Ecological risk assessment(ERA) to 5 abandoned mine drainage was investigated by using chemical measurement and bioassay experiment. From the results of chemical analysis, the high concentration of heavy metals are detected in most area. The Arsenite were mostly detected in Songcheon, Nakdong, and Dukum abandoned mine area, and various heavy metals were highly dispersed in Nakdong area. The study area have also high biological toxicity, resulted from the bioassay based on WET(Whole Effluent Toxicity) test by using Vibrio fisheri, Selenastrum copricornutum, and Daphnia magna. The maximum toxicity was shown in the point where the mine waters start to flow. The sensitivity of toxicity by S. capricornutum was relatively high considering the values of toxicity in all samples, from 1.3 to 32.0 TU. The different sensitivities of toxicity recommends the use of battery system, resulted from at least two test species for bioassay or ecological risk assessment of mine drainage. Besides, the results showed high hazard quotient(i.e., greater than 1 HQ value indicating potentially significant toxic risks) with regard to abandoned mine drainage area in this study. On the other hand, the biological toxicity results were sharply decreased by attenuation along further distance from discharging of mine waters. Therefore, environmental parameters including the dilution factor, dissolved organic matter, and hardness should be considered when the remediation and ERA of abandoned mine drainage is planned.
The objective of this study was to determine the effects of processing method and rapeseed variety on ruminal and intestinal protein digestibility of rapeseed meal in steers. Intestinal amino acid digestibility was assessed with an in situ ruminal incubation and precision-fed rooster bioassay. In this experiment one traditional rapeseed meal sample (sample A, prepress extraction) and three double low rapeseed meal samples (sample B, prepress extraction, sample C, screw press and sample D, low temperature press) were placed in polyester bags(8 cm${\times}$12 cm) and suspended in the ventral rumen of steers for 16 h. The residues of in situ incubations were intubated to roosters. Total excreta were collected for 48 h after incubation and then desiccated and amino acid concentrations were determined. Results showed that in ruminal incubation the degradation rate of amino acid and crude protein was higher for traditional rapeseed meal sample A than for double low rapeseed meal sample B, but was much lower than for double low sample C and D. In the group of double low rapeseed meal samples, sample D processed by low temperature press had the highest degradation rate of amino acids in the rumen. For all amino acids, the digestibility of the residual protein as measured by the precision-fed rooster bioassay tended to be lower for sample B than for sample A, which had the same processing method with sample B, and in the group of double low rapeseed meals, sample B had similar digestibility of amino acid in residual protein to sample D and higher than that of sample C. However, although the total amino acid availability involving the digestibility of amino acids in the rumen and rooster bioassay of double low rapeseed meal sample D (low temperature press) was higher than those of the other three samples by 7 to 9 percent, there were no significant differences. Results indicated that processing method markedly affected ruminal and post ruminal amino acid digestibility of rapeseed meal when the temperature exceeded 110$^{\circ}C$. Rapeseed meal that had a high content of fiber was not suitable for dry heat treatment at higher temperatures or the amino acids digestibility in rumen and total availability of amino acids could be reduced. Results also suggested the variety of rapeseed meal had no significant effect on the digestibility and availability of amino acids.
This experiment was performed to compare the techniques of the metabolizable energy determination and to, examine the problems associated with metabolizable energy determination of poultry feedingstuffs. The feedingstuffs evaluated herein, were corn, sorghum, barley. fish meal and soybean meal The results obtained were as followings : 1. When comparing AME and AMEn values of five feedingstuffs(measured) by Total Collection ana Index Methods there was no significant difference (P>.05) between two methods except fish meal. 2. The AME$_{F}$ values of feedingstuffs were not different (P>.05) from AME values. which implied that bioassay of AME$_{F}$ be applicable to rapid measure AME value of feedingstuffs for poultry and the problems connecting to AME$_{F}$ determination were discussed. 3. The AME$_{FC}$ and TME values of corn, sorghum and barley were significantly higher (P<.05) than AME and AME$_{F}$ values but those of fish meal and soybean meal were similar or lower than AME and AME$_{F}$ values. This fact was indicative that the techniques measuring AME$_{FC}$ and TME has some depressing factors when fish meal and soybean meal were determined. 4. Comparing of TME, AME$_{FC}$ and T-AME values tile TME values of corn sorghum and barley were significantly higher(P<.05) than AME$_{FC}$ and T- AME values. But that of fish meal and soybean meal was much lower(P< .05)comparing with AME$_{FC}$ and T-AME values. According]y, it night be concluded that the relationship among the values of TME. AME$_{FC}$ and T-AME was shown great variation according to the feedingstuffs, especially in the protein diets.
