조현병에서 나타나는 후성유전학적 나이 가속도 감속 (Slowing of the Epigenetic Clock in Schizophrenia)

  • 지난 10년 동안 인공지능의 도움으로 노화를 정량화하기 위한 수많은 연구가 수행되었다. DNA 메틸화 데이터를 사용하여 다양한 모델이 개발되었으며 흔히 후성유전학적 시계라고 불린다. 후성유전학적 나이 가속화는 일반적으로 질병 상태와도 주로 연관이 있어 보인다. 조현병은 가속 노화 가설과 관련있는 정신질병으로 심각한 정신적, 신체적 스트레스를 동반한다. 다른 심리 질환과 비교했을 때 이 질병은 젊은 사람들에서 높은 사망률과 질병률을 유발한다. 과거 연구에서는 이 질병이 가속 노화 가설과 연관있다고 알려져 있었다. 이번 연구에서는 조현병 환자의 후성유전학적 나이 가속도 변화를 통해 질병에 대한 후성유전학적 통찰을 얻고자 하였다. 후성유전학적 나이 가속화를 측정하기 위해 두 가지 다른 DNA 메틸화 시계 모델을 사용했으며 이는 범조직 모델인 Horvath clock과 Epi clock을 사용하였다. 우리는 Horvath clock과 Epi clock이 모두 호환되는 450k 어레이 데이터를 사용하였다. 그 결과, Epi clock을 사용했을 때 환자샘플에서 후성유전학적 나이 가속화가 더 느리다는 것을 발견했다. Epi clock이 질병으로 인한 DNA 메틸화 변화를 잘 감지해낼 수 있음을 알아내었다. 또한 Epi clock에서 대조군과 환자군에서 차등적으로 메틸화된 CpG 부위를 분석하고 경로 농축 분석을 수행한 결과, 대부분의 CpG가 신경 세포 과정에 관여한다는 사실을 발견했다.

  • Human body ages differently due to environmental, genetic and pathological factors. DNA methylation patterns also differs depending on various factors such as aging and several other diseases. The aging clock model, which uses these differences to predict age, analyzes DNA methylation patterns, recognizes age-specific patterns, predicts age, and grasps the speed and degree of aging. Aging occurs in everyone and causes various problems such as deterioration of physical ability and complications. Alzheimer's disease is a disease associated with aging and the most common brain degenerative disease. This disease causes various cognitive functions disabilities such as dementia and impaired judgment to motor functions, making daily life impossible. It has been reported that the incidence and progression of this disease increase with aging, and that increased phosphorylation of Aβ and tau proteins, which are overexpressed in this disease and accelerates epigenetic aging. It has also been reported that DNA methylation is significantly increased in the hippocampus and entorhinal cortex of Alzheimer's disease patients. Therefore, we calculated the biological age using the Epi clock, a pan-tissue aging clock model, and confirmed that the epigenetic age of patients suffering from Alzheimer's disease is lower than their actual age. Also, it was confirmed to slow down aging.

정서경험과 생체노화 (The Effect of Emotional Experiences on Biological Aging)

  • 연구배경: 본 연구는 인간이 평소에 부정적인 정서들을 경험하는 것과 생물학적 노화 사이의 연관성을 알아보기 위하여 고안되었다. 방법: 2013년 5월부터 9월까지 충남대학교병원 건강증진센터를 내원한 237명을 대상으로 연구하였다. 피험자들 개개인에게 구조화된 설문지를 주어 응답하도록 하였다. 피험자들이 '평소에 느끼는 우울, 불안, 분노 그리고 분노표현' (이하 '정서경험들')은 특성우울척도([the Korean adaptation of] State-Trait Depression Inventory에서 분리함), 특성불안척도(한국형 상태-특성불안척도[State-Trait Anxiety Inventory]에서 분리함) 그리고 특성분노척도와 분노표현척도([the Korean adaptation of] State-Trait Anger Expression Inventory에서 분리함)로 사정하였다. 피험자들 각각이 '생물학적으로 노화가 진행되는 정도' (이하 '생체노화')는 생체 연령(비만과 주요 장기들의 노화를 반영)과 실제 나이로 이루어진 특수한 공식으로 계산하였다. 독립 t-검정, 일원분산분석, 상관분석을 시행하여 피험자들의 일반적 특성들(정서경험들을 포함)과 생체노화들 간의 연관성들을 평가하였다. 단계적 회귀분석을 사용하여 피험자들의 일반적 특성들과 생체노화들 간의 설명력($R^2$)들을 산출하였다. 결과: 일변량분석에서 오직 특성분노와 교육수준만이 생체노화와 유의한 상관관계를 보였다; 전자의 경우는 r=0.160, P=0.014, 후자는 r=-0.189, P=0.024. 다변량분석에서는 특성 분노와 낮은 교육수준만이 생체노화와 유의한 설명력을 나타냈다; 전자의 경우는 $R^2=0.044$, P=0.020, 후자는 $R^2=0.022$, P=0.038. 결론: 본 연구는 평소에 경험하는 분노(즉 특성분노)와 낮은 교육수준이 인체의 생물학적 노화를 촉진시키는 주목할 만한 요인들임을 시사한다.

