일본 바이오매스 활용 정책 및 기술현황 (Status of Technology and Policy for the Utilization of Biomass in Japan)

  • Based on the general policy called "Green Growth", the Korean government planed to establish a biomass town in South Korea in order to recover energy from organic waste and to substitute for fossil fuel at rural region. Technical and financial support for the establishment of biomass town was insufficient so far. There are some policies to support biomass town establishment, however financial support from several Korean ministries seemed not to have been used efficiently. Some policies are planned excessively so that they cannot be realized on time. Therefore, there is a need to analyze the status of biomass utilization technology and policy in Japan from the point of view of an external biomass expert, since biomass utilization technology and policy of Japan take good achievement during the many developed countries. For the analyzing of technology and policy in Japan, literatures concerned biomass management policy and biomass town design were collected by visiting Japan Ministry of Agriculture, Forestry and Fisheries and interview of public officials in charge was carried out. There are several implications for the promotion of Korean policy concerned with biomass utilization and biomass town establishment.

Biochemical Characterization of a GDSL-Motif Esterase from Bacillus sp. K91 with a New Putative Catalytic Mechanism

  • The esterase gene Est8 from the thermophilic bacterium Bacillus sp. K91 was cloned and expressed in Escherichia coli. The monomeric enzyme exhibited a theoretical molecular mass of 24.5 kDa and an optimal activity around $50^{\circ}C$ at pH 9.0. A model of Est8 was constructed using a hypothetical YxiM precursor structure (2O14_A) from Bacillus subtilis as template. The structure showed an ${\alpha}/{\beta}$-hydrolase fold and indicated the presence of a typical catalytic triad consisting of Ser-11, Asp-182, and His-185, which were investigated by site-directed replacements coupled with kinetic characterization. Asp-182 and His-185 residues were more critical than the Ser-11 residue in the catalytic activity of Est8. A comparison of the amino acid sequence showed that Est8 could be grouped into the GDSL family and further classified as an SGNH hydrolase. Est8 is a new member of the SGNH hydrolase subfamily and may employ a different catalytic mechanism.

  • In compare with a major developed countries, Korea consumes much energy, but also eliminates much carbon dioxide. Agricultural sector eliminate much carbon dioxide than industrial sector. In Kyoto protocol, Korea needs to reduce carbon dioxide. One way to reduce carbon dioxide is utilization of biomass in rural area. This paper focus on utilization of biomass in rural area. If use 20% potential amount of biomass, it obtain 50% of TPES on agricultural sector. The condition of utilization biomass is connected with agricultural policy, environmental policy, and energy policy. And environmental restriction keep pace with economical incentive.

지역단위 미이용 바이오매스 발생량 추정 (Estimation of Community-Based Unused Biomass Generation)

  • This study was carried out to evaluation of biomass generation mechanisms and to propose the estimation method of biomass generation. Agricultural by-product biomass is generated during crops cultivation and after harvest. However these are not uniformly generated yearly and these depending on the seasons. For planning of biomass utilization, accurate information of the biomass resources is needed, especially characteristic and productivity of biomass are necessary. Agricultural by-product biomass are generated in a wide area being scattered and it is one of the major reason why agricultural biomass utilization is not activated compared with other waste biomass. In this study, estimation and evaluation biomass generation is achieved in specific spatial and temporal boundary, A-city in Gyeongi-do and september to November respectively. Quantity and quality of by-product biomass show big difference depending on the crop species and cultivation periods and these difference bring up that accurate biomass estimation should be considered during planning of biomass utilization and technology selecting for biomass converting to energy and other forms.

목질바이오매스 에너지 부산물(리그닌)이용에 관한 연구 동향 (A Research Trend on Utilization of the Byproducts(Lignin) from Bioethanol Production Process with Lignocellulosic Biomass: A Literature Review)

  • This study reviewed on the research trend of sources and utilization of the byproducts(Lignin) from bioethanol production process with lignocellulosic biomass such as wood, agri-processing by-products(corn fiber, sugarcane bagasse etc.) and energy crops(switch grass, poplar, Miscanthus etc.). During biochemical conversion process, only Cellulose and hemicellulosic fractions are converted into fermentable sugar, but lignin which represents the third largest fraction of lignocellulosic biomass is not convertible into fermentable sugars. It is therefore extremely important to recover and convert biomass-derived Lignin into high-value products to maintain economic competitiveness of cellulosic ethanol processes. It was introduced that lignin types and characteristics were different from various isolation methods and biomass sources. Also utilization and potentiality for market of those were discussed.

Identification and Characterization of a New Alkaline SGNH Hydrolase from a Thermophilic Bacterium Bacillus sp. K91

  • est19 is a gene from Bacillus sp. K91 that encodes a new esterase. A comparison of the amino acid sequence showed that Est19 has typical Ser-Gly-Asn-His (SGNH) family motifs and could be grouped into the SGNH hydrolase family. The Est19 protein was functionally cloned, and expressed and purified from Escherichia coli BL21(DE3). The enzyme activity was optimal at 60℃ and pH 9.0, and displayed esterase activity towards esters with short-chain acyl esters (C2-C6). A structural model of Est19 was constructed using phospholipase A1 from Streptomyces albidoflavus NA297 as a template. The structure showed an α/β-hydrolase fold and indicated the presence of the typical catalytic triad Ser49-Asp227-His230, which were further investigated by site-directed mutagenesis. To the best of our knowledge, Est19 is a new member of the SGNH hydrolase family identified from thermophiles, which may be applicable in the industrial production of semisynthetic β-lactam antibiotics after modification.

  • 본 연구에서는 대체에너지로서의 바이오매스에너지원의 정의와 특성 등을 살펴보고, 국내에서의 바이오매스 에너지원의 잠재력과 이용가능성에 대해서 검토하였다. 특히 임지잔재, 간벌재 등의 목질바이오매스는 자원량이 풍부하고, 임업 임산업의 활성화와 인공림의 간벌작업을 촉진하여 산림의 공익적 기능을 증대시킬 수 있는 에너지 자원으로서, 앞으로 지속적인 산림관리를 위한 새로운 임업의 과제가 될 것으로 판단된다.

  • As a carbon neutral fuel, biomass can be converted into various types of high-valued products such as synthetic natural gas (SNG), Hydrogen, Fischer - Tropsch (FT) diesel. and valuable chemicals. In order to make above mentioned products, gasificaion process is essential as energy utilization platform of solid biomass. In this study, state of the art and prospect for biomass gasification technologies are presented.

  • 임산바이오매스 자원 이용의 일환으로 율추(栗皺) 열수추출물의 디에틸에테르 가용부의 HPLC 분석결과, 표준물질과의 retention time이 일치한 것을 RI detector로 얻어진 각 피크의 스펙트럼을 표준물질과 비교하고, 순도를 확인하는 방법으로 피크의 동정을 실시하였다. 피크의 동정 결과, phenolic acids와 aldehyde류로는 gallic acid, 3,5-dihydroxybenzoic acid, 2,4,6-trihydroxybenzoic acid, 그리고 protocatechualdehyde이었으며, flavonoids는 catechin과 epicatechin으로 모두 6종이 확인되었다.
