Molecular and Biochemical Characteristics of ${\beta}$-Propeller Phytase from Marine Pseudomonas sp. BS10-3 and Its Potential Application for Animal Feed Additives

  • Phytate is an antinutritional factor that impacts the bioavailability of essential minerals such as $Ca^{2+}$, $Mg^{2+}$, $Mn^{2+}$, $Zn^{2+}$, and $Fe^{2+}$ by forming insoluble mineral-phytate salts. These insoluble mineral-phytate salts are hydrolyzed rarely by monogastric animals, because they lack the hydrolyzing phytases and thus excrete the majority of them. The ${\beta}$-propeller phytases (BPPs) hydrolyze these insoluble mineral-phytate salts efficiently. In this study, we cloned a novel BPP gene from a marine Pseudomonas sp. This Pseudomonas BPP gene (PsBPP) had low sequence identity with other known phytases and contained an extra internal repeat domain (residues 24-279) and a typical BPP domain (residues 280-634) at the C-terminus. Structure-based sequence alignment suggested that the N-terminal repeat domain did not possess the active-site residues, whereas the C-terminal BPP domain contained multiple calcium-binding sites, which provide a favorable electrostatic environment for substrate binding and catalytic activity. Thus, we overexpressed the BPP domain from Pseudomonas sp. to potentially hydrolyze insoluble mineral-phytate salts. Purified recombinant PsBPP required $Ca^{2+}$ or $Fe^{2+}$ for phytase activity, indicating that PsBPP hydrolyzes insoluble $Fe^{2+}$-phytate or $Ca^{2+}$-phytate salts. The optimal temperature and pH for the hydrolysis of $Ca^{2+}$-phytate by PsBPP were $50^{\circ}C$ and 6.0, respectively. Biochemical and kinetic studies clearly showed that PsBPP efficiently hydrolyzed $Ca^{2+}$-phytate salts and yielded myo-inositol 2,4,6-trisphosphate and three phosphate groups as final products. Finally, we showed that PsBPP was highly effective for hydrolyzing rice bran with high phytate content. Taken together, our results suggest that PsBPP has great potential in the animal feed industry for reducing phytates.

Long-Term Monitoring of Noxious Bacteria for Construction of Assurance Management System of Water Resources in Natural Status of the Republic of Korea

  • Climate change is expected to affect not only availability and quality of water, the valuable resource of human life on Earth, but also ultimately public health issue. A six-year monitoring (total 20 times) of Escherichia coli O157, Salmonella enterica, Legionella pneumophila, Shigella sonnei, Campylobacter jejuni, and Vibrio cholerae was conducted at five raw water sampling sites including two lakes, Hyundo region (Geum River) and two locations near Water Intake Plants of Han River (Guui region) and Nakdong River (Moolgeum region). A total 100 samples of 40 L water were tested. Most of the targeted bacteria were found in 77% of the samples and at least one of the target bacteria was detected (65%). Among all the detected bacteria, E. coli O157 were the most prevalent with a detection frequency of 22%, while S. sonnei was the least prevalent with a detection frequency of 2%. Nearly all the bacteria (except for S. sonnei) were present in samples from Lake Soyang, Lake Juam, and the Moolgeum region in Nakdong River, while C. jejuni was detected in those from the Guui region in Han River. During the six-year sampling period, individual targeted noxious bacteria in water samples exhibited seasonal patterns in their occurrence that were different from the indicator bacteria levels in the water samples. The fact that they were detected in the five Korea's representative water environments make it necessary to establish the chemical and biological analysis for noxious bacteria and sophisticated management systems in response to climate change.

