Development of DNA Barcode Database and Identification System of Forest Mushrooms in Korea

  • Over five thousand higher fungal specimens were collected from 32 forest areas of Chungcheong and Gyeongsang province from 2012 to 2013. We obtained 513 strains and 3,120 ITS sequences. Mushrooms were first identified with macro- and micro-scopic characters, and their identification was confirmed on the basis of ITS sequences. Voucher specimens were designated for each species found in Korea. Construction of DNA barcoding Database is currently underway with sequences of 409 species. During the development of the database, some new species were recognized, along with several Korean new records. When the system has been completed, it will provide essential molecular information for metagenomic and phylogenetic researches for higher fungi.

Long-Term Biodiversity Research Programme for Mindanao, Philippines

  • The Long-Term Biodiversity Research Programme (LTBRP) for Mindanao is envisioned to be a collaborative programme of the Philippines. It will be a programmatic research on biodiversity - its status, threats. and conservation and management. The chosen research site is Mt. Malindang in Misamis Occidental of Mindanao. The BRP will support a set of research projects that will generate knowledge on biological and ecological, socio-economic. cultural and policy aspects of biodiversity conservation. It will also enable researchers to develop and try new methods for research on these aspects, separately or crossing boundaries of academic disciplines. Moreover. the BRP will implement support programmes which will provide the linkages of research activities to development issues and needs in the research site. The support programmes will also draw from the research projects, the knowledge that can be lent or immediately available to policy and programme formulation. The support programmes will comprise: human resource development or capability-building; information, education and communication; database; networking; community organizing: and development action. The BRP will be undertaken by a group of academie and research institutions from the Philippines, in partnership with their respective government entities and local government units in Mindanao. The Biodiversity Research Programme (BRP) will comprise a set of research projects to be undertaken by small research teams in the site. Researchable areas have been identified initially through the National Biodiversity Research Agenda. and later enriched in the Participatory Rural Appraisal (PRA) Analysis workshops by the Mindanao researchers and Philippine resource persons. Results of the PRA are presented in the paper.

  • Biodiversity is a key element of ecosystem of which function provides essential product and service in human life. In the course since development projects often causes damages to biodiversity, environmental impact assessment technique must be capable of accurately assessing potential impact from flora and fauna and to entire ecosystem. Korea needs improvement of its assessment technique that is compatible with Korean environmental regulatory standard that is generally stricter than that of most countries. This study attempts to explore both domestic and overseas biodiversity assessment techniques and analyze each stage of environmental impact assessment. The data is collected from numbers of literatures selected by navigating both domestic and overseas literature database with certain keywords. Among the 44 selected papers, overseas publications outnumber those of domestics, and there are more researches on assessment methodology of biodiversity than assessment tool and model. In terms of environmental impact assessment, the number of papers on environmental impact forecast exceeds the numbers of papers on current state of environment and the impact minimizing solution. Therefore, contents and trends of those researches in the different stages of environmental impact assessment discussed in this paper not only suggest potential impact on biodiversity and minimization solutions in detail, but is also a valuable resource particularly for biodiversity relevant environmental assessment technique improvement in Korea. Proposing of a new direction of improvement in biodiversity assessment techniques makes this study significant, and further research for preservation of biodiversity should follow up to provide an improvement scheme for biodiversity assessment techniques in the future.

So bioinformation is necessary of biodiversity in broadly wide and detailed information. Among information, bioinformation of biodiversity is important and utilization of living things. Among them, the mushroom(higher fungi) are an important part in ecosystem as a decomposer responsible for recycling materials. Many living things today, however, have endangered by environmental pollution and ecological destruction. The higher fungi also are not exception. Mushroom has been used for food sources, pharmacy and forests resources from ancient times. Among biodiversity, database of mushroom is very necessary for university, institute and industry. This DB contains four items of native mushroom(higher fungi) from Korea. first item contain species, genus, family, order class, ad division according to the classification. Second item contain pharmaceutical purpose, food source, culture, toxic, anti-cancer of the application. Third item contain symbiosis, rotten trees of the ecological resources. Fourth item contain geographical distribution and illustrated literature. Information system is also available using KRISTAL II for searches on the WEB in URL http://ruby. kisti. re. kr/∼mushroom.

  • A major concern regarding the collection and storage of biodiversity information is the inefficiency of conventional taxonomic approaches in dealing with a large number of species. This inefficiency has increased the demand for automated, rapid, and reliable molecular identification systems and large-scale biological databases. DNA-based taxonomic approaches are now arguably a necessity in biodiversity studies. In particular, DNA barcoding using short DNA sequences provides an effective molecular tool for species identification. We constructed a large-scale database system that holds a collection of 5531 barcode sequences from 2429 Korean species. The Korea Barcode of Life database (KBOL, http://koreabarcode.org) is a web-based database system that is used for compiling a high volume of DNA barcode data and identifying unknown biological specimens. With the KBOL system, users can not only link DNA barcodes and biological information but can also undertake conservation activities, including environmental management, monitoring, and detecting significant organisms.

