국내 가금 농장에서 야생조류 침입 억제 장치를 이용한 야생조류 출현율 조사 및 평가 (Investigation and evaluation of appearance ratio using wildbird intrusion prevention device in domestic fowl farms)

  • The epidemiological investigation of outbreak in korea confirmed that the inflow of avian influenza (AI) is related to the migration of migratory birds. In this study, avian repellents instrument were implemented and developed using the visual effects of lasers in accordance with the situation of small domestic fowl farms, and monitoring cameras were installed around each instrument to investigate the frequency of wild birds appearing and evaluate the performance of the instrument. Observation showed that the appreance ratio was reduced by 95%, and no significant reduction in the intrusion prevention effect by adaptation was observed on all fowl farms. In conclusion, it is expected that the outbreak of wild bird-borne infectious diseases such as avian influenza will be decreased if the device is installed on domestic fowl farms.

  • More than 3.4 million people use Hong Kong metro a day. It was first constructed in 1979, and extended to 9 operating lines with 168.1km length operated by Hong Kong Mass Transit Railway Corporation (MTR). In Hong Kong, airborne viruses such as avian influenza (bird's flu) and severe acute respiratory syndrome (SARS), air pollution by high-density traffics, bacteria and mold due to relatively high humidity are major concerns on air quality management. In this study, we focused on the cases for improving indoor air quality conducted by Hong Kong MTR currently. The case includes the photocatalytic coatings on knobs (or hand straps) to prevent outbreak of bird's flu and SARS, the high voltage ionizers installed at metro platform, and the periodical radon monitoring.

Communications Protocol Used in the Wireless Token Rings for Bird-to-Bird

  • We developed a multicast communication packet radio protocol using a time-sharing tablet system ("wireless token ring") to achieve the efficient exchange of files among packet radio terminals attached to swans. This paper provides an overview of the system and the protocol of the packet communications. The packet device forming the main part of the transceiver developed is the Texas Instruments CC2500. This device consists of one call-up channel and one data transmission channel and could improve error frame correction using FEC (forward error correction) with 34.8 kbps MSK and receiving power of at least -64 dBm (output 1 dBm at distance of 200 m using 3 dBi antenna). A time-sharing framework was determined for the wireless token ring using call sign ordinals to prevent transmission right loss. Tests using eight stations showed that resend requests with the ARQ (automatic repeat request) system are more frequent for a receiving power supply of -62 dBm or less. A wireless token ring system with fixed transmission times is more effective. This communication protocol is useful in cases in which frequency resources are limited; the energy consumed is not dependent on the transmission environment (preset transmission times); multiple terminals are concentrated in a small area; and information (position data and vital data) is shared among terminals under circumstances in which direct communication between a terminal and the center is not possible. The method allows epidemiological predictions of avian influenza infection routes based on vital data and relationships among individual birds based on the network topology recorded by individual terminals. This communication protocol is also expected to have applications in the formation of multiple in vivo micromachines or terminals that are inserted into living organisms.

지리정보시스템 기반의 고병원성 조류인플루엔자 발생 위험지도 구축 (A GIS-Based Mapping to Identify Locations at Risk for Highly Pathogenic Avian Influenza Virus Outbreak in Korea)

  • Six major outbreaks of highly pathogenic avian influenza (HPAI) occurred from 2003 to 2016 in Korea. Epidemiological investigations of each outbreak revealed that migratory birds were the primary source of the HPAI virus. During the last five years, the geographic transmission pattern of domestic HPAI seems to have extended from local to nationwide; therefore, it is necessary to identify specific locations in which poultry farms are at elevated risk for HPAI outbreak to enable targeted surveillance and other mitigation strategies. Here, a geographical information system (GIS)-based analysis was used to identify geographic areas at high risk for future HPAI incidents in Korea based on historical outbreak data collected between December 2003 and April 2016. To accomplish this, seven criteria were used to identify areas at high-risk for HPAI occurrence. The first three criteria were based on defined spatial criteria buffering of 200 bird migration sites to some defined extents and the historical incidence of HPAI outbreaks at the buffering sites. The remaining criteria were based on combined attribute information such as number of birds or farms at district levels. Based on the criteria established for this study, the most-likely areas at higher risk for HPAI outbreak were located in Chungcheong, Jeolla, Gyeonggi, and Gyeongnam provinces, which are densely populated poultry regions considered major poultry-production areas that are located along bird migration sites. The proportion of areas at risk for HPAI occurrence ranged from 4.5% to 64.9%. For the worst criteria, all nine provinces, including Jeju Island, were found to be at risk of HPAI. The results of this study indicate that the number of poultry farms at risk for HPAI outbreaks is largely underestimated by current regulatory risk assessment procedures conducted for biosecurity authorization. The HPAI risk map generated in this study will enable easy use of information by policy makers to identify surveillance zones and employ targeted surveillance to reduce the impact of HPAI transmission.