Construction of Research Fronts Using Factor Graph Model in the Biomedical Literature (팩터그래프 모델을 이용한 연구전선 구축: 생의학 분야 문헌을 기반으로)

  • This study attempts to infer research fronts using factor graph model based on heterogeneous features. The model suggested by this study infers research fronts having documents with the potential to be cited multiple times in the future. To this end, the documents are represented by bibliographic, network, and content features. Bibliographic features contain bibliographic information such as the number of authors, the number of institutions to which the authors belong, proceedings, the number of keywords the authors provide, funds, the number of references, the number of pages, and the journal impact factor. Network features include degree centrality, betweenness, and closeness among the document network. Content features include keywords from the title and abstract using keyphrase extraction techniques. The model learns these features of a publication and infers whether the document would be an RF using sum-product algorithm and junction tree algorithm on a factor graph. We experimentally demonstrate that when predicting RFs, the FG predicted more densely connected documents than those predicted by RFs constructed using a traditional bibliometric approach. Our results also indicate that FG-predicted documents exhibit stronger degrees of centrality and betweenness among RFs.

A Quantitative Analysis on PLoS ONE Articles Published by Authors Affiliated with Korean Institutions (PLoS ONE 학술지 게재 국내 기관 소속 연구자 논문의 계량적 분석)

  • This research provides a quantitative analysis on research articles published in PLoS ONE, a multidisciplinary open access journal, by authors affiliated with Korean institutions. Korean authors published more than 6,500 research ariticles in the mega journal between 2006 and 2019. Korea is ranked the top 11th place in terms of article publishing in the journal. Most articles by Korean authors are concentrated in the biomedical fields. In recent years, the overall production of PLoS ONE has decreased as authors migrated to competing mega journals such as Scientific Reports and BMJ Open. The change might have been affected in part by the delay in the review period and the dropping impact factor score. The open access share of the Korean PLoS ONE authors of more than 10 articles hovers around 30%. However, there is a significant variation among researchers reaching up to 50% discrepancies. Among altmetrics provided by PLoS ONE, the saves are highly correlated with the views and the citations. On the contrary, the shares show low correlation with other use metrics. A follow up, survey questionnarie based research involving researchers who have published in PLoS ONE is planned in order to investigate author motivation and experience in the review process.

Structural Study of the Cytosolic C-terminus of Vanilloid Receptor 1

  • Vanilloid receptor I [transient receptor potential vanilloid subfamily member 1 (TRPV1), also known as VR1] is a non-selective cationic channel activated by noxious heat, vanilloids, and acid, thereby causing pain. VR1 possesses six transmembrane domain and N-and C-terminus cytosolic domains, and appears to be a homotetramer. We studied the structural properties of Cterminus of VR1 (VR1C) using CD and NMR spectroscopy. DPC micelles, with a zwitterionic surface, and SDS micelles, with a negatively charged surface, were used as a membrane mimetic model system. Both SDS and DPC micelles could increase the stability of helical structures and/or reduce the aggregation form of the VR1C. However, the structural changing mode of the VR1C induced by the SDS and DPC micelles was different. The changes according to the various pHs were also different in two micelles conditions. Because the net charges of the SDS and DPC micelles are negative and neutral, respectively, we anticipate that this difference might affect the structure of the VR1C by electrostatic interaction between the surface of the VR1C and phospholipids of the detergent micelles. Based on these similarity and dissimilarity of changing aspects of the VR1C, it is supposed that the VR1C probably has the real pI value near the pH 7. Generally, mild extracellular acidic pH ($6.5{\sim}6.8$) potentiates VRI channel activation by noxious heat and vanilloids, whereas acidic conditions directly activate the channel. The channel activation of the VRI might be related to the structural change of VR1C caused by pH (electrostatic interactions), especially near the pH 7. By measuring the $^1-^{15}N$ TROSY spectra of the VR1C, we could get more resolved and dispersed spectra at the low pH and/or detergent micelles conditions. We will try to do further NMR experiments in low pH with micelles conditions in order to get more information about the structure of VR1C.

