The group treated with non-fermented plant extract had a significantly higher (p < 0.05) aldehyde concentration than the other groups. These results demonstrate that the non-fermented medicinal plant extract helped alleviate hangovers 40 min after administration by reducing aldehyde concentrations in the blood.

청간산(淸肝散)이 흰쥐의 알코올 대사(代謝) 효소(酵素) 활성도(活性度)에 미치는 영향(影響) (Effects of Chunggansan on Detoxication of Alcohol by Activity of Enzyme in Rats)

  • Chunggansan was tested for the effects on detoxication mechanism of alcohol. Chunggansan was treated firstly into samples, and then ethanol intoxicated animal models were set with them. The administration of Chunggansan to the rats increased proportionally in alcohol dehydrogenase activities in liver in relation to the level of concentration and days of treatment. Especially, the alcohol dehydrogenase was the most active when the concentration of extract was 200mg/kg and it was 7th day. The enzyme activities of alcohol dehydrogenase and aldehyde dehydrogenase in liver highly increased in Chunggansan pre-medicating group compared to that of ethanol treated group. Also, the blood ethanol concentration in rats was considerably decreased. In conclusion, Chunggansan recovers the damage of liver due to acute alcohol intoxication by the increased enzyme activities of alcohol dehydrogenase and aldehyde dehydrogenase.

Mature silkworm powder reduces blood alcohol concentration and liver injury in ethanol-treated rats

  • Hangover due to alcohol consumption causes social and physical problems. There is a growing interest in edible insects worldwide. We have previously published a new technology to make hard mature silkworm, Bombyx mori, into edible form, steamed and freeze-dried mature silkworm larval powder (SMSP). In this study, AIN-76 or SMSP (0.1 and 1 g/kg rat body weight) containing diets in SD rats were pretreated for 2 weeks, and ethanol (3 g/kg rat body weight) was administered as an oral gavage and sacrificed after 3 hours. As a result, blood alcohol and aldehyde levels were significantly decreased in SMSP fed rats. In addition, liver injury markers, alanine aminotransferase (ALT) and alkaline phosphatase (ALP) were significantly decreased in SMSP group compared to ethanol group. $TNF-{\alpha}$, an inflammatory cytokine, and malondialdehyde (MDA), an oxidative stress marker, also showed a dose-dependent decrease in the group receiving SMSP. Conclusively, consumption of SMSP not only reduced hangover induced by ethanol, but also decreased liver damage, oxidative stress, and inflammatory response.

The Extract of Limonium tetragonum Protected Liver against Acute Alcohol Toxicity by Enhancing Ethanol Metabolism and Antioxidant Enzyme Activities

  • The protective effect of EtOAc fraction of Limonium tetragonum extract (EALT) against alcohol-induced hepatotoxicity was assessed following acute ethanol intoxication in Spraque-Dawley rats. EALT (200 mg/kg p.o.) was administrated once before alcohol intake (8 g/kg, p.o.). Blood ethanol concentration, and the activities of alcohol metabolic enzymes, alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) in the liver were measured. Also, the formation of malondialdehyde (MDA) and the activities of antioxidant enzymes, superoxide dismutase (SOD), glutathione peroxidase (GSH-px), catalase were determined after acute alcohol exposure. Pretreatment of rats received ethanol with EALT significantly decreased blood ethanol concentration and elevated the activities of ADH and ALDH in liver. The increased MDA level was decreased, and the reduced activities of SOD, GSH-px and catalase were markedly preserved by the treatment with EALT. This study suggests that EALT prevent hepatic injury induced by acute alcohol which is likely related to its modulation on the alcohol metabolism and antioxidant enzymes activities.

  • To investigate the effect of Feelch on alcohol metabolism, we measured both blood alcohol concentration in human and hepatic alcohol metabolizing enzyme activity in rats. The blood alcohol concentration in Feelch-ingested group was significantly lower than that in water-ingested group at 0, 40, and 80 minute after alcohol intake. The blood alcohol concentration between male and female taken 300ml of $21\%$ alcohol showed the significant differences; the peak value of blood alcohol concentration in male and female were $0.083\pm0.014\%\;and\;0.108\pm0.018\%$, respectively. In alcohol-fed rats, aldehyde dehydrogenase (ALDH) activity was significantly increased, whereas alcohol dehydrogenase (ADH) activity was not changed. In both Feelch-fed group and Feelch plus alcohol-fed group, ADH and ALDH activity were significantly increased as compared with each control group. Feelch decreased phospholipase $A_2$ activity and lipid peroxidation in hepatic tissue and activities of serum aminotransferases as compared with control. These results suggest that Feelch may have a hepatoprotective effects and this is likely due to lower blood alcohol concentration via the increment of hepatic ADH and ALDH activity.