Background: Korean red ginseng (KRG) is a product from ginseng roots, which is enriched with ginsenosides and has been utilized for a long time as an adaptogen to alleviate various physiological or disease conditions. While KRG is generally considered safe, conducting a thorough toxicological assessment of the spray-dried powder G1899 during the juvenile period is essential to establish its safety profile. This study aimed to assess the safety of G1899 during the juvenile period using Sprague-Dawley rats. Methods: Two studies were conducted separately: a juvenile toxicity study and a uterotrophic bioassay. To assess the potential toxicity at systemic, postnatal developmental, and reproductive levels, G1899 was orally gavaged once a day in post-weaning juvenile Sprague-Dawley (SD) rats at 0, 1250, 2500, or 5000 mg/kg/day. Estrogenicity was assessed by orally gavaging G1899 in immature female SD rats at 0, 2500, or 5000 mg/kg/day on postnatal days (PND) 19-21, followed by a uterotrophic bioassay. These studies were conducted in accordance with the Good Laboratory Practice (GLP) regulations and regulatory test guidelines. Results: Regarding juvenile toxicity, no abnormalities related to the G1899 treatment were observed in any group during the experiment. Moreover, no uterotrophic responses were observed in the dosed female group. Based on these results, the no observed adverse effect level (NOAEL) of G1899 was determined to be at least 5000 mg/kg/day for general systemic function, developmental/reproductive function, and estrogenic activity. Conclusion: Our results suggest that G1899 is not toxic to juveniles at doses of up to 5000 mg/kg/day.
This experiment was performed to study the acute toxic effects of mereuric chloride(HgCl$_{2}$) in goldfish (Carassius aurams). In the study, Lc$_{50}$ values were determined for each 24, 48, 72 ,and 96-hr and mercury bioassay during the tests were performed. in five parts gill, kidney, liver, muscle and egg. The results isummarized were as follows 1. In the range of acute lethal toxicity for goldfish was 0.15-1.50 mg/l and the Lc$_{50}$ values decreased with exposure time. 2. The Lc$_{50}$ values of HgCl$_{2}$ for goldfish at 24, 48, 72 and 96 hour were 0.64, 0.53, 0.46, and 0.39 mg/l, respectively. 3. In the 400 $\mu$g/l treatment group, the average levels of mercury residues in the gill, kidey, liver, muscle and egg during 96 hour were 107.02 $\mu$g/g, 8.65 $\mu$g/g, 6.87 $\mu$g/g, 0.61 $\mu$g/g, and 0.82 $\mu$g/g, respectively. 4. The mercuny residues of gill was up to 12-175 times greater than mercury contents found in the other tissues.
The virulence of five Verticillium lecanii strains against greenhouse whiteflies, Trialeurodes vaporariorum, was tested at various temperatures as a major environmental factor. Strain CS-626 was found to be the most durable strain at a broad temperature range, and highly virulent against greenhouse whiteflies in a detached-leaf bioassay. In a tomato plant pot experiment, the $LT_{50}\;and\;LC_{50}$ of the CS-626 strain were 6.2 days and $2.3{\times}10^6$ conidia/ml, respectively. The optimal concentration of CS-626 for successful infection was $1{\times}10^8$conidia/ml. These results indicate that the CS-626 strain of Verticillium lecanii has a strong potential for effectively controlling greenhouse whiteflies.