Nutrients and Salt Consumption of Hypertension Patients According to Treatment Status (고혈압환자에서 치료상태에 따른 영양소 및 나트륨 섭취 양상)

  • High blood pressure is an important determinant of the incidence of coronary heart disease, stroke, congestive heart failure, renal failure, and peripheral vascular disease. Recommendations for control of high blood pressure emphasize lifestyle modification, including weight control, reduced sodium intake, increased physical activity. Subjects who were normotensive (n=19, $47.2\pm9.0$ y, BP l16/81 mmHg) ,treatment hypertensive (n=33, $54.2\pm6.9$ y, BP 132/85 mmHg) and non-treatment hypertensive (n=14, $50.1\pm11.0$ y, 149/94 mmHg) recruited. Anthropometric assessment (height weight waist circumference, hip circumference, fat$\%$, fat mass, and lean body mass) and dietary assessments (using 3-days food records, daily nutrient intakes were inuysed by CAN PRO 2.0 were carried out. Blood and 24-hour urine were collected). Test of recognition for salt taste threshold were performed. In non-treatment hypertensive male subjects, weight, $\%$IBW, BMI, and waist circumference were significantly higher than those of normotensive and treatment hypertensive subjects (p<0.05) .Food habits were not significantly different among the three groups. Intakes of vitamin A, vitamin B,, and vitamin B, were significantly higher in normotensive group (p<0.05). Intakes of sodium and salt taste recognition threshold were the highest in normotensive group and the lowest in treatment hypertensive group (p<0.05). Blood levels of lipids and minerals were not significantly different among the three groups. Urinary calcium level of normotensive group were significantly higher than that of treatment hypertensive and non-treatment hypertensive groups (p<0.05). These results indicate that continuous management of hypertension by drug and non-drug treatment affects salt taste recognition threshold and reduced the consumption of sodium. However, dietary sodium intake exceed recommended sodium intake to prevent and treat hypertension. It is necessary to develop the lifestyle modification program that may have beneficial effects on hypertension treatment.

The Development of Pc Based EGG-NIBP Patient Monitor (PC 기반의 심전도-비관혈식 혈압 환자감시장치의 개발)

  • In this paper, an ECG-NIBP patient monitor is designed. This is an essential equipment to measure and monitor patient's physical condition - electrocardiogram(ECG) wave, heart rate(HR), and noninvasive blood pressure(NIBP) - in ICU, CCU, and operating room. The ECG is an electrical waveform produced by relaxation and contraction of the cardiac muscle. Most physicians diagnose patient's cardiac states from ECG pattern. A blood pressure is one of the clinical indexes measured in a emergency room or operating room. In this paper, the blood pressure is measured in artery by using the nonivasive oscillometric method. The developed patient monitor was inspected and compared with other instruments in operating rooms. The results were 1bpm of maximum difference in the heart rate, 15mmHg in the systolic pressure, 16mmHg in the diastolic pressure, and 25mmHg in the mean blood pressure. But the total results were 0.15bpm of the mean difference in the heart rate, 5mmHg in the systolic pressure, 10mmHg in the diastolic pressure, and 9mmHg in the mean blood pressure. The designed ECG-NIBP patient monitor can measure the ECG wave, HR, and BP. And the multi-tasking module of pulse oximetry . respiration . temperature monitor will be added in the near future.

Effect of Electrical Stimulation using ABR and ECochG Analysis based on Jastreboff Tinnitus Mocel (Jastreboff 이명 모델에서의 ABR과 ECochG 신호분석을 통한 전기자극의 효과)

  • Many researches have been performed whether electrical stimulation could be used for diagnosis and treatment on the auditory system impairment. Unfortunately, there were no standard methods or theoretical background for choosing stimulus conditions because of the lack of understanding on the transmission of electrical stimulation through the auditory pathway. This research was conducted to observe the effect of electrical stimulation on the tinnitus-induced animals. Nine guniea pigs were used for the experment and divided into two groups, five animals for the experimental group(A) and four animals for the control group(B). Experimental conditions were divided into four steps, before tinnitus induction and 1, 6, 12 hours after tinnitus induction using salicylate based on the Jastreboff model. In each experimental condition, ABR and ECochG were obtained, and autocorrelation coefficients were calculated from normalized waveforms based on rms values. Sum of all the autocorrelation coefficients was extracted as a parameter to observe the changes between before and after the electrical stimulation. As a result, ABR parameter values were rapidly increased 6 hours after tinnitus induction, the gradually returned back to the initial state. On the other hand, when electrical stimulation was applied, parameter values did not change compared with the initial sate. Parameter values of ECochG showed that the effect of electrical stimulation appeared 12 hours after the tinnitus induction. It was concluded that an electrical stimulation to the tinnitus-induced model changes the correlation coefficients of ABR and ECochG waveforms.

