Effects of Intermediate Metabolites on Phenanthrene Biodegradation

  • Stimulatory effects of three different intermediate metabolites (1-hydroxy-2-naphthoate, salicylate, and catechol) as potential inducers on phenanthrene degradation were investigated using two different bacteria (Pseudomonas putida ATCC 17484 and Burkholderia cepacia PB12). The relative induction capacity was high in the sequence of 1-hydroxy-2-naphthoate, salicylate, and catechol in both strains. The highest of up to 12 times increase of the induction was obtained by the addition of 1-hydroxy-2-naphthoate in the strain PB12, compared with the control where no exogenous inducer was added. The induction capacity of the potential inducers was closely related with the number of oxygenations required per electron equivalents in one mole of the inducer.

  • Monod kinetics에 관련된 주요 생분해 파라미터를 도출하기 위하여 microcosm 규모의 배치실험에서 BTEX 화합물에 대해 분해능이 우수한 Pseudomonas aeruginosa을 이용해 다양한 농도의 벤젠에 대한 분해기작을 고찰하였다. 벤젠의 생분해율(D)과 Maximumspecific growth rate ($\mu$$_{max}$)는 기질의 농도가 증가할수록 높아지다가 최고점에 도달 후에 점차적으로 감소하였으며 이것은 어느 한계점 이상의 벤젠 농도가 미생물의 생분해에 방해 요소로 작용한다는 것을 나타낸다. 그러나 미생물에 의한 벤젠 분해의 상관관계를 나타내는 yield coefficient(Y)는 벤젠의 초기 농도가 낮을수록 높은 값을 나타내었다. Microbial decay constant( b)와 half-saturation constant(K$_{c}$)는 각각 0.21~0.48day$^{-1}$와 218mg/$\ell$로서 문헌값 보다 높은 수치를 나타내었다. 실험으로부터 결정된 생분해 파라미터들은 초기 벤젠 농도에 따라 큰 차이를 보이므로 생분해 모델링에 사용할 파라미터는 기질농도에 따라 적절하게 선택되어야 한다고 사료된다.

  • NAPL이 다핵방향족화합물의 하나인 phenanthrene 의 생분해에 미치는 영향을 알아보았다. Pseudomonas putida CRE7 을 이용한 실험에서 NAPL 의 첨가로 인한 가장 큰 차이는 미생물의 소수성의 변화였다. 소수성이 증대됨으로써 phenanthrene 의 가용성이 증대되었으며, 이로 인해 더 많은 양의 오염물 분해가 이루어졌다. 생물학적 분해의 관찰은 발생되어지는 $^{14}$ $CO_2$의 radioactivity 측정을 통해 이루어졌으며, 미생물의 소수성 측정은 bacterial adhesion to hydrocarbon (BATH) assay 를 이용하였다.

생합성 플라스틱 Poly-$\beta$-Hydroxybutyrate의 생분해와 토양온도의 관계 (Relationship between Biodegradation of Biosynthetic Plastics, Poly-$\beta$-Hydroxybutyrate, and Soil Temperature)

  • The microbial degradation of $poly-{\beta}-hydroxybutyrate$ (PHB) films was studied in soil microco는 incubated at a constant temperature of 2, 10, 20, 30 and $40^{\circ}C$ for up to 49 days. The degradation rate measured through loss of weight was enhanced by incubation at a higher temperature. At the soil temperature $40^{\circ}C$, $poly-{\beta}-hydroxybutyrate$ was rapidly degraded at a decay rate of 3.5% weight loss per day. The degradation of $poly-{\beta}-hydroxybutyrate$ did not affected significantly the chemical properties of soils such as pH and electric conductivity. However, microbial activity of soil in terms of dehydrogenase activity was increased by the degradation of $poly-{\beta}-hydroxybutyrate$.

Toluene내성세균 Pseudomonas sp. BCNU 154을 이용한 방향족화합물의 분해 (Biodegradation of Aromatic Hydrocarbons by Toluene-tolerant Pseudomonas sp. BCNU 154)

  • 분리된 toluene내성세균 Pseudomonas sp. BCNU 154을 생물분해반응에 적용하고저 그 가능성을 조사하였다. 방향 족화합물 16개을 대상으로 저화능을 조사한 결과 cumene, cyclohexane, ethylbenzene, p-xylen, m-xylen, toluene 그리고 diphenylether가 공시균쥬에 의해 자화됨이 확인 되었다. Pseudomonas sp. BCNU 154는 toluene, ethylbenzene 그리고 p-xylen 그리고 cumene을 호기적으로 분해하였다. Touene은 12시간후에 완전히 분해되었고, p-xylene과 cumene은 12시간배양시 90% 분해되었으며, ethylbenzene은 12시간 배양시 75%가 분해되었다.