모의실험을 통한 한국 동해 도루묵(Arctoscopus japonicus)의 자연사망 계수 조건에 따른 가입당 생산 분석 (Simulation-based Yield-per-recruit Analysis of Sandfish Arctoscopus japonicus in the East Sea of Korea Subjected to Natural Mortality Conditions)

  • To estimate the biological reference points, suitable for fisheries management of sandfish Arctoscopus japonicas in the East Sea of Korea, we simulated the yield-per-recruit (Y/R) from age 0 to 6 (0-2,555 days). The stimulation was based on two instantaneous natural mortality conditions: size-dependent (Mt, d-1) and constant (Mcons, d-1); Subsequently, the biological reference points of the two mortality conditions was compared. Mt decreased from 0.0075 d-1 to 0.0018 d-1 depending on growth, and Mcons remained constant at 0.0011 d-1 for all ages. Our Y/R model showed that the maximum yield of Mcons was 14 times higher than that of the Mt. The length at first capture to maximize the harvest at the F0.1 points of the two natural mortality conditions was Lc,t=10.2 cm (TL) and Lc,cons=17 cm (TL). We concluded that Mt was more suitable for estimating M than Mcons; this is because Lc,t showed minimal difference from the current fishing regulations (11 cm, TL), and Mt reflected more biological characteristics than Mcons. We suggest that 10.2 cm and 0.8 as the suitable length at first capture and corresponding age, respectively for efficient fisheries management of sandfish.

Role of tea catechins in prevention of aging and age-related disorders

  • Tea polyphenols especially catechins have long been studied for their antioxidant and radical scavenging properties. Scientists throughout the world have investigated the usefulness of the regular green tea consumption in several disease conditions. In-vitro and in-vivo experiments on catechins especially epigallocatechingallate have revealed a significant role in many ways. Reactive oxygen species have been increasingly implicated in the pathogenesis of many diseases and important biological processes. Toxic effects of these oxidants, commonly referred to as oxidative stress, can cause cellular damage by oxidizing nucleic acids, proteins, and membrane lipids. Oxidative stress has been related to aging and age related disorders. It is found that in a wide variety of pathological processes, including cancer, atherosclerosis, neurological degeneration, Alzheimer's disease, ageing and autoimmune disorders, oxidative stress has its implications. Catechins have been reported to be useful in combating aging and age related disorders like cancer, cardiovascular disorders and neurodegenerative diseases. In this mini review we will discuss such studies done across the globe.

  • In this paper, a two-dimensional electron beam dose calculational algorithm implented for use in a two-dimensional radiation therapy planning system is described. The 2-D electron beam calculations have been in use clinically for a few decades. Our algorithm uses Age-diffusion model based int the Boltzman Transport Equation. Our implementation provides convenient user interface associated with electron beam therapy planning and displays radiation dose distribution according to different electron energy on patient images.

제주도 자생 왕벚나무의 연륜생장 특성 분석 (Growth characteristics by age measurement of Prunus yedoensis on Jeju Island, Korea)

  • This study conducted to analyze the growth characteristics of old Prunus yedoensis on Jeju island. The diameter growth per year was measured using a DTRS-2000 instrument. The DBH, ground DBH and height of the investigated P. yedoensis were 137 cm, 143 cm, and 15.5 m, respectively. Our analysis showed that the age of the old P. yedoensis was 93 years. An annual diameter growth of $2.85mm{\pm}0.96$ was observed. The result of age estimation, about $265{\pm}64$ years in P. yedoensis on Jeju island. This information could be useful to understand the annual diameter growth characteristics the P. yedoensis distributed on Jeju island.

Signaling Interface of Advanced Glycation Endproducts Receptor and Ubiquitin-Conjugating Enzyme Ubc9 Complex in Atherosclerosis and Cancer Cells

  • The advanced glycation endproducts receptor (AGER) is a multiligand signal transduction receptor. One of its ligands, S100b molecules activates vascular smooth muscle cells and endothelial cells via its receptor, thus triggering activation of signaling cascades and generation of cytokines and proinflammatory molecules. Ubiquitin-conjugating enzyme Ubc9 is an E2 conjugating enzyme that transfers the activated small ubiquitin-related modifier to protein substrates, and thus it plays a critical role in SUR-Mylation-mediated cellular pathways. Previous studies have shown that both AGE-R and Ubc9 play roles in diverse cellular signaling pathways. However, until recently, little attention has been paid to interactions between AGE-R and Ubc9. In this study, sequence database searches allowed us to identify a potential interaction motif between AGE-R and Ubc9. The subsequent biochemical and molecular biological analysis suggested that there may be specificity in AGE-R and Ubc9 complex signaling in atherosclerosis and cancer cells in a cell-type specific manner. Although the determinant for specificity in AGE-R and Ubc9 complex signaling in cancer cells and atherosclerosis is yet to be determined, this study provides the basis to develop a specific therapeutic application of AGE-R, SURM (small ubiquitin-related modifier)-1, and Ubc9 complex activation pathways in atherosclerosis, diabetes, cancer and inflammatory diseases.