CDR 자료를 이용한 고병원성 조류인플루엔자 분석 (The Analysis of HPAI Using CDR Data)

  • 이 연구는 2018년도 정부(농림축산식품부)의 재원으로 농림식품기술기획평가원 지원을 받아 수행된 연구이다. 고병원성 조류인플루엔자의 유입은 해외로부터 철새를 통해 유입되고 있으나 어떤 경로를 통해 감염 원인을 농장에 제공하는지 정확히 밝혀진 바 없다. 그리고 발생 농장으로부터 농장 간의 전이도 차량이 주 원인이라고 추정할 뿐, 전파 주 원인이 정확히 밝혀진 것은 아니다. 본 논문 연구에서는 KT가 제공하는 CDR(Call Detailed Record) 데이터를 기반으로, 발생후보지로 추정되는 철새도래지를 방문한 사람이 어떤 경로를 거쳐 감염 농장으로 유입이 되는지 해당 시기의 인구 흐름을 보려고 한다.

Identification of Differentially Expressed Genes in Ducks in Response to Avian Influenza A Virus Infections

  • 본 연구는 고병원성 조류 인플루엔자 바이러스(high pathogenic avian influenza virus; HPAIV)와 저병원성 조류인플루엔자 바이러스(low pathogenic avian virus; LPAIV)가 감염된 오리의 폐세포에서 보고된 기존 전사체 데이터를 재분석하여 조류 인플루엔자 감염에 대응하는 숙주의 공통 전사체를 발굴하고, 생물정보 분석을 실시하여 바이오 마커로서 가능성을 제시하기 위하여 수행하였다. 이전 연구에서 생산된 microarray 데이터 세트를 재분석하여, HPAIV와 LPAIV가 각각 감염된 오리의 폐세포에서 각각 총 731 및 439개의 차등발현 유전자를 발굴하였다. 이들 차등발현 유전자 중에서, 227개의 유전자가 HPAIV와 LPAIV가 감염된 세포에서 공통적으로 조절되어, 193개의 유전자는 발현이 증가한 반면, 34개의 유전자는 발현이 감소하였다. 생물정보 분석을 통하여 차등발현 유전자들의 기능에 대한 주석달기를 실시하여, 리보솜과 단백질 대사 및 유전자 발현 관련 GO가 풍부해짐을 확인하였다. REACTOME 분석을 통하여 단백질 및 RNA 대사 경로 및 콜라겐 생합성과 변형을 포함한 조직 복구 경로가 조절됨을 확인하였다. 보다 구체적으로, 번역 및 RNA 품질 관리 경로에 관여하는 단백질을 코딩하는 유전자는 HPAIV 및 LPAIV 감염에 반응하여 발현의 증가 또는 감소하는 방향으로 조절되어 AIV가 숙주 번역 기계를 억제함으로써 숙주 방어 시스템을 회피할 수 있거나 번역을 위해 세포질로 내보내기 전에 AIV가 억제될 수 있음을 시사한다. AIV 감염은 바이러스 감염으로 인한 조직의 병변 형성을 조절하는 경로를 활성화시킬 수 있음을 시사한다.

H7 아형 조류인플루엔자 바이러스의 유전자 특성 (Genetic Characterization of H7-subtype Avian Influenza Viruses)

  • 조류인플루엔자 바이러스 H7 subtype에 속하는 바이러스 중 일부는 가금류에 감염할 경우 고병원성이 발휘된다. 또 H7 아형 AIV중 일부는 사람에 감염하여 사망 등을 유발할 수도 있다. 본 연구는 야생조류로부터 분리된 H7 아형 조류인플루엔자 바이러스 6주(H7N7 아형 4주, H7N1 아형 2주)를 대상으로 8개 유전자 분절 전체의 염기서열을 분석하여 병원성, 사람 감염 가능성 등 그 특성을 조사하였다. 계통유전학적 분석결과, 국내에서 분리된 H7 아형 분리주들은 8개 유전자(HA, NA, PB2, PB1, PA, NP, M, NS) 모두 Eurasian lineage로 분류되었으나, Eurasian lineage 내에서도 각기 다른 sublineage로 분류되어 유전적 다양성이 있는 것으로 분석되었다. 한국 분리주 6주는 HA 단백질 분절부위 아미노산은 두 종류(PEIPKGR 및 PELPKGR)의 motif를 가지고 있었으나, 모두 저병원성 바이러스 특성을 가지고 있었다. 숙주세포 결합 특이성과 관련 있는 HA 단백질 receptor-binding site를 분석한 결과, 한국 분리주 모두는 사람 세포 수용체 결합특이성보다는 조류 세포 수용체 결합 특이성을 가지는 것으로 나타났다. 사람 감염 가능성을 높게 하는 부위에서의 아미노산 치환(PB2 단백질의 E627K 및 PB1단백질의 I368V)도 나타나지 않았고, 또한 NA stalk region에서의 결손도 관찰되지 않았다. 이상의 결과를 미루어 볼 때 한국 야생조류에서 분리된 H7 아형 6주 모두는 저병원성 바이러스로 최근 중국에서 사람 감염이 나타나고 있는 H7N9 바이러스와는 유전적으로 다른 계열의 바이러스인 것으로 판단된다.