Computer Interface for the Disabled Using Gyro-sensors and Artificial Neural Network (자이로 센서와 인공신경망을 이용한 장애인용 컴퓨터)

  • This paper aims at developing 'gyro-mouse' which provides decent and comfortable human-computer interface that supports the usage of such software as an internet-browser in PC for the people paralyzed in upper limbs. This interface operates on information collected from head movement to get the cursor control. The interface is composed of two modules. One is hardware module in which the head horizontal and vertical angular velocities are detected and transmitted into PC. The other is a PC software that translates the received data into movement and click signals of the mouse. The ANN (artificial neural network) learns the quick nodding pattern of each user as click input so that it can provide user-friendly interface. The performance of the system was evaluated by three indices that are click recognition rate. error in cursor position control. and click rate of the moving target box. The performance result of the gyro-mouse was compared with that of the optical-mouse to assess the efficiency of the gyro-mouse. The average click recognition rate was 93%, average error in cursor position control was 1.4∼5 times of optical mouse. and the click rate with 50 pixels target box was 40%(30 clicks/min) to that of optical mouse. The click rate increased monotonously with the number of trial from 35% to 44%. The suggested system is expected to provide a new possibility to communicate with the society.

Analysis of Muscle Activities and Driving Performance for Manipulating Brake and Accelerator Pedal by using Left and Right Hand Control Devices (장애인용 핸드컨트롤을 이용한 가속 및 제동 페달을 동작할 때의 상지 근육 EMG 분석 및 운전 성능 평가)

  • The purpose of this study was to investigate the EMG characteristics of driver's upper extremity and driving performance for manipulating brake and accelerator pedal by using left and right hand control devices during simulated driving. The people with disabilities in the lower limb have problems in operation of the motor vehicle because of functional loss for manipulating brake and accelerator pedal. Therefore, if hand control device is used for adaptive driving controls in people with lower limb impairments, the disabled people can improve their quality of life by driving a motor vehicle. Six subjects were participated in this study to evaluate driving performance and muscle activities for operating brake and accelerator pedal by using two different hand controls (steering column mounted hand control and floor mounted hand control) in driving simulator. We measured EMG activities of six muscles (posterior deltoid, middle deltoid, triceps, biceps, flexor carpi radialis, and extensor carpi radialis) during pushing and pulling movement with different hand controls for acceleration and braking. STISim Drive 3 software was used for the performance test of different hand control devices in straight lane course for time to reach target speed and brake reaction time. While pulling the hand control lever toward the driver, normalized EMG activities of middle deltoid, triceps and flexor carpi radialis in subjects with disabilities were significantly increased (p < 0.05) compared to the normal subjects. It was also found that muscle responses of posterior deltoid were significantly increased (p < 0.05) when using the right hand control than left hand control. While pushing the hand control lever forward away from the driver, normalized EMG activities of posterior deltoid, middle deltoid and extensor carpi radialis in subjects with disability were significantly increased (p < 0.05) compared to the normal subjects. It was shown that muscle responses of middle deltoid, biceps and extensor carpi radialis were significantly increased when using the right hand control than left hand control. Brake reaction time and time to reach target speed in subjects with disability was increased by 12% and 11.3% on average compared to normal subjects. The subjects with physical disabilities showed a tendency to relatively slow acceleration at the straight lane course.
