The effects of paeoniflorin injection on soluble triggering receptor expressed on myeloid-1 (sTREM-1) levels in severe septic rats

  • Paeoniflorin (PAE) is the most abundant compound in Xuebijing injection widely used to treat sepsis. We aimed to investigate effect of PAE on expression of soluble triggering receptor expressed on myeloid cells-1 (sTREM-1) in a rat model of sepsis. Wistar rats were divided into Normal, Model, and PAE groups (n=20 each). Endotoxin was administrated at 5 mg/ml/kg in Model and PAE rats to establish rat sepsis model. 1 h after endotoxin administration, PAE was administrated at 4 ml/kg in PAE group once per day for 3 days. Routine blood tests and biochemical indexes were assessed, including aspartate aminotransferase (AST) and creatine kinase-MB (CK-MB). The plasma sTREM-1 level was measured using quantitative ELISA. At the end of experiment, the small intestine, liver, kidney and lung were subjected to pathological examinations. A rat model of sepsis-induced multiple organ dysfunction syndrome (MODS) was established successfully with endotoxin administration (5 mg/ml/kg), evidenced by histo-pathological examinations, routine blood tests and biochemical indexes: platelet count decreased and white blood cell count increased (p<0.05), CK-MB and AST increased (p<0.05). PAE treatment significantly reduced the plasma levels of AST, CK-MB, and sTREM-1, compared to Model group (p<0.05). Meanwhile, sepsis-induced damages in the liver, lung, stomach and intestinal mucosa were also markedly ameliorated by PAE treatment. PAE demonstrated a significantly protective effect in a rat model of sepsis by decreasing plasma sTREM-1 level, reducing inflammation, preventing MODS and protecting organ functions.

매립장의 발생가스특성을 이용한 매립장 침하예측 (Prediction of Landfill Settlement Using Gas Generation Characteristics)

  • 폐기물 매립지의 침하량을 예측하는 것은 우리나라와 같은 좁은 국토를 효율적으로 관리하기 위하여 매우 중요하다. 매립장내 유기물이 장기간에 걸쳐 생화학적으로 분해되기 때문에 압밀이론으로 해석하기 곤란하다. 본 연구에서는 실내모델실험을 통하여 매립가스의 발생특성을 분석하였다. 두개의 시험매립조를 만들었는데 하나는 침출수를 재순환한 것과 다른 하나는 재순환하지 않은 것이다. 시간의 변화에 따른 가스발생량과 매립조의 침하량과의 관계를 분석하였다. 수학적 침하량예측모델을 제안하여 장기침하량을 예측하여 실험계측치와 비교하고 수정계수를 사용하도록 제안하였다. 침출수 재순환이 침하를 촉진하는 효과가 있는 것으로 나타났는데 가스모델의 수정계수가 침출수순환을 하지 않은 경우는 1.4, 재순환한 경우는 1.7으로서 약 22%의 촉진효과가 있다.

  • The main object of this study was to offer information about incoming waste in landfill and to evaluate biochemical methane and hydrogen sulfide potentials of landfill wastes. We examined brick, soil, mixed waste (C&D waste and MSW) samples for the study. The leaching experiments showed that BOD, COD and sulfate were determined in the range of 0~18,816 mg/kg, 85~21,100 mg/kg and 160~1,205 mg/kg, respectively in 6hr extraction test. An accumulated extraction tests for 140day were determined BOD 226~197,219 mg/kg, COD 436~242,588 mg/kg and Sulfate 1,090~25,140 mg/kg. Also, BMP (biochemical methane potential) tests were carried out to examine methane and hydrogen sulfide yields for the 3 different wastes. As a result, methane yield was determined to 262.68 mL $CH_4/g$ VS of MSW and 0~17.75 mL $CH_4/g$ VS in brick, soil and C&D waste. Higher hydrogen sulfide yield was observed to 0.079mL $H_2S/g$ VS in C&D waste. This result indicate that brick and soil could be sources of sulfate, and higher production of hydrogen sulfide could be odor problem and inhibitor of methane production.

Single-dose Toxicity of Guseonwangdo-go Glucose 5% Intravenous Injection in a Rat Model

  • Objectives: The purpose of this study was to examine the single-dose intravenous toxicity of Guseonwangdo-go glucose 5% pharmacopuncture (GWG5). Methods: Forty Sprague-Dawley rats were divided into four groups of five males and five females per group: an intravenous (IV) injection of 1.0 mL of normal saline solution per animal was administered to the control group; IV injections of 0.1, 0.5, and 1.0 mL of GWG5 per animal were administered to the experimental groups (G: 0.1, G: 0.5, and G: 1.0). Observation of clinical signs and body weight measurements were carried out for 14 days following the injections. At the end of the observation period, hematological, biochemical, and histopathological tests, as well as necropsy examinations, were performed on the injected parts. Results: No mortalities or adverse clinical signs were observed in any of the groups. The body weights of all groups continuously increased. In the hematological and the biochemical tests, females in G-0.1 had minimal changes, but those changes were not dose dependent. On necropsy examination, no abnormalities were observed. In the histopathological test, focal inflammatory cell infiltrations were observed in two female rats, one in the control group and one in G-1.0. Also, one female rat in the control group had an epidermis crust. These changes were concluded to have been caused by the insertion of the needle into a vein. Conclusion: The above findings suggest that the lethal dose of GWG5 administered via IV injection is more than 1.0 mL per animal in both male and female rats. Further studies are needed to establish more detailed evidence of its toxicity.