Reverse transcription loop-mediated isothermal amplification assay for the rapid and simultaneous detection of H5 and other subtypes of avian influenza viruses

  • A two-tube reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay was designed for the rapid visual detection of the M gene of all subtypes of avian influenza virus (AIV) and the H5 gene of the H5 subtype of highly pathogenic AIV (HPAIV). The reaction carried out in two tubes in a single step at $58^{\circ}C$ for 40 min, and the assay results could be visually detected by using hydroxynaphthol blue dye. Using M or H5 gene-specific primers, the assay successfully detected all subtypes or H5 subtypes of AIVs, including the Korean representative H5N1 and H5N8 HPAIVs. The detection limit of the assay was approximately $10^{2.0}$ $EID_{50}/reaction$ for the M and H5 genes of H5N1 HPAIV, respectively, and was more sensitive than that of previously reported RT-LAMP and comparable to that of real-time RT-PCR. These results suggest that the present RT-LAMP assay, with its high specificity, sensitivity, and simplicity, will be a useful diagnostic tool for surveillance of currently circulating H5 HPAIVs and other subtypes of AIV in bird population, even in under-equipped laboratories.

Combination of multiplex reverse transcription recombinase polymerase amplification assay and capillary electrophoresis provides high sensitive and high-throughput simultaneous detection of avian influenza virus subtypes

  • The pandemic of avian influenza viruses (AIVs) in Asia has caused enormous economic loss in poultry industry and human health threat, especially clade 2.3.4.4 H5 and H7 subtypes in recent years. The endemic chicken H6 virus in Taiwan has also brought about human and dog infections. Since wild waterfowls is the major AIV reservoir, it is important to monitor the diversified subtypes in wildfowl flocks in early stage to prevent viral reassortment and transmission. To develop a more efficient and sensitive approach is a key issue in epidemic control. In this study, we integrate multiplex reverse transcription recombinase polymerase amplification (RT-RPA) and capillary electrophoresis (CE) for high-throughput detection and differentiation of AIVs in wild waterfowls in Taiwan. Four viral genes were detected simultaneously, including nucleoprotein (NP) gene of all AIVs, hemagglutinin (HA) gene of clade 2.3.4.4 H5, H6 and H7 subtypes. The detection limit of the developed detection system could achieve as low as one copy number for each of the four viral gene targets. Sixty wild waterfowl field samples were tested and all of the four gene signals were unambiguously identified within 6 h, including the initial sample processing and the final CE data analysis. The results indicated that multiplex RT-RPA combined with CE was an excellent alternative for instant simultaneous AIV detection and subtype differentiation. The high efficiency and sensitivity of the proposed method could greatly assist in wild bird monitoring and epidemic control of poultry.

Mucosal Administration of Lactobacillus casei Surface-Displayed HA1 Induces Protective Immune Responses against Avian Influenza A Virus in Mice

  • Avian influenza is a serious threat to both public health and the poultry industry worldwide. This respiratory virus can be combated by eliciting robust immune responses at the site of infection through mucosal immunization. Recombinant probiotics, specifically lactic acid bacteria, are safe and effective carriers for mucosal vaccines. In this study, we engineered recombinant fusion protein by fusing the hemagglutinin 1 (HA1) subunit of the A/Aquatic bird/Korea/W81/2005 (H5N2) with the Bacillus subtilis poly γ-glutamic acid synthetase A (pgsA) at the surface of Lactobacillus casei (pgsA-HA1/L. casei). Using subcellular fractionation and flow cytometry we confirmed the surface localization of this fusion protein. Mucosal administration of pgsA-HA1/L. casei in mice resulted in significant levels of HA1-specific serum IgG, mucosal IgA and neutralizing antibodies against the H5N2 virus. Additionally, pgsA-HA1/L. casei-induced systemic and local cell-mediated immune responses specific to HA1, as evidenced by an increased number of IFN-γ and IL-4 secreting cells in the spleens and higher levels of IL-4 in the local lymphocyte supernatants. Finally, mice inoculated with pgsA-HA1/L. casei were protected against a 10LD50 dose of the homologous mouse-adapted H5N2 virus. These results suggest that mucosal immunization with L. casei displaying HA1 on its surface could be a potential strategy for developing a mucosal vaccine against other H5 subtype viruses.