Biodegradation of Bunker-A Oil by Acinetobacter sp. EL-081K

  • Bunker-A oil-degrading microorganisms were isolated from a marine environment using an enrichment culture technique. The isolated strain EL-081K was identified as the genus Acinetobacter based on the results of morphological, culture, and biochemical tests. The optimal temperature and initial pH for bunker-A oil degradation were $25^{\circ}C$ and 7.0, respectively, including aeration. The optimal medium composition for the degradation of bunker-A oil by Acinetobacter sp. EL_O81K was 10 ml/l bunker-A oil as the carbon source and 0.1% (NH$_4$)$_2$SO$_4$as the nitrogen source. Under the above conditions, the biodegradability of bunker-A oil was 38% after 96 hours of incubation. The addition of detergent did not increase the bunker-A oil degradation.

Biodegration of Pentachlorophenol by White Rot Fungi under Ligniolytic and Nonligninolytic Conditions

  • The roles of lignin peroxidase, manganese peroxidase, and laccase were inverstigated in the biodegration of pentachlorphenol (PCP) by several which rot fungi. The disappearance of pentachlorophenol from cultures of wild type strains, P. chrysosporium, Trametes sp. and of pentachlorophenol from cultures of wild type strains, P. cheysocporium, Trametes sp. and Pleurotus ap., was observed. The activities of mangnese peroxidase and laccase was detected in Trametes sp. and pleurotus sp. cultures. However, the activities showed that PCP was degraded under ligninolytic as well as nonligninoytic condicationg that lignin peroxidase, manganese peroxidase, and laccase are not essential in the biodegradation of PCP by white rot fungi.

Bioabsorbable osteofixation for orthognathic surgery

  • Orthognathic surgery requires stable fixation for uneventful healing of osteotomized bony segments and optimal remodeling. Titanium plates and screws have been accepted as the gold standard for rigid fixation in orthognathic surgery. Although titanium osteofixation is the most widely used approach, the use of bioabsorbable devices has been increasing recently. Biodegradation of bioabsorbable devices eliminates the need for a second operation to remove metal plates and screws. However, long-term stability and relapse frequency in bioabsorbable osteofixation are still insufficiently studied, especially in cases of segmental movements of great magnitude or segmental movements to a position where bony resistance exists. This paper reviews the background, techniques, and complications of bioabsorbable osteofixation and compares bioabsorbable and titanium osteofixation in orthognathic surgery in terms of skeletal stability.

  • The genus Rhodococcus is a phylogenetically and catabolically diverse group that has been isolated from diverse environments, including polar and alpine regions, for its versatile ability to degrade a wide variety of natural and synthetic organic compounds. Their metabolic capacity and diversity result from their diverse catabolic genes, which are believed to be obtained through frequent recombination events mediated by large catabolic plasmids. Many rhodococci have been used commercially for the biodegradation of environmental pollutants and for the biocatalytic production of high-value chemicals from low-value materials. Recent studies of their physiology, metabolism, and genome have broadened our knowledge regarding the diverse biotechnological applications that exploit their catabolic enzymes and pathways.

Decolorization of Azo Dyes by Aspergillus sojae B-10

  • Biodegradation of azo dyes such as Amaranth, Sudan III and Congo-Red by Aspergillus sojae B-10 was demonstrated using Aspergillus sojae B-10. Aspergillus sojae B-10 showed the greatest decolorization ability when it was cultivated in a nitrogen-limited medium containing, azo dyes(10 mg/l), 2.0% glucose, 0.06% sodium nitrate, 0.1% $KH_PO_4$, 0.5% MgSO_4$$7H_2O$ at pH 5.0 and $37^\circ{C}$ for 5 days. Under optimal conditions, Amaranth started being decolorized within 24 hr and was almost complete after decolorization of 4 days incubation. Sudan III was completely decolorized after a cultivation of 5 days. However, Congo-Red was not completely decolorized until 5 days of cultivation.