A Study of Health-Related Habit and Hematological Index of Male Workers Residing in Ulsan City

  • This study was conducted by surveying 616 male workers living in Ulsan City regarding their health status based on lifestyles such as alcohol consumption, smoking and exercising as well as physical measurements and biochemical tests. The average height, weight and BMI(body mass index, $kg/m^2$) of the subjects was 170.9cm, 70.2kg and 24.2, respectively. The rate of drinking was $80.9\%$ and the rate of smoking was $53.4\%$. Seventy four percent of subjects responded that they exercise regularly. The results of the blood biochemical tests revealed that the average hemoglobin concentration was 14.7g/dl, and the levels of GPT(glutamic pyruvic transaminase) and GOT (glutamic oxaloacetic transaminase) were 32.74unit/l, 26.99 unit/l, respectively. The average hemoglobin concentration for the subjects aged in the 50s was 14.39g/dl, which was significantly lower than those in the 20s(14.81g/dl), 30s(14.69g/dl) and 40s(14.73g/dl). The blood glucose level and the cholesterol level also increased with age. Also investigated was the blood pressure of the subjects increased with age,. and there was a significant increase(p < 0.05) for the subjects in the 50s compared to those in the 20s. The frequency of alcoholic beverages was significantly correlated with systolic/ diastolic blood pressure(p < 0.05) and $\gamma-GTP(gamma\;glutamyl\; transpeptidase)$(p<0.01). The duration of smoking showed a negative correlation(p < 0.05) with the hemoglobin and positive correlations with diastolic blood pressure, cholesterol and $\gamma-GTP(p<0.01)$. The study shows that blood pressure, blood glucose level, cholesterol level, GOT, GPT and $\gamma­GTP$ level, increase with age, which indicates higher possibility of degenerative diseases, calling for nutritional education in terms of advisable lifestyles regarding eating habits, alcohol consumption, smoking and regular exercise.

Early Liver and Kidney Dysfunction Associated with Occupational Exposure to Sub-Threshold Limit Value Levels of Benzene, Toluene, and Xylenes in Unleaded Petrol

  • Background: Unleaded petrol contains significant amounts of monocyclic aromatic hydrocarbons such as benzene, toluene, and xylenes (BTX). Toxic responses following occupational exposure to unleaded petrol have been evaluated only in limited studies. The main purpose of this study was to ascertain whether (or not) exposure to unleaded petrol, under normal working conditions, is associated with any hepatotoxic or nephrotoxic response. Methods: This was a cross-sectional study in which 200 employees of Shiraz petrol stations with current exposure to unleaded petrol, as well as 200 unexposed employees, were investigated. Atmospheric concentrations of BTX were measured using standard methods. Additionally, urine and fasting blood samples were taken from individuals for urinalysis and routine biochemical tests of kidney and liver function. Results: The geometric means of airborne concentrations of BTX were found to be $0.8mg\;m^{-3}$, $1.4mg\;m^{-3}$, and $2.8mg\;m^{-3}$, respectively. Additionally, means of direct bilirubin, alanine aminotransferase, aspartate aminotransferase, blood urea and plasma creatinine were significantly higher in exposed individuals than in unexposed employees. Conversely, serum albumin, total protein, and serum concentrations of calcium and sodium were significantly lower in petrol station workers than in their unexposed counterparts. Conclusion: The average exposure of petrol station workers to BTX did not exceed the current threshold limit values (TLVs) for these chemicals. However, evidence of subtle, subclinical and prepathologic early liver and kidney dysfunction was evident in exposed individuals.

First Report of Pectobacterium brasiliense Causing Soft Rot on Graft Cactus in Korea

  • The graft cactus (Gymnocalycium mihanovichii) continues to be exported to more than 20 countries worldwide. In April 2021, typical bacterial symptoms of soft rot were observed in the graft cactus (cv. Yeonbit) in Goyang, Gyeonggi-do, Korea, resulting in economic losses in cactus production. The stems turned dark brown and the flowers were covered with black rot. The bacterial strain designated as KNUB-01-21 was isolated from infected stems and flowers. The results of the morphological and biochemical tests of the isolate were similar to those of Pectobacterium brasiliense. For molecular analysis, the 16S rRNA region and three housekeeping genes (dnaX, leuS, and recA) of the strain KNUB-01-21 were amplified. Based on the results of the molecular analysis and morphological and biochemical tests, KNUB-01-21 was identified as P. brasiliense. The pathogenicity of KNUB-01-21 on graft cactus was confirmed by an inoculation test. Artificial inoculation using P. brasiliense KNUB-01-21 produced soft rot symptoms on the grafted cactus, and the same bacterium was re-isolated and re-identified. This is the first report of P. brasiliense causing soft rot in graft cactus in Korea.