골다공증 모델동물, SAMP6의 특성 연구 (Studies of SAMP6 as an Animal Model for Human Osteoporosis)

  • SAMP6의 성별 및 가령에 따른 생물학적인 특성을 알아보기 위하여 3, 5, 18개월령의 SAMP6 암컷, 수컷의 체중, 장기중량, 혈액학적 및 혈액생화학적 수치, 골의 무게, 비중, 두께 및 골의 화학적 분석을 위해 인과 칼슘을 측정하였다. 1. 체중은 수컷에서 7개월령부터 감소를 나타내었으나, 암컷은 18개월령부터 체중이 감소되기 시작했다. 장기중량을 보면, 간, 신장, 폐의 중량은 수컷이 암컷보다 무거웠으며, 흉선은 암컷이 수컷보다 무겁게 나타났다. 2. 혈액학적 및 혈액생화학적 검사결과 백혈구수가 수컷이 암컷보다 높게 나타났으며, 적혈구수는 암컷이 높게 나타났다. GOT는 5, 7개월령에, BUN은 전체개월령에서 수컷이 암컷보다 높게 나타났다. alkaline phosphatase 활성은 3개월령과 비교하여, 전체 개월령에서 유의성있는 감소가 관찰되었다. 3. 골두께를 측정한 결과 대퇴골에서 암수 모두 7개월령 부터 골두께의 급격한 감소가 나타났다. Cortical thickness index를 측정한 결과 대퇴골에서 암수 모두 5개월령에서 최고치에 다다랐으며, 이후 계속 감소하였다. 4. 골의 칼슘량을 정한 결과, 대퇴골에서 암수 모두 5개월령에 칼슘량이 최고치에 달했으며, 이후 계속 감소하였고, 인량은 수컷에서 5개월령, 암컷에서는 7개월령에 최고치에 달하였고, 이후 계속 감소하였다. 결론적으로 본 실험에서 사용한 골다공증 모델동물인 SAMP6의 특성발현은 5개월 이후 동물에서 나타나고 있으며 따라서 이들 동물을 골다공증 약물 개발을 위한 약효검색에 이용할 때에는 5개월령 이후 7개월령의 동물을 사용하는 것이 바람직하며 본 실험에서 유의성 있는 특성발현이 나타난 골 두께 및 골의 칼슘과 인량, alkaline phosphatase 활성 측정등을 약효 검색에 사용할 수 있을것으로 사료된다.

자동차 정비업체 도장공정의 작업환경 및 근로자 노출 실태에 관한 연구 (A Study of Working Environment for Automotive Painting in Auto Repair Shops and Workers' Exposure to Hazardous Chemicals)

  • The purpose of this paper is to evaluate 1) blood lead levels of workers at auto repair shops as Biological Exposure Indices (BEI) of toxic substances such as lead and toluene that are produced during automotive painting process, 2) the differences depending on personal characteristics of workers who have been exposed to toluene by using urine hippuric acid concentration as a marker and 3) the correlation between the concentration of hazardous chemicals in each workplace and the BEL. All subjects were male with a mean age of 36.2 years. In terms of age, most were in the 30 to 40 age group (13 persons, 48.1%). In relation to the length of work experience, the highest proportion had experience of 10 years of less (18 persons, 66.7%). Twenty three workers were cigarette smokers (85.2%) while 4 (14.8%) were non-smokers. In addition, more than 80% of the workers drank alcohol. Dust concentration and toluene exposure during automotive painting showed no significant difference with age, length of work experience, smoking and drinking while a significant difference (p<0.05) has been detected between lead concentration and smoking. The geometric mean of dust concentration, lead concentration and toluene concentration were $0.38mg/m^3,\;0.0021mg/m^3$ and 1.08ppm respectively. In addition, the geometric mean of blood lead levels and urine hippuric acid concentration were $1.70{\mu}g/dl$ and 0.25g/g respectively, which were lower than the standard levels suggested by the Ministry of Labor. To determine the influential factors on blood lead and urine hippuric acid concentrations, a correlation analysis has been conducted with variables of air, lead and toluene concentrations, age, length of work experience and amount of cigarette smoking. According to the analysis, a relatively high correlation (p<0.01) has been observed between air lead concentration and biological sample concentration.
