In this study was selected the cellulolytic microorganism and investigated optimum condition of cellulase production for the cellulosic bioethanol production. A bacterial strain Paenibacillus jamilae BRC15-1, was isolated from soil of domestic reclaimed land. For optimizing cellulase production from the selected strain, various culture parameters were investigated such as culture medium, pH (pH 4~10), temperature ($25{\sim}50^{\circ}C$) and culture time (2~72 h). As a result, P. jamilae BRC15-1 efficiently produced cellulase from cellulosic biomass under following conditions: 24 h of culture time (pH 7, $40^{\circ}C$) in manufactured media of CMC (carboxymethyl cellulose) with peptone. Optimum saccharifying condition of crude enzyme produced from P. jamilae BRC15-1 was identified on pH 6 and $40^{\circ}C$ of reaction temperature, respectively. This crude enzyme from P. jamilae BRC15-1 was used for saccharification of pretreated sweet sorghum (Sorghum bicolor var. dulciusculum Ohwi) bagasse under the optimal condition. Finally, pretreated sweet sorghum bagasse including 0.1 g of glucan was saccharified by crude enzyme of P. jamilae BRC15-1 into 2.75 mg glucose, 0.79 mg xylose and 1.12 mg arabinose.
억새와 같은 초본계 바이오매스로부터 cellulose, hemicellulose, lignin 등 주요성분을 추출하기 위해서는 알칼리 전처리가 효율적이며, 본 연구에서는 수산화칼륨(KOH)을 이용한 전처리 조건을 최적화하였다. 전처리 변수의 최적화는 반응표면분석법(RSM)을 적용하였다. RSM의 변수는 3개였으며, 변수범위는 각각 KOH 0.2~0.8M, 반응온도 110~190℃ 및 반응시간 10~90min 이었다. 억새의 알칼리 전처리를 위한 최적조건은 KOH 농도 0.47M, 반응온도 134℃ 및 반응시간 65min 이었다. 최적 전처리 조건에 따라 전처리를 수행한 후 고형물의 cellulose 함량은 66.1±1.1% 이었으며, hemicellulose 및 lignin 함량은 각각 26.4±0.4%, 3.7±0.1% 이었다. RMS 모델식에 따라 계산된 예측값은 실제값 대비 95% 범위 내에서 유효하였다. 최종적으로 전처리물을 동시당화발효를 통해 검증한 결과 에탄올 생산 수율은 96% 이었다.
본 논문에서는 감마선 조사가 NaOH 전처리를 이용한 우드칩의 당화에 미치는 영향을 살펴보았다. 당화량은 전처리된 우드칩에 효소를 이용하여 가수분해한 후 생성된 환원당과 포도당 양을 측정하여 확인하였다. 10 g/L NaOH 처리한 우드칩을 각각 50, 100, 200 kGy 세기의 감마선을 조사하여 당화량을 측정하였다. 우드칩의 가수분해 수율을 비교했을 때, 200 kGy 감마선 조사한 우드칩에서 가장 높은 환원당 함량 12.2 g/L이 얻어졌다. 또한 전처리 과정에서의 감마선 조사 단계의 영향을 평가하기 위하여 우드칩을 먼저 50, 100, 200 kGy 감마선 조사한 후, 10 g/L NaOH 로 전처리하였다. 200 kGy 감마선 조사 후 NaOH 전처리를 수행했을 때 환원당 함량은 13.4 g/L로 전처리 이후의 감마선 조사구보다 증가하였고, 7.9 g/L 포도당을 얻을 수 있었다. 이러한 결과로부터 감마선 조사가 셀루로오즈 바이오매스 전처리에 활용 가능할 것으로 기대된다.
억새의 식물학적인 분류 및 형태, 선진국의 억새 육종, 재배 및 이용 현황 그리고 우리나라의 억새 연구성과 등을 고찰함으로서 향후 우리나라 에너지 자급율 제고를 위한 억새 연구방향을 제시하고자 국내외 연구결과를 요약하면 다음과 같다. 억새속 식물에는 17개종이 존재하는데 이 중 바이오 에너지용으로 중요한 종은 한국 등 동아시아가 원산인 참억새(M. sinensis)와 물억새(M. sacchariflorus) 그리고 이들 두 종의 종간 교잡종인 3배체 억새(M. x giganteous) 등이다. 여러 가지 에너지작물 중 억새는 연간 바이오매스 수량이 많고 저온, 염해, 습해 등 열악 환경 적응성과 질소이용효율이 우수하며 에너지 산출/투입 수지가 가장 높고 지상부 줄기와 땅속줄기에 다량의 탄소를 저장하기 때문에 탄수수지가 가장 높다. 유럽, 미국 및 캐나다 에서는 1930년대부터 일본으로부터 3배체 억새를 도입하여 연료용 펠릿 생산, 열병합발전, 양액재배 배지, 건축자재 및 강화 플라스틱 원료로 이용한다. 현재 재배되는 3배체 억새 단점보완을 위한 품종육성연구와 사탕수수와 억새교잡종인 "Miscane" 육성 연구가 진행되고 있다. 우리나라에서는 2007년 억새 유전자원 수집이 착수된 이후 2009년에 바이오매스 수량이 많은 "거대억새1호"와 억새를 증식효율을 높일 수 있는 "억새대량증식방법"을 개발하였다. 향후 우리나라 억새산업 발전을 위해서는 분자육종 등 새로운 육종기법을 활용하여 우리나라뿐만 아니라 해외에서도 재배할 수 있는 우량 신품종을 육성하는 한편 바이오매스의 전처리, 당화 및 발효 등 셀룰로오스계 에탄올 생산공정에 관한 연구도 강화해야 할 것이다.