  • 본 논문에서는 국내에 산재한 생물다양성정보를 학습에 활용하기 위하여 KISTI에서 구축한 생물다양성 DB 현황과 e-Learning의 기술요소 등을 조사하였으며, 기존에 구축된 생물다양성정보 DB를 활용하여 일반인과 학생들을 위한 생물다양성 학습 콘텐트를 기획하고 설계하였다. 본 설계를 바탕으로 생물다양성 콘텐트를 개발한다면, 국토가 좁고, 네트워크 인프라가 잘 갖추어져 있는 우리나라의 실정에 맞는 사이버공간상의 학습의 장으로서 일반인과 학생들에게 양질의 생물다양성 학습 콘텐트를 제공할 수 있으리라 기대한다.

  • To distinguish manchurian clematis, Clematis terniflora var. mandshurica and three-leaf clematis, C. apiifolia, we collected 9 nuclear ITS sequences and two sequences of trnQ-trnH intergenic spacer and trnH region in GenBank database. Those sequences were aligned to find differences between those of C. terniflora var. mandshurica and C. apiifolia. Two primer pairs were newly designed base on the differences between two species and conducted multiplex PCR. The size of amplified fragments using generated primers were 380 base pairs (only three-leaf clematis) and 189 base pairs (both species). This genetic marker based on PCR is useful to discrimination of C. terniflora var. mandshurica and C. apiifolia

세계생물다양성정보기구(GBIF)에 출판된 동아시아 관속식물 생물다양성 정보 현황과 자료품질 분석 (Status and Quality Analysis on the Biodiversity Data of East Asian Vascular Plants Mobilized through the Global Biodiversity Information Facility (GBIF))

  • 생물다양성정보학(Biodiversity Informatics)은 정보과학을 생물다양성정보에 접목한 분야로 정이명으로 구성된 학명을 비롯한 종정보를 기초로 일차종발생자료를 구축하고 이를 활용한다. 본 연구에서는 생물다양성 정보의 이용적합도를 기준으로 세계생물다양성정보기구(GBIF)에 출판된 동북아시아 자료의 품질을 BRAHMS 프로그램을 이용하여 평가하고 이를 통해 생물다양성자료 정제의 필요성을 확인하였다. 국립생물자원관, 국립생태원, 국립수목원 등의 국내 생물다양성 관련기관과 더불어 일본, 중국, 대만의 출판 자료는 자료정제과정의 문제로 학명, 지리정보, 채집자, 날짜 등에 대한 오류가 확인된다. 기본적인 속성자료에서 오류가 발생하는 원인은 동아시아의 생물다양성관리기관들이 구조화되지 않은 데이터베이스를 사용하고 평면적인 스프레드시트형 정보를 사용하기 때문이다. 생물다양성 정보 특성상 다양한 정보가 구조화가되지 않을 경우 학명, 인명, 지명, 문헌, 생태정보에 대한 데이터 무결성을 해결하지 못한다. 동아시아 생물다양성정보 관리문제를 극복하기 위해서는 자료의 구조화와 함께 자료정제에 대한 이해도를 높이고, 오류 수정을 위한 지속적인 자료 관리자인 전문 분류학자 양성이 필요하다. 생물다양성 정보관리자는 오류 원인분석을 통해 문서화된 관리 지침을 수정, 추가하는 등 향후 오류 예방을 위한 대책이 필요하며 시스템에 적용시켜야 한다. 이런 모든 과정은 데이터베이스를 기반으로 진행되고 기록되어야 한다. 동아시아의 생물다양성 출판자들은 현재 수준의 단순한 자료구조보다는 생물다양성 정보 관리를 위해 전문적인 선진 프로그램의 사용 혹은 이에 준하는 수준의 고도화된 데이터베이스의 개발이 필요하다.

기타 효과적인 지역 기반 보전 수단(OECMs)의 개별 평가 - 국립수목원을 중심으로 - (Site-Level Assessment of Other Effective Area-based Conservation Measures - Focusing on the Korea National Arboretum -)

  • By delivering effective, in-situ conservation of biodiversity, OECMs can contribute to sustaining existing biodiversity values and improving biodiversity conservation outcomes. In this study, for the reporting of OECMs required by Kunming-Montreal Global Biodiversity Framework, the site-level assessment of the Korea National Arboretum and Buffer Zones were conducted using the assessment tool of IUCN. The site-level assessment was carried out in three steps(step 1: screening, step 2 : consent for full assessment, step 3 : the full assessment). It was found that the criteria were satisfied except for the consent for full assessment, sustainability of governance and management arrangements, and the equity of governance and management. Although the governing authority, rights-holders or any other stakeholders could be identified, the step of acknowledging and agreeing to the full OECM assessment was not possible because the governance was not established. As a result of the assessment of equitable governance and management in the aspect of recognition, procedure, and distribution based on criterion(the equity of governance and management), it is judged that more specific measures are needed in the aspect of recognition. And in the aspect of procedure, there is no legal basis for participation in governance and collection of opinions, so it is judged that there is a limit to listening and reflecting the opinions of stakeholders. In the aspect of distribution, it is necessary to further confirm whether it provides direct benefits to rights-holders such as landowners in the region. And it is necessary to prepare specific criteria to assess the important biodiversity values. Therefore, in order to promote OECM reporting in the future, it is necessary to conduct detailed research on various types of governance establishment and operation plans that can establish the basis for recognizing and agreeing to OECM assessment, specific criteria and reasonable measures to judge equity, and important biodiversity value.