A Comparative Study Between Cytomegalovirus Immunoglobulin M-Positive and CMV Immunoglobulin M-Negative Biliary Atresia in Infants Attending a Tertiary Care Hospital in Bangladesh

  • Purpose: Perinatal cytomegalovirus (CMV) infection can lead to biliary atresia (BA) in different entities. This study aimed to compare the clinical, hematological, biochemical, and histological features of infants with BA based on their CMV immunoglobulin M (IgM) status at presentation. Methods: This cross-sectional descriptive study was carried out between January 2019 and June 2020 at the Department of Pediatric Gastroenterology and Nutrition at the Bangabandhu Sheikh Mujib Medical University (BSMMU) in Dhaka. Forty-three patients with BA were selected purposively and categorized into either the CMV IgM-positive or CMV IgM-negative BA group. Categorical variables were compared using Fisher's exact test and chi-square tests, while the Student's t-test and Mann-Whitney U-test were used to compare continuous variables. For all statistical tests, a p-value <0.05 was considered statistically significant. Results: Thirty-three (76.7%) of the cases were between 2 and 3 months of age on admission. The clinical, hematological, and biochemical parameters did not differ significantly between the CMV IgM-positive and CMV IgM-negative BA groups. Most (50.0%) of the CMV IgM-positive cases had fibrosis stage F2, while 43.5% of the CMV IgM-negative cases had fibrosis stage F3, with no significant difference between the groups (p=0.391). Conclusion: Our data shows no significant distinction between CMV IgM-positive and CMV IgM-negative BA, suggesting that CMV does not contribute to BA pathogenesis.

  • The authors identified 833 Salmonella cultures collected from various parts of the country in 1972. The procedures for the morphological and biochemical tests and for the serological determinations were performed by means of the conventional screening methods recommended by the National Center for Disease Control in U.S.A. The results of the laboratory tests were summerized as follows: 1. Of 833 Salmonella cultures, 1 S. paratyphi A, 1 S. nitra, 1 S. kiel, 1 S. abortusequi, 6 S. paratyphi B, 5 S. abony, 4 S. caledon, 13 S. typhimurium, 2 S. coeln, 1 S. oranienburg, 1 S. thompson, 1 S. bonn, 1 S. gabon, 1 S. colorado, 1 S. richmond, 2 S. berta, 20 S. enteritidis, 1 S. regent, 1 S. london were identified besides 769 cultures of S. typhi. 2. The antibiotics sensitivity tests by means of Ericsson's disc method using seven kinds of antibiotics were carried out, i.e. chloramphenicol, ampicillin, which were widely in common use in the country and the results were compared with that of Salmonella cultures isolated in 1971 as shown in table 4.

Detection of microbial organisms on Apis mellifera L. beehives in palm garden, Eastern Thailand

  • Background: Honey bees play a crucial role in pollination and ecological balance. Apis mellifera L. colonies, especially those located in specific geographic regions, such as the palm garden in Eastern Thailand, are susceptible to potential threats from microbial contaminants. Understanding and detecting microbial organisms in these beehives is essential for the preservation of bee health, honey production, and the broader ecosystem. However, the problem of microbial infection and antibiotic-resistant bacteria is more severe and continuously increasing, resulting in a health, economic, and social crisis. The purpose of this study is to determine the prevalence of microorganisms in A. mellifera beehives in palm gardens in Rayong province, Eastern Thailand. Results: Ten swabs in transport media were swabbed and obtained from different parts of each beehive (1 swab per beehive), for a total of 10 hives. Traditional microbial culture-based methods, biochemical tests, and antimicrobial susceptibility (disc-diffusion) tests were used to detect microbial organisms and antibiotic resistance in bacteria. The swab tests from nine beehives resulted in the detection of Gram-positive bacteria (63.64%), Gram-negative bacteria (27.27%), and fungi/yeast (9.09%). These microorganisms are classified as a group of coagulase-negative Staphylococcus spp. and made up 40.91% of the bacteria discovered. Other bacteria found were Coryneform bacteria (13.64%), Pantoea spp. (13.64%), Bacillus spp. (9.09%), yeast (9.09%), glucose non-fermentative Gram-negative bacilli (9.09%), and Pseudomonas spp. (4.55%). However, due to the traditional culture-based and 0biochemical tests usually used to identify the microbial organisms in clinical specimens and the limitation of identifying some environmental microbial species, the results of the antimicrobial susceptibility test cannot reveal if the organism is resistant or susceptible to the drug. Nevertheless, drug-sensitive inhibition zones were formed with each antibiotic agent. Conclusions: Overall, the study supports prevention, healthcare, and public health systems. The contamination of microorganisms in the beehives may affect the quality of honey and other bee products or even the health of the beekeeper. To avoid this kind of contamination, it is therefore necessary to wear personal protective equipment while harvesting honey and other bee products.