해조류 중 갈조류인 미역으로부터 분리당화발효(SHF)를 위한 전처리 및 효소당화를 검토하고, 기존의 분리당화발효(SHF)를 개선하기 위해 공배양발효(co-culture)를 수행하였다. 비순치 효모와 고농도 mannitol에 순치(adaptive evolution)한 효모를 이용한 공배양발효를 실시한 결과 발효 72시간에 12.2 g/l의 에탄올과 에탄올 수율($Y_{EtOH}$) 0.41을 나타내었다. 이러한 기존의 분리당화발효(SHF)를 개선한 공배양발효를 통해 에탄올 생산 수율이 0.23에서 0.41로 35.2% 증가하였으며, 에탄올 발효시간도 108시간에서 72시간으로 33.3% 감소하였다. 이러한 연구결과는 해양 바이오매스인 해조류로부터 바이오연료 생산과정에 있어 유용한 정보를 제공하는 것으로 판단된다.

Three endoxylanase-encoding genes from the moderately themophilic chemoorganotrophic bacterium Melioribacter roseus were cloned and expressed in Escherichia coli. Genes xyl2091 (Mros_2091) and xyl2495 (Mros_2495) encode GH10 family hydrolases, whereas xyl2090 (Mros_2090) represents the GH30 family. In addition to catalytic domains, Xyl2090 and Xyl2091 contain carbohydrate-binding modules that could facilitate their binding to xylans and Por sorting domains associated with the sorting of proteins from the periplasm to the outer membrane, where they are covalently attached. Recombinant endoxylanase Xyl2495 exhibited a high specific activity of 1,920 U/mg on birchwood xylan at 40℃. It is active at low temperatures, exhibiting more than 30% of the maximal activity even at 0℃. Endoxylanases Xyl2090 and Xyl2091 have lower specific activities but higher temperature optima at 80℃ and 65℃, respectively. Analysis of xylan hydrolysis products revealed that Xyl2090 generates xylo-oligosaccharides longer than xylopentaose. Xylose and xylobiose are the major products of xylan hydrolysis by the recombinant Xyl2091 and Xyl2495. No activity against cellulose was observed for all enzymes. The presence of three xylanases ensures efficient xylan hydrolysis by M. roseus. The highly processive "free" endoxylanase Xyl2495 could hydrolyze xylan under moderate temperatures. Xylan hydrolysis at elevated temperatures could be accomplished by concerted action of two cell-bound xylanases; Xyl2090 that probably degrades xylans to long xylo-oligosaccharides, and Xyl2091 hydrolyzing them to xylose and xylobiose. The new endoxylanases could be useful for saccharification of lignocellulosic biomass in biofuels production, bleaching of paper pulp, and obtaining low molecular weight xylooligosaccharides.
섬유소계 바이오매스의 주요 구성요소 간의 관계에 의한 물리적, 화학적 장벽은 셀룰로오스를 발효 가능한 당으로 전환시키는 효소당화를 방해한다. 전처리의 주 목적은 셀룰로오스의 효소당화율을 향상시키기 위하여 기질로의 효소접근성을 높이는 것으로, 전처리 공정의 발전은 지속적으로 요구되고 있다. 본 연구에서는, 간단하고, 상대적으로 저비용인 암모니아수에 의한 침지공정을 전처리방법로 채택하였다. 기질로는 국내 농업 잔류물 중 생산량이 높은 볏짚을 채택하였다. 암모니아수에 의한 침지 공정은 3, 12, 24 그리고 72시간 동안 수행되었다. 그리고 동시당화발효에 미치는 전처리의 효과를 조사하기 위해, 효소당화와 동시당화발효를 30, 40 그리고 $50^{\circ}C$에서 수행하였다. 연구 결과에 따르면, 볏짚이 암모니아수에 의한 침지 처리 되었을 때, 기존의 보편적인 동시당화발효와 비교하여 상대적으로 적은 효소사용량과 낮은 온도($30^{\circ}C$) 조건에서도 당화와 동시당화발효가 수행될 수 있음을 확인하였다. 그리고 암모니아수에 의한 침지 처리는 초기 당화속도를 증가시킴으로써 24시간 이내에 발효를 종료시켰다.