The bioassay-guided fractionation of protective agents against sepsis-induced lethality from the root cortex of Paeonia suffruticosa ANDREWS (Ranunculaceae) led to the isolation of eight known compounds: paeonol (1), 2,5-dihydroxy-4-methoxyacetophenone (2), acetovanillone (3), paeonoside (4), paeoniflorin (5), oxypaeoniflorin (6), apiopaeonoside (7), and methyl 3-hydroxy-4-methoxybenzoate (8). Among them, 3 showed the highest survival rate (100% with a dose of 30 mg/kg versus 17% for the control experiment) and reduced alanine aminotransferase level to be a half of the control value on the sepsis model induced by lipopolysaccharide/D-galactosamine.
Several attempts have been made to protect crops against injury from herbicide quinclorac residue in soil. In this experiment, a selection of suitable crops for bioassay of the residue and a reduction of phytotoxicity by treatment with active carbon were carried out to prevent or to counteract the phytotoxicity. Cucumber (Cuumts satims) and kidney bean (Phaseolus Mgaris) were the suitable indicator plant in points of a sensitivity to the herbicide residue and an easy cultivation. The phytotoxicity was able to be observed at 20 and 30 days after seeding on kidney bean and cucumber respectively. In pot trials, application of the active carbon at 50 kg/10a protected effectively the 2-year-old ginseng plant from the injury in a paddy soil where the herbicide had been treated at 3 g a. i./10a.
The bioassay-guided fractionation of protective agents against sepsis-induced lethality from the root bark of Paeonia suffruticosa led to the isolation of ten known compounds: paeonol (1), 2,5-dihydroxy-4-methoxyacetophenone (2) methyl 3-hydroxy-4- methoxybenzoate (3), acetovanillone (4), benzoic acid (5), benzoylpaeoniflorin (6), paeonoside (7), paeoniflorin (8), oxypaeoniflorin (9) and apiopaeonoside (10). Among them, 4 exhibited the highest survival rate in a dose-dependent manner (100% with a dose of 30 mg/kg versus 16.7% for the control experiment) and showed reduction of plasma alanine aminotransferase (ALT) value on the in vivo assay model of sepsis induced by LPS/GalN.
The experiment was performed with the aim to study not only the effect of protein levels of basal diets on apparent and true metabolizable energy (AME and THM), AME of Farrell(AME$\_$F/), and corrected AME$\_$FC/ values of corn and soybean meal but also the effect of collection time of excreta on AME and TME values of corn and soybean meal. The AME$\_$F/ and AME$\_$FC/ values of test materials were determined through rapid AME bioassay, and AME and TME by the TME bioassay. The protein levels of basal diets had range from 10% crude protein(CP) to 30% CP. The results obtained were as follows; 1. The AME$\_$F/ values of basal diets showed much difference among treatments (P<0.05) but those of corn and soybean meal had no significant differences (P>0.05)and the AHE$\_$FC/ values of basal diets were proved to be variable according to level of protein of the diets. the AME$\_$FC/ values of com were not different while those of soybean meal in 20% and 30% were reduced significantly(P<0.05). 2. The protein intake/bird/day did not differ significantly due to variation of feed intake using rapid AME bioassay. 3. The protein levels of basal diets did not influence upon the AME value of basal diets, corn and soybean meal (P>0.05), and no clear trend was found in the TME values of corn and soybean meal because of the variation of metabolic fecal energy plus endogenous urinary energy (FEm+UEe) losses fed different diets. 4. Collection time of excreta affected the AME and TME values of basal diets in 10, 15, 20% CP treatments, but the AME and TME of corn and soybean meal were not affected by collection time; Thus, a time of 24 hours was mough for 24 hr to clean the digestive tract when fed corn and soybean meal substituted diets regardless of protein levels.