${\beta}$-Glucosidase, which hydrolyzes cellobiose into two glucoses, plays an important role in the process of saccharification of the lignocellulosic biomass. In this study, we optimized the activity of ${\beta}$-glucosidase of brown-rot fungus Fomitopsis pinicola KCTC 6208 using the response surface methodology (RSM) with various concentrations of glucose, yeast extract and ascorbic acid, which are the most significant nutrients for activity of ${\beta}$-glucosidase. The highest activity of ${\beta}$-glucosidase was achieved 3.02% of glucose, 4.35% of yeast extract, and 7.41% ascorbic acid where ascorbic acid was most effective. The maximum activity of ${\beta}$-glucosidase predicted by the RSM was 15.34 U/mg, which was similar to the experimental value 14.90 U/mg at the 16th day of incubation. This optimized activity of ${\beta}$-glucosidase was 23.6 times higher than the preliminary activity value, 0.63 U/mg, and was also much higher than previous values reported in other fungi strains. Therefore, a simplified medium supplemented with a cheap vitamin source, such as ascorbic acid, could be a cost effective mean of increasing ${\beta}$-glucosidase activity.

A cellulolytic fungus isolated from Agave plantation in northeastern Thailand was identified as Acrophialophora nainiana. The fungus was capable of growing at pH between 3 - 7 and 25 - 45 $^{\circ}C$, with the optimum conditions at pH 5.0 and 40 $^{\circ}C$. The wild isolate produced cellulases, comprising of exoglucanase (0.019 U/mg protein), endoglucanase (0.366 U/mg protein), and ${\beta}$-glucosidase (0.001 U/mg protein). Mutations with UV and NTG produced the UV 10-2 mutant with cellulases activities including exoglucanase (0.093 U/mg protein), endoglucanase (0.585 U/mg protein), and ${\beta}$-glucosidase (0.013 U/mg protein). Purification of the enzymes with ultrafiltration, ammonium sulfate precipitation, and ion-exchange chromatography yielded the maximal cellulase specific activities of 2.736 U/mg protein (exoglucanase), 0.235 U/mg protein (endoglucanase), and 0.008 U/mg protein (${\beta}$-glucosidase). The mutant's cellulases were the most active at pH 5.0 and 60 $^{\circ}C$. Ion-exchange chromatography revealed that A. nainiana UV 10-2 cellulases were comprised of two peaks with one peak showing the single endoglucanase activity while the other peak showed a mixture of the three enzyme activities. Production of A. nainiana UV 10-2 cellulases using banana leaf stalk as the sole carbon source gave comparable yields to that of the pure ${\alpha}$-cellulose. The enzymes were used in the simultaneous saccharification and fermentation (SSF) of plant residue (Coix aquatica) along with Kluveromyces marxianus to produce ethanol. Moreover, when the enzymes were used in the bioscouring process of fabric, the desiravle traits of textile processing including immediate water absorbency, increased in whiteness and reduction of yellowness of the treated fabric were observed.

Pleurotus ostreatus, P. eryngii, and Flammulina velutipes are major edible mushrooms that account for over 89% of total mushroom production in Korea. Recently, Agrocybe cylindracea, Hypsizygus marmoreus, and Hericium erinaceu are increasingly being cultivated in mushroom farms. In Korea, the production of edible mushrooms was estimated to be 614,224 ton in 2013. Generally, about 5 kg of mushroom substrate is needed to produce 1 kg of mushroom, and consequently about 25 million tons of spent mushroom substrate (SMS) is produced each year in Korea. Because this massive amount of SMC is unsuitable for reuse in mushroom production, it is either used as garden fertilizer or deposited in landfills, which pollutes the environment. It is reasonably assumed that SMS includes different secondary metabolites and extracellular enzymes produced from mycelia on substrate. Three major groups of enzymes such as cellulases, xylanases, and lignin degrading enzymes are involved in breaking down mushroom substrates. Cellulase and xylanase have been used as the industrial enzymes involving the saccharification of biomass to produce biofuel. In addition, lignin degrading enzymes such as laccases have been used to decolorize the industrial synthetic dyes and remove environmental pollutions such as phenolic compounds. Basidiomycetes produce a large number of biologically active compounds that show antibacterial, antifungal, antiviral, cytotoxic or hallucinogenic activities. However, most previous researches have focused on therapeutics and less on the control of plant diseases. SMS can be considered as an easily available source of active compounds to protect plants from fungal and bacterial infections, helping alleviate the waste disposal problem in the mushroom industry and creating an environmentally friendly method to reduce plant pathogens. We describe extraction of lignocellulytic enzymes and antimicrobial substance from SMSs of different edible mushrooms and their potential applications.
